A gold screen printed electrode (Au-SPE) was modified by merging Molecular Imprinting and Self-Assembly Monolayer techniques for fast screening cardiac biomarkers in point-of-care (POC). For this purpose, Myoglobin (Myo) was selected as target analyte and its plastic antibody imprinted over a glutaraldehyde (Glu)/cysteamine (Cys) layer on the gold-surface. The imprinting effect was produced by growing a reticulated polymer of acrylamide (AAM) and N,N′-methylenebisacrylamide (NNMBA) around the Myo template, covalently attached to the biosensing surface. Electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV) studies were carried out in all chemical modification steps to confirm the surface changes in the Au-SPE. The analytical features of the resulting biosensor were studied by different electrochemical techniques, including EIS, square wave voltammetry (SWV) and potentiometry. The limits of detection ranged from 0.13 to 8 μg/mL. Only potentiometry assays showed limits of detection including the cut-off Myo levels. Quantitative information was also produced for Myo concentrations ≥0.2 μg/mL. The linear response of the biosensing device showed an anionic slope of ~70 mV per decade molar concentration up to 0.3 μg/mL. The interference of coexisting species was tested and good selectivity was observed. The biosensor was successfully applied to biological fluids.

This work introduces two major changes to the conventional protocol for designing plastic antibodies: (i) the imprinted sites were created with charged monomers while the surrounding environment was tailored using neutral material; and (ii) the protein was removed from its imprinted site by means of a protease, aiming at preserving the polymeric network of the plastic antibody. To our knowledge, these approaches were never presented before and the resulting material was named here as smart plastic antibody material (SPAM). As proof of concept, SPAM was tailored on top of disposable gold-screen printed electrodes (Au-SPE), following a bottom-up approach, for targeting myoglobin (Myo) in a point-of-care context. The existence of imprinted sites was checked by comparing a SPAM modified surface to a negative control, consisting of similar material where the template was omitted from the procedure and called non-imprinted materials (NIMs). All stages of the creation of the SPAM and NIM on the Au layer were followed by both electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). AFM imaging was also performed to characterize the topography of the surface. There are two major reasons supporting the fact that plastic antibodies were effectively designed by the above approach: (i) they were visualized for the first time by AFM, being present only in the SPAM network; and (ii) only the SPAM material was able to rebind to the target protein and produce a linear electrical response against EIS and square wave voltammetry (SWV) assays, with NIMs showing a similar-to-random behavior. The SPAM/Au-SPE devices displayed linear responses to Myo in EIS and SWV assays down to 3.5 μg/mL and 0.58 μg/mL, respectively, with detection limits of 1.5 and 0.28 μg/mL. SPAM materials also showed negligible interference from troponin T (TnT), bovine serum albumin (BSA) and urea under SWV assays, showing promising results for point-of-care applications when applied to spiked biological fluids.

A novel surface molecularly-imprinted (MI) material to detect myoglobin (Myo) using gold screen printed electrodes (SPE) was developed. The sensitive detection was carry out by introducing a carboxylic polyvinyl chloride (PVC-COOH) layer on gold SPE surface. Myo was attached to the surface of gold SPE/PVC-COOH and the vacant spaces around it were filled by polymerizing acrylamide and N,N-methylenebisacrylamide (cross-linker). This polymerization was initiated by ammonium persulphate. After removing the template, the obtained material was able to rebind Myo and discriminate it among other interfering species. Various characterization techniques including electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV) confirmed the surface modification. This sensor seemed a promising tool for screening Myo in point-of-care.

A novel artificial antibody for troponin T (TnT) was synthesized by molecular imprint (MI) on the surface of multiwalled carbon nanotubes (MWCNT). This was done by attaching TnT to the MWCNT surface, and filling the vacant spaces by polymerizing under mild conditions acrylamide (monomer) in N,N′-methylenebisacrylamide (cross-linker) and ammonium persulphate (initiator). After removing the template, the obtained biomaterial was able to rebind TnT and discriminate it among other interfering species. Stereochemical recognition of TnT was confirmed by the non-rebinding ability displayed by non-imprinted (NI) materials, obtained by imprinting without a template. SEM and FTIR analysis confirmed the surface modification of the MWCNT. The ability of this biomaterial to rebind TnT was confirmed by including it as electroactive compound in a PVC/plasticizer mixture coating a wire of silver, gold or titanium. Anionic slopes of 50 mV decade−1 were obtained for the gold wire coated with MI-based membranes dipped in HEPES buffer of pH 7. The limit of detection was 0.16 μg mL−1. Neither the NI-MWCNT nor the MWCNT showed the ability to recognize the template. Good selectivity was observed against creatinine, sucrose, fructose, myoglobin, sodium glutamate, thiamine and urea. The sensor was tested successfully on serum samples. It is expected that this work opens new horizons on the design of new artificial antibodies for complex protein structures.

