1. The population characteristics and distribution of wood mice Apodemus sylvaticus (L.) were investigated along field margins of farmland dominated by grass production. 2. Turnover, sex ratio, breeding season, spatial density dependence and density dependence in reproductive activity indicated that the population ecology of A. sylvaticus is consistent in different habitats in the same geographical region. 3. Spatial variation in the abundance of A.sylvaticus was related negatively to percentage of land under pasture and distance from woodland and positively related to variables associated with food supply and cover. 4. Variation in numbers of overwintered mice at the start of the breeding season was related more closely to breeding opportunity than to environmental factors. This was particularly so in males. 5. The association of overwintered male and female A. sylvaticus remained evident in the later half of the breeding season. Young males and females of the year, however, were distributed more with respect to physical and biological features than towards adults or reproductive opportunity. 6. A. sylvaticus is an important species of field margins, even where these are poorly developed and agriculture is pastoral rather than arable. Further studies of this species in a wider range of agricultural systems are desirable.

Variation in the diet of wood mice, Apodemus sylvaticus, was investigated in two contrasting habitats, deciduous and coniferous woodland, over 26 months and in 10 additional sites trapped during winter. Stomach contents were categorized as seed, fruit, green plant, root and animal material. Diet was evaluated using the percentage occurrence of each food type. Age and sex differences in diet occurred infrequently. Seed predominated throughout but was especially prevalent in autumn and winter. There was a peak in the incidence of animal material in the spring and early summer. Animal food was generally more frequent in mice caught in conifer plantations than in deciduous woodland during the longer-term study. Further, mice from the additional coniferous habitats had greater percentage occurrence of animal food than those from the additional deciduous sites. There was a negative, non-linear association between relative population size and diet in these winter samples. This suggests that spatial variation in numbers of A. sylvaticus is dictated by food availability and density is locally food limited.

Previously developed estrogen and androgen mammalian reporter gene assays (RGAs) were assessed for their potential use as a quantitative screening method in the detection of estrogenic and androgenic endocrine disruptors (EDs) in sport supplements. The validation of both RGAs coupled with dispersive solid phase extraction (dSPE) was performed in accordance with European Commission Decision EC/2002/6579 for biological screening methods. Decision limits (CCa) and detection capabilities (CCß) were established for both the estrogen and androgen RGAs. All samples were compliant with CCa and CCß in both bioassays. Recovery rates were 96 % for 17ß-estradiol and 115 % for dihydrotestosterone as obtained in their corresponding RGA. Both estrogens and androgens were stable in samples for more than 3 weeks, when stored at -20 °C. Specificity, good repeatability (coefficients of variation (CV), 12–25 %), reproducibility and robustness of both bioassays were also observed. Four different ED modes of action were determined for estrogens and androgens in 53 sport supplements, using the validated RGAs. This study revealed that 89 % of the investigated sport supplements contained estrogenic EDs and 51 % contained androgenic compounds. In conclusion, both bioassays are suitable for sport supplement screening of estrogenic and androgenic EDs.

Gremlin1 (Grem1) is an antagonist of bone morphogenetic proteins (BMPs) that plays a critical role in embryonic and postnatal development. Grem1 has been implicated as both a promoter and an inhibitor of cell proliferation driven by BMP-4 and other mitogens in a diverse range of cell types. Recent data showed that Grem1 can trigger angiogenesis via vascular endothelial growth factor receptor (VEGFR2) binding, highlighting that the precise modalities of Grem1 signalling require further elucidation.

Intravenous (i.v.) administration of autoantigen effectively induces Ag-specific tolerance against experimental autoimmune encephalomyelitis (EAE). We and others have shown enhanced EAE severity in mice lacking IL-12 or its receptor, strongly suggesting an immunoregulatory effect of IL-12 signaling. To examine the role of IL-12 responsiveness in autoantigen-induced tolerance in EAE, we administered autoantigen i.v. in two distinct treatment regimes to wildtype and IL-12Rβ2(-/-) mice, immunized to develop EAE. Administration at the induction phase suppressed EAE in wildtype and IL-12Rβ2(-/-) mice however the effect was somewhat less potent in the absence of IL-12Rβ2. Expression of pro-inflammatory cytokines such as IFN-γ, IL-17 and IL-2, was inhibited in wild-type tolerized mice but less so in IL-12Rβ2(-/-) mice. I.v. antigen was also effective in suppressing disease in both genotypes when given during the clinical phase of disease with similar CNS inflammation, demyelination and peripheral inflammatory cytokine profiles observed in both genotypes. There was however a mild impact of a lack of IL-12 signaling on Treg induction during tolerance induction compared to WT mice in this treatment regime. These findings show that the enhanced severity of EAE that occurs in the absence of IL-12 signaling can be effectively overcome by i.v. autoantigen, indicating that this therapeutic effect is not primarily mediated by IL-12 and that i.v. tolerance could be a powerful approach in suppressing severe and aggressive MS.

Abstract Background IL-31 is a novel cytokine that has been implicated in allergic diseases such as atopic dermatitis and more recently asthma. While IL-31 has been well studied in skin conditions such as atopic dermatitis, little is known about the role IL-31 plays in asthma and specifically the differentiation process of the bronchial epithelium, which is central to the pathogenesis of allergic asthma. Methods We examined the effects of IL-13 (20 ng/ml), IL-31 (20 ng/ml) and an IL-13/IL-31 combination stimulation (20 ng/ml each) on the in vitro mucociliary differentiation of paediatric bronchial epithelial cells (PBECs) from healthy patients (n=6). IL-31 receptor (IL-31-RA) expression, markers of differentiation (goblet and ciliated cells), transepithelial electrical resistance (TEER), quantification of goblet and ciliated cells, real time PCR for MUC5AC, ELISA for VEGF, EGF and MCP-1 (CCL-2) and ELISA for MUC5AC were assessed. Results We found that well-differentiated PBECs expressed IL-31-RA however it's expression did not increase upon stimulation with IL-31 or either of the other treatments. TEER indicated good formation of tight junctions which was found to be similar across all treatment groups (p=0.9).
