We study the shape parameters of the Dπ scalar and vector form factors using as input dispersion relations and unitarity for the moments of suitable heavy-light correlators evaluated with Operator Product Expansions, including O(α 2 s) terms in perturbative QCD. For the scalar form factor, a low energy theorem and phase information on the unitarity cut are implemented to further constrain the shape parameters. We finally determine points on the real axis and isolate regions in the complex energy plane where zeros of the form factors are excluded.

The availability of a reliable bound on an integral involving the square of the modulus of a form factor on the unitarity cut allows one to constrain the form factor at points inside the analyticity domain and its shape parameters, and also to isolate domains on the real axis and in the complex energy plane where zeros are excluded. In this lecture note, we review the mathematical techniques of this formalism in its standard form, known as the method of unitarity bounds, and recent developments which allow us to include information on the phase and modulus along a part of the unitarity cut. We also provide a brief summary of some results that we have obtained in the recent past, which demonstrate the usefulness of the method for precision predictions on the form factors.

Curcumin, a principal component of turmeric, acts as an immunomodulator regulating the host defenses in response to a diseased condition. The role of curcumin in controlling certain infectious diseases is highly controversial. It is known to alleviate symptoms of Helicobacter pylori infection and exacerbate that of Leishmania infection. We have evaluated the role of curcumin in modulating the fate of various intracellular bacterial pathogens. We show that pretreatment of macrophages with curcumin attenuates the infections caused by Shigella flexneri (clinical isolates) and Listeria monocytogenes and aggravates those caused by Salmonella enterica serovar Typhi CT18 (a clinical isolate), Salmonella enterica serovar Typhimurium, Staphylococcus aureus, and Yersinia enterocolitica. Thus, the antimicrobial nature of curcumin is not a general phenomenon. It modulated the intracellular survival of cytosolic (S. flexneri and L. monocytogenes) and vacuolar (Salmonella spp., Y. enterocolitica, and S. aureus) bacteria in distinct ways. Through colocalization experiments, we demonstrated that curcumin prevented the active phagosomal escape of cytosolic pathogens and enhanced the active inhibition of lysosomal fusion by vacuolar pathogens. A chloroquine resistance assay confirmed that curcumin retarded the escape of the cytosolic pathogens, thus reducing their inter- and intracellular spread. We have demonstrated that the membrane-stabilizing activity of curcumin is crucial for its differential effect on the virulence of the bacteria.

Ag-Ni films were electrodeposited over a Cu substrate. Structural characterization revealed a fibrous microstructure with an amorphous structure for the as-deposited film. Isothermal annealing at 400 degrees C of the film inside transmission electron microscope led to amorphous-to-crystalline transition along with the evolution of nano-sized particles in the microstructure. The crystalline phase was Ni-Ag solid solution. The relative volume fraction of the nano-sized particles increased gradually with time. There was however no detectable decomposition of solid solution phase till about 4 h of annealing. Beyond 4 h phase separation initiated and pure Ag and Ni phases formed in the film. This study provides a methodology by which microstructural engineering of as-electrodeposited amorphous Ag-Ni films can be conducted to isolate a particular microstructure in order to tap specific potentially usable functionalities. Staphylococcus aureus is a commensal gram positive bacteria which causes severe and non severe infections in humans and livestock. In India, ST772 is a dominant and ST672 is an emerging clone of Staphylococcus aureus. Both cause serious human diseases, and carry type V SCCmec elements. The objective of this study was to characterize SCCmec type V elements of ST772 and ST672 because the usual PCR methods did not amplify all primers specific to the type. Whole genome sequencing analysis of seven ST772 and one ST672 S. aureus isolates revealed that the SCCmec elements of six of the ST772 isolates were the smallest of the extant type V elements and in addition have several other novel features. Only one ST772 isolate and the ST672 isolate carried bigger SCCmec cassettes which were composites carrying multiple ccrC genes. These cassettes had some similarities to type V SCCmec element from M013 isolate (ST59) from Taiwan in certain aspects. SCCmec elements of all Indian isolates had an inversion of the mec complex, similar to the bovine SCCmec type X. This study reveals that six out of seven ST772 S. aureus isolates have a novel type V (5C2) SCCmec element while one each of ST772 and ST672 isolates have a composite SCCmec type V element (5C2&5) formed by the integration of type V SCCmec into a MSSA carrying a SCC element, in addition to the mec gene complex inversions and extensive recombinations.

Objectives Based on previous screening results, the cytotoxic effect of the hexane (JDH) and ethyl acetate extracts (JDE) of the marine sponge Jaspis diastra were evaluated on HeLa cells and the present study aimed at determining their possible mechanism of cell death. Methods Nuclear staining, membrane potential change, flow cytometry analysis of cell cycle distribution and annexin V staining were undertaken to investigate the effects of JDE and JDH. Electrospray ionization mass spectrometry (ESI-MS) and nuclear magnetic resonance were used to characterize an isolated bioactive molecule. Key findings JDE displayed an IC50 25 times more significant than the JDH. Flow cytometry analysis revealed JDE induced apoptosis in HeLa cells accompanied by the collapse of mitochondrial membrane potential. Fractionation of JDE resulted in the isolation of the known cytotoxic cyclodepsipeptide, Jaspamide. Conclusions Taking our results together suggest that JDE can be valuable for the development of anticancer drugs, especially for cervical cancer. Further investigations are currently in progress with the aim to determine and isolate other bioactive compounds from this extract.