Myoglobin (Mb) is among the cardiac biomarkers playing a major role in urgent diagnosis of cardiovascular diseases. Its monitoring in point-of-care is therefore fundamental. Pursuing this goal, a novel biomimetic ionophore for the potentiometric transduction of Mb is presented. It was synthesized by surface molecular imprinting (SMI) with the purpose of developing highly efficient sensor layers for near-stereochemical recognition of Mb. The template (Mb) was imprinted on a silane surface that was covalently attached to silica beads by means of self-assembled monolayers. First the silica was modified with an external layer of aldehyde groups. Then, Mb was attached by reaction with its amine groups (on the external surface) and subsequent formation of imine bonds. The vacant places surrounding Mb were filled by polymerization of the silane monomers 3-aminopropyltrimethoxysilane (APTMS) and propyltrimethoxysilane (PTMS). Finally, the template was removed by imine cleavage after treatment with oxalic acid. The results materials were finely dispersed in plasticized PVC selective membranes and used as ionophores in potentiometric transduction. The best analytical features were found in HEPES buffer of pH 4. Under this condition, the limits of detection were of 1.3 × 10−6 mol/L for a linear response after 8.0 × 10−7 mol/L with an anionic slope of −65.9 mV/decade. The imprinting effect was tested by preparing non-imprinted (NI) particles and employing these materials as ionophores. The resulting membranes showed no ability to detect Mb. Good selectivity was observed towards creatinine, sacarose, fructose, galactose, sodium glutamate, and alanine. The analytical application was conducted successfully and showed accurate and precise results.

Robotics research in Portugal is increasing every year, but few students embrace it as one of their first choices for study. Until recently, job offers for engineers were plentiful, and those looking for a degree in science and technology would avoid areas considered to be demanding, like robotics. At the undergraduate level, robotics programs are still competing for a place in the classical engineering graduate curricula. Innovative and dynamic Master's programs may offer the solution to this gap. The Master's degree in autonomous systems at the Instituto Superior de Engenharia do Porto (ISEP), Porto, Portugal, was designed to provide a solid training in robotics and has been showing interesting results, mainly due to differences in course structure and the context in which students are welcomed to study and work.

O BIM – Building Information Modeling – é um conceito de controlo e gestão de informação desenvolvida, entre diferentes especialidades e intervenientes envolventes, durante o ciclo de vida das construções. A utilização das tecnologias BIM, no ramo da Engenharia Mecânica e Civil, tem sido uma aposta constante, e cada vez mais concisa, nos projetos de construção. A justificação para a adoção destas metodologias mais eficientes, em substituição dos processos convencionais, prende-se com o facto desses processos convencionais, ainda hoje, apresentarem muitas dificuldades e problemas associados, por exemplo, à falta de comunicação entre os intervenientes e ao ineficiente controlo na gestão de projetos. O objetivo desta dissertação centra-se na análise da interoperabilidade de softwares BIM, ou seja, na verificação da viabilidade de exportação de dados dos modelos produzidos, entre as ferramentas BIM. Para este campo de ação contribuirá a análise da passagem de informação, relativa ao modelo de uma Nave Industrial modelada, em alguns dos softwares BIM, correntemente mais utilizados. O conhecimento adquirido com a modelação do caso de estudo do presente trabalho irá permitir identificar algumas lacunas existentes ao nível da falta de recomendações práticas que sirvam de orientação na modelação recorrendo a ferramentas informáticas BIM-compatíveis. Assim, a base deste trabalho consiste na criação de uma série de recomendações ou de um roteiro de modelação em específico para a disciplina de estruturas enquanto área de aplicação concreta do estudo efetuado. Nesse sentido, a proposta deste trabalho é de apresentar alguns critérios de modelação onde são definidos os elementos a modelar em cada fase do projeto.

It is unquestionable that an effective decision concerning the usage of a certain environmental clean-up technology should be conveniently supported. Significant amount of scientific work focussing on the reduction of nitrate concentration in drinking water by both metallic iron and nanomaterials and their usage in permeable reactive barriers has been worldwide published over the last two decades. This work aims to present in a systematic review of the most relevant research done on the removal of nitrate from groundwater using nanosized iron based permeable reactive barriers. The research was based on scientific papers published between 2004 and June 2014. It was performed using 16 combinations of keywords in 34 databases, according to PRISMA statement guidelines. Independent reviewers validated the selection criteria. From the 4161 records filtered, 45 met the selection criteria and were selected to be included in this review. This study's outcomes show that the permeable reactive barriers are, indeed, a suitable technology for denitrification and with good performance record but the long-term impact of the use of nanosized zero valent iron in this remediation process, in both on the environment and on the human health, is far to be conveniently known. As a consequence, further work is required on this matter, so that nanosized iron based permeable reactive barriers for the removal of nitrate from drinking water can be genuinely considered an eco-efficient technology.

