Complex pupil filters are introduced to improve the three-dimensional resolving power of an optical imaging system. Through the design of the essential parameters of such filters, the transmittance and radius of the first zone, three-dimensional superresolution is realized. The Strehl ratio and the transverse and axial gains of such filters are analyzed in detail. A series of simulation examples of such filters are also presented that prove that three-dimensional superresolution can be realized. The advantage of such filters is that it is easy to realize three-dimensional superresolution, and the disadvantage is that the sidelobes of the axial intensity distribution are too high. But this can be overcome by the application of a confocal system. (C) 2005 Optical Society of America.

The characterization of acid-sensing ion channel (ASIC)-like currents has been reported in hippocampal neurons in primary culture. However, it is suggested that the profile of expression of ASICs changes in culture. In this study, we investigated the properties of proton-activated current and its modulation by extracellular Ca2+ and Zn2+ in neurons acutely dissociated from the rat hippocampal CA1 using conventional whole-cell patch-clamp recording. A rapidly decaying inward current and membrane depolarization was induced by exogenous application of acidic solution. The current was sensitive to the extracellular proton with a response threshold of pH 7.0-6.8 and the pH(50) Of 6.1, the reversal potential close to the Na+ equilibrium potential. It had a characteristic of acid-sensing ion channels (ASICs) as demonstrated by its sensitivity to amiloride (IC50 = 19.6 +/- 2.1 muM). Either low [Ca2+](0) or high [Zn2+](0) increased the amplitude of the current. All these characteristics are consistent with a current mediated through a mixture of homomeric ASIC1a and heteromeric ASIC1a + 2a channels and closely replicate many of the characteristics that have been previously reported for hippocampal neurons cultured for a week or more, indicating that culture artifacts do not necessarily flaw the properties of ASICs. Interestingly, we found that high [Zn2+] (>10(-4) M) slowed the decay time constant of the ASIC-like current significantly in both acutely dissociated and cultured hippocampal neurons. In addition, the facilitating effects of low [Ca2+](0) and high [Zn2+](0) on the ASIC-like current were not additive. Since tissue acidosis, extracellular Zn elevation and/or Ca2+ reduction occur concurrently under some physiological and/or pathological conditions, the present observations suggest that hippocampal ASICs may offer a novel pharmacological target for therapeutic invention. 杨树具有分布广、适应性强的特征,在生态环境治理和解决木材短缺方面均占有重要位置。青杨(Populus cathayana Rehd.)是青杨派树种的重要成员之一,也是生长较迅速、易繁殖的重要杨树资源。本研究选取了来自不同气候地区的青杨两种群为材料,采用植物生态学、生理学和生物化学的研究方法,系统地研究了青杨对干旱与遮荫、干旱与外源脱落酸(ABA)喷施的生长、形态、生理和生化响应及种群间差异,研究成果可为我国干旱半干旱地区的造林以及生态恢复提供理论依据和科学指导。主要研究结论如下:1.青杨在干旱胁迫下的适应机制为:生长性状及生物量的分配变化:干旱胁迫下虽然植株生长受抑,株高、基茎及各部分生物量都显著减小,但有相对较多的生物量向根部分配,根/冠比以及细/粗根比增加。青杨对干旱胁迫的光合作用表现为:干旱胁迫降低了青杨的净光合速率、蒸腾速率、气孔导度以及光合氮利用效率,提高了瞬时用水效率。干旱还引起了活性氧的产生,使得膜脂过氧化产物丙二醛(MDA)增加,同时也增强了植物抗氧化酶系统(如超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和抗坏血酸过氧化物酶(APX)活性的增加)及非酶系统的能力(如抗坏血酸(AsA)含量的增加)。干旱降低了植物叶片的相对含水量,而促进了渗透调节物质(游离脯氨酸及可溶性糖)的积累,增加了植物的渗调能力。干旱下青杨两种群的内源ABA含量显著增加,碳同位素组分(δ13C)也显著提高。这些结果证明植物遭受干旱胁迫后发生一系列的形态、生理和生化响应,这些变化能提高植物在干旱下的存活和生长能力。2.青杨两种群对干旱胁迫反应的种群差异:与来自湿润地区的汉源种群相比,来自干旱地区的乐都种群在干旱条件下生物量向根系分配的可塑性更强,同时具有更强的抗氧化系统能力,所受到活性氧的伤害也更少,并且累积更多的脯胺酸和ABA,具有更高的δ13C。这些都说明了乐都种群对干旱的适应性比汉源种群更强。两种群对干旱的响应差异应归于它们的用水策略的不同:汉源种群来自湿润地区,采用了耗水型的用水策略,抗旱能力较弱;而乐都种群,来自干旱地区,通常采用节水型的用水策略,有更强的抗旱能力。3.遮荫对青杨两种群抗旱性的影响:遮荫对青杨抗旱性的影响决定于遮荫程度的不同,我们的结果表明中度的遮荫可以有效的提高干旱下植物的生长,对干旱胁迫有明显的缓解作用,具体体现在中度遮荫下受旱植物的叶片相对含水量得到提高,使得植物体内水分状况得到了改善;光合速率并未降低,植物光合氮利用效率增加,说明中度的遮荫并未明显限制植物的碳获得;抗氧化酶活性与膜脂过氧化产物MDA含量的同时降低,说明中度遮荫下所受到的活性氧伤害减少;中度遮荫下的ABA及δ13C的变化也不如在全光下变化明显,这也说明中度遮荫缓解了干旱胁迫。但是重度的遮荫却对干旱胁迫有明显的加剧作用,主要表现在重度遮荫降低了植物的光合速率,严重抑制了植物的生长;同时重度遮荫下脯胺酸含量和抗氧化酶活性的急剧下降,导致了植物渗调能力的下降及膜脂过氧化产物MDA的显著升高;重度遮荫还显著降低了内源ABA的累积和δ13C,降低了植物的抗旱能力。