Este estágio foi realizado em clínica de animais de companhia. Observou e acompanhou-se tanto consultas com carácter preventivo como de clínica médica e cirúrgica. As consultas de medicina preventiva consistiram essencialmente em vacinações e desparasitações, enquanto outras foram meramente informativas. As consultas de clínica médica incidiram em diversas áreas, incluíndo dermatologia, gastroenterologia e cardiologia. As cirurgias efetuadas foram sobretudo eletivas, mas também se assistiu a cirurgias corretivas. Um dos casos observados foi um cão com disfunção cognitiva. Este foi o caso escolhido para ser alvo do estudo mais aprofundado que é apresentado na segunda secção deste relatório. A disfunção cognitiva canina é o equivalente à doença humana de Alzheimer. Esta condição é relativamente desconhecida e os seus sinais clínicos são frequentemente considerados normais da idade. No entanto, quando detetada e tratada atempadamente é possível abrandar a sua evolução, devendo os proprietários de cães afetados ser sensibilizados a procurar assistência profissional; Internship in small animal practice: canine cognitive dysfunction syndrome Abstract: This internship was conducted in small animal clinic. The attended appointments consisted mostly of cases of preventive and internal medicine and also surgical assistance. The appointments of preventive medicine included vaccinations, deworming and some merely informative sessions. The cases of internal medicine concerned areas such as dermatology, gastroenterology and cardiology. The surgical interventions were mainly optional but also corrective procedures. One of the observed cases was a dog with cognitive dysfunction. This case was elected to be the target of further study which is presented in the second section of this report. The canine cognitive dysfunction is the equivalent to the human Alzheimer’s disease. This condition is still largely unknown and its clinical presentation is often considered normal of aging. However, if detected and treated early it is possible to slow down its progress, thus the owners of affected dogs should be encouraged to seek professional assistance.

The focus of this thesis was the study of a recently developed class of picolinamide cinchona alkaloid derivatives for the synthesis of Rivastigmine, a biologically active compound used for the treatment of Alzheimer’s disease. Six 9-picolinamide-cinchona alkaloid derivatives were successfully synthesized through simple and effective methods. These catalysts were then applied in the enantioselective reduction of O-protected ketimines (intermediates of Rivastigmine). The hydrosilylation of the N-phenyl ketimines afforded good results with excellent yields and high enantioselectivities, while much lower values, in terms of both enantioselectivity and yield, were obtained in the reduction of N-benzyl ketimines. Preliminary studies on the immobilization of these organocatalysts to different solid supports were conducted, with the purpose of applying them in continuous flow systems, which to date has never been reported; RESUMO: No âmbito deste trabalho, foi estudada a síntese de um composto biologicamente ativo usado para o tratamento da doença de Alzheimer, Rivastigmina, usando uma classe de picolinamidas derivadas de alcaloides de cinchona recentemente desenvolvida. Seis 9-picolinamida derivados de alcaloides de cinchona foram preparados com êxito através de metodologias simples e eficazes. Os organocatalisadores foram posteriormente aplicados na redução enantiosseletiva de cetiminas O-protegidas (intermediários de Rivastigmina). Foram obtidos bons resultados na hidrossililação de N-fenilo cetiminas, com rendimentos excelentes e elevadas enantiosseletividades, enquanto a redução de N-benzilo cetiminas proporcionou valores muito mais baixos, tanto em termos de rendimento como de enantiosseletividade. Com o objetivo de serem aplicados em sistemas de fluxo contínuo, realizaram-se estudos preliminares sobre a imobilização destes organocatalisadores em diferentes suportes sólidos, a qual, ate à data, ainda não foi descrita na literatura.

