A 5-unit polyubiquitin gene, TTU3, was isolated from a T. thermophila genomic library and sequenced. This gene presents an extra triplet coding for Phe, a AGAGA motif and a putative HSE element in its 5'-non-coding region. The ubiquitin gene expression in this ciliate was investigated by Northern blot hybridization in conjugating cells or cells under stress conditions. Exponentially growing cells express two ubiquitin mRNAs of 0.75 and 1.8 kb and a new species of 1.4 kb is induced under hyperthermic stress. During sexual reproduction of the cells (conjugation) the 1.8-kb mRNA is still transcribed whereas the steady-state population of the 0.75 mRNA transcripts is strongly diminished. Southern blot analysis suggests that ubiquitin in T. thermophila constitutes a large family of about ten members.

We report here the cloning and the characterization of the T. pyriformis CCT eta gene (TpCCT eta) and also a partial sequence of the corresponding T. thermophila gene (TtCCT eta). The TpCCt eta gene encodes a protein sharing a 60.3% identity with the mouse CCT eta. We have studied the expression of these genes in Tetrahymena exponentially growing cells, cells regenerating their cilia for different periods and during different stages of the cell sexual reproduction. These genes have similar patterns of expression to those of the previously identified TpCCt gamma gene. Indeed, the Tetrahymena CCT eta and CCT gamma genes are up-regulated at 60-120 min of cilia recovery, and in conjugation when vegetative growth was resumed and cell division took place. Our results seem to indicate that both CCT subunits play an important role in the biogenesis of the newly synthesized cilia of Tetrahymena and during its cell division.

O abuso sexual de crianças, não se tratando de um fenómeno novo, caracteriza-se, no entanto, pela dificuldade e demora no reconhecimento social e científico, enquanto problemática, sendo ainda mais recente a preocupação da investigação científica de caraterizar e compreender o abuso sexual de crianças a partir das suas vozes. Os estudos sociológicos sobre o abuso sexual têm sido escassos. Não obstante, o facto de se tratar de um fenómeno complexo e multidimensional, onde se acumulam diversos fatores e consequências, urge reconhecer e estudar sociologicamente o fenómeno, porque (i) possibilitará discutir a forma como a dominação sexual e geracional persiste nas sociedades contemporâneas e, ainda, se reveste de outros contornos; (ii) encarará o fenómeno como uma violação sistemática dos direitos humanos e dos direitos da criança na discussão do fenómeno; (iii) contribuirá para a discussão sobre a necessidade de uma intervenção política e social mais adequada e eficaz. Este texto desdobra-se ao longo de três dimensões de análise. Em primeiro lugar, olha o abuso sexual de crianças a partir do enfoque dos direitos da criança, explicitando os esforços que têm vindo a ser desenvolvidos para proteger os direitos das crianças à escala nacional e internacional. Segue-se a análise quantitativa e qualitativa dos dados públicos sobre o abuso sexual de crianças produzidos por três organismos nacionais: o Ministério de Administração Interna (MAI), a Comissão Nacional de Proteção de Crianças e Jovens em Risco (CNPCJR) e a Associação Portuguesa de Apoio à Vítima (APAV). Por fim, convocam-se as vozes de crianças sobre o fenómeno, no sentido de atribuir à análise desta problemática uma tessitura mais densa, reveladora daquelas que são as suas complexidades, o que na nossa opinião, contribui com novos olhares sobre uma velha-nova problemática.

Cada vez mais os profissionais de saúde aspiram contribuir para o maior bem-estar possível dos seus doentes. Neste contexto, torna-se indispensável identificar variáveis, idealmente passíveis de ser modificadas, que estejam relacionadas com o bem-estar dos doentes. Assim, o objectivo do presente estudo é explorar a relação entre bem-estar geral (BEG), adesão ao tratamento (AT) e funcionamento sexual (FS) em indivíduos com epilepsia. Foram avaliados 79 indivíduos com epilepsia (59,5% do sexo feminino; idade: M = 36,10, DP = 11,09; diagnóstico (anos): M = 19,72, DP = 11,50), recorrendo a um Questionário Sócio-demográfico e Clínico, ao Índice de Bem-estar Pessoal, à Medida de Adesão aos Tratamentos e à Escala de Funcionamento Sexual da Multiple Sclerosis Quality of Life Scale com 54 itens (MSQOL-54). Verificaram-se bons indicadores globais de BEG, AT e FS na amostra. Verificaram-se correlações estatisticamente significativas entre o BEG e: a AT (rs(75) = 0,37, p ≤ 0,001) e o FS feminino (rs(35) = -0,44, p ≤ 0,008). Não se verificou uma correlação estatisticamente significativa entre BEG e FS masculino. Ao analisar os indicadores parciais, verificou-se que o FS masculino se correlacionava apenas com a satisfação com a segurança do seu futuro (rs(31) = -0,40, p ≤ 0,03), enquanto o FS feminino se correlacionava com este indicador (rs(35) = -0,36, p ≤ 0,04), com a satisfação com o seu nível de vida (rs(34) = -0,37, p ≤ 0,04) e com a satisfação com a sua vida espiritual ou com a sua religião (rs(35) = -0,40, p ≤ 0,02). Sem esquecer o efectivo da amostra e os indicadores genericamente positivos da mesma, os presentes resultados sugerem que a promoção da AT pode saldar-se por melhorias no BEG dos indivíduos com epilepsia. Já a intervenção ao nível do FS parece apenas promissora, ao nível do BEG, para as doentes. Novas variáveis devem ser exploradas, para que se consigam identificar os melhores preditores do BEG (de amostras mais amplas) destes doentes.

