Monitoring and visualizing specimens at a large penetration depth is a challenge. At depths of hundreds of microns, several physical effects (such as, scattering, PSF distortion and noise) deteriorate the image quality and prohibit a detailed study of key biological phenomena. In this study, we use a Bessel-like beam in-conjugation with an orthogonal detection system to achieve depth imaging. A Bessel-like penetrating diffractionless beam is generated by engineering the back-aperture of the excitation objective. The proposed excitation scheme allows continuous scanning by simply translating the detection PSF. This type of imaging system is beneficial for obtaining depth information from any desired specimen layer, including nano-particle tracking in thick tissue. As demonstrated by imaging the fluorescent polymer-tagged-CaCO3 particles and yeast cells in a tissue-like gel-matrix, the system offers a penetration depth that extends up to 650 mu m. This achievement will advance the field of fluorescence imaging and deep nano-particle tracking.

Classical literature on solid mechanics claims existence of radial deformation due to torsion but there is hardly any literature on analytic solutions capturing this phenomenon. This paper tries to solve this problem in an asymptotic sense using the variational asymptotic method (VAM). The method makes no ad hoc assumptions and hence asymptotic correctness is assured. The VAM splits the 3D elasticity problem into two parts: A 1D problem along the length of the cylinder which gives the twist and a 2D cross-sectional problem which gives the radial deformation. This enables closed form solutions, even for some complex problems. Starting with a hollow cylinder, made up of orthotropic but transversely isotropic material, the 3D problem has been formulated and solved analytically despite the presence of geometric nonlinearity. Design of a dual band pass filter employing microstrip line with defected ground is presented in this paper. A dual band filter at 2.45GHz and 3.5GHz (covering WLAN and WiMAX) with 6% bandwidth has been designed at each frequency. Apertures in ground plane were used to improve the stop band rejection characteristics and coupling levels in the filter. Measured results of the experimental filter were compared against the simulation results for the purpose of validation.

We consider the speech production mechanism and the asso- ciated linear source-filter model. For voiced speech sounds in particular, the source/glottal excitation is modeled as a stream of impulses and the filter as a cascade of second-order resonators. We show that the process of sampling speech signals can be modeled as filtering a stream of Dirac impulses (a model for the excitation) with a kernel function (the vocal tract response),and then sampling uniformly. We show that the problem of esti- mating the excitation is equivalent to the problem of recovering a stream of Dirac impulses from samples of a filtered version. We present associated algorithms based on the annihilating filter and also make a comparison with the classical linear prediction technique, which is well known in speech analysis. Results on synthesized as well as natural speech data are presented.

We propose a new method for design of computationally efficient nonsubsampled multiscale multidirectional filter bank with perfect reconstruction (PR). This filter bank is composed of two nonsubsampled filter banks, for multiscale decomposition and for directional expansion. For multiscale decomposition, we transform the 1-D equivalent subband filters directly into 2-D equivalent subband filters. The computational cost is considerably reduced by avoiding the computation of 2-D convolutions. The multidirectional decomposition utilizes fan filters. A new method for design of 2-D zero phase FIR fan filter transformation function is developed. This method also aids the transformation of a 1-D filter bank to a 2-D multidirectional filter bank. The potential application of the proposed filter bank is illustrated by comparing the image denoising performance of the proposed filter bank with other design method that exist in available literature.

Earlier studies have indicated that variability in size, surface texture and charge greatly influence the contaminant removal process in granular media. Based on surface characteristics of montmorillonite, it is anticipated that small addition of this clay would increase adhesion sites for bacterial growth and extracellular polymer production in the slow sand filter and thereby enhance its contaminant removal ability. Experiments were performed by permeating groundwater contaminated with pathogens (total coliform and E. Coli) and inorganic contaminants through the bentonite amended slow sand filter (BASSF). Surprisingly, the BASSF retained inorganic contaminants besides pathogens. Water-leach tests (pH of water leachate ranged from 2 to 9) with spent BASSF specimen indicated that the inorganic contaminants are irreversibly adsorbed to a large extent. It is considered that the combined effects of enhanced-organic matter mediated adhesion sites and increased hydraulic retention time enables the BASSF specimen to retain inorganic contaminants. It is envisaged that BASSF filters could find use in treating contaminated groundwater for potable needs at household and community level.

