No presente trabalho estudaram-se três espécies do género Lavandula, do sul de Portugal: L. luisieri e L. pedunculata (Alentejo) e L.viridis (Algarve), com vista caraterizar a composição química dos óleos essenciais das diferentes espécies, parte vegetativa (folha) e espiga floral, e dos respetivos extratos de hidrodestilação, e avaliar as propriedades antioxidante, antimicrobiana, toxicológica, analgésica e antiinflamatória de alguns dos seus óleos essenciais e extratos aquosos, mais promissores. Os resultados mostraram importantes diferenças na composição química dos óleos essenciais, quanto à diversidade e à proporção dos seus constituintes. Os óleos essenciais e hidrolatos, em estudo, apresentaram importantes propriedades antioxidantes e antimicrobianas. Os estudos toxicológicos e farmacológicos mostraram que os óleos apresentaram citotoxicidade em Artemia salina, apresentaram valores de DL50 muito superiores a 2000 mg/kg em ratinhos Swiss, e que possuem importantes propriedades analgésicas e anti-inflamatória. Estes resultados sugerem o seu potencial uso para aplicações farmacológicas como agentes nutracêuticos e/ou fitoterapêuticos; ABSTRACT:The aim of present work was to develop a set of studies of three species of the genera Lavandula, at the South of Portugal: L. luisieri e L. pedunculata (Alentejo) e L.viridis (Algarve), in order to characterize the chemical composition of the essential oils (leaves and flowers) and to evaluate the antioxidant, antimicrobial, toxicological and pharmacological properties of selected essential oils and aqueous extracts. Results show important differences in chemical composition of essential oils, both in diversity as the proportion of their constituents. Essential oils and aqueous extracts of different Lavandula spp. showed important antimicrobial and antioxidant properties. Pharmacological studies have shown that essential oils showed cytotoxicity against Artemia salina, low acute toxicity, with LD50 >> 2000 mg/kg for mice, and important analgesic and anti-inflammatory properties. These results suggest their potential use for pharmacological applications as nutraceutical and/or phytotherapeutic agents.

