Chitosan grafted poly(alkyl methacrylate)s (namely chitosan grafted poly(methyl methacrylate) (ChgPMMA), chitosan grafted poly(ethyl methacrylate)(ChgPEMA), chitosan grafted poly(butyl methacrylate) (ChgPBMA) and chitosan grafted poly(hexyl methacrylate) (ChgPHMA)) were synthesized and characterized by using FT-IR and C-13 NMR techniques. The adsorption batch experiments on these grafted copolymers were conducted by using an anionic sulfonated dye. Orange-G. A pseudo-second-order kinetic model was used to determine the kinetics of adsorption. The effect of grafting, effect of process variables and the effect of different sulfonated anionic dyes (Orange-C, Congo Red, Remazol Brill Blue R and Methyl Blue) on the adsorption kinetics was determined. The Langmuir and Freundlich models were used to fit the adsorption isotherms and from the values of correlation coefficients (R-2), it was observed that the experimental data fits very well to the Langmuir model. The values of the maximum adsorption capacity of the adsorbents follow the order: ChgPMMA > ChgPEMA > ChgPBMA > ChgPHMA > chitosan. (C) 2010 Elsevier B.V. All rights reserved.

The seeds of Lathyrus sativus contain the unusual amino acid homoarginine. The possible breakdown of homoarginine to lysine and urea has been investigated with enzyme extracts prepared from the seedlings of L. sativus. The results indicate that there is no separate homoarginase enzyme but that the arginase present has about 5 per cent activity towards Image -homoarginine as compared to that obtained with Image -arginine. The enzyme does not show an absolute dependence on Mn2+ for activity and maximal activation of the enzyme has been realized with Fe3+. It is concluded that the breakdown of homoarginine through the urea cycle may only represent a minor pathway for the catabolism of this compound in this plant.

Sesbania mosaic virus (SeMV),a single-strand positive-sense RNA plant virus, belongs to the genus Sobemoviruses. Mechanism of replication in Sobemoviruses is poorly understood. In the present study, SeMV RNA-dependent RNA polymerase (RdRp) was overexpressed and purified as a thioredoxin-tagged protein. The recombinant SeMV RdRp could synthesize RNA from genomic or subgenomic RNA templates, even in the absence ofthe protein primer, VPg. Analysis of the product indicated that it was double-stranded and that the mode of initiation was de novo. Mutational analysis of the 3' UTR of subgenomic RNA revealed that a stem-loop structure at the 3' end was important. Further, analysis of this stem-loop showed that the SeMV RdRp was capable of recognizing stem-loop structures of various lengths and forms. These results demonstrate that the SeMV RdRp is capable of primer-independent RNAsynthesis in vitro. (C) 2010 Elsevier Inc. All rights reserved.

Chloramine-T has been found to bring about the rupture of S-S link in polythionates in acid medium and oxidise all the sulphur present in the chain into sulphuric acid. Quantitative estimation of a polythionate may be made on the basis of this oxidation reaction.

Traditionally, laminar separation bubbles have been characterised as being 'long' or 'short' on the basis of a two parameter 'bursting' criterion involving a pressure gradient parameter and Reynolds Number at separation. In the present work we suggest a refined bursting criterion, which takes into account not just the length of the bubble but also the maximum height of the bubble, thereby shedding some light on the less understood phenomenon of 'bursting' in laminar separation bubbles.

We report data from two related assay systems (isolated enzyme assays and whole blood assays) that C-phycocyanin a biliprotein from Spirulina platensis is a selective inhibitor of cyclooxygenase-a (COX-2) with a very low IC50 COX-2/IC50 COX-1 ratio (0.04). The extent of inhibition depends on the period of preincubation of phycocyanin with COX-2, but without any effect on the period of preincubation with COX-1. The IC50 value obtained for the inhibition of COX-2 by phycocyanin is much lower (180 nM) as compared to those of celecoxib (255 nM) and rofecoxib (401 nM), the well-known selective COX-2 inhibitors. In the human whole blood assay, phycocyanin very efficiently inhibited COX-2 with an IC50 value of 80 nM. Reduced phycocyanin and phycocyanobilin, the chromophore of phycocyanin are poor inhibitors of COX-2 without COX-2 selectivity. This suggests that apoprotein in phycocyanin plays a key role in the selective inhibition of COX-2. The present study points out that the hepatoprotective, anti-inflammatory, and anti-arthritic properties of phycocyanin reported in the literature may be due, in part, to its selective COX-2 inhibitory property, although its ability to efficiently scavenge free radicals and effectively inhibit lipid peroxidation may also be involved. Silver salts of hexafluorophosphates, tetrafluoro-borates and hexafluorosilicates have been prepared by a metathetic reaction between the respective ammonium salts and silver nitrate in acetonitrile medium. This one step procedure at room temperature offers salts of high purity in good yields. The salts (AgpF6, AgBF4 and Ag2SiF6) have been characterised by IR spectral data analysis and chemical analysis.

