Streptococcus agalactiae –juverinflammation var tidigare ett stort problem i många länder, inklusive Finland. I och med förbättrad mjölkningshygien och antibiotikabehandlingar har bakterien så gott som eradikerats från mjölkbesättningarna. Nu verkar bakterien i viss mån ha kommit tillbaka till våra stora mjölkgårdar. Avhandlingens experimentella del utfördes på en mjölkgård, med ca 180 mjölkande och tre mjölkningsrobotar, som haft problem med Str. agalactiae. Man hoppades utreda hur stort problemet på gården var samt möjliga smittovägar. Man undersökte också möjligheten att använda mjölkningsrobotens automatiska provtagningsutrustning för provtagning av bakteriella prov. PCRmetoden jämfördes med konventionell odling vid diagnostik av juverinflammationer orsakade av Str. agalactiae. På gården gick man igenom anteckningar samt hälso- och seminkort för att få en bild över situationen. Man gjorde en uppföljning av mjölkningen för tolv kor vid den ena mjölkningsroboten. Man tog 47 stycken kospecifika mjölkprov samt ett prov från mjölktanken. Mjölkprov i tre serier både mjölkade för hand och direkt från mjölkuppsamlaren på mjölkningsroboten togs. Man tog sammanlagt 23 renlighetsprov från mjölkningsroboten, tre från den automatiska provtagningsutrustningen samt två från djurskötarnas händer. Från den automatiska provtagningsutrustningen togs även ett genomsköljningsprov. Av mjölkprov som tidigare tagits på gården hade man hittat Str. agalactiae i ca 17%. I denna studie hittades Str. agalactiae i tre kospecifika mjölkprov, vilket motsvarar en prevalens på ca 2%. Vid uppföljningen av mjölkningarna upptäcktes inget alarmerande, men spenarnas hälsa samt tommjölkningar är något som bör följas upp. Av renlighetsproven hittades Str. agalactiae i ett prov taget från borsthållaren. Svaren från mjölkproven tagna i serier tyder på att den automatiska provtagningsutrustningen inte går att använda till bakteriella prov, eftersom mjölken från en Str. agalactiae –infekterad ko verkar påverka resultatet också hos följande kor. Resultatet är väntat, eftersom mjölkprov alltid skall tas aseptiskt och det går inte med den automatiska provtagningsutrustningen så som den i dagsläget är utvecklad. Från sju av nio mjölkprov, där man hittat Str. agalactiae med PCR-metoden, hittades bakterien också med konventionell odling. Från tankmjölksprovet kunde man inte hitta Str. agalactiae med konventionell odling. PCR-metoden verkar enligt den här studien vara mer känslig att upptäcka Str. agalactiae jämfört med konventionell odling.

Earlier school text book studies (eg Tainio and Teräs 2010, Blumberg 2007; Ohlander 2010) have shown that women are underrepresented in school books, both in illustrations and texts, and the genders are represented stereotyped. The study will examine how gender in seen on pre-school education materials. The aim of this study is to determine what kind of representations, discourses and the subject positions of the gender are presented in pre-school materials. This study utilizes a feminist research method. The theoretical starting points are the social constructivist, poststructuralist theory and gender studies. The concept of gender as a social construct. The research used content analysis as well as discource analysis and deconstructive reading. The material was used four different publishers, WSOY, Tammi, Otava and Lasten Keskus preschool integrated material packages, which contain the child's exercise book or booklet, and teacher's guide. The analysis examined the quantity of gender-specific images and gendered words and phrases, and representations of gender, subject position, and discourses, and what linguistic means had been used for representation of masculinity and femininity. Based on the results there were on average more masculine characters and words as feminine in the illustrations and stories of pre-school materials. Feminine and masculine characters representations emphasized traditional gender stereotypes, especially in external characteristics and clothing. Genders had the highest available, with the subject position of stereotypes with reduced mobility, but also other kinds of subject position was observed. The data found in the following gender discources: difference discource, diversity discource and similarity discource. The highest number occurred in the difference and diversity discources. However, there were differences between the different materials. In some materials there were more diverse gender representations and other materials highlighted the differences between genders. Overall, the genders were represented stereotypically in the pre-school materials.

The bacterial genus Stenotrophomonas comprises 12 species. They are widely found throughout the environment and particularly S. maltophilia, S. rhizophila and S. pavanii are closely associated with plants. Strains of the most common Stenotrophomonas species, S. maltophilia, promote plant growth and health, degrade natural and man-made pollutants and produce biomolecules of biotechnological and economical value. Many S. maltophilia –strains are also multidrug resistant and can act as opportunistic human pathogens. During an INCO-project (1998-2002) rhizobia were collected from root nodules of the tropical leguminous tree Calliandra calothyrsus Meisn. from several countries in Central America, Africa and New Caledonia. The strains were identified by the N2-group (Helsinki university) and some strains turned out to be members of the genus Stenotrophomonas. Several Stenotrophomonas strains induced white tumor- or nodule-like structures on Calliandra?s roots in plant experiments. The strains could, besides from root nodules, also be isolated from surface sterilized roots and stems. The purpose of my work was to investigate if the Stenotrophomonas strains i) belong to a new Stenotrophomonas species, ii) have the same origin, iii) if there are other differences than colony morphology between phase variations of the same strain, iv) have plant growth-promoting (PGP) activity or other advantageous effects on plants, and v) like rhizobia have ability to induce root nodule formation. The genetic diversity and clustering of the Stenotrophomonas strains were analyzed with AFLP fingerprinting to get indications about their geographical origin. Differences in enzymatic properties and ability to use different carbon and energy sources were tested between the two phases of each strain with commercial API tests for bacterial identification. The ability to infect root hairs and induce root nodule formation was investigated both using plant tests with the host plant Calliandra and PCR amplification of nodA and nodC genes for nodulation. The PGP activity of the strains was tested in vitro mainly with plate methods. The impact on growth, nitrogen content and nodulation in vivo was investigated through greenhouse experiments with the legumes Phaseolus vulgaris and Galega orientalis. Both the genetic and phenotypic diversity among the Stenotrophomonas strains was small, which proposes that they have the same origin. The strains brought about changes on the root hairs of Calliandra and they also increased the amount of root hairs. However, no root nodules were detected. The strains produced IAA, protease and lipase in vitro. They also showed plant a growth-promoting effect on G. orientalis, both alone and together with R. galegae HAMBI 540, and also activated nodulation among efficient rhizobia on P. vulgaris in greenhouse. It requires further research to get a better picture about the mechanisms behind the positive effects. The results in this thesis, however, confirm earlier studies concerning Stenotrophomonas positive impact on plants.
