High levels of mortality in the Mediterranean bath sponge industry have raised concerns for the future of sponge farms. Healthy sponges feed predominantly on bacteria, and many harbour a wide diversity of inter- and extra-cellular symbiotic bacteria. Here we describe the first isolation and description of a pathogenic bacterium from an infected marine sponge. Microbiological examination of tissue necrosis in the Great Barrier Reef sponge Rhopaloeides odorabile resulted in isolation of the bacterial strain NW4327. Sponges infected with strain NW4327 exhibited high levels of external tissue necrosis, and the strain was re-isolated from infected sponges. A single morphotype, which had burrowed through the collagenous spongin fibres causing severe necrosis, was observed microscopically. Strain NW4327 was capable of degrading commercial preparations of azo-collagen, providing further evidence of its involvement in spongin fibre necrosis, Strain NW4327 disrupted the microbial community associated with R. odorabile and was able to infect and kill healthy sponge tissue. 16S rRNA sequence analysis revealed that strain NW4327 is a novel member of the alpha-proteobacteria.

Investigation of two southern Australian marine sponges, Mycale spp., resulted in isolation of the known norsesterterpene mycaperoxide F methyl ester (5) together with a new norsesterterpene mycaperoxide G methyl ester (10) and a new norterpene ketone 11. All structures were secured by spectroscopic analysis and chemical derivatization. The absolute stereochemistry previously assigned to 5 by application of the Horeau procedure has been revised by application of the Mosher procedure.

In thin sections of resin-embedded samples of glutaraldehyde- and osmium tetroxide-fixed tissue from five genera of marine sponges, Stromatospongia, Astrosclera, Jaspis, Pseudoceratina and Axinyssa, cells of a bacteria-like symbiont microorganism which exhibit a membrane-bounded nuclear region encompassing the fibrillar nucleoid have been observed within the sponge mesohyl. The nuclear region in these cells is bounded by a single bilayer membrane, so that the cell cytoplasm is divided into two distinct regions. The cell wall consists of subunits analogous to those in walls of some Archaea. Cells of the sponge symbionts observed here are similar to those of the archaeal sponge symbiont Cenarchaeum symbiosum. Bioassay-directed fractionation of the ethanol extracts of two Amphimedon spp. collected during trawling operations in the Great Australian Eight yielded four new macrocyclic lactone/lactams, amphilactams A-D (1-4). The amphilactams possess potent in vitro nematocidal properties, and their structures were assigned on the basis of detailed spectroscopic analysis and comparison with synthetic model compounds. The amphilactams feature both carbon skeletons and an enamino lactone/lactam moiety unprecedented in the natural products literature.

Two sponge's belonging to the family Latrunculiidae (Negombata and Latrunculia sp.) collected during scientific trawling operations in Prydz Bay, Antarctica, and by scuba off Port Campbell, Victoria, have yielded a new antibacterial pyrroloiminoquinone, discorhabdin R (2). The structure was assigned as 2 on the basis of detailed, spectroscopic analysis and comparison with the known co-metabolite discorhabdin B (3).

Bioassay-directed fractionation of two southern Australian sponges, Phoriospongia sp. and Callyspongia bilamellata, yielded two new nematocidal depsipeptides, identified as phoriospongins A (1) and B (2). The structures of the phoriospongins were determined by detailed spectroscopic analysis and comparison with the previously reported sponge depsipeptide cyclolithistide A (3), as well as ESIMS and HPLC analysis of acid hydrolysates. It is noteworthy that the unique and yet structurally related metabolites 1-3 are found in sponges spanning three taxonomic orders, Poescilosclerida, Haplosclerida, and Lithistida.

The biosynthetic origins of the isocyanide and isothiocyanate functional groups in the marine sponge metabolites diisocyanoadociane (1), 9-isocyanopupukeanane (10) and 9- isothiocyanatopupukeanane (11) are probed by the use of [C-14]-labelled precursor experiments. Incubation of the sponge Amphimedon terpenensis with [C-14]-labelled thiocyanate resulted in radioactive diisocyanoadociane ( 1) in which the radiolabel is specifically associated with the isocyanide carbons. As expected, cyanide and thiocyanate were confirmed as precursors to the pupukeananes 10 and 11 in the sponge Axinyssa n. sp.; additionally these precursors labelled 2-thiocyanatoneopupukeanane ( 12) in this sponge. To probe whether isocyanide-isothiocyanate interconversions take place at the secondary metabolite level, the advanced precursor bisisothiocyanate 17 was supplied to A. terpenensis, but did not result in significant labelling in the natural product isocyanide 1. In contrast, in the sponge Axinyssa n. sp., feeding of [C-14]-9-isocyanopupukeanane (10) resulted in isolation of radiolabelled 9- isothiocyanatopupukeanane 11, while the feeding of [C-14]-11 resulted in labelled isocyanide 10. These results show conclusively that isocyanides and isothiocyanates are interconverted in the sponge Axinyssa n. sp., and confirm the central role that thiocyanate occupies in the terpene metabolism of this sponge.

