Combined effect of cyclosporine and sirolimus on improving the longevity of recombinant adenovirus-mediated transgene expression in the retina

Objectives: To reevaluate the longevity and intraocular safety of recombinant adenovirus (rAd)-mediated gene delivery after subretinal injection, and to prolong transgene expression through the combination of 2 synergistic immunosuppressants. Methods: An rAd vector carrying green fluorescent protein (GFP) gene was delivered subretinally in the rat eye. The GFP expression was monitored in real time by fundus fluorescent photography. Intraocular safety was examined by observation of changes of retinal pigmentation, cell infiltration in virus-contacted area, immunophenotyping for CD4(+) and CD8(+) cytotoxic T lymphocytes, and CD68(+) macrophages, histologic findings, and dark-adapted electroretinography. Two synergistic immunosuppressants, cyclosporine and sirolimus, were used alone or in combination to prolong transgene expression by temporary immunosuppression. Results: The GFP expression peaked on day 4, dramatically decreased on day 10, and was not detectable on day 14. The decreased GFP expression was coincident with cell infiltration in virus-contacted area. Immunostaining showed that the infiltrating cells were CD4(+) and CD8(+) cytotoxic T lymphocytes and CD68(+) macrophages. Clumped retinal pigmentation and decreased b wave of dark-adapted electroretinogram were observed at 3 to 4 weeks after injection. Histologic examination confirmed rAd-induced retinal degeneration. Transient immunosuppression by cyclosporine and sirolimus, either alone or in combination, improved transgene expression, with the combination being the most efficient. The combined immunosuppression attenuated but did not retard the rAd-induced retinal damage. Conclusions: Transgene expression mediated by rAd after subretinal delivery is short-term and toxic to the retina. Combination of cyclosporine and sirolimus may act as an immunosuppressive adjunct to prolong rAd-mediated gene transfer. Clinical Relevance: The intraocular safety of rAd should be carefully considered before clinical trials are performed.

The City as Territory: Aerial Photography and the Everyday

For most of us, the modern city is somewhat unintelligible, both in terms of its structure and its significance. As an urbanised population, the city should be our natural territory, but the pace and scope of its change leads to an illegibility of its form and character. There is infinite, seemingly significant, activity - demolition, building and makeover - but there is little sense of the city as a site of meaning.

Advances in technologies such as mass spectrometry and capillary electrophoresis have encouraged the study of ancient lipids and other ancient biomolecules. Now, microarray technology looks set to revolutionise the study of ancient DNA, perhaps with as much impact as that of PCR.

The archaeology of the Peel Island Lazaret: Part 1: Survey

An archaeological survey on Peel Island in Moreton Bay, southeast Queensland, was conducted to assist the conservation planning for the Peel Island Lazaret (PIL), one of a number of institutions housed on the island during the nineteenth and twentieth centuries. The survey revealed a patteming of artefacts across the island as well as landscape modification related to its Aboriginal and European institutional uses.

Marine and estuarine reservoir effects in central Queensland, Australia: Determination of Delta R values

As a component of archaeological investigations on the central Queensland coast, a series of five marine shell specimens live-collected between A.D. 1904 and A.D. 1929 and 11 shell/ charcoal paired samples from archaeological contexts were radiocarbon dated to determine local DeltaR values. The object of the study was to assess the potential influence of localized variation in marine reservoir effect in accurately determining the age of marine and estuarine shell from archaeological deposits in the area. Results indicate that the routinely applied DeltaR value of -5 +/- 35 for northeast Australia is erroneously calculated. The determined values suggest a minor revision to Reimer and Reimer's (2000) recommended value for northeast Australia from DeltaR = +11 +/- 5 to + 12 +/- 7, and specifically for central Queensland to DeltaR = +10 +/- 7, for near-shore open marine environments. In contrast, data obtained from estuarine shell/charcoal pairs demonstrate a general lack of consistency, suggesting estuary-specific patterns of variation in terrestrial carbon input and exchange with the open ocean. Preliminary data indicate that in some estuaries, at some time periods, a DeltaR value of more than - 155 +/- 55 may be appropriate, In estuarine contexts in central Queensland, a localized estuary-specific correction factor is recommended to account for geographical and temporal variation in C-14 activity. Strange bedfellows: Green Hill Fort, archaeology and tourism

Green Hill Fort, Thursday Islalld was constructed between 1891-1893 to defend the Australian colonies against a feared Russian invasion. It retained an operational role until the 1920.'1 and played a minor role in World /t'ar 2. From 1954 to 1993 the site, but not the facilities, was used as a weather station. More recently it has been home ofthe Ton-es Strait Historical Society and Museum Association museum. It is a major attraction during the tourist season and an important local icon. For archaeologists it has sign~ficance as a relatively intact nineteenth-century military installation. Two 'Centenary of Federation' grants have proVided the impetus to undertake conservation and presentation works involving various task -spec~fic, archaeological activities. At the management level archaeologists play the lead role in the project. The project has demonstrated the value oJarchaeology and tourism joining forces. The danger ofa 'theme park 'presentation has been avoided. Technical accuracy and careful site planning has ensured a high degree ofaccuracy is retained. Provided these qualities can be assured then, it is argued, there is an opportunity for archaeology to be a majOl; long-term beneficiary. But to achieve that, the discipline must move from being entrenched in its academic mould and become Jar more receptive to the broader needs ofthe twentyfirst century.
