Tutkimuksen tavoitteena oli selvittää suomalaisten viininkuluttajien luomuviinien valintaa sekä niihin liitettyjä mielikuvia. Erityisen kiinnostuneita oltiin luomuviinien koetusta laadusta sekä siitä, mitkä mielikuvat ja motiivit edistävät ja mitkä estävät luomuviinien valintaa. Teoriaosassa tarkasteltiin viinimarkkinoita kansainvälisellä sekä kansallisella tasolla, viinintuotannon vaikutuksia ympäristöön sekä ympäristöystävällisyyden merkitsemistä. Ympäristöystävällisiin viineihin luettiin tässä tutkimuksessa luomuviinit, biodynaamiset viinit ja viinit, joiden tuotannossa noudatetaan kestävän kehityksen periaatteita. Päähuomio tutkimuksessa oli Alkon luomuhyllyntunnuksilla merkityissä luonnonmukaisista rypäleistä viljellyillä viineillä, joita arkikielessä kutsutaan luomuviineiksi. Tutkimus suoritettiin kvantitatiivisiin tutkimusmenetelmiin kuuluvalla kyselylomakkeella. Teoriaosion kirjallisuus antoi viitekehyksen, jonka pohjalta empiirinen tutkimus toteutettiin. Aineisto kerättiin pääkaupunkiseudulla kolmessa eri Alkon myymälässä, ja aineisto koostuu 145 viiniä kuluttavasta vastaajasta. Tutkimuksen tulosten mukaan luomuviiniä suositaan valinnoissa, mutta valintaan tai hylkäämiseen johtavat mielikuvat eivät ole yksiselitteisiä. Valintaan johtavia motiiveja ja mielikuvia oli helpompi löytää kuin hylkäämiseen johtavia. Kokeilunhalu / mielenkiinto, ympäristönäkökulmat sekä puhtauteen ja terveyteen liittyvät mielikuvat olivat motiiveja, jotka tässä tutkimuksessa edistivät eniten luomuviinien valintaa. Luomuviinien laatu ja hinnat koettiin tasavertaiseksi suhteessa tavanomaisin viineihin. Luomuviinien laatu liittyi erityisesti luonnollisuuteen ja puhtauteen. Markkinoinnin ja viestinnän keinoin on mahdollista lisätä kuluttajien tietämystä luomuviineistä, ja siten vaikuttaa niihin liitettyihin mielikuviin.

In Finland, barley, Hordeum vulgare L., covers 50 % of the total acreage devoted to cereal cultivation. The most common disease of barley in Finland is net blotch, a foliar disease caused by the ascomycete Pyrenophora teres Drechsler. Disease resistance based on plant genes is an environmentally friendly and economical way to manage plant diseases caused by biotic stresses. Development of a disease resistance breeding programme is dependent on knowledge of the pathogen. In addition to information on the epidemiology and virulence of a pathogen, knowledge on how the pathogen evolves and the nature of the risks that might arise in the future are essential issues that need to be taken into account to achieve the final breeding aims. The main objectives of this study were to establish reliable and efficient testing methods for Pyrenophora teres f. teres virulence screening, and to understand the role of virulence of P. teres f. teres in Finland from a disease resistance breeding point of view. The virulence of P. teres was studied by testing 239 Finnish P. teres f. teres isolates collected between 1994 2007 originating from 19 locations, and 200 P. teres progeny isolates originating from artificially produced P. teres matings. According to the results of this study, screening for P. teres f. teres isolates on barley seedlings under greenhouse conditions is a feasible and cost efficient method to describe the virulence spectrum of the pathogen. Inoculum concentration and the seedling leaf used to gauge virulence had significant effects. Barley grain size, morphological traits of P. teres isolates, spore production and growth rate on agar did not affect the expression of virulence. A common barley differential set to characterize the P. teres virulence was developed and is recommended to be used globally. The virulence spectrum of Finnish P. teres f. teres isolates collected in 1994-2007 was constant both within and between the years. The results indicated differences in the pathogen s aggressiveness and in barley genotypes resistance. However, differences in virulence were rarely significant. Unlike in laboratory conditions, no indications of changes in virulence caused by the sexual reproduction have been observed in Finnish barley fields. In Finland, durable net blotch resistance has been achieved by introducing resistance from other barley varieties using traditional crossing methods, including wide crossing, and testing the breeding material at early generations at several sites under natural infection pressure. Novel resistance is available, which is recommended to minimize the risk of selection of virulent isolates and breakdown of currently deployed resistance.

