Era objectivo do presente trabalho o desenvolvimento de um biossensor baseado na inibição da amidase de Pseudomonas aeruginosa para a quantificação de ureia em diversas amostras com recurso a um eléctrodo selectivo de iões amónio (ISE). A ureia é um poderoso inibidor do centro activo da amidase (Acilamida hidrolase EC 3.5.1.4) de Pseudomonas aeruginosa a qual catalisa a hidrólise de amidas alifáticas produzindo o ácido correspondente e amónia. O extracto celular de Pseudomonas aeruginosa L10 contendo actividade de amidase foi imobilizado em membranas de poliétersulfona modificadas (PES) e em membranas de nylon Porablot NY Plus na presença de gelatina e de glutaraldeído (GA) como agente bifuncional. Estas membranas foram posteriormente utilizadas na construção do biossensor baseado no ISE, utilizando acetamida como substrato, a reacção enzimática foi seguida medindo os iões amónio produzidos pela hidrólise da amida alifática, e a resposta do biossensor apresentada como a velocidade inicial da reacção (mV.min-1). A optimização dos parâmetros de imobilização foi efectuada de acordo com a metodologia ANOVA. Assim, a mistura de 30μL extracto celular, 2μL GA (5%) e 10 μL Gelatina 15% (p/v) foi a que conduziu a uma melhor resposta do biossensor. Efectuou-se ainda o estudo de optimização de alguns parâmetros experimentais pH e tempo de incubação em ureia, este conduziu ao valor pH=7,2 como pH óptimo de resposta do biossensor e 20 min como tempo óptimo de incubação das membranas nas soluções de ureia, sendo neste caso a resposta do biossensor dada pela diferença das respostas do biossensor antes e após incubação. A calibração do biossensor foi efectuada em soluções contendo concentrações conhecidas de ureia preparadas em tampão Tris, leite e vinho caseiro, exibindo um limite de detecção de 2,0 ×10-6 M de ureia. A incubação das membranas em hidroxilamina 2M por um período de 2h permitiu a recuperação de 70% da actividade enzimática da membrana. O biossensor apresentou uma elevada estabilidade de armazenamento por um período de 55 dias revelando uma perda de apenas 15% da sua resposta. O biossensor desenvolvido apresenta uma sensibilidade de 58,245 mV.min-1 e um tempo de resposta de aproximadamente 20s. A resposta do biossensor foi linear para concentrações de ureia presentes no vinho na gama de 4-10 μM de ureia.

The crustal and lithospheric mantle structure at the south segment of the west Iberian margin was investigated along a 370 km long seismic transect. The transect goes from unthinned continental crust onshore to oceanic crust, crossing the ocean-continent transition (OCT) zone. The wide-angle data set includes recordings from 6 OBSs and 2 inland seismic stations. Kinematic and dynamic modeling provided a 2D velocity model that proved to be consistent with the modeled free-air anomaly data. The interpretation of coincident multi-channel near-vertical and wide-angle reflection data sets allowed the identification of four main crustal domains: (i) continental (east of 9.4 degrees W); (ii) continental thinning (9.4 degrees W-9.7 degrees W): (iii) transitional (9.7 degrees W-similar to 10.5 degrees W); and (iv) oceanic (west of similar to 10.5 degrees W). In the continental domain the complete crustal section of slightly thinned continental crust is present. The upper (UCC, 5.1-6.0 km/s) and the lower continental crust (LCC, 6.9-7.2 km/s) are seismically reflective and have intermediate to low P-wave velocity gradients. The middle continental crust (MCC, 6.35-6.45 km/s) is generally unreflective with low velocity gradient. The main thinning of the continental crust occurs in the thinning domain by attenuation of the UCC and the LCC. Major thinning of the MCC starts to the west of the LCC pinchout point, where it rests directly upon the mantle. In the thinning domain the Moho slope is at least 13 degrees and the continental crust thickness decreases seaward from 22 to 11 km over a similar to 35 km distance, stretched by a factor of 1.5 to 3. In the oceanic domain a two-layer high-gradient igneous crust (5.3-6.0 km/s; 6.5-7.4 km/s) was modeled. The intra-crustal interface correlates with prominent mid-basement, 10-15 km long reflections in the multi-channel seismic profile. Strong secondary reflected PmP phases require a first order discontinuity at the Moho. The sedimentary cover can be as thick as 5 km and the igneous crustal thickness varies from 4 to 11 km in the west, where the profile reaches the Madeira-Tore Rise. In the transitional domain the crust has a complex structure that varies both horizontally and vertically. Beneath the continental slope it includes exhumed continental crust (6.15-6.45 km/s). Strong diffractions were modeled to originate at the lower interface of this layer. The western segment of this transitional domain is highly reflective at all levels, probably due to dykes and sills, according to the high apparent susceptibility and density modeled at this location. Sub-Moho mantle velocity is found to be 8.0 km/s, but velocities smaller than 8.0 km/s confined to short segments are not excluded by the data. Strong P-wave wide-angle reflections are modeled to originate at depth of 20 km within the lithospheric mantle, under the eastern segment of the oceanic domain, or even deeper at the transitional domain, suggesting a layered structure for the lithospheric mantle. Both interface depths and velocities of the continental section are in good agreement to the conjugate Newfoundland margin. A similar to 40 km wide OCT having a geophysical signature distinct from the OCT to the north favors a two pulse continental breakup.

