Cross-strand disulfides bridge two cysteines in a registered pair of antiparallel beta-strands. A nonredundant data set comprising 5025 polypeptides containing 2311 disulfides was used to study cross-strand disulfides. Seventy-six cross-strand disulfides were found of which 75 and 1 occurred at non-hydrogen-bonded (NHB) and hydrogen-bonded (HB) registered pairs, respectively. Conformational analysis and modeling studies demonstrated that disulfide formation at HB pairs necessarily requires an extremely rare and positive chi(1) value for at least one of the cysteine residues. Disulfides at HB positions also have more unfavorable steric repulsion with the main chain. Thirteen pairs of disulfides were introduced in NHB and HB pairs in four model proteins: leucine binding protein (LBP), leucine, isoleucine, valine binding protein (LIVBP), maltose binding protein (MBP), and Top7. All mutants LIVBP T247C V331C showed disulfide formation either on purification, or on treatment with oxidants. Protein stability in both oxidized and reduced states of all mutants was measured. Relative to wild type, LBP and MBP mutants were destabilized with respect to chemical denaturation, although the sole exposed NHB LBP mutant showed an increase of 3.1 degrees C in T-m. All Top7 mutants were characterized for stability through guanidinium thiocyanate chemical denaturation. Both exposed and two of the three buried NHB mutants were appreciably stabilized. All four HB Top7 mutants were destabilized (Delta Delta G(0) = -3.3 to -6.7 kcal/mol). The data demonstrate that introduction of cross-strand disulfides at exposed NHB pairs is a robust method of improving protein stability. A method for the preparation of acicular hydrogoethite (alpha -FeOOH.xH(2)O, 0.1 < x < 0.22) particles of 0.3-1 mm length has been optimized by air oxidation of Fe( II) hydroxide gel precipitated from aqueous (NH4)(2)Fe(SO4)(2) solutions containing 0.005-0.02 atom% of cationic Pt, Pd or Rh additives as morphology controlling agents. Hydrogoethite particles are evolved from the amorphous ferrous hydroxide gel by heterogeneous nucleation and growth. Preferential adsorption of additives on certain crystallographic planes thereby retarding the growth in the perpendicular direction, allows the particles to acquire acicular shapes with high aspect ratios of 8-15. Synthetic hydrogoethite showed a mass loss of about 14% at similar to 280 degreesC, revealing the presence of strongly coordinated water of hydration in the interior of the goethite crystallites. As evident from IR spectra, excess H2O molecules (0.1- 0.22 per formula unit) are located in the strands of channels formed in between the double ribbons of FeO6 octahedra running parallel to the c- axis. Hydrogoethite particles constituted of multicrystallites are formed with Pt as additive, whereas single crystallite particles are obtained with Pd (or Rh). For both dehydroxylation as well as H-2 reduction, a lower reaction temperature (similar to 220 degreesC) was observed for the former (Pt treated) compared to the latter (Pd or Rh) (similar to 260 degreesC). Acicular magnetite (Fe3O4) was prepared either by reducing hydrogoethite (magnetite route) or dehydroxylating hydrogoethite to hematite and then reducing it to magnetite (hematite- magnetite route). We report the temperature evolution of coherently excited acoustic and optical phonon dynamics in the superconducting iron pnictide single crystal Ca(Fe0.944Co0.056)(2)As-2 across the spin density wave transition at T-SDW similar to 85 K and the superconducting transition at T-SC similar to 20 K. The strain pulse propagation model applied to the generation of the acoustic phonons yields the temperature dependence of the optical constants, and longitudinal and transverse sound velocities in the temperature range from 3.1 K to 300 K. The frequency and dephasing times of the phonons show anomalous temperature dependence below T-SC indicating a coupling of these low-energy excitations with the Cooper-pair quasiparticles. A maximum in the amplitude of the acoustic modes at T similar to 170 is seen, attributed to spin fluctuations and strong spin-lattice coupling before T-SDW. Copyright (c) EPLA, 2012

We develop a communication theoretic framework for modeling 2-D magnetic recording channels. Using the model, we define the signal-to-noise ratio (SNR) for the channel considering several physical parameters, such as the channel bit density, code rate, bit aspect ratio, and noise parameters. We analyze the problem of optimizing the bit aspect ratio for maximizing SNR. The read channel architecture comprises a novel 2-D joint self-iterating equalizer and detection system with noise prediction capability. We evaluate the system performance based on our channel model through simulations. The coded performance with the 2-D equalizer detector indicates similar to 5.5 dB of SNR gain over uncoded data.