Standards have been placed to regulate the microbial and preservative contents to assure that foods are safe to the consumer. In a case of a food-related disease outbreak, it is crucial to be able to detect and identify quickly and accurately the cause of the disease. In addition, for every day control of food microbial and preservative contents, the detection methods must be easily performed for numerous food samples. In this present study, quicker alternative methods were studied for identification of bacteria by DNA fingerprinting. A flow cytometry method was developed as an alternative to pulsed-field gel electrophoresis, the golden method . DNA fragment sizing by an ultrasensitive flow cytometer was able to discriminate species and strains in a reproducible and comparable manner to pulsed-field gel electrophoresis. This new method was hundreds times faster and 200,000 times more sensitive. Additionally, another DNA fingerprinting identification method was developed based on single-enzyme amplified fragment length polymorphism (SE-AFLP). This method allowed the differentiation of genera, species, and strains of pathogenic bacteria of Bacilli, Staphylococci, Yersinia, and Escherichia coli. These fingerprinting patterns obtained by SE-AFLP were simpler and easier to analyze than those by the traditional amplified fragment length polymorphism by double enzyme digestion. Nisin (E234) is added as a preservative to different types of foods, especially dairy products, around the world. Various detection methods exist for nisin, but they lack in sensitivity, speed or specificity. In this present study, a sensitive nisin-induced green fluorescent protein (GFPuv) bioassay was developed using the Lactococcus lactis two-component signal system NisRK and the nisin-inducible nisA promoter. The bioassay was extremely sensitive with detection limit of 10 pg/ml in culture supernatant. In addition, it was compatible for quantification from various food matrices, such as milk, salad dressings, processed cheese, liquid eggs, and canned tomatoes. Wine has good antimicrobial properties due to its alcohol concentration, low pH, and organic content and therefore often assumed to be microbially safe to consume. Another aim of this thesis was to study the microbiota of wines returned by customers complaining of food-poisoning symptoms. By partial 16S rRNA gene sequence analysis, ribotyping, and boar spermatozoa motility assay, it was identified that one of the wines contained a Bacillus simplex BAC91, which produced a heat-stable substance toxic to the mitochondria of sperm cells. The antibacterial activity of wine was tested on the vegetative cells and spores of B. simplex BAC91, B. cereus type strain ATCC 14579 and cereulide-producing B. cereus F4810/72. Although the vegetative cells and spores of B. simplex BAC91 were sensitive to the antimicrobial effects of wine, the spores of B. cereus strains ATCC 14579 and F4810/72 stayed viable for at least 4 months. According to these results, Bacillus spp., more specifically spores, can be a possible risk to the wine consumer.
Development of Sample Pretreatment and Liquid Chromatographic Techniques for Antioxidative Compounds

In this study, novel methodologies for the determination of antioxidative compounds in herbs and beverages were developed. Antioxidants are compounds that can reduce, delay or inhibit oxidative events. They are a part of the human defense system and are obtained through the diet. Antioxidants are naturally present in several types of foods, e.g. in fruits, beverages, vegetables and herbs. Antioxidants can also be added to foods during manufacturing to suppress lipid oxidation and formation of free radicals under conditions of cooking or storage and to reduce the concentration of free radicals in vivo after food ingestion. There is growing interest in natural antioxidants, and effective compounds have already been identified from antioxidant classes such as carotenoids, essential oils, flavonoids and phenolic acids. The wide variety of sample matrices and analytes presents quite a challenge for the development of analytical techniques. Growing demands have been placed on sample pretreatment. In this study, three novel extraction techniques, namely supercritical fluid extraction (SFE), pressurised hot water extraction (PHWE) and dynamic sonication-assisted extraction (DSAE) were studied. SFE was used for the extraction of lycopene from tomato skins and PHWE was used in the extraction of phenolic compounds from sage. DSAE was applied to the extraction of phenolic acids from Lamiaceae herbs. In the development of extraction methodologies, the main parameters of the extraction were studied and the recoveries were compared to those achieved by conventional extraction techniques. In addition, the stability of lycopene was also followed under different storage conditions. For the separation of the antioxidative compounds in the extracts, liquid chromatographic methods (LC) were utilised. Two novel LC techniques, namely ultra performance liquid chromatography (UPLC) and comprehensive two-dimensional liquid chromatography (LCxLC) were studied and compared with conventional high performance liquid chromatography (HPLC) for the separation of antioxidants in beverages and Lamiaceae herbs. In LCxLC, the selection of LC mode, column dimensions and flow rates were studied and optimised to obtain efficient separation of the target compounds. In addition, the separation powers of HPLC, UPLC, HPLCxHPLC and HPLCxUPLC were compared. To exploit the benefits of an integrated system, in which sample preparation and final separation are performed in a closed unit, dynamic sonication-assisted extraction was coupled on-line to a liquid chromatograph via a solid-phase trap. The increased sensitivity was utilised in the extraction of phenolic acids from Lamiaceae herbs. The results were compared to those of achieved by the LCxLC system.

