The correlation of soil fertility x seed physiological potential is very important in the area of seed technology but results published with that theme are contradictory. For this reason, this study to evaluate the correlations between soil chemical properties and physiological potential of soybean seeds. On georeferenced points, both soil and seeds were sampled for analysis of soil fertility and seed physiological potential. Data were assessed by the following analyses: descriptive statistics; Pearson's linear correlation; and geostatistics. The adjusted parameters of the semivariograms were used to produce maps of spatial distribution for each variable. Organic matter content, Mn and Cu showed significant effects on seed germination. Most variables studied presented moderate to high spatial dependence. Germination and accelerated aging of seeds, and P, Ca, Mg, Mn, Cu and Zn showed a better fit to spherical semivariogram: organic matter, pH and K had a better fit to Gaussian model; and V% and Fe showed a better fit to the linear model. The values for range of spatial dependence varied from 89.9 m for P until 651.4 m for Fe. These values should be considered when new samples are collected for assessing soil fertility in this production area.

The software Seed Vigor Imaging System (SVIS®), has been successfully used to evaluate seed physiological potential by automated analyses of scanned seedlings. In this research, the efficiency of this system was compared to other tests accepted for assessing cucumber (Cucumis sativus L.) seed vigor of distinct seed lots of Supremo and Safira cultivars. Seeds were subjected to germination, traditional and saturated salt accelerated aging, seedling emergence, seedling length and SVIS analyses (determination of vigor indices and seedling growth uniformity, lengths of primary root, hypocotyl and whole seedlings). It was also determined whether the definition of seedling growth/uniformity ratios affects the sensitivity of the SVIS®. Results showed that analyses SVIS have provided consistent identification of seed lots performance, and have produced information comparable to those from recommended seed vigor tests, thus demonstrating a suitable sensitivity for a rapid and objective evaluation of physiological potential of cucumber seeds. Analyses of four-days-old cucumber seedlings using the SVIS® are more accurate and growth/uniformity does not affect the precision of results.

Nowadays, image analysis is one of the most modern tools in evaluating physiological potential of seeds. This study aimed at verifying the efficiency of the seedling imaging analysis to assess physiological potential of wheat seeds. The seeds of wheat, cultivars IAC 370 and IAC 380, each of which represented by five different lots, were stored during four months under natural environmental conditions of temperature (T) and relative humidity (RH), in municipality of Piracicaba, Stated of São Paulo, Brazil. For this, bimonthly assessments were performed to quantify moisture content and physiological potential of seeds by means of tests of: germination, first count, accelerated aging, electrical conductivity, seedling emergence, and computerized analysis of seedlings, using the Seed Vigor Imaging System (SVIS®). It has been concluded that the computerized analyses of seedling through growth indexes and vigor, using the SVIS®, is efficient to assess physiological potential of wheat seeds.

A colheita é uma das principais etapas no processo de produção de sementes de soja. O momento adequado para se efetuar a colheita pode variar em função do tipo de colheita e do grau de umidade das sementes. Neste sentido, o objetivo dessa pesquisa foi determinar as alterações fisiológicas de sementes de soja colhidas de forma manual e mecânica com diferentes teores de água. Para tanto, utilizaram-se sementes das cultivares Embrapa 48 e FTS Águia, que têm diferenças no teor de lignina do tegumento, colhidas de forma manual e mecanicamente com 18,0; 15,0 e 12,0% de água. Após a colheita, as sementes foram submetidas à secagem, e a qualidade das sementes foi avaliada pelos testes de germinação, emergência de plântulas, tetrazólio (viabilidade e vigor), envelhecimento acelerado e índice de velocidade de emergência de plântula. Em seguida, as sementes foram armazenadas (20 °C e 45% UR do ar) e analisadas logo após a colheita e aos seis meses de armazenamento. Os resultados obtidos indicam que o parâmetro fisiológico das sementes de soja, não é afetado, quando: se realiza colheita manual com grau de umidade das sementes de 11,4% a 18,4%; e para sementes colhidas à máquina, com grau de umidade entre 12,0% a 15,9%; mantendo a qualidade fisiológica mesmo após seis meses de armazenamento.

The development of new procedures for quickly obtaining accurate information on the physiological potential of seed lots is essential for developing quality control programs for the seed industry. In this study, the effectiveness of an automated system of seedling image analysis (Seed Vigor Imaging System - SVIS) in determining the physiological potential of sun hemp seeds and its relationship with electrical conductivity tests, were evaluated. SVIS evaluations were performed three and four days after sowing and data on the vigor index and the length and uniformity of seedling growth were collected. The electrical conductivity test was made on 50 seed replicates placed in containers with 75 mL of deionised water at 25 ºC and readings were taken after 1, 2, 4, 8 and 16 hours of imbibition. Electrical conductivity measurements at 4 or 8 hours and the use of the SVIS on 3-day old seedlings can effectively detect differences in vigor between different sun hemp seed lots.

