O tratamento de água para consumo humano tem por objectivos não só a sua qualidade em termos de parâmetros químicos e físicos, como também microbiológicos. Considerando que a MON pode afectar os sistemas de tratamento, a sua redução minimiza a formação de subprodutos de desinfecção, como por exemplo os trihalometanos e diminui o crescimento de microrganismos ao longo do sistema de distribuição. Nesse sentido tem havido recentemente uma grande evolução na investigação relativamente à remoção de matéria orgânica natural. O principal objectivo deste trabalho foi o de caracterizar a matéria orgânica presente na água bruta e ao longo do processo de tratamento, assim como a sua evolução sazonal. Os dados obtidos a partir desta caracterização poderão contribuir para uma futura optimização no processo de tratamento de águas de consumo. O processo utilizado baseou-se na sorção da matéria orgânica dissolvida em diferentes tipos de resinas de permuta iónica, DAX-8, DAX-4 e IRA-958, permitindo a sua separação em várias fracções: ácidos muito hidrofóbicos (VHA), ácidos ligeiramente hidrofóbicos (SHA), compostos hidrofílicos carregados (CHA) e hidrofílicos neutros (NEU). De acordo com os resultados obtidos apenas a fracção NEU demonstrou ter uma tendência sazonal, apresentando valores máximos no Verão, nos pontos de amostragem referentes à água bruta superficial (PA802) e água bruta superficial após pré-tratamento por filtração (PA800). Os valores de COD não mostraram uma variação sazonal para as amostras de água bruta superficial que rondaram os 2 mg C/L ao longo do período de amostragem (Julho a Outubro), durante o qual se verificou uma baixa pluviosidade e temperaturas médias muito semelhantes. Os compostos NEU predominam em todos os pontos de amostragem não apresentando uma tendência definida ao longo do tratamento. Observou-se uma diminuição das fracções SHA e CHA ao longo do tratamento. Não se pode indicar uma tendência definida relativamente à fracção VHA. Verifica-se globalmente uma diminuição do teor de MON ao longo do tratamento. Este trabalho demonstrou que na água bruta superficial existe uma predominância dos compostos NEU, seguidos dos SHA, dos VHA e finalmente dos compostos CHA. Na água bruta captada no sub-leito do rio, verifica-se apenas a existência dos compostos NEU, sendo as restantes fracções praticamente nulas. Os valores mais elevados de TSUVA254nm foram obtidos para as amostras que não sofreram qualquer tratamento, água bruta do sub-leito (PA903) e superficial (PA802), e água bruta após pré-filtração (PA800). Nos restantes pontos de amostragem, apesar de se verificarem valores inferiores, não se observa uma diminuição deste parâmetro ao longo do tratamento, nem uma variação sazonal. Os valores de TSUVA254nm obtidos são geralmente inferiores a 3 L.mgC-1.m-1, correspondendo a materiais não húmicos, que são considerados biodegradáveis. Para as amostras de água tratada os valores oscilam entre os 1,23 e 1,58 L.mgC-1.m-1, valores inferiores a 2 L.mgC-1.m-1, o que é considerado um valor de referência ao nível do tratamento, segundo a USEPA.

A indústria da construção, nomeadamente no sector da edificação, baseia-se essencialmente em métodos de construção tradicional. Esta indústria é caracterizada pelo consumo excessivo de matérias-primas, de recursos energéticos não renováveis e pela elevada produção de resíduos. Esta realidade é de todo incompatível com os desígnios do desenvolvimento sustentável, nos quais se procura a conveniência harmoniosa entre as dimensões ambiental, social e económica. O desafio da sustentabilidade, colocado à actividade da construção, tem motivado abordagens distintas, não só por parte das várias especialidades da engenharia, como também da arquitectura. É nesta perspectiva, que o presente modelo pretende ser um contributo para uma abordagem inovadora, introduzindo linhas de intervenção e de orientação, para apoiar e estimular o desenvolvimento de soluções sustentáveis em edifícios habitacionais, em qualquer fase do ciclo de evolução de um projecto e das várias especialidades do mesmo. Assim, no sentido de optimizar os recursos envolvidos no projecto são expostas estratégias de intervenção, com os seguintes objectivos: optimização do potencial do local, preservação da identidade regional e cultural, minimização do consumo de energia, utilização de materiais e produtos de baixo impacto ambiental, redução do consumo de água, redução da produção de emissões, resíduos e outros poluentes, adequada qualidade do ambiente interior e optimização das fases de operação e manutenção. A ferramenta apresentada surge como um instrumento facilitador para a equipa de projectistas, e que se esta adaptada para o desenvolvimento de projectos de edifícios de habitação, dada a génese dos métodos utilizados. As soluções de sustentabilidade apresentadas neste manual emanam dos sistemas de certificação LíderA, LEED, BREEAM e SBToolpt. O modelo encontra-se estruturado, no que às fases de projecto diz respeito, de acordo com os requisitos expressos na Portaria 701-H/2008 de 29 de Julho, tendo sido igualmente seguido o descrito para os respectivos intervenientes.

