We have developed a new method for single-drop microextraction (SDME) for the preconcentration of organochlorine pesticides (OCP) from complex matrices. It is based on the use of a silicone ring at the tip of the syringe. A 5 μL drop of n-hexane is applied to an aqueous extract containing the OCP and found to be adequate to preconcentrate the OCPs prior to analysis by GC in combination with tandem mass spectrometry. Fourteen OCP were determined using this technique in combination with programmable temperature vaporization. It is shown to have many advantages over traditional split/splitless injection. The effects of kind of organic solvent, exposure time, agitation and organic drop volume were optimized. Relative recoveries range from 59 to 117 %, with repeatabilities of <15 % (coefficient of variation) were achieved. The limits of detection range from 0.002 to 0.150 μg kg−1. The method was applied to the preconcentration of OCPs in fresh strawberry, strawberry jam, and soil.

The cyanobacteria are known to be a rich source of metabolites with a variety of biological activities in different biological systems. In the present work, the bioactivity of aqueous and organic (methanolic and hexane) crude extracts of cyanobacteria isolated from estuarine ecosystems was studied using different bioassays. The assessment of DNA damage on the SOS gene repair region of mutant PQ37 strain of Escherichia coli was performed. Antiviral activity was evaluated against influenza virus, HRV-2, CVB3 and HSV-1 viruses using crystal violet dye uptake on HeLa, MDCK and GMK cell lines. Cytotoxicity evaluation was performed with L929 fibroblasts by MTT assay. Of a total of 18 cyanobacterial isolates studied, only the crude methanolic extract of LEGE 06078 proved to be genotoxic (IF > 1.5) in a dose-dependent manner and other four were putative candidates to induce DNA damage. Furthermore, the crude aqueous extract of LEGE 07085 showed anti- herpes type 1 activity (IC50 = 174.10 μg dry extract mL−1) while not presenting any cytotoxic activity against GMK cell lines. Of the 54 cyanobacterial extracts tested, only the crude methanolic and hexane ones showed impair on metabolic activity of L929 fibroblasts after long exposure (48–72 h). The inhibition of HSV-1 and the strong cytotoxicity against L929 cells observed emphasizes the importance of evaluating the impact of those estuarine cyanobacteria on aquatic ecosystem and on human health. The data also point out their potential application in HSV-1 treatment and pharmacological interest.

Specific marine macro algae species abundant at the Portuguese coast (Laminaria hyperborea, Bifurcaria bifurcata, Sargassum muticum and Fucus spiralis) were shown to be effective for removing toxic metals (Cd(II), Zn(II) and Pb(II)) from aqueous solutions. The initial metal concentrations in solution were about 75–100 mg L−1. The observed biosorption capacities for cadmium, zinc and lead ions were in the ranges of 23.9–39.5, 18.6–32.0 and 32.3–50.4 mg g−1, respectively. Kinetic studies revealed that the metal uptake rate was rather fast, with 75% of the total amount occurring in the first 10 min for all algal species. Experimental data were well fitted by a pseudo-second order rate equation. The contribution of internal diffusion mechanism was significant only to the initial biosorption stage. Results indicate that all the studied macro algae species can provide an efficient and cost-effective technology for eliminating heavy metals from industrial effluents.

Modern real-time systems, with a more flexible and adaptive nature, demand approaches for timeliness evaluation based on probabilistic measures of meeting deadlines. In this context, simulation can emerge as an adequate solution to understand and analyze the timing behaviour of actual systems. However, care must be taken with the obtained outputs under the penalty of obtaining results with lack of credibility. Particularly important is to consider that we are more interested in values from the tail of a probability distribution (near worst-case probabilities), instead of deriving confidence on mean values. We approach this subject by considering the random nature of simulation output data. We will start by discussing well known approaches for estimating distributions out of simulation output, and the confidence which can be applied to its mean values. This is the basis for a discussion on the applicability of such approaches to derive confidence on the tail of distributions, where the worst-case is expected to be.