O presente relatório de estágio foi elaborado no âmbito da Unidade Curricular de DIPRE, Dissertação/Projeto/Estágio, do 2.º ano de Mestrado em Engenharia Civil do Instituto Superior de Engenharia do Porto, do ramo de estruturas, tendo como principal foco, a análise e dimensionamento de madres de aço enformado a frio e sua utilização com painéis do tipo sandwich em coberturas. Seções enformadas a frio são cada vez mais utilizadas em construções modernas, especificamente como estrutura secundária em coberturas, onde geralmente são fixas a painéis através de ligações aparafusadas. A presença de painéis e fixações através de parafusos permitem a estabilização lateral e torsional de madres aumentando desta forma a capacidade resistente, mas por serem elementos estruturais de espessura reduzida, os enformados a frio são suscetíveis a fenómenos de instabilidade associados. Desta forma, a norma EN 1993-1-3 [4] permite a análise e dimensionamento deste tipo de elementos através das disposições regulamentares preconizadas nas partes 1-1 [3] e 1-5 [5] da mesma norma. Num primeiro estudo, o presente trabalho tem como objetivo o dimensionamento e verificação de segurança de elementos enformados a frio com base na seção efetiva determinada com o auxílio das normas EN 1993-1-1 (regras gerais) e EN 1993-1-5 (regras para elementos estruturais constituídos por placas). Numa segunda fase, este trabalho pretende apresentar um estudo do comportamento de interação entre os sistemas madres-painéis. Para tal, são quantificadas as rigidezes das conexões dos sistemas e dos painéis para se realizarem a análise relativamente à restrição lateral e restrição torsional de madres. Neste contexto, concluiu-se que os painéis, quando fixos de forma adequada às madres, contribuem para a estabilidade.

O presente relatório diz respeito ao trabalho desenvolvido durante os seis meses do estágio curricular, realizado no âmbito do Mestrado em Engenharia Civil, Ramo de Construções, na empresa Porto Vivo, SRU - Sociedade de Reabilitação Urbana da Baixa Portuense, mais precisamente no Núcleo de Gestão de Obras (NGO). Ao longo do período de estágio, foram realizadas diversas atividades, tais como: o acompanhamento das obras de reabilitação no Centro Histórico do Porto, a análise dos projetos em curso, a realização de vistorias para determinação do nível de conservação de edifícios e a participação em reuniões de obra com empreiteiros e projetistas. Todas estas atividades serão abordadas ao longo deste relatório. Para além da descrição de todas as tarefas desenvolvidas na empresa, este relatório aborda também uma temática muito importante na Reabilitação de Edifícios, o comportamento acústico. Será realizado um estudo sobre o desempenho acústico de alguns edifícios, caracterizando as soluções construtivas adotadas em obra.

Neste trabalho foi considerada a possibilidade de incorporar serviços remotos, normalmente associados a serviços web e cloud computing, numa solução local que centralizasse os vários serviços num único sistema e permitisse aos seus utilizadores consumir e configurar os mesmos, quer a partir da rede local, quer remotamente a partir da Internet. Desta forma seria possível conciliar o acesso a partir de qualquer local com internet, característico nas clouds, com a simplicidade de concentrar num só sistema vários serviços que são por norma oferecidos por entidades distintas e ainda permitir aos seus utilizadores o controlo e configuração sobre os mesmos. De forma a validar que este conceito é viável, prático e funcional, foram implementadas duas componentes. Um cliente que corre nos dispositivos dos utilizadores e que proporciona a interface para consumir os serviços disponíveis e um servidor que irá conter e prestar esses serviços aos clientes. Estes serviços incluem lista de contactos, mensagens instantâneas, salas de conversação, transferência de ficheiros, chamadas e conferências de voz e vídeo, pastas remotas, pastas sincronizadas, backups, pastas partilhadas, VoD (Video-on Demand) e AoD (Audio-on Demand). Para o desenvolvimento do cliente e do servidor foi utilizada a framework Qt que recorre à linguagem de programação C++ e ao conjunto de bibliotecas que possui, para o desenvolvimento de aplicações multiplataforma. Para as comunicações entre clientes e servidor, foi utilizado o protocolo XMPP (Extensible Messaging and Presence Protocol), pela forma da biblioteca qxmpp e do servidor XMPP ejabberd. Pelo facto de conter um conjunto de centenas de extensões atualmente ativas que auferem funcionalidades como salas de conversação, transferências de ficheiros e até estabelecer sessões multimédia, graças à sua flexibilidade permitiu ainda a criação de extensões personalizada necessárias para algumas funcionalidades que se pretendeu implementar. Foi ainda utilizado no servidor a framework ffmpeg para suportar algumas funcionalidades multimédia. Após a implementação do cliente para Windows e Linux, e de implementar o servidor em Linux foi realizado um conjunto de testes funcionais para perceber se as funcionalidades e seus mecanismos funcionam corretamente. No caso onde a análise da performance e do consumo de recursos era importante, foram realizados testes de performance e testes de carga.