此外,青杨两种群在对干旱和遮荫的响应中,也表现出种群差异。汉源种群,来自湿润且年日照辐射较少的地区,表现出相对更强的耐荫性和需水性。而乐都种群,来自干旱且年日照辐射丰富的地区,表现出相对更强的耐旱性和需光性。这说明了植物对环境胁迫的耐受性是其长期适应原生境的结果,并且来自不同气候地区的两种群在面临环境胁迫时会采取不同的生存策略。4. 外源ABA喷施对青杨两种群抗旱性的影响:外源ABA的喷施可以提高两种群的抗旱性,具体表现为外源ABA喷施促进了青杨根系的生长,显著提高了干旱下植物的根/冠比和细/粗根比,减少了比叶面积;在生理生化方面,外源ABA降低了干旱下植物叶片的气孔导度,降低了蒸腾速率和净光合速率,但提高了瞬时用水效率,提高了叶片的相对含水量,增加了干旱下植物的保水能力。外源ABA进一步增加了干旱下植物内源ABA的积累,促进了植物渗调物质如脯胺酸和可溶性糖的积累,增加了抗氧化酶系统(如SOD、APX、CAT)的活性和非酶系统AsA的含量,降低了活性氧(如超氧阴离子(O2和过氧化氢(H2O2))对植株的伤害。此外,外源ABA还进一步提高了干旱下植物的δ13C,提高了植物的长期用水效率,由此提高了植物的抗旱能力。另一方面,两种群对外源ABA和干旱的响应也有所差别。来自湿润地区的汉源种群,对干旱较为敏感,所受干旱的影响也较大,而外源ABA的喷施对汉源种群抗旱性的提高作用也更为突出。乐都种群,由于其长期适应干旱地区的生长,本身已具有较强的抗旱能力,因此外源ABA喷施对其抗旱性的提高不如对汉源种群的效果明显。由此我们可以得出对于一些抗性弱或干旱敏感的物种或者种群,可以采用外施ABA的方法来提高其抗性。Poplars play an important role in lumber supply, and are important component ofecosystems due to their wide distribution and well adaptation. Populus cathayana Rehd.,which belongs to Populus Sect. Tacamahaca Spach, is one of the most important resources ofpoplars for its fast growth and reproductive. In this study, different populations of P.cathayana were used as experiment material to investigate the adaptability to drought stressand population differences in adaptability, and the effects of shade and exogenous abscisicacid (ABA) application on the drought tolerance. Our results could provide a strongtheoretical evidence and scientific direction for the afforestation, and rehabilitation ofecosystem in the arid and semi-arid area, and provide a strong evidence for adaptivedifferentiation of different populations, and so may be used as criteria for species selectionand tree improvement. The results are as follows:1. A large set of parallel response to drought stress: Drought stress caused pronouncedinhibition of the growth and increased relatively dry matter allocation into the root. For thetwo populations, the shoot height, basal diameter and total biomass were decreased but theroot/shoot ratio and fine root/coarse root ratio were increased under drought conditions;Drought stress caused pronounced inhibition of photosynthesis, decreased the stomatalconductance, transpiration rate, and photosynthetic nitrogen-use efficiency (PNUE) butincreased the instantaneous water use efficiency. Drought significantly improved the levels ofreactive oxygen species and malondialdehyde (MDA) and to induce the entire set ofantioxidative systems including the increase of activities of superoxide dismutase (SOD),ascorbate peroxidase (APX), catalase (CAT) and ascorbate (AsA) content. Drought decreased the leaf relative water content (RWC) but improved the capability of osmotic adjustmentindicated by the higher proline accumulation. Drought also increased the ABA content andcarbon isotope composition (δ13C), which indicating the long period water use efficiency wasimproved under drought. These results demonstrate that there are a large set of parallelchanges in the morphological, physiological and biochemical responses when plants areexposed to drought stress; these changes may enhance the capability of plants to survive andgrow during drought periods.2. Difference in adaptation to drought stress between contrasting populations of P.cathayana: Compared with the Hanyuan population (wet climate), the Ledu population (dryclimate) showed higher root/shoot ratio and water use efficiency, exhibited higherantioxidative systems capability thus resulting in less oxidative damage, accumulated moreABA and free proline content under drought conditions. The results suggested that there weredifferent water-use strategies between the two populations. The Ledu population, whichcomes from dry climate region, with higher drought tolerance, may employ a conservativewater-use strategy, whereas the Hanyuan population, which comes from wet climate, withlower drought tolerance, may employ a prodigal water-use strategy. These variations indrought responses may be used as criteria for species selection and tree improvement.3. The effects of shade on the drought tolerance: The reduction in the