The human pathogens enteropathogenic (EPEC) and enterohemorrhagic Escherichia coli and the related mouse pathogen Citrobacter rodentium subvert a variety of host cell signaling pathways via their plethora of type III secreted effectors, including triggering of an early apoptotic response. EPEC-infected cells do not develop late apoptotic symptoms, however. In this study we demonstrate that the NleH family effectors, homologs of the Shigella effector kinase OspG, blocks apoptosis. During EPEC infection, NleH effectors inhibit elevation of cytosolic Ca(2+) concentrations, nuclear condensation, caspase-3 activation, and membrane blebbing and promote cell survival. NleH1 alone is sufficient to prevent procaspase-3 cleavage induced by the proapoptotic compounds staurosporine, brefeldin A, and tunicamycin. Using C. rodentium, we found that NleH inhibits procaspase-3 cleavage at the bacterial attachment sites in vivo. A yeast two-hybrid screen identified the endoplasmic reticulum six-transmembrane protein Bax inhibitor-1 (BI-1) as an NleH-interacting partner. We mapped the NleH-binding site to the N-terminal 40 amino acids of BI-1. Knockdown of BI-1 resulted in the loss of NleH's antiapoptotic activity. These results indicate that NleH effectors are inhibitors of apoptosis that may act through BI-1 to carry out their cytoprotective function.

The insertion of a DNA copy of its RNA genome into a chromosome of the host cell is mediated by the viral integrase with the help of mostly uncharacterized cellular cofactors. We have recently described that the transcriptional co-activator LEDGF/p75 strongly interacts with HIV-1 integrase. Here we show that interaction of HIV-1 integrase with LEDGF/p75 is important for viral replication. Using multiple approaches including two-hybrid interaction studies, random and directed mutagenesis, we could demonstrate that HIV-1 virus harboring a single mutation that disrupts integrase-LEDGF/p75 interaction, resulted in defective HIV-1 replication. Furthermore, we found that LEDGF/p75 tethers HIV-1 integrase to chromosomes and that this interaction may be important for the integration process and the replication of HIV-1.

The HIV-1 genome contains several genes coding for auxiliary proteins, including the small Vpr protein. Vpr affects the integrity of the nuclear envelope and participates in the nuclear translocation of the preintegration complex containing the viral DNA. Here, we show by photobleaching experiments performed on living cells expressing a Vpr-green fluorescent protein fusion that the protein shuttles between the nucleus and the cytoplasm, but a significant fraction is concentrated at the nuclear envelope, supporting the hypothesis that Vpr interacts with components of the nuclear pore complex. An interaction between HIV-1 Vpr and the human nucleoporin CG1 (hCG1) was revealed in the yeast two-hybrid system, and then confirmed both in vitro and in transfected cells. This interaction does not involve the FG repeat domain of hCG1 but rather the N-terminal region of the protein. Using a nuclear import assay based on digitonin-permeabilized cells, we demonstrate that hCG1 participates in the docking of Vpr at the nuclear envelope. This association of Vpr with a component of the nuclear pore complex may contribute to the disruption of the nuclear envelope and to the nuclear import of the viral DNA.

The nonlinear coupling between two perpendicularly propagating ( with respect to the external magnetic field direction) upper-hybrid ( UH) waves in a uniform magnetoplasma is considered, taking into account quasi-stationary density perturbations which are driven by the UH wave ponderomotive force. This interaction is governed by a pair of coupled nonlinear Schrodinger equations ( CNLSEs) for the UH wave envelopes. The CNLSEs are used to investigate the occurrence of modulational instability. Waves in the vicinity of the UH resonance are considered, so that the group dispersion terms for both waves are approximately equal, but the UH wave group velocities may be different. It is found that a pair of unstable UH waves ( obeying anomalous group dispersion) yields an increased instability growth rate, while a pair of stable UH waves ( individually obeying normal group dispersion) remains stable for equal group velocities, although it is destabilized by a finite group velocity mismatch. Stationary nonlinear solutions of the CNLSEs are presented.