Aflatoxin B1 (AFB1) has been recognized to produce cancer in human liver. In addition, epidemiological and laboratory studies demonstrated that the respiratory system was a target for AFB1. Exposure occurs predominantly through the food chain, but inhalation represents an additional route of exposure. The present study aimed to examine AFB1 exposure among poultry workers in Portugal. Blood samples were collected from a total of 31 poultry workers from six poultry farms. In addition, a control group (n = 30) was included comprised of workers who undertook administrative tasks. Measurement of AFB1 in serum was performed by enzyme-linked immunosorbent assay (ELISA). For examining fungi contamination, air samples were collected through an impaction method. Air sampling was obtained in pavilion interior and outside the premises, since this was the place regarded as the reference location. Using molecular methods, toxicogenic strains (aflatoxin-producing) were investigated within the group of species belonging to Aspergillus flavus complex. Eighteen poultry workers (59%) had detectable levels of AFB1 with values ranging from <1 ng/ml to4.23 ng/ml and with a mean value of 2 ± 0.98ng/ml. AFB1 was not detected in the serum sampled from any of the controls. Aspergillus flavus was the fungal species third most frequently found in the indoor air samples analyzed (7.2%) and was the most frequently isolated species in air samples containing only Aspergillus genus (74.5%). The presence of aflatoxigenic strains was only confirmed in outdoor air samples from one of the units, indicating the presence of a source inside the building in at least one case. Data indicate that AFB1 inhalation represents an additional risk in this occupational setting that needs to be recognized, assessed, and prevented.

A biosensor for urea has been developed based on the observation that urea is a powerful active-site inhibitor of amidase, which catalyzes the hydrolysis of amides such as acetamide to produce ammonia and the corresponding organic acid. Cell-free extract from Pseudomonas aeruginosa was the source of amidase (acylamide hydrolase, EC 3.5.1.4) which was immobilized on a polyethersulfone membrane in the presence of glutaraldehyde; anion-selective electrode for ammonium ions was used for biosensor development. Analysis of variance was used for optimization of the biosensorresponse and showed that 30 mu L of cell-free extract containing 7.47 mg protein mL(-1), 2 mu L of glutaraldehyde (5%, v/v) and 10 mu L of gelatin (15%, w/v) exhibited the highest response. Optimization of other parameters showed that pH 7.2 and 30 min incubation time were optimum for incubation ofmembranes in urea. The biosensor exhibited a linear response in the range of 4.0-10.0 mu M urea, a detection limit of 2.0 mu M for urea, a response timeof 20 s, a sensitivity of 58.245 % per mu M urea and a storage stability of over 4 months. It was successfully used for quantification of urea in samples such as wine and milk; recovery experiments were carried out which revealed an average substrate recovery of 94.9%. The urea analogs hydroxyurea, methylurea and thiourea inhibited amidase activity by about 90%, 10% and 0%, respectively, compared with urea inhibition.

La estructura temporal de los ejercicios de técnica de danza ejecutados por los estudiantes (bailarines en contexto académico) suele estar organizada para responder a un marco métrico musical a menudo sostenido por un acompañante musical de danza. La consigna de estos ejercicios es transmitida multimodalmente por un profesor de danza. A pesar de haber entre ellos un acuerdo tácito para codi car y decodi car métricamente esta información multimodal, se producen frecuentes desentendimientos. Adoptamos como marco teórico un modelo de interacción triádica que describe la relación interpersonal en situación ecológica de clase. Planteamos la hipótesis de que los desentendimientos referidos se producen debido a divergencias en la información multimodal que pueden presentar las consignas. Tomando una metodología microgenética, analizamos una unidad de danza según criterios taxonómicos previamente estudiados. Los resultados muestran que la consigna conlleva información métrica y proposicional contradictoria. Concluimos que, como consecuencia de estas divergencias informacionales, los estudiantes produjeron respuestas motrices altamente desajustadas.

Anaemia is known to have an impact on child development and mortality and is a severe public health problem in most countries in sub-Saharan Africa. We investigated the consistency between ecological and individual-level approaches to anaemia mapping by building spatial anaemia models for children aged ≤15 years using different modelling approaches. We aimed to (i) quantify the role of malnutrition, malaria, Schistosoma haematobium and soil-transmitted helminths (STHs) in anaemia endemicity; and (ii) develop a high resolution predictive risk map of anaemia for the municipality of Dande in northern Angola. We used parasitological survey data for children aged ≤15 years to build Bayesian geostatistical models of malaria (PfPR≤15), S. haematobium, Ascaris lumbricoides and Trichuris trichiura and predict small-scale spatial variations in these infections. Malnutrition, PfPR≤15, and S. haematobium infections were significantly associated with anaemia risk. An estimated 12.5%, 15.6% and 9.8% of anaemia cases could be averted by treating malnutrition, malaria and S. haematobium, respectively. Spatial clusters of high risk of anaemia (>86%) were identified. Using an individual-level approach to anaemia mapping at a small spatial scale, we found that anaemia in children aged ≤15 years is highly heterogeneous and that malnutrition and parasitic infections are important contributors to the spatial variation in anaemia risk. The results presented in this study can help inform the integration of the current provincial malaria control programme with ancillary micronutrient supplementation and control of neglected tropical diseases such as urogenital schistosomiasis and STH infections.
