Proteins are biochemical entities consisting of one or more blocks typically folded in a 3D pattern. Each block (a polypeptide) is a single linear sequence of amino acids that are biochemically bonded together. The amino acid sequence in a protein is defined by the sequence of a gene or several genes encoded in the DNA-based genetic code. This genetic code typically uses twenty amino acids, but in certain organisms the genetic code can also include two other amino acids. After linking the amino acids during protein synthesis, each amino acid becomes a residue in a protein, which is then chemically modified, ultimately changing and defining the protein function. In this study, the authors analyze the amino acid sequence using alignment-free methods, aiming to identify structural patterns in sets of proteins and in the proteome, without any other previous assumptions. The paper starts by analyzing amino acid sequence data by means of histograms using fixed length amino acid words (tuples). After creating the initial relative frequency histograms, they are transformed and processed in order to generate quantitative results for information extraction and graphical visualization. Selected samples from two reference datasets are used, and results reveal that the proposed method is able to generate relevant outputs in accordance with current scientific knowledge in domains like protein sequence/proteome analysis.

É neste contexto que assume especial relevância a recente aprovação, no nosso ordenamento jurídico, de nova normativa reguladora do crédito ao consumo. Por outro lado, será de realçar o comando constitucional que prevê a protecção dos consumidores. De facto, prescreve o artigo 60° da Constituição da República Portuguesa que "os consumidores têm direito à qualidade dos bens e serviços consumidos, à formação e à informação, à protecção da saúde, da segurança e dos seus interesses económicos, bem como à reparação dos danos". lmporta, pois, conhecer em rigor as normas que actualmente regem o acesso e a regulação dos contratos de crédito ao consumo, num contexto de harmonização comunitária deste instituto jurídico-económico. Será oportuno, sobretudo, compreender de que forma o diploma que actualmente rege a matéria em questão protege de modo mais vincado a parte considerada economicamente mais débil.

β-lactamases are hydrolytic enzymes that inactivate the β-lactam ring of antibiotics such as penicillins and cephalosporins. The major diversity of studies carried out until now have mainly focused on the characterization of β-lactamases recovered among clinical isolates of Gram-positive staphylococci and Gram-negative enterobacteria, amongst others. However, only some studies refer to the detection and development of β-lactamases carriers in healthy humans, sick animals, or even in strains isolated from environmental stocks such as food, water, or soils. Considering this, we proposed a 10-week laboratory programme for the Biochemistry and Molecular Biology laboratory for majors in the health, environmental, and agronomical sciences. During those weeks, students would be dealing with some basic techniques such as DNA extraction, bacterial transformation, polymerase chain reaction (PCR), gel electrophoresis, and the use of several bioinformatics tools. These laboratory exercises would be conducted as a mini research project in which all the classes would be connected with the previous ones. This curriculum was compared in an experiment involving two groups of students from two different majors. The new curriculum, with classes linked together as a mini research project, was taught to a major in Pharmacy and an old curriculum was taught to students from environmental health. The results showed that students who were enrolled in the new curriculum obtained better results in the final exam than the students who were enrolled in the former curriculum. Likewise, these students were found to be more enthusiastic during the laboratory classes than those from the former curriculum.

The great majority of the courses on science and technology areas where lab work is a fundamental part of the apprenticeship was not until recently available to be taught at distance. This reality is changing with the dissemination of remote laboratories. Supported by resources based on new information and communication technologies, it is now possible to remotely control a wide variety of real laboratories. However, most of them are designed specifically to this purpose, are inflexible and only on its functionality they resemble the real ones. In this paper, an alternative remote lab infrastructure devoted to the study of electronics is presented. Its main characteristics are, from a teacher's perspective, reusability and simplicity of use, and from a students' point of view, an exact replication of the real lab, enabling them to complement or finish at home the work started at class. The remote laboratory is integrated in the Learning Management System in use at the school, and therefore, may be combined with other web experiments and e-learning strategies, while safeguarding security access issues.

