Resumo:
Crustacean color change results partly from granule aggregation induced by red pigment concentrating hormone (RPCH). In shrimp chromatophores, both the cyclic GMP (3', 5'-guanosine monophosphate) and Ca2+ cascades mediate pigment aggregation. However, the signaling elements upstream and downstream from cGMP synthesis by GC-S (cytosolic guanylyl cyclase) remain obscure. We investigate post-RPCH binding events in perfused red ovarian chromatophores to disclose the steps modulating cGMP concentration, which regulates granule translocation. The inhibition of calcium/calmodulin complex (Ca2+/CaM) by N-(6-aminohexyl)-5-chloro-1-naphthalenesulphonamide (W7) induces spontaneous aggregation but inhibits RPCH-triggered aggregation, suggesting a role in pigment aggregation and dispersion. Nitric oxide synthase inhibition by N omega-nitro-L-arginine methyl ester hydrochloride (L-NAME) strongly diminishes RPCH-induced aggregation; protein kinase G inhibition (by rp-cGMPs-triethylamine) reduces RPCH-triggered aggregation and provokes spontaneous dispersion, disclosing NO/PKG participation in aggregation signaling. Myosin light chain phosphatase inhibition (by cantharidin) accelerates RPCH-triggered aggregation, whereas Rho-associated protein kinase inhibition (by Y-27632, H-11522) reduces RPCH-induced aggregation and accelerates dispersion. MLCP (myosin light chain kinase) and ROCK (Rho-associated protein kinase) may antagonistically regulate myosin light chain (MLC) dephosphorylation/phosphorylation during pigment dispersion/aggregation. We propose the following general hypothesis for the cGMP/Ca2+ cascades that regulate pigment aggregation in crustacean chromatophores: RPCH binding increases Ca2+ (int), activating the Ca2+/CaM complex, releasing NOS-produced nitric oxide, and causing GC-S to synthesize cGMP that activates PKG, which phosphorylates an MLC activation site. Myosin motor activity is initiated by phosphorylation of an MLC regulatory site by ROCK activity and terminated by MLCP-mediated dephosphorylation. Qualitative comparison reveals that this signaling pathway is conserved in vertebrate and invertebrate chromatophores alike.

The spectral reflectance of the sea surface recorded using ocean colour satellite sensors has been used to estimate chlorophyll-a concentrations for decades. However, in bio-optically complex coastal waters, these estimates are compromised by the presence of several other coloured components besides chlorophyll, especially in regions affected by low-salinity waters. The present work aims to (a) describe the influence of the freshwater plume from the La Plata River on the variability of in situ remote sensing reflectance and (b) evaluate the performance of operational ocean colour chlorophyll algorithms applied to Southwestern Atlantic waters, which receive a remarkable seasonal contribution from La Plata River discharges. Data from three oceanographic cruises are used, in addition to a historical regional bio-optical dataset. Deviations found between measured and estimated concentrations of chlorophyll-a are examined in relation to surface water salinity and turbidity gradients to investigate the source of errors in satellite estimates of pigment concentrations. We observed significant seasonal variability in surface reflectance properties that are strongly driven by La Plata River plume dynamics and arise from the presence of high levels of inorganic suspended solids and coloured dissolved materials. As expected, existing operational algorithms overestimate the concentration of chlorophyll-a, especially in waters of low salinity (S<33.5) and high turbidity (Rrs(670)>0.0012 sr−1). Additionally, an updated version of the regional algorithm is presented, which clearly improves the chlorophyll estimation in those types of coastal environment. In general, the techniques presented here allow us to directly distinguish the bio-optical types of waters to be considered in algorithm studies by the ocean colour community.

O sistema estuarino-lagunar da Bacia do Pina e Rio Capibaribe situa-se no Recife (PE) Brasil, formado pela confluência dos rios Capibaribe, Tejipió, Jiquiá, Jordão e Pina, de grande importância para a Região Metropolitana do Recife. A variação vertical da porcentagem de carbonato de cálcio (CaCO3) no ambiente e a susceptibilidade magnética são ferramentas utilizadas para identificar o processo sedimentar dominante durante a deposição em ambientes costeiros e marinhos, podendo ser de origem marinha ou continental. Foi recuperado um testemunho, com 109 cm, localizado no setor médio do sistema estuarino. Os valores de susceptibilidade foram obtidos com o medidor da marca Bartington MS 2C, descritos visualmente quanto a granulometria e estruturas sedimentares, sub-amostrados em intervalos de 2 cm para a %CaCO3. Os valores de susceptibilidade variaram de 1 a 24 SI, os valores de carbonato variaram de 1,56 a 41,17 %. De acordo com a descrição visual foi observado a presença de granocrescência ascendente entre areia fina a média, cascalho biodetríticos, fragmentos de CaCO3, camadas de lama com matéria orgânica, fragmentos plásticos em algumas profundidades e a presença de uma camada de cascalho terrígeno. Por ser um ambiente de transição continental/marinho, a variação da sedimentação possivelmente está associada ao regime pluviométrico, onde em períodos chuvosos o sedimento fino presente no ambiente é remobilizado, e no período seco a pouca vazão fluvial favorece a deposição de finos. No intervalo 14 – 16 cm foi observado a maior porcentagem de CaCO3 indicando influência marinha durante a sedimentação. Diversos pulsos de sedimentação predominantemente terrígena foram observados na coluna sedimentar, representados por picos mais altos de susceptibilidade magnética provavelmente associados à ciclicidade das cheias do rio Capibaribe. As variações verticais de carbonato e da susceptibilidade obtidas foram importantes na identificação da variabilidade de influência terrígena ou marinha durante o processo de sedimentação recente no estuário