availability of lightand water affected the morphological and physiological responses of the two P. cathayanapopulations. In addition, the light environment modified the growth responses of P.cathayana seedlings to varying water environments in different ways depending upon theintensity of the light levels considered. There is an apparent alleviation to drought effects bymoderate shade in P. cathayana seedlings, as indicated by the higher leaf RWC, and unchanged net photosynthesis and PNUE, as well as by the lower antioxditative enzymeactivity, MDA, ABA and δ13C levels, which implied moderate shade did not significantlylimited the carbon acquisition or inhibited the plant growth, but ameliorated the detrimentaleffects of drought. On the other hand, an apparent aggravation to drought effects by severeshade was also observed, as indicated by the pronounced decrease of plant growth and net photosynthesis, the lower total biomass, ABA level, δ13C, free proline content andantioxditative enzyme activity and higher MDA accumulation. By contrast, the twopopulations showed different responses to shade and drought. The Hanyuan population,which comes from a riparian basin having a relatively wet climate and less annual solarradiation, is more sensitive to drought but more tolerant to shade. The Ledu population, whichcomes from a mountainous plateau with less rainfall and with more annual solar radiation, ismore tolerant to drought but more sensitive to shade. The results demonstrated that theendurance of plants to stress is a result of long-term evolution and adaptation to theenvironment, as suggested by the different strategies employed by the P. cathayanapopulations originating from contrasting habitats when they were exposed to drought andshade.4. The effects of exogenous ABA application on the drought tolerance: For bothpopulations under drought conditions tested, exogenous ABA application significantlyimproved the root/shoot ratio, fine root/coarse root ratio, and decreased the specifical leaf area.On the physiological and biochemical traits, exogenous ABA application significantlydecreased stomatal conductance, transpiration rate and net photosythesis but increased theinstance water use efficiency and leaf RWC. On the other hand, exogenous ABA applicationsignificantly increased endogenous ABA, proline, solube sugar and AsA content, as well asSOD, APX and CAT activities, thus reduced the damage of reactive oxygen species. Moreover,the long period water use efficiency as indicated by δ13C was also improved by exogenousABA application. In additionally, there was different responsive between the two populationsto drought and exogenous ABA application. The Hanyuan population, which comes from wetclimate region, is more sensitive to drought, and the effect of exogenous ABA is moreobviously than that in the Ledu population, which comes from dry climate region and is moredrought-responsive. Therefore, we can use exogenous ABA application to improve theresistance of plants, especially for the drought- sensitive species or populations.

A new method for prolidase (PLD, EC 3.4.13.9) activity assay was developed based on the determination of proline produced from enzymatic reaction through capillary electrophoresis (CE) with tris(2,2'-bipyridyl)ruthenium(11) [Ru(bpy)(3)(2+)] electrochemiluminescence detection (ECL). A detection limit of 12.2 fmol (S/N = 3) for proline, corresponding to 1.22 x 10(-8) units of prolidase catalyzing for 1 min was achieved. PLD activity determined by CE-ECL method was in agreement with that obtained from the classical Chinard's one. CE-ECL showed its powerful resolving ability and selectivity as no sample pretreatmentwas needed and no interference existed. The clinical utility of this method was successfully demonstrated by its application to assay PLD activity in the serum of diabetic patients in order to evaluate collagen degradation in diabetes mellitus (DM). The results indicated that enhanced collagen degradation occurred in DM.