The objective of the present study was to evaluate the efficiency of X-rays in identifying fissures in artificially dried rice seeds and the relationship between damage and seed performance in the germination test. Irrigated rice seeds of the IRGA 417 and IRGA 420 cultivars were harvested with 23.3 and 24.5% water content respectively and submitted to stationary drying treatments at 32, 38, 44 and 50 °C. X-rays were taken of subsamples of 100 seeds for each treatment, using an MX-20 X-ray equipment. The X-rayed seeds were classified from 1 to 3, where 1 corresponded to seeds without fissures, 2 to seeds with non-severe fissures and 3 to seeds with severe fissures. The same X-rayed seeds were planted and on the seventh day the seedlings (normal or abnormal) and dead seeds were photographed and evaluated to verify any relationship between the fissures and physiological potential. Higher drying temperature increased the percentage of fissures in the two cultivars, which can adversely affect their germination. Seeds with fissures can be identified using X-rays.

O vigor das sementes pode influenciar a emergência, o desempenho e produtividade das plantas, dependendo de cada espécie e de fatores ambientais. Dessa forma, um dos principais desafios das pesquisas com sementes é averiguar a influência do potencial fisiológico das sementes sobre o ciclo das plantas. O objetivo deste trabalho foi avaliar o efeito do vigor de sementes sobre as características morfofisiológicas de plantas de algodão, bem como, sobre o rendimento de caroço e de fibra. Três lotes de sementes da cultivar FMT 701 (safra 2005/2006) foram selecionados com base em resultados de testes realizados em laboratório. O efeito do vigor foi avaliado em campo, nas safras 2006/2007 e 2007/2008, no município de Jaciara, Mato Grosso. Determinou-se a porcentagem de plântulas emergidas, índice de velocidade de emergência, altura das plantas, número de ramos reprodutivos, número de nós, número de posições frutíferas, número de capulhos, rendimento de fibras e caroço. O desempenho inicial e reprodutivo de plantas de algodão em campo depende, além de outros fatores, do nível de vigor das sementes. Plantas com maior vigor apresentam maior rendimento de fibras e de caroço.

The objective of this study was to investigate the possibility of using hydric restriction as a method for evaluating vigor of soybean seeds. The soybean seeds, cultivar BRS 245RR, represented by four different seed lots, were characterized by germination and vigor. For the treatment of hydric restriction and temperature, the combination of substrate water potential and temperature were the following: deionized water (0.0 MPa); polyethylene glycol (PEG 6000) aqueous solution (-0.1, -0.3 and -0.5 MPa); and four temperatures (20 ºC, 25 ºC, 30 ºC, and 35 ºC), respectively. A completely randomized experimental design was used, with four replications per treatment, and the ANOVA was performed individually for each combination of temperature and water potential of substrate. According to results obtained, the test of hydric restriction has the same efficiency of the accelerated aging test in estimating vigor of soybean seeds, cv. BRS 245RR, when water potentials of -0.1 MPa or -0.3 MPa at a temperature of 25 ºC, or -0.3 MPa at a temperature of 30 ºC are used.

O presente trabalho foi conduzido com o objetivo de verificar a eficiência do sistema computadorizado de avaliação do vigor de sementes (SVIS®), em comparação com outros procedimentos disponíveis, em sementes de milho superdoce. Foram utilizadas sementes dos híbridos SWB 551 e SWB 585, cada uma representada por cinco lotes, conduzindo-se testes de germinação, envelhecimento acelerado (tradicional e com solução saturada de NaCl) e emergência de plântulas em campo. Para a determinação do vigor das sementes por meio do SVIS®, as plântulas foram avaliadas aos três dias após a semeadura do teste de germinação, sendo obtidos dados referentes ao índice de vigor, à uniformidade de desenvolvimento e ao comprimento de plântulas. Para ambos os híbridos avaliados, os resultados fornecidos pelo SVIS® foram coerentes com os do teste de envelhecimento acelerado tradicional. Concluiu-se que o SVIS® é eficiente para avaliação do vigor de sementes de milho superdoce, constituindo alternativa viável para a determinação do potencial fisiológico.