This paper presents a collaborative virtual learning environment, which includes technologies such as 3D virtual representations, learning and content management systems, remote experiments, and collaborative learning spaces, among others. It intends to facilitate the construction, management and sharing of knowledge among teachers and students, in a global perspective. The environment proposes the use of 3D social representations for accessing learning materials in a dynamic and interactive form, which is regarded to be closer to the physical reality experienced by teachers and students in a learning context. A first implementation of the proposed extended immersive learning environment, in the area of solid mechanics, is also described, including the access to theoretical contents and a remote experiment to determine the elastic modulus of a given object.These instructions give you basic guidelines for preparing camera-ready papers for conference proceedings. Nos últimos anos, o avanço da tecnologia e a miniaturização de diversos componentes têm permitido o aparecimento de novos conceitos, ideias e projetos, que até aqui não passariam de filmes de ficção científica. Com a tecnologia atual, podem ser desenvolvidos pequenos dispositivos wearable com diversas interfaces, múltiplas conectividades, poder de processamento e autonomia. Permitindo desta forma, dar resposta à crescente necessidade de interação com os mais diversos equipamentos eletrónicos do dia-a-dia, melhorando o acesso e o fornecimento de informação. O principal objetivo deste trabalho passa assim por demonstrar e implementar um conceito que permita estreitar e facilitar a interação entre o utilizador e o mundo que o rodeia, quer em ambientes domésticos quer industriais. Para isso foi projetado e implementado um dispositivo wearable (para utilização no pulso) baseado numa arquitetura de hardware e software capaz de correr diferentes aplicações, tais como extensão de alertas de um smartphone, crowdsourcing de informações meteorológicas, manutenção e inspeção industrial e monitorização remota de forças de segurança. Os resultados obtidos demonstram que este conceito é viável tanto do ponto de vista técnico como funcional, evidenciando boas hipóteses para que estes conceitos, métodos e tecnologias possam ser integradas em plataformas robóticas desenvolvidas no âmbito de projetos do Laboratório de Sistemas Autónomos (LSA) bem como nos contextos industrial e de lazer.

Mathematical models and statistical analysis are key instruments in soil science scientific research as they can describe and/or predict the current state of a soil system. These tools allow us to explore the behavior of soil related processes and properties as well as to generate new hypotheses for future experimentation. A good model and analysis of soil properties variations, that permit us to extract suitable conclusions and estimating spatially correlated variables at unsampled locations, is clearly dependent on the amount and quality of data and of the robustness techniques and estimators. On the other hand, the quality of data is obviously dependent from a competent data collection procedure and from a capable laboratory analytical work. Following the standard soil sampling protocols available, soil samples should be collected according to key points such as a convenient spatial scale, landscape homogeneity (or non-homogeneity), land color, soil texture, land slope, land solar exposition. Obtaining good quality data from forest soils is predictably expensive as it is labor intensive and demands many manpower and equipment both in field work and in laboratory analysis. Also, the sampling collection scheme that should be used on a data collection procedure in forest field is not simple to design as the sampling strategies chosen are strongly dependent on soil taxonomy. In fact, a sampling grid will not be able to be followed if rocks at the predicted collecting depth are found, or no soil at all is found, or large trees bar the soil collection. Considering this, a proficient design of a soil data sampling campaign in forest field is not always a simple process and sometimes represents a truly huge challenge. In this work, we present some difficulties that have occurred during two experiments on forest soil that were conducted in order to study the spatial variation of some soil physical-chemical properties. Two different sampling protocols were considered for monitoring two types of forest soils located in NW Portugal: umbric regosol and lithosol. Two different equipments for sampling collection were also used: a manual auger and a shovel. Both scenarios were analyzed and the results achieved have allowed us to consider that monitoring forest soil in order to do some mathematical and statistical investigations needs a sampling procedure to data collection compatible to established protocols but a pre-defined grid assumption often fail when the variability of the soil property is not uniform in space. In this case, sampling grid should be conveniently adapted from one part of the landscape to another and this fact should be taken into consideration of a mathematical procedure.

The recent changes on power systems paradigm requires the active participation of small and medium players in energy management. With an electricity price fluctuation these players must manage the consumption. Lowering costs and ensuring adequate user comfort levels. Demand response can improve the power system management and bring benefits for the small and medium players. The work presented in this paper, which is developed aiming the smart grid context, can also be used in the current power system paradigm. The proposed system is the combination of several fields of research, namely multi-agent systems and artificial neural networks. This system is physically implemented in our laboratories and it is used daily by researchers. The physical implementation gives the system an improvement in the proof of concept, distancing itself from the conventional systems. This paper presents a case study illustrating the simulation of real-time pricing in a laboratory.

