End-stopped cells in cortical area V1, which combine out- puts of complex cells tuned to different orientations, serve to detect line and edge crossings (junctions) and points with a large curvature. In this paper we study the importance of the multi-scale keypoint representa- tion, i.e. retinotopic keypoint maps which are tuned to different spatial frequencies (scale or Level-of-Detail). We show that this representation provides important information for Focus-of-Attention (FoA) and object detection. In particular, we show that hierarchically-structured saliency maps for FoA can be obtained, and that combinations over scales in conjunction with spatial symmetries can lead to face detection through grouping operators that deal with keypoints at the eyes, nose and mouth, especially when non-classical receptive field inhibition is employed. Al- though a face detector can be based on feedforward and feedback loops within area V1, such an operator must be embedded into dorsal and ventral data streams to and from higher areas for obtaining translation-, rotation- and scale-invariant face (object) detection.

Object categorisation is linked to detection, segregation and recognition. In the visual system, these processes are achieved in the ventral \what"and dorsal \where"pathways [3], with bottom-up feature extractions in areas V1, V2, V4 and IT (what) in parallel with top-down attention from PP via MT to V2 and V1 (where). The latter is steered by object templates in memory, i.e. in prefrontal cortex with a what component in PF46v and a where component in PF46d.

Models of visual perception are based on image representations in cortical area V1 and higher areas which contain many cell layers for feature extraction. Basic simple, complex and end-stopped cells provide input for line, edge and keypoint detection. In this paper we present an improved method for multi-scale line/edge detection based on simple and complex cells. We illustrate the line/edge representation for object reconstruction, and we present models for multi-scale face (object) segregation and recognition that can be embedded into feedforward dorsal and ventral data streams (the “what” and “where” subsystems) with feedback streams from higher areas for obtaining translation, rotation and scale invariance.

Nos últimos anos, a rede de estações GPS permanentes tem crescido de forma sustentada em todo o mundo. Também em Portugal existe um número cada vez maior de estações permanentes, a maior parte das quais ligadas a instituições públicas e universidades. O volume de dados produzido por estas redes implica a criação de procedimentos automáticos de processamento de observações com metodologias adequadas. Tendo como objectivo principal processar os dados GPS provenientes de uma rede de estações da Faculdade de Ciências da Universidade de Lisboa (FCUL), foi elaborado um sistema de processamento automático de observações para um conjunto alargado de estações. A rede é constituída por estações IGS (Madrid, Maspalomas, Ponta Delgada, San Fernando e Villa Franca del Campo), por estações EUREF (Cascais, Vila Nova de Gaia e Lagos) e estações da FCUL (Flores, Graciosa, Santa Maria, Instituto de Meteorologia da Madeira, Observatório Astronómico de Lisboa Norte, Observatório Astronómico de Lisboa Sul). O processamento de observações GPS foi feito com recurso ao Bernese Processing Engine, uma componente do Bernese GPS Software 4.2, e foram utilizadas diferentes estratégias que incluem a modificação do ângulo de máscara, a determinação de gradientes troposféricos e a inclusão do efeito de carga oceânica. Neste trabalho são analisadas várias séries temporais e discutidas as vantagens e desvantagens de cada uma das estratégias utilizadas.

Dissolved oxygen (DO) is one of the most important environmental variables of water quality, especially for marine life. Consequently, oxygen is one of the Chemical Quality Elements required for the implementation of European Union Water Framework Directive. This study uses the example of the Ria Formosa, a meso-tidal lagoon on the south coast of Portugal to demonstrate how monitoring of water quality for coastal waters must be well designed to identify symptoms of episodic hypoxia. New data from the western end of the Ria Formosa were compared to values in a database of historical data and in the published literature to identify long-term trends. The dissolved oxygen concentration values in the database and in the literature were generally higher than those found in this study, where episodic hypoxia was observed during the summer. Analysis of the database showed that the discrepancy was probably related with the time and the sites where the samples had been collected, rather than a long-term trend. The most problematic situations were within the inner lagoon near the city of Faro, where episodic hypoxia (<2 mg dm3 DO) occurred regularly in the early morning. These results emphasise the need for a balanced sampling strategy for oxygen monitoring which includes all periods of the day and night, as well as a representative range of sites throughout the lagoon. Such a strategy would provide adequate data to apply management measures to reduce the risk of more persistent hypoxia that would impact on the ecological, important natural resource. economic and leisure uses of this important natural resource.