Solar radiation management (SRM) geoengineering has been proposed as a potential option to counteract climate change. We perform a set of idealized geoengineering simulations using Community Atmosphere Model version 3.1 developed at the National Center for Atmospheric Research to investigate the global hydrological implications of varying the latitudinal distribution of solar insolation reduction in SRM methods. To reduce the solar insolation we have prescribed sulfate aerosols in the stratosphere. The radiative forcing in the geoengineering simulations is the net forcing from a doubling of CO2 and the prescribed stratospheric aerosols. We find that for a fixed total mass of sulfate aerosols (12.6 Mt of SO4), relative to a uniform distribution which nearly offsets changes in global mean temperature from a doubling of CO2, global mean radiative forcing is larger when aerosol concentration is maximum at the poles leading to a warmer global mean climate and consequently an intensified hydrological cycle. Opposite changes are simulated when aerosol concentration is maximized in the tropics. We obtain a range of 1 K in global mean temperature and 3% in precipitation changes by varying the distribution pattern in our simulations: this range is about 50% of the climate change from a doubling of CO2. Hence, our study demonstrates that a range of global mean climate states, determined by the global mean radiative forcing, are possible for a fixed total amount of aerosols but with differing latitudinal distribution. However, it is important to note that this is an idealized study and thus not all important realistic climate processes are modeled.

Histones regulate a variety of chromatin templated events by their post-translational modifications (PTMs). Although there are extensive reports on the PTMs of canonical histones, the information on the histone variants remains very scanty. Here, we report the identification of different PTMs, such as acetylation, methylation, and phosphorylation of a major mammalian histone variant TH2B. Our mass spectrometric analysis has led to the identification of both conserved and unique modifications across tetraploid spermatocytes and haploid spermatids. We have also computationally derived the 3-dimensional model of a TH2B containing nucleosome in order to study the spatial orientation of the PTMs identified and their effect on nucleosome stability and DNA binding potential. From our nucleosome model, it is evident that substititution of specific amino acid residues in TH2B results in both differential histone-DNA and histone-histone contacts. Furthermore, we have also observed that acetylation on the N-terminal tail of TH2B weakens the interactions with the DNA. These results provide direct evidence that, similar to somatic H2B, the testis specific histone TH2B also undergoes multiple PTMs, suggesting the possibility of chromatin regulation by such covalent modifications in mammalian male germ cells.

The tropical easterly jet (TEJ) is a prominent atmospheric circulation feature observed during the Asian summer monsoon. It is generally assumed that sensible heating over the Tibetan Plateau directly influences the location of the TEJ. However, other studies have suggested the importance of latent heating in determining the jet location. In this paper, the relative importance of latent heating on the maintenance of the TEJ is explored through simulations with a general circulation model. The simulation of the TEJ by the Community Atmosphere Model, version 3.1 is discussed in detail. These simulations showed that the location of the TEJ is well correlated with the location of the precipitation. Significant zonal shifts in the location of the precipitation resulted in similar shifts in the zonal location of the TEJ. These zonal shifts had minimal effect on the large-scale structure of the jet. Further, provided that precipitation patterns were relatively unchanged, orography did not directly impact the location of the TEJ. These changes were robust even with changes in the cumulus parameterization. This suggests the potential important role of latent heating in determining the location and structure of the TEJ. These results were used to explain the significant differences in the zonal location of the TEJ in the years 1988 and 2002. To understand the contribution of the latitudinal location of latent heating on the strength of the TEJ, aqua-planet simulations were carried out. It has been shown that for similar amounts of net latent heating, the jet is stronger when heating is in the higher tropical latitudes. This may partly explain the reason for the jet to be very strong during the JJA monsoon season.
