The effect of organic and conventional agricultural systems on the physicochemical parameters, bioactive compounds content, and sensorial attributes of tomatoes (‘‘Redondo’’ cultivar) was studied. The influence on phytochemicals distribution among peel, pulp and seeds was also accessed. Organic tomatoes were richer in lycopene (+20%), vitamin C (+30%), total phenolics (+24%) and flavonoids (+21%) and had higher (+6%) in vitro antioxidant activity. In the conventional fruits, lycopene was mainly concentrated in the pulp, whereas in the organic ones, the peel and seeds contained high levels of bioactive compounds. Only the phenolic compounds had a similar distribution among the different fractions of both types of tomatoes. Furthermore, a sensorial analysis indicated that organic farming improved the gustative properties of this tomato cultivar.

Eight tropical fruit pulps from Brazil were simultaneously characterised in terms of their antioxidant and antimicrobial properties. Antioxidant activity was screened by DPPH radical scavenging activity (126–3987 mg TE/100 g DW) and ferric reduction activity power (368–20819 mg AAE/100 g DW), and complemented with total phenolic content (329–12466 mg GAE/100 g DW) and total flavonoid content measurements (46–672 mg EE /100 g DW), whereas antimicrobial activity was tested against the most frequently found food pathogens. Acerola and açaí presented the highest values for the antioxidant-related measurements. Direct correlations between these measurements could be observed for some of the fruits. Tamarind exhibited the broadest antimicrobial potential, having revealed growth inhibition of Pseudomonas aeruginosa. Escherichia coli, Listeria monocytogenes, Salmonella sp. and Staphylococcus aureus. Açaí and tamarind extracts presented an inverse relationship between antibacterial and antioxidant activities, and therefore, the antibacterial activity cannot be attributed (only) to phenolic compounds.

Folk medicine is a relevant and effective part of indigenous healthcare systems which are, in practice, totally dependent on traditional healers. An outstanding coincidence between indigenous medicinal plant uses and scientifically proved pharmacological properties of several phytochemicals has been observed along the years. This work focused on the leaves of a medicinal plant traditionally used for therapeutic benefits (Angolan Cymbopogon citratus), in order to evaluate their nutritional value. The bioactive phytochemical composition and antioxidant activity of leaf extracts prepared with different solvents (water, methanol and ethanol) were also evaluated. The plant leaves contained ~60% of carbohydrates, protein (~20%), fat (~5%), ash (~4%) and moisture (~9%). The phytochemicals screening revealed the presence of tannins, flavonoids, and terpenoids in all extracts. Methanolic extracts also contained alkaloids and steroids. Several methods were used to evaluate total antioxidant capacity of the different extracts (DPPH; NO; and H2O2 scavenging assays, reducing power, and FRAP). Ethanolic extracts presented a significantly higher antioxidant activity (p < 0.05) except for FRAP, in which the best results were achieved by the aqueous extracts. Methanolic extracts showed the lowest radical scavenging activities for both DPPH; and NO; radicals.

The effect of peel and seed removal, two commonly practiced procedures either at home or by the processing industry, on the physicochemical properties, bioactive compounds contents and antioxidant capacity of tomato fruits of four typical Portuguese cultivars (cereja, chucha, rama and redondo) were appraised. Both procedures caused significant nutritional and antioxidant activity losses in fruits of every cultivar. In general, peeling was more detrimental, since it caused a higher decrease in lycopene, bcarotene, ascorbic acid and phenolics contents (averages of 71%, 50%, 14%, and 32%, respectively) and significantly lowered the antioxidant capacity of the fruits (8% and 10%, using DPPH. and b-carotene linoleate model assays, correspondingly). Although seeds removal favored the increase of both color and sweetness, some bioactive compounds (11% of carotenoids and 24% of phenolics) as well as antioxidant capacity (5%) were loss. The studied cultivars were differently influenced by these procedures. The fruits most affected by peeling were those from redondo cultivar (-66% lycopene, -44% b-carotene, -26% ascorbic acid and -38% phenolics). Seeds removal, in turn, was more injurious for cereja tomatoes (-10% lycopene, -38% b-carotene, -25% ascorbic acid and -63% phenolics). Comparatively with the remaining ones, the rama fruits were less affected by the trimming procedures.