Closed-form expressions for the propagation characteristics of coupled microstrip lines with a symmetrical aperture in the ground plane are derived. Expressions for the regular microstrip coupled lines have been modified using physical insights to incorporate the effect of the aperture. The accuracy of these expressions has been verified by full-wave simulations and compared with conformal mapping analysis. These expressions are accurate within 5% for a substrate whose thickness varies from 0.2 to 1.6mm and permittivity in the range of 210. Designing a broadband filter based on planar multi-conductor coupled lines with aperture in the ground plane is demonstrated in this paper using the proposed expressions for its practical use.

A common-mode (CM) filter based on the LCL filter topology is proposed in this paper, which provides a parallel path for ground currents and which also restricts the magnitude of the EMI noise injected into the grid. The CM filter makes use of the components of a line to line LCL filter, which is modified to address the CM voltage with minimal additional components. This leads to a compact filtering solution. The CM voltage of an adjustable speed drive using a PWM rectifier is analyzed for this purpose. The filter design is based on the CM equivalent circuit of the drive system. The filter addresses the adverse effects of the PWM rectifier in an adjustable speed drive. Guidelines are provided on the selection of the filter components. Different variants of the filter topology are evaluated to establish the effectiveness of the proposed circuit. Experimental results based on EMI measurement on the grid side and the CM current measurement on the motor side are presented. These results validate the effectiveness of the filter.

Fastest curve-fitting procedures are proposed for vertical and radial consolidations for rapid loading methods. In vertical consolidation, the next load increment can be applied at 50-60% consolidation (or even earlier if the compression index is known). In radial consolidation, the next load increment can be applied at just 10-15% consolidation. The effects of secondary consolidation on the coefficient of consolidation and ultimate settlement are minimized in both cases. A quick procedure is proposed in vertical consolidation that determines how far is calculated from the true , where is coefficient of consolidation. In radial consolidation no such procedure is required because at 10-15% the consolidation effects of secondary consolidation are already less in most inorganic soils. The proposed rapid loading methods can be used when the settlement or time of load increment is not known. The characteristic features of vertical, radial, three-dimensional, and secondary consolidations are given in terms of the rate of settlement. A relationship is proposed between the coefficient of the vertical consolidation, load increment ratio, and compression index. (C) 2013 American Society of Civil Engineers.

A power filter is necessary to connect the output of a power converter to the grid so as to reduce the harmonic distortion introduced in the line current and voltage by the power converter. Many a times, a transformer is also present before the point of common coupling. Magnetic components often constitute a significant part of the overall weight, size and cost of the grid interface scheme. So, a compact inexpensive design is desirable. A higher-order LCL-filter and a transformer are increasingly being considered for grid interconnection of the power converter. This study proposes a design method based on a three-winding transformer, that generates an integrated structure that behaves as an LCL-filter, with both the filter inductances and the transformer that are merged into a single electromagnetic component. The parameters of the transformer are derived analytically. It is shown that along with a filter capacitor, the transformer parameters provide the filtering action of an LCL-filter. A single-phase full-bridge power converter is operated as a static compensator for performance evaluation of the integrated filter transformer. A resonant integrator-based single-phase phase locked loop and stationary frame AC current controller are employed for grid frequency synchronisation and line current control, respectively.

Study of Oceans dynamics and forecast is crucial as it influences the regional climate and other marine activities. Forecasting oceanographic states like sea surface currents, Sea surface temperature (SST) and mixed layer depth at different time scales is extremely important for these activities. These forecasts are generated by various ocean general circulation models (OGCM). One such model is the Regional Ocean Modelling System (ROMS). Though ROMS can simulate several features of ocean, it cannot reproduce the thermocline of the ocean properly. Solution to this problem is to incorporates data assimilation (DA) in the model. DA system using Ensemble Transform Kalman Filter (ETKF) has been developed for ROMS model to improve the accuracy of the model forecast. To assimilate data temperature and salinity from ARGO data has been used as observation. Assimilated temperature and salinity without localization shows oscillations compared to the model run without assimilation for India Ocean. Same was also found for u and v-velocity fields. With localization we found that the state variables are diverging within the localization scale.