O vigor da semente tem sido avaliado por meio de testes fisiológicos, bioquímicos e de tolerância a estresse. Mais recentemente, com o uso da análise computadorizada de imagens de plântulas, podem ser obtidas informações objetivas, em período relativamente curto, com menor interferência humana. O objetivo nesta pesquisa foi identificar testes eficientes para avaliação do vigor de sementes de alface, com ênfase para a análise de imagens digitais de plântulas. Sementes de 12 lotes (seis da cultivar Vanda e seis da Vera) foram avaliadas por meio de testes de germinação, germinação a baixa temperatura (15ºC), emergência de plântulas e envelhecimento acelerado tradicional e com solução salina (NaCl 40%) saturada, a 41ºC/48 h e 41ºC/72 h. Utilizou-se, também, o sistema de análise computadorizada de imagens de plântulas (SVIS®), desenvolvido pela Ohio State University (EUA), que forneceu dados referentes ao índice de vigor, uniformidade e crescimento de plântulas. Os testes foram realizados a cada três meses, durante 12 meses de armazenamento, sob condições ambiente. Com base nos resultados obtidos, concluiu-se que os testes de envelhecimento acelerado com uso de solução saturada de NaCl, a 41ºC por 48 e 72 horas e de velocidade de germinação, e os parâmetros obtidos com o software SVIS, índice de crescimento de plântulas e comprimento da raiz primária, são eficientes na avaliação do vigor de lotes de sementes de alface.

A injúria mecânica é um dos mais importantes fatores na redução da qualidade de sementes de soja. Neste contexto, este trabalho objetivou avaliar a influência da pressão de impacto e o teor de água em sementes, bem como a aplicabilidade da técnica de análise de imagens, na ocorrência de danos mecânicos em sementes de soja. Sementes das cultivares Vencedora e Monsoy 8001, com teores de água de 13% e 18%, foram submetidas a danos mecânicos, com a utilização de equipamento simulador de impactos (551,6 KPa e 965,3 KPa). Posteriormente, as sementes foram secas a 32ºC, até atingirem 12% de teor de água. Imediatamente após os impactos e após cinco meses de armazenamento, as sementes foram avaliadas pelos testes de tetrazólio e raios X. A pressão de impacto e o teor de água nas sementes exerceram efeitos diretos na severidade do dano mecânico, porém, o local do impacto exerceu maior influência do que aqueles fatores. A técnica de análise de imagens foi de grande utilidade na avaliação de danos mecânicos em sementes de soja, possibilitando a análise detalhada dos efeitos de diferentes fatores.

Goat breeding in Sardinia constitutes an important source of income for farming and shepherding activities. In this study 170 LAB strains were isolated from Sardinian goat's milk and tested for bacteriocins production against several food-borne pathogenic microorganisms. Four isolates (SD1, SD2, SD3 and SD4) were selected for their effective inhibition on Listeria monocytogenes. The strains were classified as members of Enterococcus genus, according to their biochemical and physiological characteristics, and then genetically identified as Enterococcus faecium. In MRS broth at 37 degrees C, bacteriocins SD1 and SD2 were produced at much higher levels (51200 AU/ml) compared to bacteriocin SD3 (3200 AU/ml) and bacteriocin SD4 (800 AU/ml). Their peptides were inactivated by proteolytic enzymes, but not when treated with alpha-amylase, catalase and lipase. The four bacteriocins remained stable at pH from 2.0 to 12.0, after exposure to 100 degrees C for 120 min and were not affected by the presence of surfactants and salts (N-Laourylsarcosine, NaCl, SDS, Triton X-100, Tween 20, Tween 80 and urea). Their molecular size was determined to be approximately 5 kDa by tricine-SDS-PAGE. Since the strains exhibited a strong antimicrobial activity against 21 L monocytogenes strains and 6 Salmonella spp. isolates, they should be considered as potential bio-preservatives cultures for fermented food productions. Moreover, due to their technological features, the four strains could be taken in account for using as adjunct NSLAB (non-starter lactic acid bacteria) rather than as starter culture. Insects are able to combat infection by initiating an efficient immune response that involves synthesizing antimicrobial peptides and a range of other defense molecules. These responses may be costly to the organism, resulting in it exploiting endogenous resources to maintain homeostasis or support defense to the detriment of other physiological needs. We used queenless worker bees on distinct dietary regimes that may alter hemolymph protein storage and ovary activation to investigate the physiological costs of infection with Serratia marcescens. The expression of the genes encoding the storage proteins vitellogenin and hexamerin 70a, the vitellogenin receptor, and vasa (which has a putative role in reproduction), was impaired in the infected bees. This impairment was mainly evident in the bees fed beebread, which caused significantly higher expression of these genes than did royal jelly or syrup, and this was confirmed at the vitellogenin and hexamerin 70a protein levels. Beebread was also the only diet that promoted ovary activation in the queenless bees, but this activation was significantly impaired by the infection. The expression of the genes encoding the storage proteins apolipophorins-I and -III and the lipophorin receptor was not altered by infection regardless the diet provided to the bees. Similarly, the storage of apolipophorin-I in the hemolymph was only slightly impaired by the infection, independently of the supplied diet. Taken together these results indicate that, infection demands a physiological cost from the transcription of specific protein storage-related genes and from the reproductive capacity.