O nemátode da madeira do pinheiro (NMP), Bursaphelenchus xylophiius, tem uma extensa distribuição na América do Norte, e encontra-se atualmente distribuído ao longo da maioria dos territórios de Canadá e dos Estados Unidos. Durante o último século, esta espécie foi transportada pelo Homem para outras regiões do mundo (não-nativas), associadas com o comércio e o fluxo global de produtos de origem florestal. Atualmente, esta espécie invasiva está reportada para algumas regiões do SE asiático (China, Japão, Coreia e Taiwan) e mais recentemente para a Europa (Portugal). Devido ao impacto que este organismo agente da doença da murchidão dos pinheiros causa nas florestas nativas destas regiões esta espécie assume uma elevada importância económica a nível mundial Em Portugal, a distribuição do NMP encontra-se confinada a uma área restrita e limitada (500 000 ha), a sul de Lisboa (península de Setúbal); contudo, constitui uma das maiores ameaças às florestas de pinheiro do país e da UE. Ate recentemente, nenhum consenso existia quanto à origem do NMP em Portugal. Diversas hipóteses têm sido colocadas para explicar esta introdução, nomeadamente a partir de zonas onde o nematode ocorre naturalmente (América do Norte), ou de outras áreas (não-nativas) onde o nematode se comporta como uma espécie invasiva (Leste da Ásia). A fim de avaliar a variabilidade genética do NMP proveniente da área afetada em Portugal, foram utilizadas várias técnicas moleculares, designadamente o random amplified polymorphic DNA (RAPD-PCR) e o satellite DNA (satDNA). No caso do RAPD-PCR, foram utilizados 24 isolados do NMP provenientes de Portugal, 1 proveniente da América do Norte e 1 da Ásia, tendo sido utilizado como out-group um isolado de B. mucronatus. A partir dos 28 RAPD primers utilizados obtiveram-se 640 fragmentos. No caso do satDNA, foram utilizados 21 isolados do NMP provenientes de Portugal, obtendo-se no total 206 sequências da família MspI. Ambos os métodos revelaram uma elevada similaridade genética entre os vários isolados do NMP da área afetada em Portugal O nível reduzido de diversidade genética obtido entre os isolados portugueses do NMP, permite concluir que se trata de uma única introdução deste organismo em Portugal, e proveniente de uma região asiática. A inexistência de uma de correlação entre a variabilidade genética e a distribuição geográfica do NMP dentro da área afetada em Portugal, indica que o NMP se encontra distribuído de forma uniforme ao longo de toda a área afetada, provavelmente relacionado com a distribuição e a expansão natural do inseto vector. The pinewood nematode (PWN), Bursaphelenchus xylophilus, has a wide distribution in North America, and is present throughout most of the territories of Canada and the United Stata. During the last century, this species has been transported by man to several non-native regions of the world, associated with trade and the global flow of forest products. Up to date, this invasive species has been reported from Asia (PR China, Japan, Korea and Taiwan) and more recently in Europe (Portugal). Due to the impact on native pine forests of these regions, this nematode species, the causal agent of pine wilt disease, is of great economic importance worldwide. In Portugal, the distribution of the PWN has been constrained to a relatively small area (500 000 ha) in the south of Lisbon (Setúbal Peninsula); however, it has become the most serious threat to pine forests in the country. Until recently, no consensus had emerged on the possible pathway of the PWN introduction in Portugal. Several hypotheses have been put forward to explain this introduction, such as an origin from endemic areas where the nematode naturally occurs (North America), or non-endemic areas where the nematode behaves as an exotic pest (East Asia). Random amplified polymorphic DNA (RAPD-PCR) and satellite DNA (satDNA) techniques were used in order to assess the level of genetic variability and genetic relationships, among several isolates of the PWN, representative of the entire affected area in Portugal. In the case of RAPD-PCR, 24 Portuguese isolates, plus two additional isolates of B. xylophilus, representing North America and East Asia were included. B. mucronatus was used as an out-group. Twenty-eight random primers generated a total of 640 DNA fragments. With satDNA, 206 Mspl sequence repeats were obtained from 21 Portuguese isolates of B. xylophilus. Both molecular methods revealed a high genetic similarity among the Portuguese isolates, and the low level of genetic diversity strongly suggests that they were dispersed recently from a single introduction, and from East Asia. The lack of apparent relationship between the genetic variability and the geographic distribution of the PWN within the affected area, suggests that the recent introduction of this pest (and pathogen) in Portugal has been uniformly distributed since its establishment, probably following the natural distribution and expansion of the insect vector.

Esta dissertação pretende contribuir para um melhor conhecimento da complexidade das redes de transferência de conhecimentos e técnicas, no domínio da engenharia civil e mais concretamente através dos caminhos-de-ferro, nos séculos XIX e XX. Em Portugal, os caminhos-de-ferro estiveram no cerne de um vasto debate, sobretudo político, concomitante com uma instabilidade crescente no cenário político e uma fase de fragilidade económica. É neste contexto que a Linha do Sul e Sueste vai ser construída (seguida pela sua extensão até Vila Real de Santo António e pela construção do ramal de Portimão, que chegará a Lagos). Este empreendimento é uma clara ilustração da realidade portuguesa de então, no que concerne ao desenvolvimento desta rede de transportes, que nos permite, igualmente, conhecer e compreender quem interveio no processo de construção da linha (os engenheiros, as empresas, entre outros aspectos) e assim determinar quais as influências e transferências técnicas que tiveram lugar; RESUMEE: Cette mémoire attire à la contribution pour une meilleure connaissance de la complexité des réseaux de transfert de techniques et connaissances qui ont eu lieu dans le domaine de l’ingénierie civile, surtout dans les chemins de fer, au XIXème et XXème siècles. Au Portugal, les chemins de fer sont été le cerne d’un très vaste débat, coïncidant avec une croissante instabilité dans le scenario politique et aussi une phase économique fragile. C’est dans ce contexte que la Ligne du Sud et Sud-est va être bâti (suivi par l’extension jusqu’à Vila Real de Santo António et la construction de l’embranchement ferroviaire Portimão). Cette entreprise c’est une illustration claire de la réalité portugaise, en concernant l’implémentation de cette réseau de transport, que nous permettre de comprendre et également bien connaitre qui a intervenu dans le processus de construction de la ligne (les ingénieurs, entreprises, etcetera), ainsi que déterminer les influences et les transferts techniques qui ont eu lieu; ABSTRACT: With this master’s thesis, the aim is to be able to contribute to a better understanding of the complex network of technique’s and knowledge transfers, that took place within the field of civil engineering, in the 19th and 20th centuries, namely on the railways. In Portugal, railways take-up was a wide and ample debate, coinciding with an uprising turmoil on the Portuguese political outskirt and a phase of economic frailty. It’s in this context that the construction of the South and Southeast Line took place (followed, later on, by its extension until Vila Real de Santo António and by the construction of the Portimão’s branch). This enterprise is, as we pretend to prove in this master’s thesis, a clear example of the Portuguese reality, enabling us to understand and to get to know those who intervened in the construction’s process (the engineers and the companies) as well as determining influences and technique transfers that have taken place.