This research has been prompted by an interest in the atmospheric processes of hydrogen. The sources and sinks of hydrogen are important to know, particularly if hydrogen becomes more common as a replacement for fossil fuel in combustion. Hydrogen deposition velocities (vd) were estimated by applying chamber measurements, a radon tracer method and a two-dimensional model. These three approaches were compared with each other to discover the factors affecting the soil uptake rate. A static-closed chamber technique was introduced to determine the hydrogen deposition velocity values in an urban park in Helsinki, and at a rural site at Loppi. A three-day chamber campaign to carry out soil uptake estimation was held at a remote site at Pallas in 2007 and 2008. The atmospheric mixing ratio of molecular hydrogen has also been measured by a continuous method in Helsinki in 2007 - 2008 and at Pallas from 2006 onwards. The mean vd values measured in the chamber experiments in Helsinki and Loppi were between 0.0 and 0.7 mm s-1. The ranges of the results with the radon tracer method and the two-dimensional model were 0.13 - 0.93 mm s-1 and 0.12 - 0.61 mm s-1, respectively, in Helsinki. The vd values in the three-day campaign at Pallas were 0.06 - 0.52 mm s-1 (chamber) and 0.18 - 0.52 mm s-1 (radon tracer method and two-dimensional model). At Kumpula, the radon tracer method and the chamber measurements produced higher vd values than the two-dimensional model. The results of all three methods were close to each other between November and April, except for the chamber results from January to March, while the soil was frozen. The hydrogen deposition velocity values of all three methods were compared with one-week cumulative rain sums. Precipitation increases the soil moisture, which decreases the soil uptake rate. The measurements made in snow seasons showed that a thick snow layer also hindered gas diffusion, lowering the vd values. The H2 vd values were compared to the snow depth. A decaying exponential fit was obtained as a result. During a prolonged drought in summer 2006, soil moisture values were lower than in other summer months between 2005 and 2008. Such conditions were prevailing in summer 2006 when high chamber vd values were measured. The mixing ratio of molecular hydrogen has a seasonal variation. The lowest atmospheric mixing ratios were found in the late autumn when high deposition velocity values were still being measured. The carbon monoxide (CO) mixing ratio was also measured. Hydrogen and carbon monoxide are highly correlated in an urban environment, due to the emissions originating from traffic. After correction for the soil deposition of H2, the slope was 0.49±0.07 ppb (H2) / ppb (CO). Using the corrected hydrogen-to-carbon-monoxide ratio, the total hydrogen load emitted by Helsinki traffic in 2007 was 261 t (H2) a-1. Hydrogen, methane and carbon monoxide are connected with each other through the atmospheric methane oxidation process, in which formaldehyde is produced as an important intermediate. The photochemical degradation of formaldehyde produces hydrogen and carbon monoxide as end products. Examination of back-trajectories revealed long-range transportation of carbon monoxide and methane. The trajectories can be grouped by applying cluster and source analysis methods. Thus natural and anthropogenic emission sources can be separated by analyzing trajectory clusters.

Tutkimuksen tavoitteena oli tutkia, millaisia vaikutuksia sanoma- ja aikakausilehdissä julkaistuilla viiniarvosteluilla on kyseisten arvosteltujen viinien myyntiin. Voidaanko siis suhdetoiminnan avulla viinikriitikoiden kautta vaikuttaa kuluttajan ostokäyttäytymiseen? Tutkimuskysymystä lähestyttiin tarkastelemalla ensin kuluttajan ostokäyttäytymistä ja ostoprosessin eri vaiheita. Samalla käytiin läpi myös kuluttajan sitoutumista ja tuotteen oston yhteydessä ilmeneviä riskejä. Sen jälkeen perehdyttiin suhdetoimintaan, jonka avulla pyritään vaikuttamaan siihen, mitä yrityksestä ja sen tuotteista kirjoitetaan medioissa. Empiirisessä osassa tehtiin tilastollinen tutkimus, jossa tutkittiin, onko eri sanoma- ja aikakausilehtien viiniarvosteluilla vaikutusta arvostellun viinin myyntiin. Lisäksi tutkittiin, löytyykö arvosteluista yhteisiä tekijöitä, jotka ovat vaikuttaneet myynnin muutokseen. Aineisto kerättiin kahden suomalaisen viinien maahantuonti- ja markkinointiyrityksen viineistä kirjoitetuista arvosteluista ja näiden kyseisten arvosteltujen viinien myyntiluvuista. Arvosteluja kerättiin helmikuun 18. päivän ja lokakuun 19. päivän 2010 väliseltä ajalta. Yhteensä hyväksyttyjä arvosteluja kertyi 90 kappaletta. Tutkimuksen analysointiin käytettiin regressioanalyysiä. Tämän tutkimusten tulosten mukaan viiniarvosteluilla näyttäisi olevan vaikutusta viinien myyntiin eli kuluttajien ostopäätökseen. Teoriaosioon peilaten tämä oli odotettavissa, sillä myös aikaisemmat tutkimukset ovat osoittaneet mielipidejohtajien ja viinikriitikoiden positiivisen vaikutuksen kuluttajiin. Kuluttajat kokevat viinin ostopäätöksen yhteydessä riskiä, jota he pyrkivät välttämään viiniarvostelujen avulla. Tuloksien mukaan myös arvostelun julkaisseella sanoma- tai aikakausilehdellä on vaikutusta kuluttajan ostopäätökseen.