Two-dimensional (2D) materials have generated great interest in the last few years as a new toolbox for electronics. This family of materials includes, among others, metallic graphene, semiconducting transition metal dichalcogenides (such as MoS2) and insulating Boron Nitride. These materials and their heterostructures offer excellent mechanical flexibility, optical transparency and favorable transport properties for realizing electronic, sensing and optical systems on arbitrary surfaces. In this work, we develop several etch stop layer technologies that allow the fabrication of complex 2D devices and present for the first time the large scale integration of graphene with molybdenum disulfide (MoS2) , both grown using the fully scalable CVD technique. Transistor devices and logic circuits with MoS2 channel and graphene as contacts and interconnects are constructed and show high performances. In addition, the graphene/MoS2 heterojunction contact has been systematically compared with MoS2-metal junctions experimentally and studied using density functional theory. The tunability of the graphene work function significantly improves the ohmic contact to MoS2. These high-performance large-scale devices and circuits based on 2D heterostructure pave the way for practical flexible transparent electronics in the future. We present a fast and efficient hybrid algorithm for selecting exoplanetary candidates from wide-field transit surveys. Our method is based on the widely used SysRem and Box Least-Squares (BLS) algorithms. Patterns of systematic error that are common to all stars on the frame are mapped and eliminated using the SysRem algorithm. The remaining systematic errors caused by spatially localized flat-fielding and other errors are quantified using a boxcar-smoothing method. We show that the dimensions of the search-parameter space can be reduced greatly by carrying out an initial BLS search on a coarse grid of reduced dimensions, followed by Newton-Raphson refinement of the transit parameters in the vicinity of the most significant solutions. We illustrate the method's operation by applying it to data from one field of the SuperWASP survey, comprising 2300 observations of 7840 stars brighter than V = 13.0. We identify 11 likely transit candidates. We reject stars that exhibit significant ellipsoidal variations caused indicative of a stellar-mass companion. We use colours and proper motions from the Two Micron All Sky Survey and USNO-B1.0 surveys to estimate the stellar parameters and the companion radius. We find that two stars showing unambiguous transit signals pass all these tests, and so qualify for detailed high-resolution spectroscopic follow-up.

In this paper, we present a random iterative graph based hyper-heuristic to produce a collection of heuristic sequences to construct solutions of different quality. These heuristic sequences can be seen as dynamic hybridisations of different graph colouring heuristics that construct solutions step by step. Based on these sequences, we statistically analyse the way in which graph colouring heuristics are automatically hybridised. This, to our knowledge, represents a new direction in hyper-heuristic research. It is observed that spending the search effort on hybridising Largest Weighted Degree with Saturation Degree at the early stage of solution construction tends to generate high quality solutions. Based on these observations, an iterative hybrid approach is developed to adaptively hybridise these two graph colouring heuristics at different stages of solution construction. The overall aim here is to automate the heuristic design process, which draws upon an emerging research theme on developing computer methods to design and adapt heuristics automatically. Experimental results on benchmark exam timetabling and graph colouring problems demonstrate the effectiveness and generality of this adaptive hybrid approach compared with previous methods on automatically generating and adapting heuristics. Indeed, we also show that the approach is competitive with the state of the art human produced methods.

The coefficients of an echo canceller with a near-end section and a far-end section are usually updated with the same updating scheme, such as the LMS algorithm. A novel scheme is proposed for echo cancellation that is based on the minimisation of two different cost functions, i.e. one for the near-end section and a different one for the far-end section. The approach considered leads to a substantial improvement in performance over the LMS algorithm when it is applied to both sections of the echo canceller. The convergence properties of the algorithm are derived. The proposed scheme is also shown to be robust to noise variations. Simulation results confirm the superior performance of the new algorithm.

Mode-mixing of coherent excitations of a trapped Bose-Einstein condensate is modeled using the Bogoliubov approximation. Calculations are presented for second-harmonic generation between the two lowest-lying even-parity m=0 modes in an oblate spheroidal trap. Hybridization of the modes of the breather (l=0) and surface (l=4) states leads to the formation of a Bogoliubov dark state near phase-matching resonance so that a single mode is coherently populated. Efficient harmonic generation requires a strong coupling rate, sharply-defined and well-separated frequency spectrum, and good phase matching. We find that in all three respects the quantal results are significantly different from hydrodynamic predictions. Typically the second-harmonic conversion rate is half that given by an equivalent hydrodynamic estimate.