In eubacteria, ribosome recycling factor (RRF) and elongation factor G (EFG) function together to dissociate posttermination ribosomal complexes. Earlier studies, using heterologous factors from Mycobacterium tuberculosis in Escherichia coli revealed that specific interactions between RRF and EFG are crucial for their function in ribosome recycling. Here, we used translation factors from E.coli,Mycobacterium smegmatis and M. tuberculosis, and polysomes from E. coli and M. smegmatis, and employed in vivo and in vitro experiments to further understand the role of EFG in ribosome recycling. We show thatE. coli EFG (EcoEFG) recycles E. coli ribosomes with E. coli REF (EcoRRF), but not with mycobacterial RRFs. Also, EcoEFG fails to recycle M. smegmatis ribosomes with either EcoRRF or mycobacterial RRFs. On the other hand, mycobacterial EFGs recycle both E. coli and M. smegmatis ribosomes with either of the RRFs. These observations suggest that EFG establishes distinct interactions with REF and the ribosome to carry out ribosome recycling. Furthermore, the EFG chimeras generated by swapping domains betweenmycobacterial EFGs and EcoEFG suggest that while the residues needed to specify the EFG interaction with REF arelocated in domains IV and V. those required to specify its interaction with the ribosome are located throughout the molecule. Hydrogen plasma can be used for deoxidation of functional materials containing reactive metals in both bulk and thin film forms. Since the different species in the plasma are not in thermodynamic equilibrium, application of classical thermodynamics to the analysis of such a system is associated with some difficulties. While global equilibrium approaches have been tried, with and without additional approximations or constraints, there is some ambiguity in the results obtained. Presented in this article is the application of a local equilibrium concept to assess the thermodynamic limit of the reaction of each species present in the gas with oxides or oxygen dissolved in metals. Each reaction results in a different pal tial pressure of H2O. Because of the higher reactivity of the dissociated and ionized species and the larger thermodynamic driving force for reactions involving these species, they act as powerful reducing agents. It is necessary to remove the products of reaction from the plasma to prevent back reaction and gradual approach to global equilibrium. A quantitative description using the framework of the Ellingham-Richardson-Jeffes diagrams is presented.

The asymmetric unit contains one nucleoside molecule with a disordered solvent Me2S_O molecule. The geometry about the C(4')-C(5') bond is gauche-gauche. The guanosine base is in the anti conformation with the furanose ring having C(3')-exo (E 3) puckering. The bases do not show any stacking in contrast to other guanosine-containing structures. The crystal structure is stabilized by N--H...N and N--H...O hydrogen bonding.

An examination of radiation-damage processes consequent to high-energy irradiation in certain ammonium salts studied using ESR of free radical together with the structural information available from neutron diffraction studies shows that, other factors being equal/nearly equal, symmetry-related bonds are preserved in preference to those unrelated to one another by any symmetry.

A simple and rapid affinity chromatographic method for the isolation of aspartate transcarbamylase from germinated seedlings of mung bean (Phaseolus aureus) was developed. A partially purified preparation of the enzyme was chromatographed on an affinity column containing aspartate linked to CNBr-activated Sepharose 4B. Aspartate transcarbamylase was specifically eluted from the column with 10 mImage aspartate or 0.5 Image KCl. The enzyme migrated as a single sharp band during disc electrophoresis at pH 8.6 on polyacrylamide gels. Electrophoresis of the sodium dodecyl sulfate-treated enzyme showed two distinct protein bands, suggesting that the mung bean aspartate transcarbamylase was made up of nonidentical subunits. Like the enzyme purified by conventional procedures, this enzyme preparation also exhibited positive homotropic interactions with carbamyl phosphate and negative heterotropic interactions with UMP. This method was extended to the purification of aspartate transcarbamylase from Lathyrus sativus, Eleucine coracona, and Trigonella foenum graecum.
