Resumo:
This article studies the Drama and Music Conservatory of São Paulo as an acting school since its foundation in 1906. Although the institution is currently dedicated exclusively to music education, we need to remember that it was the first drama school in South America and represented an important aspect of the growth and modernization of theater in Sao Paulo in the early 1900s. It is on this latter aspect that this study focuses on as it provides background information and examines the role of the institution in the context of São Paulo and Brazilian theater of that period. The essay attempts to understand the artistic orientation of the acting program (until the 1940s when the course was eliminated) and to evaluate its merits and shortcomings throughout the years

Numerous steatotic livers are discarded for transplantation because of their poor tolerance to ischemia-reperfusion (I/R). We examined whether tauroursodeoxycholic acid (TUDCA), a known inhibitor of endoplasmic reticulum (ER) stress, protects steatotic and nonsteatotic liver grafts preserved during 6 h in University of Wisconsin (UW) solution and transplanted. The protective mechanisms of TUDCA were also examined. Neither unfolded protein response (UPR) induction nor ER stress was evidenced in steatotic and nonsteatotic liver grafts after 6 h in UW preservation solution. TUDCA only protected steatotic livers grafts and did so through a mechanism independent of ER stress. It reduced proliferator-activated receptor-gamma(PPAR gamma) and damage. When PPAR gamma was activated, TUDCA did not reduce damage. TUDCA, which inhibited PPAR gamma, and the PPAR gamma antagonist treatment up-regulated toll-like receptor 4 (TLR4), specifically the TIR domain-containing adaptor inducing IFN beta (TRIF) pathway. TLR4 agonist treatment reduced damage in steatotic liver grafts. When TLR4 action was inhibited, PPAR gamma antagonists did not protect steatotic liver grafts. In conclusion, TUDCA reduced PPAR gamma and this in turn up-regulated the TLR4 pathway, thus protecting steatotic liver grafts. TLR4 activating-based strategies could reduce the inherent risk of steatotic liver failure after transplantation.

Executive dysfunction is reported in juvenile myoclonic epilepsy (JME). However, batteries employed in previous studies included no more than three tests of executive function. In this study, we aimed to assess executive and attentional functions in JME using a comprehensive battery of eight tests (encompassing fifteen subtests). We also evaluated neuropsychological profiles using a clinical criterion of severity and correlated these findings with epilepsy clinical variables and the presence of psychiatric disorders. We prospectively evaluated 42 patients with JME and a matched control group with Digit Span tests (forward and backward), Stroop Color-Word Test, Trail Making Test, Wisconsin Card-Sorting Test, Matching Familiar Figures Test and Word Fluency Test. We estimated IQ with the Matrix Reasoning and Vocabulary subtests of the Wechsler Abbreviated Intelligence Scale. The patients with JME showed specific deficits in working memory, inhibitory control, concept formation, goal maintenance, mental flexibility, and verbal fluency. We observed attentional deficits in processes such as alertness and attention span and those requiring sustained and divided attention. We found that 83.33% of the patients had moderate or severe executive dysfunction. In addition, attentional and executive impairment was correlated with higher frequency of seizures and the presence of psychiatric disorders. Furthermore, executive dysfunction correlated with a longer duration of epilepsy. Our findings indicate the need for comprehensive neuropsychological batteries in patients with JME, in order to provide a more extensive evaluation of attentional and executive functions and to show that some relevant deficits have been overlooked. Hepatitis C virus (HCV) is the leading cause of liver disease worldwide. In this study, we analyzed four treatment-naive patients infected with subtype 1a and performed Roche/454 pyrosequencing across the coding region. We report the presence of low-level drug resistance mutations that would most likely have been missed using conventional sequencing methods. The approach described here is broadly applicable to studies of viral diversity and could help to improve the efficacy of direct-acting antiviral agents (DAA) in the treatment of HCV-infected patients.

