A transformation technique for the introduction of transgenes to control blackheart by particle bombardment has been developed for pineapple cv. Smooth Cayenne. Leaf callus cultures capable of high frequency organogenesis with a short regeneration time were used as explant material. Gus and gfp reporter genes were used to observe and determine transient and stable expression. The ppo gene, isolated from pineapple, was introduced to control blackheart. Co-transformation occurred with constructs containing the nptII gene conferring geneticin resistance. We have recovered 15 independent transgenic gus and gfp lines each from 8 separate experiments and 22 ppo lines from 11 experiments. Gus, gfp, ppo and nptII positive plants have been regenerated, which have been shown by Southern blot analysis to be stable transgenics containing multiple copies of the introduced genes. These results show that biolistic gene delivery in pineapple can be successfully achieved at an acceptable efficiency of 0.21-1.5% for genetic improvement of 'Smooth Cayenne', the industry standard throughout the world.

Two new cercosporoid hyphomycetes from Vanuatu, Cercospora sphathulata and Pseudocercospora pometiae, are described and illustrated. Six new records of cercosporoid hyphomycetes that cause plant diseases from New Caledonia, Samoa and Vanuatu are given. Four new hosts of Cercospora apii s. lat. from the region are reported.

Growing agricultural crops in wide row spacings has been widely adopted to conserve water, to control pests and diseases, and to minimise problems associated with sowing into stubble. The development of herbicide resistance combined with the advent of precision agriculture has resulted in a further reason for wide row spacings to be adopted: weed control. Increased row spacing enables two different methods of weed control to be implemented with non-selective chemical and physical control methods utilised in the wide inter-row zone, with or without selective chemicals used on the on-row only. However, continual application of herbicides and tillage on the inter-row zone brings risks of herbicide resistance, species shifts and/or changes in species dominance, crop damage, increased costs, yield losses, and more expensive weed management technology.

Open-pollination: originated as a chance seedling from Z44 (maternal clonal parent), obtained from Beltsville MD in 1981, with an unknown pollen source from a zoysia grass germplasm field nursery at the Texas Agricultural Experiment Station in Dallas. ‘Palisades’ was selected over the parent Z44 on the basis of its lower tendency to produce thatch, its excellent lateral growth habit and its superior mowing qualities. ‘Palisades’ has been vegetatively propagated, and is uniform in growth expression. No seedling establishment from ‘Palisades’ has been noticed in either greenhouse or field studies. Selection criteria: rapid regrowth and spread by, and/or from, stolons and rhizomes; turf colour and density; tolerance to low mowing; winter hardiness; shade tolerance; low water use requirements. This study examined whether element: Ca ratios within the otoliths of juvenile brown trout could provide accurate trace element signatures for specific natal tributaries, and attempted to match these to trace element natal signatures found within the otoliths of adult trout caught in the main stem rivers of the same catchment. The trace element signatures of juvenile trout otoliths were analysed from a sample of eight tributaries representing the main sub-catchments of the Motueka River catchment, New Zealand. Trace element signatures were determined using laser ablation inductively coupled plasma mass spectrometry, and differentiated using linear discriminant function analysis with an overall cross-validated classification success of 96.8%. Temporal stability in element: Ca ratios was investigated by repeat collections of juvenile fish over two years. Natal signatures from 11 of 23 adult trout sampled from the catchment main stems were matched to one of the eight tributary signatures showing recruitment sources to be spread relatively evenly throughout the catchment. This study demonstrates the potential of using otolith trace element analysis to determine the natal origins of freshwater fish within a catchment.