Over the last two decades the research and development of legged locomotion robots has grown steadily. Legged systems present major advantages when compared with ‘traditional’ vehicles, because they allow locomotion in inaccessible terrain to vehicles with wheels and tracks. However, the robustness of legged robots, and especially their energy consumption, among other aspects, still lag behind mechanisms that use wheels and tracks. Therefore, in the present state of development, there are several aspects that need to be improved and optimized. Keeping these ideas in mind, this paper presents the review of the literature of different methods adopted for the optimization of the structure and locomotion gaits of walking robots. Among the distinct possible strategies often used for these tasks are referred approaches such as the mimicking of biological animals, the use of evolutionary schemes to find the optimal parameters and structures, the adoption of sound mechanical design rules, and the optimization of power-based indexes.

A Box–Behnken factorial design coupled with surface response methodology was used to evaluate the effects of temperature, pH and initial concentration in the Cu(II) sorption process onto the marine macroalgae Ascophyllum nodosum. The effect of the operating variables on metal uptake capacitywas studied in a batch system and a mathematical model showing the influence of each variable and their interactions was obtained. Study ranges were 10–40ºC for temperature, 3.0–5.0 for pH and 50–150mgL−1 for initial Cu(II) concentration. Within these ranges, the biosorption capacity is slightly dependent on temperature but markedly increases with pH and initial concentration of Cu(II). The uptake capacities predicted by the model are in good agreement with the experimental values. Maximum biosorption capacity of Cu(II) by A. nodosum is 70mgg−1 and corresponds to the following values of those variables: temperature = 40ºC, pH= 5.0 and initial Cu(II) concentration = 150mgL−1.

Solvent extraction is considered as a multi-criteria optimization problem, since several chemical species with similar extraction kinetic properties are frequently present in the aqueous phase and the selective extraction is not practicable. This optimization, applied to mixer–settler units, considers the best parameters and operating conditions, as well as the best structure or process flow-sheet. Global process optimization is performed for a specific flow-sheet and a comparison of Pareto curves for different flow-sheets is made. The positive weight sum approach linked to the sequential quadratic programming method is used to obtain the Pareto set. In all investigated structures, recovery increases with hold-up, residence time and agitation speed, while the purity has an opposite behaviour. For the same treatment capacity, counter-current arrangements are shown to promote recovery without significant impairment in purity. Recycling the aqueous phase is shown to be irrelevant, but organic recycling with as many stages as economically feasible clearly improves the design criteria and reduces the most efficient organic flow-rate.

Although Mobility is a trendy and an important keyword in education matters, it has been a knowledge tool since the beginning of times, namely the Classical Antiquity, when students were moving from place to place following the masters. Over the time, different types of academic mobility can be found and this tool has been taken both by the education and business sector as almost a compulsory process since the world has gone global. Mobility is, of course, not an end but a means. And as far as academic mobility is concerned it is above all a means to get knowledge, being it theoretical or practical. But why does it still make sense to move from one place to another to get knowledge if never as before we have heaps of information and experiences available around us, either through personal contacts, in books, journals, newspapers or online? With this paper we intend to discuss the purpose of international mobility in the global world of the 21st century as a means to the development of world citizens able to live, work and learn in different and unfamiliar contexts. Based on our own experience as International Coordinator in a Higher Education Institution (HEI) over the last 8 years, on the latest research on academic mobility and still on studies on employability we will show how and why academic mobility can develop skills either in students or in other academic staff that are hardly possible to build in a classroom, or in a non-mobile academic or professional experience and that are highly valued by employers and society in general.