This paper reports the first observations of transition from a pre-vortex breakdown (Pre-VB) flowreversal to a fully developed central toroidal recirculation zone in a non-reacting, double-concentric swirling jet configuration and its response to longitudinal acoustic excitation. This transition proceeds with the formation of two intermediate, critical flow regimes. First, a partially penetrated vortex breakdown bubble (VBB) is formed that indicates the first occurrence of an enclosed structure as the centre jet penetration is suppressed by the growing outer roll-up eddy; resulting in an opposed flow stagnation region. Second, a metastable transition structure is formed that marks the collapse of inner mixing vortices. In this study, the time-averaged topological changes in the coherent recirculation structures are discussed based on the non-dimensional modified Rossby number (Ro(m)) which appears to describe the spreading of the zone of swirl influence in different flow regimes. Further, the time-mean global acoustic response of pre-VB and VBB is measured as a function of pulsing frequency using the relative aerodynamic blockage factor (i.e., maximum radial width of the inner recirculation zone). It is observed that all flow modes except VBB are structurally unstable as they exhibit severe transverse radial shrinkage (similar to 20%) at the burner Helmholtz resonant modes (100-110 Hz). In contrast, all flow regimes show positional instability as seen by the large-scale, asymmetric spatial shifting of the vortex core centres. Finally, the mixing transfer function M (f) and magnitude squared coherence lambda(2)(f) analysis is presented to determine the natural couplingmodes of the system dynamic parameters (u', p'), i.e., local acoustic response. It is seen that the pre-VB flow mode exhibits a narrow-band, low pass filter behavior with a linear response window of 100-105 Hz. However, in the VBB structure, presence of critical regions such as the opposed flow stagnation region alters the linearity range with the structure showing a response even at higher pulsing frequencies (100-300 Hz). The pore of sodium channels contains a selectivity filter made of 4 amino acids, D/E/K/A. In voltage sensitive sodium channel (Nav) channels from jellyfish to human the fourth amino acid is Ala. This Ala, when mutated to Asp, promotes slow inactivation. In some Nav channels of pufferfishes, the Ala is replaced with Gly. We studied the biophysical properties of an Ala-to-Gly substitution (A1529G) in rat Nav1.4 channel expressed in Xenopus oocytes alone or with a beta 1 subunit. The Ala-to-Gly substitution does not affect monovalent cation selectivity and positively shifts the voltage-dependent inactivation curve, although co-expression with a beta 1 subunit eliminates the difference between A1529G and WT. There is almost no difference in channel fast inactivation, but the beta 1 subunit accelerates WT current inactivation significantly more than it does the A1529G channels. The Ala-to-Gly substitution mainly influences the rate of recovery from slow inactivation. Again, the beta 1 subunit is less effective on speeding recovery of A1529G than the WT. We searched Nav channels in numerous databases and noted at least four other independent Ala-to-Gly substitutions in Nav channels in teleost fishes. Thus, the Ala-to-Gly substitution occurs more frequently than previously realized, possibly under selection for alterations of channel gating.

Comparator based switched capacitor circuits provide an excellent opportunity to design sampled data systems where the virtual ground condition is detected rather than being continuously forced with negative feedback in Opamp based circuits. This work is an application of this concept to design a 1 st order 330 KHz cutoff frequency Lowpass filter operating at 10 MHz sampling frequency in 0.13μm technology and 1.2 V supply voltage. The Comparator Based Switched Capacitor (CBSC) filter is compared with conventional Two stage Miller compensated Operational amplifier based switched capacitor filter. It is shown that CBSC filter relaxes the constraints like speed ,linearity, gain, stability which would otherwise be hard to satisfy in scaled technologies in Opamp based circuits. The designed CBSC based lowpass filter provides significant power savings compared to traditional Opamp based switched capacitor filter.

Impoverishment of particles, i.e. the discretely simulated sample paths of the process dynamics, poses a major obstacle in employing the particle filters for large dimensional nonlinear system identification. A known route of alleviating this impoverishment, i.e. of using an exponentially increasing ensemble size vis-a-vis the system dimension, remains computationally infeasible in most cases of practical importance. In this work, we explore the possibility of unscented transformation on Gaussian random variables, as incorporated within a scaled Gaussian sum stochastic filter, as a means of applying the nonlinear stochastic filtering theory to higher dimensional structural system identification problems. As an additional strategy to reconcile the evolving process dynamics with the observation history, the proposed filtering scheme also modifies the process model via the incorporation of gain-weighted innovation terms. The reported numerical work on the identification of structural dynamic models of dimension up to 100 is indicative of the potential of the proposed filter in realizing the stated aim of successfully treating relatively larger dimensional filtering problems.