Two distinct subsets of γδ T cells that produce interleukin 17 (IL-17) (CD27(-) γδ T cells) or interferon-γ (IFN-γ) (CD27(+) γδ T cells) develop in the mouse thymus, but the molecular determinants of their functional potential in the periphery remain unknown. Here we conducted a genome-wide characterization of the methylation patterns of histone H3, along with analysis of mRNA encoding transcription factors, to identify the regulatory networks of peripheral IFN-γ-producing or IL-17-producing γδ T cell subsets in vivo. We found that CD27(+) γδ T cells were committed to the expression of Ifng but not Il17, whereas CD27(-) γδ T cells displayed permissive chromatin configurations at loci encoding both cytokines and their regulatory transcription factors and differentiated into cells that produced both IL-17 and IFN-γ in a tumor microenvironment.

Ainda que a saúde mental dos doentes crónicos seja cada vez mais uma preocupação dos profissionais de saúde, não é generalizada a avaliação rotineira do seu funcionamento sexual, que poderá ter impacto sobre a sua saúde mental. O objectivo do presente estudo é explorar a relação entre funcionamento sexual e saúde mental em doentes crónicos. Foram avaliados 77 adultos com diabetes tipo 1, 40 com diabetes tipo 2, 100 com esclerose múltipla, 79 com epilepsia, 205 com obesidade e 106 com cancro, recorrendo a um Questionário Sócio-demográfico e Clínico, à Escala de Função Sexual do MSQOL-54 e à Escala de Saúde Mental do SF-36. Na amostra total, verificaram-se correlações lineares estatisticamente significativas entre Funcionamento Sexual e Saúde Mental nos dois sexos (homens: r(161)=-0,35, p<0,0001; mulheres: r(387)=-0,36, p<0,0001). Entre os homens, as correlações oscilaram entre rs(33)=-0,58 (p<0,0001) e rs(34)=-0,21 (p=0,23); entre as mulheres, entre rs(161)=-0,49 (p<0,0001) e rs(19)=-0,03 (p=0,89). Mais concretamente, nos indivíduos com diabetes tipo 1 e cancro verificaram-se correlações lineares estatisticamente significativas entre Funcionamento Sexual e Saúde Mental nos dois sexos; nos indivíduos com diabetes tipo 2 e esclerose múltipla não se verificaram correlações significativas em nenhum dos sexos; nos indivíduos com obesidade só se verificaram correlações significativas nos indivíduos do sexo feminino; nos indivíduos com epilepsia só se verificaram correlações significativas nos indivíduos do sexo masculino. Os resultados sugerem que a promoção do funcionamento sexual de doentes crónicos poderá saldar-se por uma melhoria na sua saúde mental (e vice-versa), mas não em todas as doenças crónicas analisadas.

Alzheimer Disease (AD) is characterized by progressive cognitive decline and dementia. Earlier diagnosis and classification of different stages of the disease are currently the main challenges and can be assessed by neuroimaging. With this work we aim to evaluate the quality of brain regions and neuroimaging metrics as biomarkers of AD. Multimodal Imaging Brain Connectivity Analysis (MIBCA) toolbox functionalities were used to study AD by T1weighted, Diffusion Tensor Imaging and 18FAV45 PET, with data obtained from the AD Neuroimaging Initiative database, specifically 12 healthy controls (CTRL) and 33 patients with early mild cognitive impairment (EMCI), late MCI (LMCI) and AD (11 patients/group). The metrics evaluated were gray-matter volume (GMV), cortical thickness (CThk), mean diffusivity (MD), fractional anisotropy (FA), fiber count (FiberConn), node degree (Deg), cluster coefficient (ClusC) and relative standard-uptake-values (rSUV). Receiver Operating Characteristic (ROC) curves were used to evaluate and compare the diagnostic accuracy of the most significant metrics and brain regions and expressed as area under the curve (AUC). Comparisons were performed between groups. The RH-Accumbens/Deg demonstrated the highest AUC when differentiating between CTRLEMCI (82%), whether rSUV presented it in several brain regions when distinguishing CTRL-LMCI (99%). Regarding CTRL-AD, highest AUC were found with LH-STG/FiberConn and RH-FP/FiberConn (~100%). A larger number of neuroimaging metrics related with cortical atrophy with AUC>70% was found in CTRL-AD in both hemispheres, while in earlier stages, cortical metrics showed in more confined areas of the temporal region and mainly in LH, indicating an increasing of the spread of cortical atrophy that is characteristic of disease progression. In CTRL-EMCI several brain regions and neuroimaging metrics presented AUC>70% with a worst result in later stages suggesting these indicators as biomarkers for an earlier stage of MCI, although further research is necessary.