O Neem (Azadirachta indica) é uma árvore indiana conhecida pela atividade pesticida e por várias atividades farmacológicas. De entre os vários compostos já isolados e estudados, a Azadiractina (AZA) foi identificada como o principal composto bioativo desta planta. Este composto apresenta uma grande diversidade de localizações nesta planta, porém assume a sua máxima concentração ao nível das sementes, porção que se apresenta também como a principal fonte de obtenção do óleo de Neem. O óleo apresenta-se como a porção menos estudada do Neem, quer ao nível do seu teor em AZA, quer ao nível das suas propriedades, nomeadamente antimicrobianas. Neste sentido, os objetivos primordiais deste estudo foram o doseamento da Azadiractina e a avaliação da atividade antimicrobiana em produtos contendo óleo de Neem. Um método analítico rápido, sensível e seletivo utilizando HPLC-UV foi desenvolvido para a identificação e quantificação da Azadiractina-A (AZA-A) e 3-tigloylazadirachtol (AZA-B) em diferentes amostras de óleo de Neem. O teor de AZA-A, B e A+B determinado nas amostras de óleo de Neem apresentou valores entre 58,53-843,42 mg/kg, 12,52-800,223 mg/kg e 104,20-1642,17 mg/kg, respetivamente. Na generalidade, os valores obtidos foram inferiores aos descritos na literatura. A partir dos resultados obtidos, verificou-se ainda que o teor destes compostos não é similar em todas as amostras, sendo este condicionado pela qualidade das sementes que deram origem ao óleo e pelo processo extrativo utilizado. Para além disso, foi possível inferir que duas das amostras testadas teriam qualidade inferior, dados os teores reduzidos de AZA que apresentavam. As diferentes amostras de óleo de Neem, bem como formulações comerciais contendo óleo de Neem, foram testadas em 14 microrganismos de forma a avaliar o seu potencial antimicrobiano. Após a análise, verificou-se atividade antimicrobiana de todas as amostras sobre todos os microrganismos testados, observando-se atividade tanto em bactérias Gram+ como Gram-. Os resultados alcançados mostraram que o óleo de Neem e as formulações comerciais contendo óleo de Neem têm um potencial antimicrobiano interessante, principalmente sobre bactérias comuns em patologias da pele. Para além disso, foi possível comprovar que, no caso do óleo de Neem, a AZA não será a principal responsável por esta atividade. Por outro lado, verificou-se que a atividade antimicrobiana das formulações comerciais não se deverá exclusivamente à presença do óleo de Neem, Doseamento da Azadiractina e avaliação da atividade antimicrobiana em produtos contendo óleo de Neem X uma vez que os valores dos halos de inibição obtidos com as formulações tenderam a ser superiores aos verificados apenas com o óleo, além de que os valores de inibição mais elevados foram observados para as formulações contendo menor percentagem de óleo de Neem incorporado. Em suma, os resultados alcançados para os diferentes produtos analisados são promissores e, na sua maioria, convergem com o que está descrito na literatura. No entanto, apesar destes resultados serem um grande contributo, mais estudos são necessários e importantes para conhecer melhor os produtos analisados e assim poder tirar o maior proveito deles.

The effect of boiling (10 minutes) on eleven green vegetables frequently consumed in the Mediterranean diet was evaluated. For that, some physicochemical parameters and the contents of vitamin C, phenolics and carotenoids, as well as the antioxidant activity, were determined in raw and boiled samples. The raw vegetables analysed in this study were good sources of vitamin C, carotenoids and phenolic compounds, with contents ranging from 10.6 to 255.1 mg/100 g, 0.03 to 3.29 mg/100 g and 202.9 to 1010.7 mg/100 g, respectively. Boiling promoted losses in different extensions considering both the different bioactive compounds and the distinct vegetables analysed. Contrary to phenolics (more resistant), vitamin C was the most affected compound. Boiling also originated significant losses in the antioxidant activity of the vegetables. Considering all the parameters analysed, the vegetables most affected by boiling were broccoli and lettuce. The least affected ones were collard and tronchuda cabbage.

The purpose of this study was to develop a bone substitute material capable of preventing or treating osteomyelitis through a sustainable release of vancomycin and simultaneously inducing bone regeneration. Porous heparinized nanohydroxyapatite (nanoHA)/collagen granules were characterized using scanning electron microscopy, micro-computed tomography and attenuated total reflectance Fourier transform infrared spectroscopy. After vancomycin adsorption onto the granules, its releasing profile was studied by UV molecular absorption spectroscopy. The heparinized granules presented a more sustainable release over time, in comparison with nonheparinized nanoHA and nanoHA/collagen granules. Vancomycin was released for 360 h and proved to be bioactive until 216 h. Staphylococcus aureus adhesion was higher on granules containing collagen, guiding the bacteria to the material with antibiotic, improving their eradication. Moreover, cytotoxicity of the released vancomycin was assessed using osteoblast cultures, and after 14 days of culture in the presence of vancomycin, cells were able to remain viable, increasing their metabolic activity and colonizing the granules, as observed by scanning electron microscopy and confocal laser scanning microscopy. These findings suggest that heparinized nanoHA/collagen granules are a promising material to improve the treatment of osteomyelitis, as they are capable of releasing vancomycin, eliminating the bacteria, and presented morphological and chemical characteristics to induce bone regeneration.