Greenhouse production is a very important activity in the West region of Portugal, with an area of approximately 800 ha where the regular production consists in two crops per year, one in winter-spring and the other in summer-autumn. Many growers are now prepared to better exploit market opportunities, since they know that the big export window opportunity is from June to September, when the production is difficult in other regions of south due to high temperatures. Grower’s use new and more productive varieties, either in soil or hydroponic systems, mostly in unheated greenhouses, naturally ventilated, and equipped with modern fertigation systems. Greenhouse production causes some environmental impacts due to the high use of inputs. Several improvements in technologies and crop practices may contribute to increase the use efficiency of resources, decreasing the negative environmental impacts. Greenhouse vegetable production in Northern EU countries is based on the supply of heating and differs significantly from the production system in the Southern EU countries. In the Northern countries, direct energy inputs, mostly for heating, are predominant while in the South the indirect energy input is also important, mainly associated with fertilizers, plastic cover materials and other auxiliary materials. The main objective of this work was to characterise the greenhouse production systems in the West region of Portugal, in order to evaluate the energetic consumptions (direct and indirect), the GHH emissions, the production costs and the farmer’s income. With this work the mostly important inputs were identified, allowing proposing alternative measures to improve efficiency and sustainability. All the data was obtained by surveys performed directly with growers, previously selected to be representative of the crop practices and greenhouse type of the region. However, more research should be performed in order to develop and to test technologies capable to improve resources use efficiency in greenhouse production.

Solar resource assessment is essential for the different phases of solar energy projects, such as preliminary design engineering, financing including due diligence and, later, insurance phases. An important aspect is the long term resource estimation. This kind of estimation can only be obtained through the statistical analysis of long-term data series of solar radiation measurements, preferably ground measurements. This paper is a first step in this direction, with an initial statistical analysis performed over the radiation data from a national measurement network, consisting of eighty-nine meteorological stations. These preliminary results are presented in figures that represent the annual average values of Global Horizontal Irradiation (GHI) and its Variability in the Portuguese continental territory. These results show that the South of Portugal is the most suitable area for the implementation of medium to large scale solar plants.

Changes in fish assemblage structure caused by human activities, such as fishing, can alter trophic relations in fish assemblages. In this context, Marine Protected Areas (MPA) are efficient tools for habitat recovery and ideal environments for evaluating changes on the trophic structure resulting from human activities. The present work targeted fish assemblages from two no-take MPAs from the northern half of South Alentejo and Costa Vicentina Marine Park, established in 2011. Previous works reported positive effects on local fish assemblages after no-take MPA designation, and it is therefore important to further study its impact on local fish assemblages, especially concerning trophic interactions. Local fish assemblages were sampled (summer 2011, winter 2012, summer 2013 and winter 2013) using trammel nets. Diets were characterized and digestive tract contents of the 10 most abundant fish species were compared between the no take MPAs (treatment) and adjacent areas (controls), and changes evaluated as a function of time since protection. Results revealed significant differences between the diets of fish from protected and non protected areas, with crabs being the preferential prey in both protected and control areas but being more ingested outside the no-take areas. However, these differences were evident since the beginning of the study. Fish assemblages from the northern area presented significantly larger niche breadth and significantly increasing with time. This way, the main effects of no-take MPA implementation were directly visible on the niche breadth but did not directly impact the diet composition of the sampled fish assemblages, contributing however to reinforce the already naturally existent differences. This work provides important information regarding the effect of changes in the fish assemblage caused by MPA designation on the trophic ecology of fish.