In recent years several countries have set up policies that allow exchange of kidneys between two or more incompatible patient–donor pairs. These policies lead to what is commonly known as kidney exchange programs. The underlying optimization problems can be formulated as integer programming models. Previously proposed models for kidney exchange programs have exponential numbers of constraints or variables, which makes them fairly difficult to solve when the problem size is large. In this work we propose two compact formulations for the problem, explain how these formulations can be adapted to address some problem variants, and provide results on the dominance of some models over others. Finally we present a systematic comparison between our models and two previously proposed ones via thorough computational analysis. Results show that compact formulations have advantages over non-compact ones when the problem size is large.

An optimised version of the Quick, Easy, Cheap, Effective, Rugged and Safe (QuEChERS) method for simultaneous determination of 14 organochlorine pesticides in carrots was developed using gas chromatography coupled with electron-capture detector (GC-ECD) and confirmation by gas chromatography tandem mass spectrometry (GC-MS/MS). A citrate-buffered version of QuEChERS was applied for the extraction of the organochlorine pesticides, and for the extract clean-up, primary secondary amine, octadecyl-bonded silica (C18), magnesium sulphate (MgSO4) and graphitized carbon black were used as sorbents. The GC-ECD determination of the target compounds was achieved in less than 20 min. The limits of detection were below the EUmaximum residue limits (MRLs) for carrots, 10–50 μg kg−1, while the limit of quantification did exceed 10 μg kg−1 for hexachlorobenzene (HCB). The introduction of a sonication step was shown to improve the recoveries. The overall average recoveries in carrots, at the four tested levels (60, 80, 100 and 140 μg kg−1), ranged from 66 to 111% with relative standard deviations in the range of 2– 15 % (n03) for all analytes, with the exception of HCB. The method has been applied to the analysis of 21 carrot samples from different Portuguese regions, and β-HCH was the pesticide most frequently found, with concentrations oscillating between less than the limit of quantification to 14.6 μg kg−1. Only one sample had a pesticide residue (β-HCH) above the MRL, 14.6 μg kg−1. This methodology combines the advantages of both QuEChERS and GC-ECD, producing a very rapid, sensitive and reliable procedure which can be applied in routine analytical laboratories.

The present work describes the optimization of a short-term assay, based on the inhibition of the esterase activity of the alga Pseudokirchneriella subcapitata, in a microplate format. The optimization of the staining procedure showed that the incubation of the algal cells with 20 μmolL−1 fluorescein diacetate (FDA) for 40 min allowed discrimination between metabolic active and inactive cells. The shortterm assay was tested using Cu as toxicant. For this purpose, algal cells, in the exponential or stationary phase of growth, were exposed to the heavy metal in growing conditions. After 3 or 6 h, cells were subsequently stained with FDA, using the optimized procedure. For Cu, the 3- and 6-h EC50 values, based on the inhibition of the esterase activity of algal cells in the exponential phase of growth, were 209 and 130 μg L−1, respectively. P. subcapitata cells, in the stationary phase of growth, displayed higher effective concentration values than those observed in the exponential phase. The 3- and 6-h EC50 values for Cu, for cells in the stationary phase, were 443 and 268 μgL−1, respectively. This short-term microplate assay showed to be a rapid endpoint for testing toxicity using the alga P. subcapitata. The small volume required, the simplicity of the assay (no washing steps), and the automatic reading of the fluorescence make the assay particularly well suited for the evaluation of the toxicity of a high number of environmental samples.