Lactate dehydrogenase (LDH) of the malaria parasite, Plasmodium vivax (Pv), serves as a drug target and immunodiagnostic marker. The LDH cDNA generated from total RNA of a clinical isolate of the parasite was cloned into pRSETA plasmid. Recombinant his-tagged PvLDH was over-expressed in E. coli Rosetta2DE3pLysS and purified using Ni2+-NTA resin giving a yield of 25-30 mg/litre bacterial culture. The recombinant protein was enzymatically active and its catalytic efficiency for pyruvate was 5.4 x 10(8) min(-1) M-1, 14.5 fold higher than a low yield preparation reported earlier to obtain PvLDH crystal structure. The enzyme activity was inhibited by gossypol and sodium oxamate. The recombinant PvLDH was reactive in lateral flow immunochromatographic assays detecting pan- and vivax-specific LDH. The soluble recombinant PvLDH purified using heterologous expression system can facilitate the generation of vivax LDH-specific monoclonals and the screening of chemical compound libraries for PvLDH inhibitors.

Quantifying the isolated and integrated impacts of land use (LU) and climate change on streamflow is challenging as well as crucial to optimally manage water resources in river basins. This paper presents a simple hydrologic modeling-based approach to segregate the impacts of land use and climate change on the streamflow of a river basin. The upper Ganga basin (UGB) in India is selected as the case study to carry out the analysis. Streamflow in the river basin is modeled using a calibrated variable infiltration capacity (VIC) hydrologic model. The approach involves development of three scenarios to understand the influence of land use and climate on streamflow. The first scenario assesses the sensitivity of streamflow to land use changes under invariant climate. The second scenario determines the change in streamflow due to change in climate assuming constant land use. The third scenario estimates the combined effect of changing land use and climate over the streamflow of the basin. Based on the results obtained from the three scenarios, quantification of isolated impacts of land use and climate change on streamflow is addressed. Future projections of climate are obtained from dynamically downscaled simulations of six general circulation models (GCMs) available from the Coordinated Regional Downscaling Experiment (CORDEX) project. Uncertainties associated with the GCMs and emission scenarios are quantified in the analysis. Results for the case study indicate that streamflow is highly sensitive to change in urban areas and moderately sensitive to change in cropland areas. However, variations in streamflow generally reproduce the variations in precipitation. The combined effect of land use and climate on streamflow is observed to be more pronounced compared to their individual impacts in the basin. It is observed from the isolated effects of land use and climate change that climate has a more dominant impact on streamflow in the region. The approach proposed in this paper is applicable to any river basin to isolate the impacts of land use change and climate change on the streamflow.

With increasing energy demand, it necessitates to generate and transmit the electrical power with minimal losses. High voltage power transmission is the most economical way of transmitting bulk power over long distances. Transmission insulator is one of the main components used as a mechanical support and to electrically isolate the conductor from the tower. Corona from the hardware and conductors can significantly affect the performance of the polymeric insulators. In the present investigation a methodology is presented to evaluate the corona performance of the polymeric shed material under different environment conditions for both ac and dc excitation. The results of the comprehensive analysis on various polymeric samples and the power released from the corona electrode for both the ac and dc excitation are presented. Some interesting results obtained from the chemical analysis confirmed the presence of nitric acid species on the treated sample which in long term will affect the strength of the insulator, also the morphological changes were found to be varying for different experimental conditions. Despite high vulnerability, the impact of climate change on Himalayan ecosystem has not been properly investigated, primarily due to the inadequacy of observed data and the complex topography. In this study, we mapped the current vegetation distribution in Kashmir Himalayas from NOAA AVHRR and projected it under A1B SRES, RCP-4.5 and RCP-8.5 climate scenarios using the vegetation dynamics model-IBIS at a spatial resolution of 0.5A degrees. The distribution of vegetation under the changing climate was simulated for the 21st century. Climate change projections from the PRECIS experiment using the HADRM3 model, for the Kashmir region, were validated using the observed climate data from two observatories. Both the observed as well as the projected climate data showed statistically significant trends. IBIS was validated for Kashmir Himalayas by comparing the simulated vegetation distribution with the observed distribution. The baseline simulated scenario of vegetation (1960-1990), showed 87.15 % agreement with the observed vegetation distribution, thereby increasing the credibility of the projected vegetation distribution under the changing climate over the region. According to the model projections, grasslands and tropical deciduous forests in the region would be severely affected while as savannah, shrubland, temperate evergreen broadleaf forest, boreal evergreen forest and mixed forest types would colonize the area currently under the cold desert/rock/ice land cover types. The model predicted that a substantial area of land, presently under the permanent snow and ice cover, would disappear by the end of the century which might severely impact stream flows, agriculture productivity and biodiversity in the region.
