The development of devices based on heterostructured thin films of biomolecules conveys a huge contribution on biomedical field. However, to achieve high efficiency of these devices, the storage of water molecules into these heterostructures, in order to maintain the biological molecules hydrated, is mandatory. Such hydrated environment may be achieved with lipids molecules which have the ability to rearrange spontaneously into vesicles creating a stable barrier between two aqueous compartments. Yet it is necessary to find conditions that lead to the immobilization of whole vesicles on the heterostructures. In this work, the conditions that govern the deposition of open and closed liposomes of 1.2-dipalmitoyl-sn-Glycero-3-[Phospho-rac-(1-glycerol)] (sodium Salt) (DPPG) onto polyelectrolytes cushions prepared by the layer-by-layer (LbL) method were analyzed. Electronic transitions of DPPG molecules as well as absorption coefficients were obtained by vacuum ultraviolet spectroscopy, while the elemental composition of the heterostructures was characterized by x-ray photoelectron spectroscopy (XPS). The presence of water molecules in the films was inferred by XPS and infrared spectroscopy. Quartz crystal microbalance (QCM) data analysis allowed to conclude that, in certain cases, the DPPG adsorbed amount is dependent of the bilayers number already adsorbed. Moreover, the adsorption kinetics curves of both adsorbed amount and surface roughness allowed to determine the kinetics parameters that are related with adsorption processes namely, electrostatic forces, liposomes diffusion and lipids re-organization on surface. Scaling exponents attained from atomic force microscopy images statistical analysis demonstrate that DPPG vesicles adsorption mechanism is ruled by the diffusion Villain model confirming that adsorption is governed by electrostatic forces. The power spectral density treatment enabled a thorough description of the accessible surface of the samples as well as of its inner structural properties. These outcomes proved that surface roughness influences the adsorption of DPPG liposomes onto surfaces covered by a polyelectrolyte layer. Thus, low roughness was shown to induce liposome rupture creating a lipid bilayer while high roughness allows the adsorption of whole liposomes. In addition, the fraction of open liposomes calculated from the normalized maximum adsorbed amounts decreases with the cushion roughness increase, allowing us to conclude that the surface roughness is a crucial variable that governs the adsorption of open or whole liposomes. This conclusion is fundamental for the development of well-designed sensors based on functional biomolecules incorporated in liposomes. Indeed, LbL films composed of polyelectrolytes and liposomes with and without melanin encapsulated were successfully applied to sensors of olive oil.

O conhecimento do comportamento das barragens de aterro é essencial para garantir o seu funcionamento adequado e para que os objetivos de utilização delineados inicialmente para o respetivo aproveitamento hidráulico possam ser cumpridos. Os fatores de maior relevância num estudo deste tipo de barragens, considerando que apenas estão sob solicitações estáticas, são as pressões de água, registadas em piezómetros, os caudais percolados e os deslocamentos superficiais, geralmente medidos em marcas de nivelamento ou em alvos colocados em peças de centragem forçada. Na presente dissertação pretende-se, com base no conhecimento dos registos dessas medições feitas anteriormente e recorrendo a modelos de inteligência artificial, predizer o valor que se obteria em próximas medições, ajudando assim a decidir qual o melhor procedimento para remediar ou tratar um problema de comportamento relacionado com as variáveis em estudo. Esta dissertação divide-se essencialmente em três partes. Primeiramente, introduzem-se os conceitos relativos à segurança de barragens de aterro, de acordo com o regulamento de segurança adotado em Portugal, dando relevo ao tipo de observação a que são submetidas. Seguidamente, introduz-se o conceito de redes neuronais artificiais e apresenta-se a base de dados, criada com o intuito de uniformizar e melhorar a organização dos valores em estudo das barragens de aterro, que têm sido acompanhadas pelo Laboratório Nacional de Engenharia Civil. Com esta pretende-se facilitar a utilização destes elementos por programas de inteligência artificial. Por último, é feito o enquadramento de um caso de estudo, uma barragem de aterro no Norte de Portugal – barragem de Valtorno-Mourão. Utilizando o Neuroph Studio, os dados relativos à observação desta barragem são aplicados numa rede neuronal artificial, Multi Layer Perceptron Backpropagation Neural Network, permitindo antever comportamentos futuros. Os resultados obtidos são discutidos e perspetivam-se trabalhos para continuar a desenvolver a investigação efetuada.

Shape Memory Alloy (SMA) Ni-Ti films have attracted much interest as functional and smart materials due to their unique properties. However, there are still important issues unresolved like formation of film texture and its control as well as substrate effects. Thus, the main challenge is not only the control of the microstructure, including stoichiometry and precipitates, but also the identification and control of the preferential orientation since it is a crucial factor in determining the shape memory behaviour. The aim of this PhD thesis is to study the optimisation of the deposition conditions of films of Ni-Ti in order to obtain the material fully crystallized at the end of the deposition, and to establish a clear relationship between the substrates and texture development. In order to achieve this objective, a two-magnetron sputter deposition chamber has been used allowing to heat and to apply a bias voltage to the substrate. It can be mounted into the six-circle diffractometer of the Rossendorf Beamline (ROBL) at the European Synchrotron Radiation Facility (ESRF), Grenoble, France, enabling an in-situ characterization by X-ray diffraction(XRD) of the films during their growth and annealing. The in-situ studies enable us to identify the different steps of the structural evolution during deposition with a set of parameters as well as to evaluate the effect of changing parameters on the structural characteristics of the deposited film. Besides the in-situ studies, other complementary ex-situ characterization techniques such as XRD at a laboratory source, Rutherford backscattering spectroscopy(RBS), Auger electron spectroscopy (AES), cross-sectional transmission electron microscopy (X-TEM), scanning electron microscopy (SEM), and electrical resistivity (ER) measurements during temperature cycling have been used for a fine structural characterization. In this study, mainly naturally and thermally oxidized Si(100) substrates, TiN buffer layers with different thicknesses (i.e. the TiN topmost layer crystallographic orientation is thickness dependent) and MgO(100) single crystals were used as substrates. The chosen experimental procedure led to a controlled composition and preferential orientation of the films. The type of substrate plays an important role for the texture of the sputtered Ni-Ti films and according to the ER results, the distinct crystallographic orientations of the Ni-Ti films influence their phase transformation characteristics.
