A better method for determination of shikimate in plant tissues is needed to monitor exposure of plants to the herbicide glyphosate [N-(phosphonomethyl)glycine] and to screen the plant kingdom for high levels of this valuable phytochemical precursor to the pharmaceutical oseltamivir. A simple, rapid, and efficient method using microwave-assisted extraction (MWAE) with water as the extraction solvent was developed for the determination of shikimic acid in plant tissues. High performance liquid chromatography was used for the separation of shikimic acid, and chromatographic data were acquired using photodiode array detection. This MWAE technique was successful in recovering shikimic acid from a series of fortified plant tissues at more than 90% efficiency with an interference-free chromatogram. This allowed the use of lower amounts of reagents and organic solvents, reducing the use of toxic and/or hazardous chemicals, as compared to currently used methodologies. The method was used to determine the level of endogenous shikimic acid in several species of Brachiaria and sugarcane (Saccharum officinarum) and on B. decumbens and soybean (Glycine max) after treatment with glyphosate. The method was sensitive, rapid and reliable in all cases.

The water availability for flood irrigated rice (Oryza sativa L.) is decreasing worldwide. Therefore, developing technologies to allow growing rice in aerobic condition, such as a no-tillage system (NTS) can contribute to produce upland rice grains without yield losses and also in saving more water. The objective of this study was to determine the effect of soil management, seed treatment and compaction on the sowing furrow on grain yield of upland rice genotypes. We made two trials, one in an NTS and another using conventional tillage, CT (one plowing and two diskings). The field experiments were performed in the Central Region of Brazil in Cerrado soils. For each trial, the experimental design was a randomized block design in a factorial scheme, with three replications. The treatments consisted of a combination of 10 genotypes with 2 compaction pressures on the sowing furrow (25 kPa and 126kPa) and 2 types of seed treatment (with and without pesticide). Under CT, the seed treatment did not contribute to increase upland rice grain yields. However, under NTS the grain yield of some genotypes [BRS Esmeralda (from 723 to 1,766 kg ha-1), BRS Pepita (from 930 to 1,874 kg ha-1), AB072044 (from 523 to 1,579 kg ha-1), and AB072085 (from 632 to 1,636 kg ha-1) at 25 kPA soil compaction pressure, and Sertaneja (from 994 to 2,167 kg ha-1), BRS Pepita (from 1,161 to 2,100 kg ha-1), and AB072085 (from 958 to 2,213 kg ha-1), at 126 kPA soil compaction pressure] increased with the use of this practice. At CT the higher soil compaction pressure on the sowing furrow (from 25 kPa to 126 kPa) increased rice grain yield only when it was used seed treatment and the genotypes Serra Dourada (from 1,239 to 2,178 kg ha-1), Sertaneja (from 1,510 to 2,379 kg ha-1), and Cambará (from 1,877 to 2,831 kg ha-1). On the other hand, under NTS, increasing soil compaction pressure on the sowing furrow allowed for an increased rice grain yield of Serra Dourada (from 1,553 to 2,347 kg ha-1), Esmeralda (from 723 to 1,643 kg ha-1), AB072044 (from 523 to 2,040 kg ha-1), and Cambará (from 1,243 to 2,032 kg ha-1) without seed treatment and Sertaneja (from 1,385 to 2,167 kg ha-1) and AB072044 (from 1,579 to 2,356 kg ha-1) with seed treatment. In CT the most productive genotypes were AB062008 (2,714 kg ha-1) and BRSMG Caravera (2,479 kg ha-1), while at NTS were the genotypes: BRSGO Serra Dourada (2,118 kg ha-1), AB072047 (1,888 kg ha-1), AB062008 (1,823 kg ha-1), BRSMG Caravera (1,737 kg ha-1), Cambará (1,716 kg ha-1), AB072044 (1,625 kg ha-1), BRS Esmeralda (1,604 kg ha-1), and BRS Pepita (1,516 kg ha-1).

O objetivo deste trabalho foi avaliar a localização e o numero de bactérias endolíticas em quatro genótipos de cana-de-açúcar e investigar sobre a possível existência de correlação com os resultados apresentados em trabalhos de quantificação da fixação biológica de nitrogênio (FBN). Fez-se um levantamento das bactérias diazotróficas presentes, e quantificou-se a população de Herbaspirillum spp. E Acetobacter diazotrophicus, em genótipos de cana-de-açúcar contrastantes quanto a capacidade de obter N da FBN. De acordo com o levantamento realizado neste trabalho, as bactérias estudadas (Azospirillum lipoferum, A. brasilense, A. amazonense, Herbaspirillum spp. e Acetobacter diazotrophicus) estavam presentes nos quatro genotipos avaliados e em todas as partes da planta, exceto A. amazonense, que nao foi isolado de amostras de folhas. A quantificaçãoo das bactérias Herbaspirillum spp. e A. diazotrophicus mostrou não haver diferenças significativas entre os genótipos, e que, geralmente, elas estão presentes em maior numero nas raízes. Enquanto Herbaspirillum spp. mantêm-se mais estável ao longo do ciclo da cultura, a população de A. diazotrophicus decresce com a aproximação do final do ciclo comercial. Pode-se sugerir que as diferenças entre as taxas de FBN encontradas nos diversos genótipos não e causada por diferenças na presença ou no numero das bactérias aqui estudadas The objective of this work was to find out the localization and number of endophytic bacteria in four sugar cane genotypes and investigate upon the possible existence of correlation to the results obtained in some studies about quantification of biological nitrogen fixation (BNF). A survey of the diazotrophic bacteria present in sugar cane genotypes differingin their capacity to obtain nitrogen through BNF was performed, and population of Herbaspirillum spp. and Acetobacter diazotrophicus was quantified. The bacteria tested in the survey were Azospirillum lipoferum, A. brasilense, A. amazonense, Herbaspirillum spp. and Acetobacter diazotrophicus. All these bacteria were present in the four genotypes and were found in all parts of the plants, except A. amazonense which was not isolated from leaf samples. The quantification of Herbaspirillum spp. and A. diazotrophicus showed that there were no significant differences among the sugar cane genotypes and, generally, the bacteria were in greater number in roots. While number of Herbaspirillum spp. remained stable during the life-cycle of the culture, the population of A. diazotrophicus suffer a decrease with the approach of the end of the commercial cycle. It is suggested that the differences in the rates of BNF found in sugar cane genotypes are not caused by differences in the presence or the number of the bacterial species studied here.