We present the modeling efforts on antenna design and frequency selection to monitor brain temperature during prolonged surgery using noninvasive microwave radiometry. A tapered log-spiral antenna design is chosen for its wideband characteristics that allow higher power collection from deep brain. Parametric analysis with the software HFSS is used to optimize antenna performance for deep brain temperature sensing. Radiometric antenna efficiency (eta) is evaluated in terms of the ratio of power collected from brain to total power received by the antenna. Anatomical information extracted from several adult computed tomography scans is used to establish design parameters for constructing an accurate layered 3-D tissue phantom. This head phantom includes separate brain and scalp regions, with tissue equivalent liquids circulating at independent temperatures on either side of an intact skull. The optimized frequency band is 1.1-1.6 GHz producing an average antenna efficiency of 50.3% from a two turn log-spiral antenna. The entire sensor package is contained in a lightweight and low-profile 2.8 cm diameter by 1.5 cm high assembly that can be held in place over the skin with an electromagnetic interference shielding adhesive patch. The calculated radiometric equivalent brain temperature tracks within 0.4 degrees C of the measured brain phantom temperature when the brain phantom is lowered 10. C and then returned to the original temperature (37 degrees C) over a 4.6-h experiment. The numerical and experimental results demonstrate that the optimized 2.5-cm log-spiral antenna is well suited for the noninvasive radiometric sensing of deep brain temperature.

Since the first in vivo studies of cerebral function with radionuclides by Ingvar and Lassen, nuclear medicine (NM) brain applications have evolved dramatically, with marked improvements in both methods and tracers. Consequently it is now possible to assess not only cerebral blood flow and energy metabolism but also neurotransmission. Planar functional imaging was soon substituted by single-photon emission computed tomography (SPECT) and positron emission tomography (PET); it now has limited application in brain imaging, being reserved for the assessment of brain death.

Brain dopamine transporters imaging by Single Emission Tomography (SPECT) with 123I-FP-CIT (DaTScanTM) has become an important tool in the diagnosis and evaluation of Parkinson syndromes.This diagnostic method allows the visualization of a portion of the striatum – where healthy pattern resemble two symmetric commas - allowing the evaluation of dopamine presynaptic system, in which dopamine transporters are responsible for dopamine release into the synaptic cleft, and their reabsorption into the nigrostriatal nerve terminals, in order to be stored or degraded. In daily practice for assessment of DaTScan TM, it is common to rely only on visual assessment for diagnosis. However, this process is complex and subjective as it depends on the observer’s experience and it is associated with high variability intra and inter observer. Studies have shown that semiquantification can improve the diagnosis of Parkinson syndromes. For semiquantification, analysis methods of image segmentation using regions of interest (ROI) are necessary. ROIs are drawn, in specific - striatum - and in nonspecific – background – uptake areas. Subsequently, specific binding ratios are calculated. Low adherence of semiquantification for diagnosis of Parkinson syndromes is related, not only with the associated time spent, but also with the need of an adapted database of reference values for the population concerned, as well as, the examination of each service protocol. Studies have concluded, that this process increases the reproducibility of semiquantification. The aim of this investigation was to create and validate a database of healthy controls for Dopamine transporters with DaTScanTM named DBRV. The created database has been adapted to the Nuclear Medicine Department’s protocol, and the population of Infanta Cristina’s Hospital located in Badajoz, Spain.

Brain dopamine transporters imaging by Single Photon Emission Tomography (SPECT) with 123I-FP-CIT has become an important tool in the diagnosis and evaluation of parkinsonian syndromes, since this radiopharmaceutical exhibits high affinity for membrane transporters responsible for cellular reabsorption of dopamine on the striatum. However, Ordered Subset Expectation Maximization (OSEM) is the method recommended in the literature for imaging reconstruction. Filtered Back Projection (FBP) is still used due to its fast processing, even if it presents some disadvantages. The aim of this work is to investigate the influence of reconstruction parameters for FBP in semiquantification of Brain Studies with 123I-FPCIT compared with those obtained with OSEM recommended reconstruction.
