DNA obtained from a human sputum isolate of Mycobacterium tuberculosis, NTI-64719, which showed extensive dissemination in the guinea pig model resulting in a high score for virulence was used to construct an expression library in the lambda ZAP vector. The size of DNA inserts in the library ranged from 1 to 3 kb, and recombinants represented 60% of the total plaques obtained. When probed with pooled serum from chronically infected tuberculosis patients, the library yielded 176 recombinants with a range of signal intensities. Among these, 93 recombinants were classified into 12 groups on the basis of DNA hybridization experiments, The polypeptides synthesized by the recombinants were predominantly LacZ fusion proteins, Serum obtained from patients who were clinically diagnosed to be in the early phase of M. tuberculosis infection was used to probe the 176 recombinants obtained. interestingly, some recombinants that gave very strong signals in the original screen did not react with early-phase serum; conversely, others whose signals were extremely weak in the original screen gave very intense signals with serum from recently infected patients, This indicates the differential nature of either the expression of these antigens or the immune response elicited by them as a function of disease progression.

The governing differential equation of linear, elastic, thin, circular plate of uniform thickness, subjected to uniformly distributed load and resting on Winkler-Pasternak type foundation is solved using ``Chebyshev Polynomials''. Analysis is carried out using Lenczos' technique, both for simply supported and clamped plates. Numerical results thus obtained by perturbing the differential equation for plates without foundation are compared and are found to be in good agreement with the available results. The effect of foundation on central deflection of the plate is shown in the form of graphs.

Die Vorliegende Arbeit beschäftigt sich mit den Spannungen und Verschiebungen an einem elastischen Halbraum unter einem kreisförmigen biegsamen Fundament, wenn an der Kontaktfläche vollkommenes Haften besteht. Das gemischte Randwertproblem wird mit Hilfe von Hankel-Transformationen auf duale Integralgleichungen von Titchmarsh- Typ zurückgeführt. Für die Berechnung der Spannungen und Verschiebungen werden Gaußsche Quadraturformeln benutzt. Die Ergebnisse werden mit denen verglichen, die man bei glattem Fundament erhält, und der Einfluß der Poisson-Zahl auf die Spannungen und Verschiebungen wird deutlich gemacht. Schließlich werden die Ergebnisse für den praktischen Gebrauch in Diagrammen und Tabellen zusammengefaßt.

The natural frequencies of a reservoir-foundation system are calculated by treating the foundation as a system of linear springs with inertia. The reservoir is treated as consisting of compressible liquid, and the influence of waves at the free surface is included. It is shown that the natural frequencies decrease monotonically as the depth of foundation participating in the motion increases. The influence of waves at the reservoir surface is negligible for the cases normally occurring in practice. It is also shown that the wavelength of motion along the reservoir has no influence on the frequencies when the foundation depth is large compared to the reservoir depth.

Ten new cyclic hexadepsipeptides, six isariins and four isaridins, from the fungus Isaria have been identified and characterized by high-performance liquid chromatography, coupled to tandem electrospray ionization mass spectrometry (LC-ESIMS/MS). The isariins possess a beta-hydroxy acid residue and five alpha-amino acids, while isaridins contain a beta-amino acid, an alpha-hydroxy acid, and four alpha-amino acids. One- and two-dimensional NMR spectroscopy confirmed the chemical identity of some of the isariin fractions. Mass spectral fragmentation patterns of [M + H](+) ions reveal clear diagnostic fragment ions for the isariins and isaridins. Previously described cyclic depsipeptides, isarfelins from Isaria felina (Guo, Y. X.; Liu, Q. H.; Ng, T. B.; Wang H. X. Peptides 2005, 26, 2384), are now reassigned as members of the isaridin family. Examination of isaridin sequences revealed significant similarities with cyclic hexadepsipeptides such as destruxins and roseotoxins. The structure of an isariin (isariin A) investigated by NMR spectroscopy indicated the presence of a hybrid alpha beta C-11 turn, formed by the beta-hydroxy acid and glycine residues and a (D)Leu-(L)Ala type II' beta-turn. Additionally, the inhibitory effect of isariins and an isaridin on the intra-erythrocytic growth of Plasmodium falciparum is presented.

The interaction of a framed structure with a foundation beam resting on an elastic medium, representing the soil, has been studied using the photoelastic method. The contact pressure distribution, the fibre stress in the foundation beam and frame structure, as well as the stresses in the elastic medium, have been obtained. These have been compared with theoretical results obtained by idealizing the soil as (a) elastic half plane, and (b) elastic half space. It is shown that the photoelastic method can provide an easy solution to this type of problem if the soil can be idealized as an elastic continuum.

In this paper, dynamic response of an infinitely long beam resting on a foundation of finite depth, under a moving force is studied. The effect of foundation inertia is included in the analysis by modelling the foundation as a series of closely spaced axially vibrating rods of finite depth, fixed at the bottom and connected to the beam at the top. Viscous damping in the beam and foundation is included in the analysis. Steady state response of the beam-foundation system is obtained. Detailed numerical results are presented to study the effect of various parameters such as foundation mass, velocity of the moving load, damping and axial force on the beam. It is shown that foundation inertia can considerably reduce the critical velocity and can also amplify the beam response.

Nonlinear static and dynamic response analyses of a clamped. rectangular composite plate resting on a two-parameter elastic foundation have been studied using von Karman's relations. Incorporating the material damping, the governing coupled, nonlinear partial differential equations are obtained for the plate under step pressure pulse load excitation. These equations have been solved by a one-term solution and by applying Galerkin's technique to the deflection equation. This yields an ordinary nonlinear differential equation in time. The nonlinear static solution is obtained by neglecting the time-dependent variables. Thc nonlinear dynamic damped response is obtained by applying the ultraspherical polynomial approximation (UPA) technique. The influences of foundation modulus, shear modulus, orthotropy, etc. upon the nonlinear static and dynamic responses have been presented.

Analysis of rectangular plates resting on a Winkler-type, one-parameter foundation is studied. The finite element method is applied and a 12-degree-of-freedom, nonconforming rectangular plate element is adopted. Based on shape functions of the plate element, an energy approach is used to derive a closed-form, 12-by-12, consistent foundation stiffness matrix for a rectangular plate on an elastic subgrade. A commonly used method of modeling structural elements on an elastic foundation is the application of discrete springs at the element nodes. The model developed in this paper is compared with the discrete spring model and the convergence of both models is discussed. The convergence of the models is compared with the well-known classical solution of plates on elastic foundation developed in the 1950s. Both models show good convergence to the classical solution. The continuous subgrade response model converges in a manner more consistent with the flexibility of the plate element.