针对90°全息记录结构的特殊性,在记录和读出阶段分别施加不同极性的外加电场,实现了Fe:LiNbO3晶体中高衍射效率的全息记录和读出。在库赫塔列夫(Kukhtarev)方程基础上引人两维耦合波理论,对组合外加电场提高掺铁铌酸锂晶体光折变特性的机理进行了探讨。实验和理论计算结果都表明这种分别在记录和读出过程施加不同极性组合的外加电场是在Fe:LiNbO3中实现高衍射效率90°记录结构体全息的有效技术方案。

For a four-port microracetrack channel drop filter, unexpected transmission characteristics due to strong dispersive coupling are demonstrated by the light tunneling between the input-output waveguides and the resonator, where a large dropping transmission at off-resonance wavelengths is observed by finite-difference time-domain simulation. It causes a severe decline of the extinction ratio and finesse. An appropriate decrease of the coupling strength is found to suppress the dispersive coupling and greately increase the extinction ratio and finesse, a decreased coupling strength can be realized by the application of an asymmetrical coupling waveguide structure. In addition, the profile of the coupling dispersion in the transmission spectra can be predicted based on a coupled mode theory analysis of an equivalent system consisting of two coupling straight waveguides. The effects of structure parameters on the transmission spectra obtained by this method agree well with the numerical results. It is useful to avoid the strong dispersive coupling region in the filter design. (c) 2007 Optical Society of America.

The most biological diversity on this planet is probably harbored in soils. Understanding the diversity and function of the microbiological component of soil poses great challenges that are being overcome by the application of molecular biological approaches. This review covers one of many approaches being used: separation of polymerase chain reaction (PCR) amplicons using denaturing gradient gel electrophoresis (DGGE). Extraction of nucleic acids directly from soils allows the examination of a community without the limitation posed by cultivation. Polymerase chain reaction provides a means to increase the numbers of a target for its detection on gels. Using the rRNA genes as a target for PCR provides phylogenetic information on populations comprising communities. Fingerprints produced by this method have allowed spatial and temporal comparisons of soil communities within and between locations or among treatments. Numerous samples can be compared because of the rapid high throughput nature of this method. Scientists now have the means to begin addressing complex ecological questions about the spatial, temporal, and nutritional interactions faced by microbes in the soil environment.

目的解决B样条曲面重建问题中矩形拓扑网自动生成和参数化两大难点问题,提出一种基于逆向参数化的B样条曲面重建算法.方法首先构建基曲面,在基曲面上根据参数(u,v)进行采样,沿其法线方向进行数据的滤波和精简,求得参数(u,v)对应的精简点,然后对采样求取的精简点集进行B样条曲面拟合,该方法提供了B样条曲面重建的一个新思路.结果新算法突破了传统密集散乱点云数据的B样条曲面重建基本过程,采用与正向参数化相反的过程进行参数化,解决了B样条曲面重建问题中矩形拓扑网自动生成和参数化的难题;具体试验分析表明新算法不仅在参数化的同时完成了数据滤波和精简,而且在时间和迭代效率方面都具有优势.结论新算法避免了求取法线的迭代过程,并且可以较容易的实现矩形拓扑网的自动生成,新算法在自主开发的智能测量建模加工一体化装备中得到了应用验证.