Background: Little is known about the diversity, phylogenetic relationships, and biogeography of trypanosomes infecting non-mammalian hosts. In this study, we investigated the influence of host species and biogeography on shaping the genetic diversity, phylogenetic relationship, and distribution of trypanosomes from South American alligatorids and African crocodilids. Methods: Small Subunit rRNA (SSU rRNA) and glycosomal Glyceraldehyde Phosphate Dehydrogenase (gGAPDH) genes were employed for phylogenetic inferences. Trypanosomes from crocodilians were obtained by haemoculturing. Growth behaviour, morphology, and ultrastructural features complement the molecular description of two new species strongly supported by phylogenetic analyses. Results: The inferred phylogenies disclosed a strongly supported crocodilian-restricted clade comprising three subclades. The subclade T. grayi comprised the African Trypanosoma grayi from Crocodylus niloticus and tsetse flies. The subclade T. ralphi comprised alligatorid trypanosomes represented by Trypanosoma ralphi n. sp. From Melanosuchus niger, Caiman crocodilus and Caiman yacare from Brazilian river basins. T. grayi and T. ralphi were sister subclades. The basal subclade T. terena comprised alligatorid trypanosomes represented by Trypanosoma terena n. sp. from Ca. yacare sharing hosts and basins with the distantly genetic related T. ralphi. This subclade also included the trypanosome from Ca. crocodilus from the Orinoco basin in Venezuela and, unexpectedly, a trypanosome from the African crocodilian Osteolaemus tetraspis. Conclusion: The close relationship between South American and African trypanosomes is consistent with paleontological evidence of recent transoceanic dispersal of Crocodylus at the Miocene/Pliocene boundaries (4–5 mya), and host-switching of trypanosomes throughout the geological configuration of South American hydrographical basins shaping the evolutionary histories of the crocodilians and their trypanosomes.

Over the last decade, molecular phylogenetics has called into question some fundamental aspects of coral systematics. Within the Scleractinia, most families composed exclusively by zooxanthellate species are polyphyletic on the basis of molecular data, and the second most speciose coral family, the Caryophylliidae (most members of which are azooxanthellate), is an unnatural grouping. As part of the process of resolving taxonomic affinities of caryophylliids', here a new Robust' scleractinian family (Deltocyathiidae fam. n.) is proposed on the basis of combined molecular (CO1 and 28S rDNA) and morphological data, accommodating the early-diverging clade of traditional caryophylliids (represented today by the genus Deltocyathus). Whereas this family captures the full morphological diversity of the genus Deltocyathus, one species, Deltocyathus magnificus, is an outlier in terms of molecular data, and groups with the Complex coral family Turbinoliidae. Ultrastructural data, however, place D.magnificus within Deltocyathiidae fam. nov. Unfortunately, limited ultrastructural data are as yet available for turbinoliids, but D.magnificus may represent the first documented case of morphological convergence at the microstructural level among scleractinian corals. Topologies of motor drive systems are studied, aiming the reduction of common-mode (CM) currents. Initially, the aspects concerning the CM currents circulation are analysed. The reason of common-mode voltages generation, the circulating paths for the resulting CM currents and their effects are discussed. Then, a non-conventional drive system configuration is proposed in order to reduce the CM currents and their effects. This configuration comprehends a non-conventional inverter module wired to a motor with an unusual connection. The cables arrangement differs from the standard solution, too. The proposed topology is compared with other ones, like the active circuit for common-mode voltages compensation. The contribution of the configuration to the reduction of CM voltages and currents and their related interferences are evaluated, based on numerical simulations. Some results are presented and discussed regarding the suitability of the proposed configuration as a potential solution to reduce the CM currents effects, when the state of art and implementation cost of drives are taken into account.

Os oceanos e as praias têm recebido todo o tipo de fragmentos, que podem ter impactos econômicos, estéticos e ecológicos (USEPA, 1992). Dentre esses fragmentos, estão os pellets ou grânulos plásticos, que são utilizados na indústria para manufatura de diversos produtos (Ogata et al, 2009). Esses grânulos entram no ambiente de forma acidental ou intencional durante o transporte ou produção de plástico (ITF, 1988) e representam um importante constituinte da poluição marinha por resíduos sólidos, estando presentes em todos os oceanos e praias do mundo, com relatos desde a década de 70 (Turra, 2008). Efeitos adversos que podem ocorrer em organismos que ingerem esses grânulos incluem bloqueio do trato intestinal, redução de consumo alimentar e aumento à exposição dos produtos químicos (Endo et al., 2005). Segundo Karapanagioti e Klontza (2008), os grânulos de baixa densidade flutuam na superfície do oceano e chegam nas praias. Sendo orgânicos, os grânulos adsorvem contaminantes hidrofóbicos, como hidrocarbonetos, presentes ao longo percurso até chegar na praia (Mato et al., 2001). Esses compostos estão entre contaminantes que podem afetar o ambiente marinho e podem ser provenientes de vazamentos de óleo e queima incompleta de combustíveis fósseis (NRC, 1985). O objetivo deste trabalho é apresentar resultados desses compostos em grânulos de plástico coletados na superfície do sedimento da Praia do Embaré, Baixada Santista, SP, região muito afetada pela introdução de hidrocarbonetos.