The abundance and community composition of the endofauna in 2 species of sponge, Haliclona sp. 1 and Haliclona sp. 2 (phylum Porifera: order Haplosclerida), were examined at different sites on the slope at Heron Island Reef, in the southern Great Barrier Reef, on 2 separate occasions. Both species of Haliclona Occupy Similar habitats on the reef slope and are often found living adjacent to each other, but the major groups of secondary metabolites and the gross external morphology in the 2 species of sponge are different. The 2 species of sponge supported significantly different endofaunal communities, with Haliclona sp. 2 Supporting 3 to 4 times more individuals than Haliclona sp. 1. Fewer demersal zooplankton (copepods), nematodes and some peracarid crustaceans were found in Haliclona sp. I compared with Haliclona sp. 2. There were also differences in the numbers of spionid, nereidid and syllid. polychaetes living in the 2 species of sponge. The only taxon that was more abundant in Haliclona sp. 1 than Haliclona sp. 2 was the spionid Polydorella prolifera, and this difference was only evident on 1. of the 2 occasions. The amount of free space (pores, channels, cavities) for a given weight of sponge was only 19% greater in Haliclona sp. 2 than in Haliclona sp. 1, suggesting other factors, such as the differences in the allelochemicals, may have a role in determining the numbers and types of animals living in these 2 species of sponge.

Contact with sponges (Phylum Porifera) usually results in minimal effects or abrasions, except for species that produce crinitoxins and can cause irritation and dermatitis. There are few reports of sponge stings, mainly in divers or collectors. We report a group of sponge stings from handling flame red/orange sponges on the beach, confirmed to be Tedania anhelans in five cases. All seven patients suffered immediate effects ranging from mild to severe pain, and local inflammation. A 38-year-old female and three children had delayed skin involvement including itchiness, pain, swelling and redness. Blistering and desquamation occurred in the female adult and limited desquamation in one child. Similar delayed effects have been reported in Tedania spp. stings previously. (c) 2005 Elsevier Ltd. A new bioeroding sponge belonging to the genus Cliona is described from the Australian Great Barrier Reef, Cliona minuscula, sp. nov. As the sponge lacked microscleres, comparison with existing clionaid species was difficult. We considered 15 other species of Cliona with only tylostyles: C. alderi, C. arenosa. C. caesia nov. comb., C. californiana, C. celata, C. delitrix, C. dissimilis, C. ecaudis, C. insidiosa, C. janitrix, C. kempi, C. laticavicola, C. macgeachii, C. millepunctata and C. peponaca. Characters of all species are presented in table-form to facilitate comparison during future studies. We listed additional species of Cliona that were not directly compared to the new species, because they were either invalid, insufficiently described, or they may not be obligate bioeroders. The form and dimensions of the megascleres of C. minuscula, sp. nov. indicated that it is distinct from all considered species. Its mean tylostyle dimensions were 225.3 mu m length, 4.5 mu m shaft width and 6.8 mu m tyle width, which is comparatively small. Because other morphological features were small as well ( erosion chambers, papillar diameter), this species was named C. minuscula. The species record for sponges of the genus Cliona reported from Australia is now 11.

Many attempts have been made by ichnologists to match bioerosion traces to their respective tracemakers. This task has been considered difficult, especially for fossil samples. The present study demonstrates that the Australian bioeroding sponge Aka paratypica can generate a cavity similar to the ichnospecies Entobia devonica. The modern sponge and its cavity are redescribed and compared to the fossil boring. A. paratypica has white fistules and soft, mucoid endosomal tissue. Spicules are stout oxeas with often telescoped or mucronate tips. Observed borings of A. paratypica are rounded and cavernous, with canals and apertures radiating from the chambers in all directions. It was noted that the internal openings of such canals are covered with porous nodules, which may act as sieves against larger particles or intruding endofauna. No obvious microsculpturing was observed in the erosion scars. A. paratypica borings are analogous to ancient E. devonica borings, which to date were only known from the fossil record.