This study aimed to carry out experimental work to determine, for Newtonian and non-Newtonian fluids, the friction factor (fc) with simultaneous heat transfer, at constant wall temperature as boundary condition, in fully developed laminar flow inside a vertical helical coil. The Newtonian fluids studied were aqueous solutions of glycerol, 25%, 36%, 43%, 59% and 78% (w/w). The non-Newtonian fluids were aqueous solutions of carboxymethylcellulose (CMC), a polymer, with concentrations of 0.2%, 0.3%, 0.4% and 0.6% (w/w) and aqueous solutions of xanthan gum (XG), another polymer, with concentrations of 0.1% and 0.2% (w/w). According to the rheological study done, the polymer solutions had shear-thinning behavior and different values of viscoelasticity. The helical coil used has an internal diameter, curvature ratio, length and pitch, respectively: 0.00483 m, 0.0263, 5.0 m and 11.34 mm. It was concluded that the friction factors, with simultaneous heat transfer, for Newtonian fluids can be calculated using expressions from literature for isothermal flows. The friction factors for CMC and XG solutions are similar to those for Newtonian fluids when the Dean number, based in a generalized Reynolds number, is less than 80. For Dean numbers higher than 80, the friction factors of the CMC solutions are lower those of the XG solutions and of the Newtonian fluids. In this range the friction factors decrease with the increase of the viscometric component of the solution and increase for increasing elastic component. The change of behavior at Dean number 80, for Newtonian and non-Newtonian fluids, is in accordance with the study of Ali [4]. There is a change of behavior at Dean number 80, for Newtonian and non-Newtonian fluids, which is in according to previous studies. The data also showed that the use of the bulk temperature or of the film temperature to calculate the physical properties of the fluid has a residual effect in the friction factor values.

The Newtonian fluids studied were aqueous solutions of glycerol, 25%, 36%, 43%, 59% and 78% (w/w) and the non-Newtonian fluids aqueous solutions of carboxymethylcellulose (CMC), a polymer, with concentrations 0.1%, 0.2%, 0.3%, 0.4% and 0.6% (w/w) and aqueous solutions of xanthan gum (XG), another polymer, with concentrations 0.1% and 0.2% (w/w). According to the rheological study performed, the polymer solutions had shear thinning behavior and different values of elasticity. The helical coil used has internal diameter, curvature ratio, length and pitch, respectively: 0.004575 m, 0.0263, 5.0 m and 11.34 mm. The Nusselt numbers for the CMC solutions are, on average, slightly higher than those for Newtonian fluids, for identical Prandtl and generalized Dean numbers. As outcome, the viscous component of the shear thinning polymer tends to potentiate the mixing effect of the Dean cells. The Nusselt numbers of the XG solutions are significant lower than those of the Newtonian solutions, for identical Prandtl and generalized Dean numbers. Therefore, the elastic component of the polymer tends to diminish the mixing effect of the Dean cells. A global correlation, for Nusselt number as a function of Péclet, generalized Dean and Weissenberg numbers for all Newtonian and non-Newtonian solutions studied, is presented.

Background: Current therapeutic strategies for advanced prostate cancer (PCa) are largely ineffective. Because aberrant DNA methylation associated with inappropriate gene-silencing is a common feature of PCa, DNA methylation inhibitors might constitute an alternative therapy. In this study we aimed to evaluate the anti-cancer properties of RG108, a novel non-nucleoside inhibitor of DNA methyltransferases (DNMT), in PCa cell lines. Methods: The anti-tumoral impact of RG108 in LNCaP, 22Rv1, DU145 and PC-3 cell lines was assessed through standard cell viability, apoptosis and cell cycle assays. Likewise, DNMT activity, DNMT1 expression and global levels of DNA methylation were evaluated in the same cell lines. The effectiveness of DNA demethylation was further assessed through the determination of promoter methylation and transcript levels of GSTP1, APC and RAR-β2, by quantitative methylation-specific PCR and RT-PCR, respectively. Results: RG108 led to a significant dose and time dependent growth inhibition and apoptosis induction in LNCaP, 22Rv1 and DU145. LNCaP and 22Rv1 also displayed decreased DNMT activity, DNMT1 expression and global DNA methylation. Interestingly, chronic treatment with RG108 significantly decreased GSTP1, APC and RAR-β2 promoter hypermethylation levels, although mRNA re-expression was only attained GSTP1 and APC. Conclusions: RG108 is an effective tumor growth suppressor in most PCa cell lines tested. This effect is likely mediated by reversion of aberrant DNA methylation affecting cancer related-genes epigenetically silenced in PCa. However, additional mechanism might underlie the anti-tumor effects of RG108. In vivo studies are now mandatory to confirm these promising results and evaluate the potential of this compound for PCa therapy.
