Exogenous treatments with cytokinins, such as 6-benzylaminopurine (BA), can delay senescence of some plant tissues. Grevillea 'Sylvia' inflorescences have a short vase life. BA supplied in vase solutions at up to 0.1 mM did not delay senescence of G. 'Sylvia' in florescences. However, BA applied by dipping at concentrations up to 10 mM extended their vase life (longevity). Senescence parameters of relative fresh weight, flower abscission, flower opening, flower discolouration and flower wilting were all suppressed by BA dips. Dip treatment with BA (1 mM) was effective on G. 'Sylvia' in florescences at three different maturity stages.

Rhizoctonia solani AG-2-2 was isolated from wilting and dying plants of sulla ( Hedysarum coronarium), which is currently being assessed in eastern and southern Australia for its potential as a pasture and forage legume. Infected plants in the field had extensive rotting of the taproot, lateral roots and crown. Koch's postulates were fulfilled using three inoculation methods. The disease may pose a considerable threat to the potential use of H. coronarium in the dryland, grazing farming systems of Australia, with resistance offering the most viable option for minimising its impact.

Fusarium oxysporum is a soilborne fungal pathogen that causes major economic losses by inducing necrosis and wilting symptoms in many crop plants. In this study, the interaction between F. oxysporum and the model plant Arabidopsis thaliana has been investigated to better understand the nature of host defences that are effective against the Fusarium wilt pathogen. The expression of salicylate- and jasmonate-responsive defence genes in F. oxysporum-challenged roots of A. thaliana plants as well as in the roots of plants whose leaves were treated with salicylate or jasmonate was analysed. Unexpectedly, genes (e.g. PR1, PDF1.2, and CHIB) encoding proteins with defensive functions or transcription factors (e.g. ERF1, AtERF2, AtERF4 and AtMYC2) known to positively or negatively regulate defences against F. oxysporum were not activated in F. oxysporum-inoculated roots. In contrast, the jasmonate-responsive defence gene PDF1.2 was induced in the leaves of plants whose roots were challenged with F. oxysporum, but the salicylate- responsive PR1 gene was not induced in the leaves of inoculated plants. Exogenous salicylic acid treatment prior to inoculation, however, activated PR1 and BGL2 defence gene expression in leaves and provided increased F. oxysporum resistance as evidenced by reduced foliar necrosis and plant death. Exogenous salicylic acid treatment of the foliar tissue did not activate defence gene expression in the roots of plants. This suggests that salicylate- dependent defences may function in foliar tissue to reduce the development of pathogen-induced wilting and necrosis. Despite the induction of defence gene expression in the leaves by jasmonate, this treatment did not lead to increased resistance to F. oxysporum. Overall, the results presented here suggest that the genetic manipulation of plant defence signalling pathways is a useful strategy to provide increased Fusarium wilt resistance.

Grevillea 'Crimson Yul-lo' inflorescences have cut flower potential, but their vase life is short. End of vase life is characterized by early wilting. The possibility of physiologically mediated stem end blockage was investigated. Hydraulic conductance of 2 cm long stem end segments declined rapidly and remained lower throughout vase life than that of 2 cm long stem segments from immediately above. Recutting daily to remove basal 2 cm stem ends increased solution uptake, delayed declines in inflorescence water potential and water content, and improved inflorescence vase life. S-carvone is a potential inhibitor of wound related suberin formation, via inhibition of phenylalanine ammonia-lyase. Vase solution treatments with S-carvone (0.318 and 0.636 mM) delayed the decline in hydraulic conductance of basal 2 cm long stem end segments and decreases in vase solution uptake and relative fresh weight of cut stems, and extended vase life. Treatments with the catechol oxidase inhibitor 4-hexylresorcinol (2.5-10 mM) also delayed stem end blockage. These findings suggest that stem end blockage in cut G. 'Crimson Yul-lo' stems is physiologically mediated. (C) 2006 Elsevier B.V. All rights reserved.

Salinity acts to inhibit plant access to soil water by increasing the osmotic strength of the soil solution. As the soil dries, the soil solution becomes increasingly concentrated, further limiting plant access to soil water. An experiment was conducted to examine the effect of salt on plant available water in a heavy clay soil, using a relatively salt tolerant species, wheat ‘Kennedy’, and a more salt sensitive species, chickpea ‘Jimbour’. Sodium chloride was applied to Red Ferrosol at 10 rates from 0 to 3 g/kg. Plants were initially maintained at field capacity. After 3 weeks, plants had become established and watering was ceased. The plants then grew using the water stored in the soil. Once permanent wilting point was reached plants were harvested, and soil water content was measured. The results showed that without salt stress, wheat and chickpea extracted approximately the same amount of water. However, as the salt concentration increased, the ability of chickpea to extract water was severely impaired, while wheat’s ability to extract water was not affected over the range of concentrations examined. Growth of both wheat and chickpea was reduced even from low salt concentrations. Possible explanations for this are that the effect on growth is due to Cl- toxicity and that this occurs at lower concentrations than the osmotic effect of salinity, or that the metabolic demands of maintaining plant water balance and extracting soil water under saline conditions result in reduced growth.