A Salmonella é um microrganismo responsável por grande parte das doenças alimentares, podendo por em causa a saúde pública da área contaminada. Uma deteção rápida, eficiente e altamente sensível e extremamente importante, sendo um campo em franco desenvolvimento e alvo de variados e múltiplos estudos na comunidade cientifica atual. Foi desenvolvido um método potenciométrico para a deteção de Salmonellas, com elétrodos seletivos de iões, construídos em laboratório com pontas de micropipetas, fios de prata e sensores com composição otimizada. O elétrodo indicador escolhido foi um ESI seletivo a cadmio, para redução da probabilidade de interferências no método, devido a pouca abundancia do cadmio em amostras alimentares. Elétrodos seletivos a sódio, elétrodos de Ag/AgCl de simples e de dupla juncão foram também construídos e caracterizados para serem aplicados como elétrodos de referência. Adicionalmente otimizaram-se as condições operacionais para a analise potenciométrica, nomeadamente o elétrodo de referencia utilizado, condicionamento dos elétrodos, efeito do pH e volume da solução amostra. A capacidade de realizar leituras em volumes muito pequenos com limites de deteção na ordem dos micromolares por parte dos ESI de membrana polimérica, foi integrada num ensaio com um formato nao competitivo ELISA tipo sanduiche, utilizando um anticorpo primário ligado a nanopartículas de Fe@Au, permitindo a separação dos complexos anticorpo-antigénio formados dos restantes componentes em cada etapa do ensaio, pela simples aplicação de um campo magnético. O anticorpo secundário foi marcado com nanocristais de CdS, que são bastante estáveis e é fácil a transformação em Cd2+ livre, permitindo a leitura potenciométrica. Foram testadas várias concentrações de peroxido de hidrogénio e o efeito da luz para otimizar a dissolução de CdS. O método desenvolvido permitiu traçar curvas de calibração com soluções de Salmonellas incubadas em PBS (pH 4,4) em que o limite de deteção foi de 1100 CFU/mL e de 20 CFU/mL, utilizando volumes de amostra de 10 ƒÊL e 100 ƒÊL, respetivamente para o intervalo de linearidade de 10 a 108 CFU/mL. O método foi aplicado a uma amostra de leite bovino. A taxa de recuperação media obtida foi de 93,7% } 2,8 (media } desvio padrão), tendo em conta dois ensaios de recuperação efetuados (com duas replicas cada), utilizando um volume de amostra de 100 ƒÊL e concentrações de 100 e 1000 CFU/mL de Salmonella incubada.

Com o decorrer dos tempos e com a evolução da indústria, acresce a necessidade de aparecimento de novas construções e consequentemente de novos desafios geotécnicos. Para responder a estes novos reptos que a construção nos lança é necessário aprofundar o conhecimento acerca dos materiais que constituem o subsolo e estudar o seu comportamento quando sujeito a novas solicitações, como por exemplo sobrecargas, pois parte do cálculo de dimensionamento de fundações das novas construções dependerá destas características. Existem duas possíveis formas de identificar os materiais existentes no subsolo e caracterizá‐los: através de ensaios de laboratório e de ensaios in situ, sendo nestes últimos que nos iremos focar. Os ensaios in situ, para além de outros dados, permitem‐nos estimar com algum grau de incerteza a profundidade a que se encontra o horizonte com capacidade de suporte pretendida, quer se trate ou não do bedrock, e identificar o processo mais indicado para a retirada do material: através de um meio mecânico de escavação ou por recurso a explosivos. Com a realização de ensaios de sísmica de refração e igualmente recorrendo a ensaios executados com o penetrómetro dinâmico médio (DPM), tentamos dar resposta a estas incógnitas através do cruzamento dos dados obtidos. No entanto, este cruzamento de dados pode não ser completamente esclarecedor, levando ao aparecimento de novas incógnitas às quais não seja possível dar resposta recorrendo‐se unicamente à informação recolhida por meio destes dois métodos, pois poderão revelar‐se insuficientes. Contudo, existem muitos outros ensaios in situ que podem ser realizados e capazes de nos ajudar a complementar os dados inicialmente obtidos no sentido de diminuir ou até mesmo eliminar as incógnitas existentes, e desta forma poder identificar e caraterizar o material existente com o maior grau de segurança possível.

Intelligent wheelchairs (IW) are technologies that can increase the autonomy and independence of elderly people and patients suffering from some kind of disability. Nowadays the intelligent wheelchairs and the human-machine studies are very active research areas. This paper presents a methodology and a Data Analysis System (DAS) that provides an adapted command language to an user of the IW. This command language is a set of input sequences that can be created using inputs from an input device or a combination of the inputs available in a multimodal interface. The results show that there are statistical evidences to affirm that the mean of the evaluation of the DAS generated command language is higher than the mean of the evaluation of the command language recommended by the health specialist (p value = 0.002) with a sample of 11 cerebral palsy users. This work demonstrates that it is possible to adapt an intelligent wheelchair interface to the user even when the users present heterogeneous and severe physical constraints.