The concept of frequency steerable two-dimensional electromagnetic focusing by using a tapered leaky-wave line source embedded in a parallel-plate medium is presented. Accurate expressions for analyzing the focusing pattern of a rectilinear leaky-wave lens (LWL) from its constituent leaky-mode tapered propagation constant are described. The influence of the main LWL structural parameters on the synthesis of the focusing pattern is discussed. The ability to generate frequency steerable focusing patterns has been demonstrated by means of an example involving a LWL in hybrid waveguide printed-circuit technology and the results are validated by a commercial full-wave solver.

This paper proposes a hybrid transmission technique based on adaptive code-to-user allocation and linear precoding for the downlink of phase shift keying (PSK) based multi-carrier code division multiple access (MC-CDMA) systems. The proposed scheme is based on the separation of the instantaneous multiple access interference (MAI) into constructive and destructive components taking into account the dependency on both the channel variation and the instantaneous symbol values of the active users. The first stage of the proposed technique is to adaptively distribute the available spreading sequences to the users on a symbol-by-symbol basis in the form of codehopping with the objective to steer the users' instantaneous crosscorrelations to yield a favourable constructive to destructive MAI ratio. The second stage is to employ a partial transmitter based zero forcing (ZF) scheme specifically designed for the exploitation of constructive MAI. The partial ZF processing decorrelates destructive interferers, while users that interfere constructively remain correlated. This results in a signal to interference-plus-noise ratio (SINR) enhancement without the need for additional power-per-user investment. It will be shown in the results section that significant bit error rate (BER) performance benefits can be achieved with this technique.

Reduction of proton acceleration in the interaction of a high-intensity, picosecond laser with a 50-mu m aluminum target was observed when 0.1-6 mu m of plastic was deposited on the back surface (opposite side of the laser). The maximum energy and number of energetic protons observed at the back of the target were greatly reduced in comparison to pure aluminum and plastic targets of the same thickness. This is attributed to the effect of the interface between the layers. Modeling of the electron propagation in the targets using a hybrid code showed strong magnetic-field generation at the interface and rapid surface heating of the aluminum layer, which may account for the results. 1. Recent proliferation of hybridisation in response to anthropogenic ecosystem change, coupled with increasing evidence of the importance of ancient hybridisation events in the formation of many species, has moved hybridisation to the forefront of evolutionary theory.
2. In spite of this, the mechanisms (e. g. differences in trophic ecology) by which hybrids co-exist with parental taxa are poorly understood. A unique hybrid zone exists in Irish freshwater systems, whereby hybrid offspring off two non-native cyprinid fishes often outnumber both parental species.
3. Using stable isotope and gut content analyses, we determined the trophic interactions between sympatric populations of roach (Rutilus rutilus), bream (Abramis brama) and their hybrid in lacustrine habitats.
4. The diet of all three groups displayed little variation across the study systems, and dietary overlap was observed between both parental species and hybrids. Hybrids displayed diet, niche breadth and trophic position that were intermediate between the two parental species while also exhibiting greater flexibility in diet across systems.

The consumption of paddy rice (Oryza sativa L.) is a major inorganic arsenic exposure pathway in S.E. Asia. A multi-location survey was undertaken in Guangdong Province, South China to assess arsenic accumulation and speciation in 2 rice cultivars, one an Indica and the other a hybrid Indica. The results showed that arsenic concentrations in rice tissue increased in the order grain <husk <straw <root. Rice grain arsenic content of 2 rice cultivars was significant different and correlated with phosphorus concentration and molar ratio of P/As in shoot, being higher for the Indica cultivar than for the hybrid Indica, which suggests altering shoot phosphorus status as a promising route for breeding rice cultivars with reduced grain arsenic. Speciation of grain arsenic, performed using HPLC-ICP-MS, identified inorganic arsenic as the dominant arsenic species present in the rice grain.