A doença de Alzheimer constitui uma ameaça significativa a nível mundial. Estima-se que, mundialmente existam cerca de 35 milhões de pessoas afetadas por este tipo de demência. Os compostos contendo um esqueleto benzocicloalcanol (que incluem benzofuranos e di-hidrobenzofuranóis) mostram atividades biológicas significativas e possuem muito potencial no tratamento das doenças neurodegenerativas. Nos últimos anos têm havido avanços significativos no campo das reações catalisadas por metais. As reações de adição nucleófila intramolecular e a de Heck intramolecular constituem metodologias importantes para a síntese de benzocicloalcanóis. No âmbito deste trabalho, pretendia-se sintetizar uma biblioteca de compostos contendo um esqueleto benzocicloalcanol. A estratégia adotada para a síntese de dihidrobenzofuranóis envolveu um método de ciclização catalítica de cetonas aril-éteres e para a síntese de benzofuranos, um método de ciclização catalítico de enoatos e enamidas (amidas de Weinreb). Várias condições foram estudadas; Abstract: Studies on Synthetic Catalytic Pathways to Benzocycloalkanols and Derivatives – Potential Drugs for Alzheimer’s Disease Alzheimer's disease constitutes a significant threat worldwide. It is estimated that are about 35 million people worldwide suffering from this type of dementia. The compounds containing a benzocycloalkanol scaffold (including benzofurans and dihydrobenzofurans) show significant biological activity and have great potential in the treatment of neurodegenerative diseases. In recent years there have been many advances in the field of catalyzed reactions by transition-metals. The intramolecular nucleophilic addition and the intramolecular Heck reactions constitute important methods for the synthesis of benzocycloalkanols. Within this work, the main goal was to synthesize a library of compounds containing a benzocycloalkanol scaffold. The adopted strategy for the synthesis of dihydrobenzofurans was the catalytic cyclization of aryl ether ketones and for the synthesis of benzofurans, the catalytic cyclization of enoates and enamides (Weinreb amides). In Alzheimer’s disease, the most common form of dementia, the loss of cholinergic neurons leads to the progressive reduction of acetylcholine in the brain, resulting cognitive impairment. Inhibition of the hydrolysis of acetylcholine by blocking acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) has been considered as a potential target in the treatment of Alzheimer’s disease. Essential oils and extracts of aromatic plants may have an important role in the oxidative stress protection. Traditionally, in Alentejo (Portugal), aromatic herbs Calamintha nepeta, Foeniculum vulgare, Mentha spicata and Thymus mastichina are often used by local population as condiments in food preparations. In this study, essential oils (EOs) and aqueous extracts (decoction waters) of these flavouring herbs were selected in order to evaluate its antioxidant potential and ability to inhibit AChE and BChE activities. Results suggest the potential use of EOs and extracts as nutraceutical or pharmaceutical preparations in the prevention of the oxidative stress and degenerative diseases.

In the chemical composition of olive oil (Olea europaea L.) it is emphasized the massive presence of oleic acid (over 70%), monounsaturated fatty acid part of the family of omega 9, a 7-8% linoleic acid (omega 6) and a small presence (0.5-1%) of linolenic acid (omega 3). For its high content of monounsaturated fatty acids, olive oil is the most stable and therefore the most suitable for heating, compared to oils with a dominance of polyunsaturated fatty acids. Interest in vitamin E has increased in recent years, thanks to its high antioxidant power and its role against related diseases with age-related, visual, dermatological, cardiovascular disorders Alzheimer’s disease and more. Vegetable oils are a major source of vitamin E through diet (Sayago et al., 2007), especially with the variety of olives “Hojiblanca”. Thanks to unsaturated fatty acids cell oxidation can be prevented: this helps prevent many illness, and even premature aging. So far, the advantages acknowledged to olive oil are those of lowering cholesterol, preventing cardiovascular disease, diabetes and cancer. Among the most recent researches it is important to distinguish the studies carried out on their contribution to the prevention and treatment of breast cancer and Alzheimer’s disease. Researchers found that in addition to the benefi ts that give monounsaturated fats, in extra virgin olive oil, there is a substance called “oleocanthal”, which helps protect nerve cells damaged in Alzheimer’s disease. The importance of this discovery is enormous when one considers that only Alzheimer’s disease affects 30 million people around the world, with a different distribution depending on the type of oil in the diet (Olguín Cordero, 2012). The latest research endorses that oleocanthal works by destroying cancer cells without affecting the healthy ones, as it is stated in the Molecular and Cellular Oncology Journal. Studies carried out in different Spanish universities have concluded that thanks to the antioxidant power of olive oil, a disease such as Alzheimer can be prevented. In conclusion, we can say that the Mediterranean diet rich in extra virgin olive oil greatly infl uences on human health, reducing, delaying or even eliminating several diseases.
