Analisando os resultados, concluímos que o perfil de expressão de Lp Paris foi modificado pela interacção com A. castellanii, no entanto essa mudança foi dependente da fase do seu ciclo de vida.-------ABSTRACT: Legionella is a pathogenic Gram-negative bacterium that replicates not only within aquatic protozoa like Acanthamoeba castellanii (A. castellanii), but also within human alveolar macrophages, which can result in a severe pneumonia. Legionella has a biphasic life cycle in broth, where exponential phase cultures display replicative traits and stationary bacteria express transmissive factors. Recent studies demonstrated that for successful infection of host cells, Legionella needs to ensure a precise timing of its life cycle. Many models of study were developed in order to learn about the intracellular life cycle and to identify the genes necessary for the host cell modulation. Although knowledge about the bacteria-host interaction is still limited, it appears that this interaction generate a pool of virulence traits, allowing the bacterium to infect human phagocytic cells and cause disease. The purpose of the present study was to investigate and select de critical genes for the infectivity of Legionella pneumophila strain Paris (Lp Paris), design and optimize a real time PCR technique for gene expression study and compare the expression profile of Lp Paris before and after co- culture of A. castellanii. The results show that eight of 12 genes in study changed its relative expression after coculture in A. castellanii when we performed the intracellular assays with early stationary phase Lp Paris cultures (five were induced and tree were repressed). When we performed the intracellular assays with late stationary phase Lp Paris cultures 11 genes showed a repressed relative expression. Analysing the results, we conclude that the expression profile of Lp Paris was modified by interaction with A. castellanii but this change was dependent of the timing of its life cycle.

The processes of mobilization of land for infrastructures of public and private domain are developed according to proper legal frameworks and systematically confronted with the impoverished national situation as regards the cadastral identification and regularization, which leads to big inefficiencies, sometimes with very negative impact to the overall effectiveness. This project report describes Ferbritas Cadastre Information System (FBSIC) project and tools, which in conjunction with other applications, allow managing the entire life-cycle of Land Acquisition and Cadastre, including support to field activities with the integration of information collected in the field, the development of multi-criteria analysis information, monitoring all information in the exploration stage, and the automated generation of outputs. The benefits are evident at the level of operational efficiency, including tools that enable process integration and standardization of procedures, facilitate analysis and quality control and maximize performance in the acquisition, maintenance and management of registration information and expropriation (expropriation projects). Therefore, the implemented system achieves levels of robustness, comprehensiveness, openness, scalability and reliability suitable for a structural platform. The resultant solution, FBSIC, is a fit-for-purpose cadastre information system rooted in the field of railway infrastructures. FBSIC integrating nature of allows: to accomplish present needs and scale to meet future services; to collect, maintain, manage and share all information in one common platform, and transform it into knowledge; to relate with other platforms; to increase accuracy and productivity of business processes related with land property management.

The purpose of this work is to understand the internal and external structure in which the company operates to provide an idea of the strategic actions needed to accomplish their organizational objectives. A strategic software was employed to build up phase one and phase two, phase one involved analysing internal and external factors that influence the company, comprehending their core competences, factors that influence the market and identification of strengths and weaknesses. Phase two consisted on providing an idea of their real competitive position and the suggestion of a development strategy, given the possible limitations in the external factors, the company should carefully analyse some of the opportunities present in the industry overseas to continue to develop their business and increase its profitability. Furthermore, a source of competitive advantage was found in their outbound logistics which could serve a differentiator between their competitors.

H3 is a fast-food chain that introduced the concept of gourmet hamburgers in the Portuguese market. This case-study illustrates its financing strategy that supported an exponential growth represented by opening 33 restaurants within approximately 3 years of its inception. H3 is now faced with the challenge of structuring its foreign ventures and change its financial approach. The main covered topics are the options an entrepreneur has for financing a new venture and how it evolves along the life cycle and different business approaches, namely franchising. It aims to be used as a learning tool in courses such as entrepreneurial finance.

Cities develop according to different patterns, undergoing population growth during some periods and decline (shrinkage) during others. Theories attempting to understand these behaviours include: 1) shrinkage is a natural process in the life cycle of a city, alternating with periods of growth, or 2) shrinkage is an extreme event that places cities into a continuous decline process with no return to population growth. We use retrospective data over a period of 130 years to study 25 Portuguese cities currently facing population decline, and show that both theories coexist in time and space. Five types of shrinking city are revealed: “Persistent Early Shrinkage” due to exodus fromthe rural periphery, “Metropolitan Shrinkage” due to the challenges of urban sprawl, “Recent Shrinkage” in de-industrialisation hotspots, “Cyclic Shrinkage” occurring in political transformation cores, and “Mild Shrinkage” due to life-style disamenity. As diversity of city population trajectories appears to be the norm in both Portugal and other Western European countries, the incorporation of this range into the management of urban transitions is recommended in order to reinforce city resilience.

The considerable amount of energy consumed on Earth is a major cause for not achieving sustainable development. Buildings are responsible for the highest worldwide energy consumption, nearly 40%. Strong efforts have been made in what concerns the reduction of buildings operational energy (heating, hot water, ventilation, electricity), since operational energy is so far the highest energy component in a building life cycle. However, as operational energy is being reduced the embodied energy increases. One of the building elements responsible for higher embodied energy consumption is the building structural system. Therefore, the present work is going to study part of embodied energy (initial embodied energy) in building structures using a life cycle assessment methodology, in order to contribute for a greater understanding of embodied energy in buildings structural systems. Initial embodied energy is estimated for a building structure by varying the span and the structural material type. The results are analysed and compared for different stages, and some conclusions are drawn. At the end of this work it was possible to conclude that the building span does not have considerable influence in embodied energy consumption of building structures. However, the structural material type has influence in the overall energetic performance. In fact, with this research it was possible that building structure that requires more initial embodied energy is the steel structure; then the glued laminated timber structure; and finally the concrete structure.

The evolution of a technology and the understanding of the moment in its life cycle is of the utmost importance to the entry strategy devised by any company. Having the entry of EDP Brazil on the micro-generation market as background, the present workproject attempts to summarize the most important topics in management literature concerning the theory of technology life-cycles and the updated literature on developments of photovoltaic technology to infer the current positioning of this technology in the theoretical models. The need for this type of work stems from the very common lack of bridging between the academic research of economic aspects relevant to the evolution of technologies and the agents of research on specific technological issues. When this occurs, namely due to the external nature of research to companies, thereby escaping the harsh economic controls of a profit seeking enterprise, the evolution many times lacks the appropriate framework to be studied on a more forward looking manner and to allow for management decisions to be based on.