The North Atlantic intertidal community provides a rich set of organismal and environmental material for the study of ecological genetics. Clearly defined environmental gradients exist at multiple spatial scales: there are broad latitudinal trends in temperature, meso-scale changes in salinity along estuaries, and smaller scale gradients in desiccation and temperature spanning the intertidal range. The geology and geography of the American and European coasts provide natural replication of these gradients, allowing for population genetic analyses of parallel adaptation to environmental stress and heterogeneity. Statistical methods have been developed that provide genomic neutrality tests of population differentiation and aid in the process of candidate gene identification. In this paper, we review studies of marine organisms that illustrate associations between an environmental gradient and specific genetic markers. Such highly differentiated markers become candidate genes for adaptation to the environmental factors in question, but the functional significance of genetic variants must be comprehensively evaluated. We present a set of predictions about locus-specific selection across latitudinal, estuarine, and intertidal gradients that are likely to exist in the North Atlantic. We further present new data and analyses that support and contradict these simple selection models. Some taxa show pronounced clinal variation at certain loci against a background of mild clinal variation at many loci. These cases illustrate the procedures necessary for distinguishing selection driven by internal genomic vs. external environmental factors. We suggest that the North Atlantic intertidal community provides a model system for identifying genes that matter in ecology due to the clarity of the environmental stresses and an extensive experimental literature on ecological function. While these organisms are typically poor genetic and genomic models, advances in comparative genomics have provided access to molecular tools that can now be applied to taxa with well-defined ecologies. As many of the organisms we discuss have tight physiological limits driven by climatic factors, this synthesis of molecular population genetics with marine ecology could provide a sensitive means of assessing evolutionary responses to climate change.

Epithelial tissues are essential during morphogenesis and organogenesis. During development, epithelial tissues undergo several different remodeling processes, from cell intercalation to cell change shape. An epithelial cell has a highly polarized structure, which is important to maintain tissue integrity. The mechanisms that regulate and maintain apicobasal polarity and epithelial integrity are mostly conserved among all species and in different tissues within the same organism. aPKC-PAR complex localizes in the apical domain of polarized cells, and its function is essential for apicobasal polarization and epithelial integrity. In this work we characterized two novel alleles of aPKC: a temperature sensitive allele (aPKCTS), which has a point mutation on a kinase domain, and another allele with a point mutation on a highly conserved amino acid within the PB1 domain of aPKC (aPKCPB1). Analysis of the aPKCTS mutant phenotypes, lead us to propose that during development different epithelial tissues have differential requirements of aPKC activity. More specifically, our work suggests de novo formation of adherens junctions (AJs) is particularly sensitive to sub-optimal levels of apkc activity. Analysis of the aPKCPB1 allele, suggests that aPKC is likely to have an apical structural function mostly independent of its kinase activity. Altogether our work suggests that although loss of aPKC function is associated to similar epithelial phenotypes (e.g., loss of apicobasal polarization and epithelial integrity), the requirements of aPKC activity within these tissues are nevertheless likely to vary.

O presente estudo tem a sua localização na Ria Formosa, mais concretamente na Península do Ancão e Ilha da Barreta. A barra do Ancão (ou de S. Luís) é a que tem influência directa sobre a área de estudo em causa. Ecossistema é todo o sistema (no sentido físico) incluindo não só a complexidade de organismos mas também a complexidade de factores físicos a que se chama de ambiente. Neste conceito de ecossistema, os componentes biológicos (bióticos) e físicos (abióticos) são um único sistema interactivo. Na Ria Formosa as dunas formam-se na praia, na zona que imediatamente se segue ao domínio das marés, dando origem a uma crista, mais ou menos contínua, relativamente baixa e sensivelmente paralela à linha de costa. A sucessão ecológica pode ser definida como o padrão contínuo não sazonal, direcional, de colonização e extinção num local por populações de espécies. Assim os sistemas dunares consistem num mosaico zonal, determinado pelas características ambientais definidas pela topografia. Os objectivos deste estudo foram caracetrizar a sucessão dunar da Ria Formosa em termos de associações que relacionam as espécies com elevada correlação e validá-las. Relacionar as correlações e associações com a idade dos locais e as outras variáveis ambientais (pH, condutividade, matéria orgânica e azoto), estabelecendo os tipos de composição florística e a sua sequência espacial e comparar os tipos composição florística obtidos com a idade e os propostos na bibliografia. Pela utilização do método de transecto em banda foram amostrados sete transectos, com um espaçamento de 3 m entre estações de 1 m2. Adaptou-se a escala de abundância-dominância proposta por Braun Blanquet (1979). Analizaram-se os resultados recorrendo à análise multivariada com metodologia Monte Carlo. Utilizou-se a análise de componentes principais, análise de agrupamento, análise discriminante e regressão múltipla para analisar os dados obtidos. Verificou-se então que a análise dos factores bióticos leva a concluir que a zonação do sistema dunar está dependente quer da idade do mesmo, quer das características que o próprio habitat apresenta (gradiente de recursos e gradiente directo). A zonação completa do sistema dunar necessita de pelo menos mais de 60 anos para ocorrer; idade com a qual as dunas apresentam uma série de relações inter-específicas que tendem a ser estabilizadas. As espécies ditas pioneiras, à medida que o tempo evolui, restringem a sua ocupação à vertente oceânica da duna, uma vez que sofrem uma pressão competitiva de outras espécies que se lhes sucedem com o tempo.
