Chromosome sorting by flow cytometry is the main source of chromosome-specific DNA for the production of painting probes. These probes have been used for cross-species in situ hybridization in the construction of comparative maps, in the study of karyotype evolution and phylogenetics, in delineating territories in interphase nuclei, and in the analysis of chromosome breakpoints. We review here the contributions that this technology has made to the analysis of primate genomes. Copyright (C) 2005 S. Karger AG, Basel.

As one of the most powerful tools in biomedical research, DNA sequencing not only has been improving its productivity in an exponential growth rate but also been evolving into a new layout of technological territories toward engineering and physical disciplines over the past three decades. In this technical review, we look into technical characteristics of the next-gen sequencers and provide prospective insights into their future development and applications. We envisage that some of the emerging platforms are capable of supporting the $1000 genome and $100 genome goals if given a few years for technical maturation. We also suggest that scientists from China should play an active role in this campaign that will have profound impact on both scientific research and societal healthcare systems.

An extensive literature survey of over 17 Journals was carried out on Chinese sponges and their natural products in the period from 1980 to 2001. This review is thus intended to provide the first thorough overview of research on marine sponges from China Ocean territories. Information is provided about the rather-limited taxonomic study of Chinese marine sponges, with an analysis on their distribution and diversity. Research findings on the natural products and their bioactivity screening from Chinese sponges are summarized. The weaknesses, gaps and problems in the past R&D program of Chinese sponges are identified, which point to the future opportunities in exploiting these huge untapped sponge resources. The report is expected to serve as an entry point for understanding Chinese sponges and for furthering R&D on their bioactive compounds for new drug development. (C) 2003 Elsevier Science B.V. Empirical Orthogonal Function (EOF) analysis is used in this study to generate main eigenvector fields of historical temperature for the China Seas (here referring to Chinese marine territories) and adjacent waters from 1930 to 2002 (510 143 profiles). A good temperature profile is reconstructed based on several subsurface in situ temperature observations and the thermocline was estimated using the model. The results show that: 1) For the study area, the former four principal components can explain 95% of the overall variance, and the vertical distribution of temperature is most stable using the in situ temperature observations near the surface. 2) The model verifications based on the observed CTD data from the East China Sea (ECS), South China Sea (SCS) and the areas around Taiwan Island show that the reconstructed profiles have high correlation with the observed ones with the confidence level > 95%, especially to describe the characteristics of the thermocline well. The average errors between the reconstructed and observed profiles in these three areas are 0.69A degrees C, 0.52A degrees C and 1.18A degrees C respectively. It also shows the model RMS error is less than or close to the climatological error. The statistical model can be used to well estimate the temperature profile vertical structure. 3) Comparing the thermocline characteristics between the reconstructed and observed profiles, the results in the ECS show that the average absolute errors are 1.5m, 1.4 m and 0.17A degrees C/m, and the average relative errors are 24.7%, 8.9% and 22.6% for the upper, lower thermocline boundaries and the gradient, respectively. Although the relative errors are obvious, the absolute error is small. In the SCS, the average absolute errors are 4.1 m, 27.7 m and 0.007A degrees C/m, and the average relative errors are 16.1%, 16.8% and 9.5% for the upper, lower thermocline boundaries and the gradient, respectively. The average relative errors are all < 20%. Although the average absolute error of the lower thermocline boundary is considerable, but contrast to the spatial scale of average depth of the lower thermocline boundary (165 m), the average relative error is small (16.8%). Therefore the model can be used to well estimate the thermocline.

Of the present estimated world population of 14.2 million yaks, approximately 13.3 million occur within Chinese territories (Food and Agriculture Organization of the United Nations, 2003). Although there is an extensive bibliography covering the species, few studies have been conducted in the area of foraging behaviour. The present study was conducted at pasture during the spring, transitional, summer and winter seasons to determine the daily temporal patterns of grazing and ruminating behaviour by yaks. During each study period, two 24 h recordings were undertaken with each of six mature dairy yaks. One study period was conducted on each of the transitional, summer and winter pastures, whereas, due to the considerable changes occurring in the morphology of the spring pasture, three separate studies were completed during March, April and May. During the second of these studies (April), the effect of level of concentrate supplementation on grazing and ruminating behaviour was also examined. Behaviour recordings were made using solid-state behaviour recorders. Short-term intake rates (IR, g min(-1)) were calculated by weighing yaks before and after approximately 1 h of grazing, retaining the faeces and urine excreted and applying a correction for insensible weight loss. Yaks spent less time grazing during the dry season (the early period on the spring pasture) compared with the later green swards (the later period on the spring pasture, the transitional pasture and the summer pasture) (P < 0.05). When the forage quality improved, but there was still insufficient mass (the later period on the spring pasture), the yaks extended their grazing time at the expense of other activities. During the early periods on the spring pasture, the short-term IR by yaks was up to 53 g DM min(-1), significantly higher than at other times (P < 0.05). The level of concentrate offered had little or no effect on grazing or ruminating time. The total eating time of the yaks offered 0.5 or 1.0 kg concentrate was 2.9 and 4.5 h day(-1) respectively, significantly lower than unsupplemented yaks (6.8 h) (P < 0.05). In general, yaks can regulate their foraging behaviour according to the changes of sward conditions in order to achieve optimal grazing strategies. (C) 2007 Published by Elsevier B.V.