The study of pore structure in reservoir was paid attention to in the early reservoir research and now a systematic research methodology is set up. On the limits of tools and conditions, methodologies and technologies on formation condition and distribution laws of pore structure and the relationship between remaining oil distribution and pore structure are uncertain and some knownage about it is also uncertain. As the development of petroleum industry, the characterization of pore structure and the prediction of remaining oil are the hot spot and difficult point in the research of oil development. The author pays a close attention to this subject and has done much research on it. In a case study in Linnan oilfield Huimin sag Jiyang Depression Bohai Bay basin by using a new method, named varied scale comprehensive modeling of pore structure, the author builds pore structure models for delta reservoir, reveals the remaining oil distribution laws in delta facies, and predicts the distribution of remaining oil in Linnan oilfield. By the application of stratigraphy, sedimentology and structure geology. the author reveals the genetic types of sandbody and its distribution laws, builds the reservoir geological models for delta sandstone reservoir in Shahejie group in Linnan oilfield and points out the geological Factors that control the development of pores and throats. Combining petrology and the reservoir sensitive analysis, the author builds the rock matrix models. It is the first time to state that rocks in different sentimental micro facies have different sensitive .response to fluid pressed into the rocks. Normally. the reservoirs in the delta front have weaker sensitivity to fluid than the reservoirs in delta plain, In same subfacies, the microfacies that have fine grain, such as bank and crevasse splay, have stronger reservoir sensitivity than the microfacies that have coarse grains, such as under-water branched channel and debauch bar. By the application of advanced testing, such as imagine analysis, scan electronic microscope, and morphology method, the author classifies the pore structure and set up the distribution models of pore, throat and pore structure. By the application of advanced theory in well-logging geology, the author finds the relationship between microscope pore structure and macroscopic percolation characteristics, and then builds the well-logging interpretation formulae for calculating pore structure parameters. By using the geostatistics methods, the author reveals the spatial correlative characteristics of pore structure. By application of conditional stochastic simulation methods, the author builds the 3D models of pore structure in delta reservoir. It is the base of predicting remaining oil distribution. By a great deal of experiments and theoretical deduction, The author expounds the laws of percolation flow in different pore structures, and the laws by which the pore structure controls the micro distribution of remaining oil, and then, states the micro mechanism of remaining oil distribution. There are two types of remaining oil. They are by-pass flow caused by micro-fingering and truncation caused by non-piston movement. By new method, the author states the different pore structure has different replacement efficiency, reveals the formation condition and distribution laws of remaining oil. predicts the remaining oil distribution in Linnan oil field, and put forward some idea about how to adjust the oil production. The study yielded good results in the production in Linnan oilfield.

At present the main object of the exploration and development (E&D) of oil and gas is not the structural oil-gas pools but the subtle lithological oil-gas reservoir. Since the last 90's, the ratio of this kind of pools in newly-added oil reserves is becoming larger and larger, so is the ratio in the eastern oilfields. The third oil-gas resource evaluation indicates the main exploration object of Jiyang depression is the lithological oil-gas pools in future. However, lack of effective methods that are applied to search for this kind of pool makes E&D difficult and the cost high. In view of the urgent demand of E&D, in this paper we deeply study and analyze the theory and application in which the seismic attributes are used to predict and describe lithological oil-gas reservoirs. The great results are obtained by making full use of abundant physics and reservoir information as well as the remarkable lateral continuity involved in seismic data in combination with well logging, drilling-well and geology. ①Based on a great deal of research and different geological features of Shengli oilfield, the great progresses are made some theories and methods of seismic reservoir prediction and description. Three kinds of extrapolation near well seismic wavelet methods-inverse distance interpolation, phase interpolation and pseudo well reflectivity-are improved; particularly, in sparse well area the method of getting pseudo well reflectivity is given by the application of the wavelet theory. The formulae for seismic attributes and coherent volumes are derived theoretically, and the optimal method of seismic attributes and improved algorithms of picking up coherent data volumes are put forward. The method of making sequence analysis on seismic data is put forward and derived in which the wavelet transform is used to analyze not only qualitatively but also quantitatively seismic characteristics of reservoirs.② According to geologic model and seismic forward simulation, from macro to micro, the method of pre- and post-stack data synthetic analysis and application is put forward using seismic in close combination with geology; particularly, based on making full use of post-stack seismic data, "green food"-pre-stack seismic data is as possible as utilized. ③ In this paper, the formative law and distributing characteristic of lithologic oil-gas pools of the Tertiary in Jiyang depression, the knowledge of geological geophysics and the feasibility of all sorts of seismic methods, and the applied knowledge of seismic data and the geophysical mechanism of oil-gas reservoirs are studied. Therefore a series of perfect seismic technique and software are completed that fit to E&D of different categories of lithologic oil-gas reservoirs. ④ This achievement is different from other new seismic methods that are put forward in the recent years, that is multi-wave multi-component seismic, cross hole seismic, vertical seismic, and time-lapse seismic etc. that need the reacquisition of seismic data to predict and describe the oil-gas reservoir. The method in this paper is based on the conventional 2D/3D seismic data, so the cost falls sharply. ⑤ In recent years this technique that predict and describe lithologic oil-gas reservoirs by seismic information has been applied in E&D of lithologic oil-gas reservoirs on glutenite fans in abrupt slop and turbidite fans in front of abrup slop, slump turbidite fans in front of delta, turbidite fans with channel in low slope and channel sanbody, and a encouraging geologic result has been gained. This achievement indicates that the application of seismic information is one of the most effective ways in solving the present problem of E&D. This technique is significant in the application and popularization, and positive on increasing reserves and raising production as well as stable development in Shengli oilfield. And it will be directive to E&D of some similar reservoirs