Abstract Background Despite recent advances in the understanding of lignocellulolytic enzyme regulation, less is known about how different carbon sources are sensed and the signaling cascades that result in the adaptation of cellular metabolism and hydrolase secretion. Therefore, the role played by non-essential protein kinases (NPK) and phosphatases (NPP) in the sensing of carbon and/or energetic status was investigated in the model filamentous fungus Aspergillus nidulans. Results Eleven NPKs and seven NPPs were identified as being involved in cellulase, and in some cases also hemicellulase, production in A. nidulans. The regulation of CreA-mediated carbon catabolite repression (CCR) in the parental strain was determined by fluorescence microscopy, utilising a CreA: GFP fusion protein. The sensing of phosphorylated glucose, via the RAS signalling pathway induced CreA repression, while carbon starvation resulted in derepression. Growth on cellulose represented carbon starvation and derepressing conditions. The involvement of the identified NPKs in the regulation of cellulose-induced responses and CreA derepression was assessed by genome-wide transcriptomics (GEO accession 47810). CreA:GFP localisation and the restoration of endocellulase activity via the introduction of the ∆creA mutation, was assessed in the NPK-deficient backgrounds. The absence of either the schA or snfA kinase dramatically reduced cellulose-induced transcriptional responses, including the expression of hydrolytic enzymes and transporters. The mechanism by which these two NPKs controlled gene transcription was identified, as the NPK-deficient mutants were not able to unlock CreA-mediated carbon catabolite repression under derepressing conditions, such as carbon starvation or growth on cellulose. Conclusions Collectively, this study identified multiple kinases and phosphatases involved in the sensing of carbon and/or energetic status, while demonstrating the overlapping, synergistic roles of schA and snfA in the regulation of CreA derepression and hydrolytic enzyme production in A. nidulans. The importance of a carbon starvation-induced signal for CreA derepression, permitting transcriptional activator binding, appeared paramount for hydrolase secretion.

Objetivo: Caracterizar os desempenhos neuropsicológicos de um caso com sequência de Robin. Método: Participou um sujeito com 11 anos e 11 meses, sexo masculino, com Sequência de Robin e fissura completa de palato e queixa de baixo aproveitamento acadêmico. Foram utilizados: Matrizes Progressivas Coloridas, Teste Gestaltico Bender, Escala de Inteligência para Crianças - WISC III; Wisconsin Card Sorting Test-WCST, Teste de Atenção Visual-Tavis-4. Resultados: O raciocínio espaço-temporal lógico apresentou-se na média para a idade. A análise das funções cognitivas para a aprendizagem revelou desempenhos com classificação na média em escalas verbal (QIV=112), execução (QIE=99) e global (QIT=106). No entanto, apresentou desempenhos inferiores em tarefas com sobrecarga, que exigiam atenção sustentada, agilidade e flexibilidade mental, memória de trabalho visual e gerenciamento de estratégias para resolução de problemas. O desempenho em provas perceptivas visiomotoras foi inferior ao esperado, compatível a oito anos de idade, com evidentes dificuldades na construção de ângulos, organização espacial e posição relativa. As habilidades de atenção mostraram-se preservadas em provas de sustentadas e alternadas; naquelas envolvendo a atenção seletiva, cujo estímulo exigiu capacidade de rastreamento e velocidade da resposta, o desempenho foi significativamente inferior ao esperado para a idade. Conclusão: Apesar do nível intelectual satisfatório, a avaliação evidenciou déficits neuropsicológicos importantes interferentes na aprendizagem, tais como, a atenção, memória de trabalho, percepção visomotora e gerenciamento executivo. Estes achados justificam os prejuízos do sujeito em atividades acadêmicas que envolvem as habilidades de leitura, escrita e aritmética.

Objetivo: Verificar os efeitos da remediação neuropsicológica em um sujeito com Síndrome Velocardiofacial. Método: Participou do estudo um sujeito do sexo masculino, 13 anos de idade, queixa de baixo rendimento escolar, diagnosticado com Síndrome Velocardiofacial (Del22q11.2) e fissura de palato submucosa. Na avaliação pré e pós programa remediativo utilizou-se: BANI-TS; Matrizes Progressivas, Teste Gestáltico Visomotor; Escala de Inteligência Wechsler para Crianças- WISC-III e o Teste Wisconsin de Classificação de Cartas. O programa de remediação neuropsicológica, com ênfase nas habilidades de execução, atenção, organização perceptual utilizou do delineamento comportamental, treino sistemático de ensino-aprendizagem viso-construtivas, e estratégias diferenciadas na tarefa. Resultado: No pré-teste, as habilidades intelectuais mostraram classificação rebaixado à idade. Na prova cognitiva verbal (QIV=91) apresentou classificação média e execução (QI = 71), limítrofe. Os índices atencionais e de processamento da informação indicaram perfis limítrofes e os de organização perceptual, deficientes. Na avaliação gráfico-percepto-motora obteve resultados inferiores a 6 anos. Na pós-testagem, as habilidades intelectuais mantiveram classificação rebaixada, porém, com aumento do número de respostas corretas. Na escala de execução (QIE) os resultados foram na média, com escore elevado em 77%; a organização perceptual elevou-se para nível médio-inferior. Os resultados percepto-motores demonstraram aumento da pontuação, de 06 para 07 anos. Conclusão: Os resultados obtidos no presente estudo de caso demonstraram que programas remediativos, focados em habilidades específicas, pode ser uma opção a mais na reabilitação de sujeitos com fissura de palato, com queixas na aprendizagem.