O recente surgimento de nanopartículas de ferro valente-zero (nFZV), um material com elevada capacidade de remediação de solos por via de reacções de oxidação/redução pode ser uma opção viável para a remoção de fármacos do solo. A sua aplicação já é uma realidade em alguns tipos de solos contaminados por compostos específicos e, com este trabalho, procura-se estudar a sua capacidade de remediação de solos contaminados por compostos farmacêuticos, recorrendo-se a uma tecnologia “verde” de síntese destas nanopartículas. Esta tecnologia é bastante recente, ainda não aplicada no campo de trabalho, que se baseia no uso de folhas de certas árvores para produzir extratos naturais que reduzem o ferro (III) a ferro zero valente, formando nFZV. Desta forma procedeu-se, à escala laboratorial, ao estudo da eficiência das nFZV na degradação de um fármaco – paracetamol – e comparou-se com a eficiência demonstrada por oxidantes, muito utilizados hoje em dia em casos de remediação in situ como o permanganato de potássio, o peróxido de hidrogénio, o persulfato de sódio e o reagente de Fenton. O estudo foi efectuado em dois meios diferentes: solução aquosa e solo arenoso. De forma muito sucinta, o estudo baseou-se na introdução dos oxidantes/nFZV em soluções/solos contaminados com paracetamol e consequente monitorização do processo de remediação através de cromatografia líquida de alta eficiência. Nos ensaios com soluções aquosas contaminadas com paracetamol, o permanganato de potássio e o reagente de Fenton revelaram capacidade para degradar o paracetamol, atingindo mesmo um grau de degradação de 100%. O persulfato de sódio também demonstrou uma capacidade de degradação do paracetamol, chegando a atingir 99% de degradação, mas apenas recorrendo ao uso de um volume de oxidante elevado quando comparado com os outros dois oxidantes já referidos. Por outro lado, o peróxido de hidrogénio não demonstrou qualquer capacidade de degradação do paracetamol, pelo que o seu uso não passou desta fase. Verificou-se também que o uso de ferro granulado para o tratamento de água contaminada com paracetamol revelou resultados diferentes dos observados no uso de nFZV, obtendo-se eficiências de 87%. Existiram dificuldades analíticas na quantificação do paracetamol, especificamente relacionadas com o uso do extracto de folhas de amoreira, cuja composição continha substâncias que causaram dificuldades acentuadas na análise dos cromatogramas. Por fim, um pequeno teste de combinação do reagente de Fenton com os fenómenos de biodegradação resultantes dos microrganismos presentes em folhas do extracto de chá preto demonstrou que este pode ser uma área que pode e deve ser mais estudada. Desta forma, a utilização das nFZV para o tratamento de água contaminada com paracetamol não permitiu a retirada de conclusões seguras sobre a capacidade que as nFZV produzidas com extractos de folhas de amoreira e de chá preto têm de degradação do paracetamol. Nos testes de remediação de solos contaminados os resultados demonstraram que, mais uma vez, tanto o permanganato de potássio como o reagente de Fenton se revelam como os melhores oxidantes para a degradação do paracetamol, obtendo-se a degradação total do paracetamol. Por outro lado, voltou a ser necessário uma elevada quantidade de persulfato de sódio quando comparada com os dois anteriores, para que ocorra a degradação desta mesma quantidade de paracetamol, demonstrando mais uma vez que, apesar de não ideal, o persulfato demonstra capacidade de degradação do paracetamol.

The study of Electricity Markets operation has been gaining an increasing importance in the last years, as result of the new challenges that the restructuring produced. Currently, lots of information concerning Electricity Markets is available, as market operators provide, after a period of confidentiality, data regarding market proposals and transactions. These data can be used as source of knowledge, to define realistic scenarios, essential for understanding and forecast Electricity Markets behaviour. The development of tools able to extract, transform, store and dynamically update data, is of great importance to go a step further into the comprehension of Electricity Markets and the behaviour of the involved entities. In this paper we present an adaptable tool capable of downloading, parsing and storing data from market operators’ websites, assuring actualization and reliability of stored data.

Context: Inclusion of antioxidants in topical formulations can contribute to minimize oxidative stress in the skin, which has been associated with photoaging, several dermatosis and cancer. Objective: A Castanea sativa leaf extract with established antioxidant activity was incorporated into a semisolid surfactant-free formulation. The objective of this study was to perform a comprehensive characterization of this formulation. Materials and methods: Physical, microbiological and functional stability were evaluated during 6 months storage at 20 °C and 40 °C. Microstructure elucidation (cryo-SEM), in vitro release and in vivo moisturizing effect (Corneometer® CM 825) were also assessed. Results and discussion: Minor changes were observed in the textural and rheological properties of the formulation when stored at 20 °C for 6 months and the antioxidant activity of the plant extract remained constant throughout the storage period. Microbiological quality was confirmed at the end of the study. Under accelerated conditions, higher modifications of the evaluated parameters were observed. Cryo-SEM analysis revealed the presence of oil droplets dispersed into a gelified external phase. The release rate of the antioxidant compounds (610 ± 70 µgh−0.5) followed Higuchi model. A significant in vivo moisturizing effect was demonstrated, that lasted at least 4 h after product’s application. Conclusion: The physical, functional and microbiological stability of the antioxidant formulation was established. Specific storage conditions should be recommended considering the influence of temperature on the stability. A skin hydration effect and good skin tolerance were also found which suggests that this preparation can be useful in the prevention or treatment of oxidative stress-mediated dysfunctions.

Toxic effects of ultraviolet (UV) radiation on skin include protein and lipid oxidation, and DNA damage. The latter is known to play a major role in photocarcinogenesis and photoaging. Many plant extracts and natural compounds are emerging as photoprotective agents. Castanea sativa leaf extract is able to scavenge several reactive species that have been associated to UV-induced oxidative stress. The aim of this work was to analyze the protective effect of C. sativa extract (ECS) at different concentrations (0.001, 0.01, 0.05 and 0.1 μg/mL) against the UV mediated-DNA damage in a human keratinocyte cell line (HaCaT). For this purpose, the cytokinesis-block micronucleus assay was used. Elucidation of the protective mechanism was undertaken regarding UV absorption, influence on 1O2 mediated effects or NRF2 activation. ECS presented a concentration-dependent protective effect against UV-mediated DNA damage in HaCaT cells. The maximum protection afforded (66.4%) was achieved with the concentration of 0.1 μg/mL. This effect was found to be related to a direct antioxidant effect (involving 1O2) rather than activation of the endogenous antioxidant response coordinated by NRF2. Electrochemical studies showed that the good antioxidant capacity of the ECS can be ascribed to the presence of a pool of different phenolic antioxidants. No genotoxic or phototoxic effects were observed after incubation of HaCaT cells with ECS (up to 0.1 μg/mL). Taken together these results reinforce the putative application of this plant extract in the prevention/minimization of UV deleterious effects on skin.
