本文报道了在育性转换敏感期光周期处理对光敏核不育水稻(农垦58S)及农垦58最新全展叶中光敏色素Ⅰ(PhyA)水平的影响PartI).在10个光周期处理的最后一个暗期结束前,收获每株水稻的最上部二叶。PhyA用酶联免疫吸附测定法(ELISA)测定。 结果表明:0.5%(v/v)聚乙烯亚胺(PEI)可除去水稻叶片粗提液中干扰ELISA的物质;所用的ELISA专一性地检测水稻PhyA。和长日照(LD)处理相比,短日照(SU)处理导致农垦58S中PhyA的相对含量增加38.5%;而农垦58只增加18.5%。显然,在较长的暗期条件下(SD),农垦58S中PhyA的合成比农垦58快。SD处理下大量增加的PhyA可能和农垦58S的育性恢复有关。 上述结果也说明:在同一品种甚至不同品种的植株间,PhyA水平均易受光周期影响而剧烈变化。 为了进一步验证农垦58S中PhyA较快积累的推论,比较了农垦58s和农垦58幼苗(三叶期)在一延长暗期(24h)中PhyA的积累时程。和育性转换敏感期的植株相似,农垦58S幼苗中PhyA积累速度快于农垦58。在暗期开始6h后,这种差异更明显。这一结果证实了过去的假设:甲基化水平低的农垦58PhyA基因可能比农垦58PhyA基因更活跃地表达。 PhyA和PhyB同时存在于水稻叶片中。为了探讨PhyB是否参与农垦58S雄性不育的调节,在育性转换敏感期每日光期结束、暗期开始开始前进行短暂的FR照射实验(即end-of- dayFR irradiations)。EOD FR反应应由PhyB介导。和SD下的对照相比,经过10次EODFR处理(EOD FR+SD)的农垦58S植株抽穗和开花期都相应地推迟2天,而花粉败育率和种子结实率都没有变化。 EODFR处理抑制了农垦58的开花,但花粉育性几乎不受影响。 综上所述,可能是PhyA而不是PhyB参与调节农垦58S的雄性不育。 另外,本文采用免疫印迹(Immunoblotting or Western blotting)比较了农垦58S和农垦58黄化苗(3天龄)中PhyA的相对含量(PartⅡ)。 结果表明,RPA可以专一性地检测两品种中120KD多肽。该肽在照射R或FR后对内源蛋白酶水解的敏感性不同,照射FR后,该肽易降解产生116KD的片段;照射R后,相对较稳定。因此,上述120KD多肽是水稻PhyA。未观察到农垦58S和农垦58的PhyA在免疫原性、分子量及内源蛋白酶解水解带型有差异。定量分析表明农垦58s黄化苗中PhyA的相对含量比农垦58多40%。这一结果和上述光周期处理的结果是相辅相成的。由于干种子、以及吸涨36h以前的水稻胚中均检测不PhyA的存在,因此两品种间PhyA含量的差异是PhyA蛋白重新合成的结果。 活体低温(80K)荧光光谱分析表明:农垦58黄化苗(3天龄)具有典型光敏色素(主要为PhyA)的荧光发射,其最大波长为683.8nm,而农垦58S以及由其转育来的培矮64s都缺少明显的光敏色素峰。显然,农垦58S和农垦58的PhyA荧光光谱特性有所不同。这一差异是否和雄性不育有关仍待深入研究。 本文第三部分比较了农垦58S和农垦58黄化苗(6天龄)最初转到白光下(4h)合成叶绿素的情况。无论是短暂红光(R)处理或对照,农垦58幼苗合成叶绿素的量(在白光下4h)都多于农垦58S。由于R促进叶绿素合成的效果可被随后的远红光照射(FR)逆转,因此水稻幼苗中叶绿素合成是在光敏色素的控制下。FR逆转性在农垦58S中似乎更完全。连续FR(12h最有效)促进叶绿素合成的效果在农垦58S中更明显,但叶绿素合成的量(在白光下4h)仍是农垦58多。然而,对于自然光周期下生长的幼苗(2-4叶期),农垦58S的叶绿素含量明显高于农垦58。文中讨论了这种差异的可能原因。

A marine green alga, Platymonas subcordiformis, was demonstrated to photobiologically evolve hydrogen (H-2) after the first stage of photosynthesis, when subjected to a two-phase incubation protocol in a second stage of H2 production: anaerobic incubation in the dark followed by the exposure to light illumination. The anaerobic incubation induced hydrogenase activity to catalyse H? evolution in the following phase of light illumination. H,) evolution strongly depended upon the duration of anaerobic incubation, deprivation of sulphur (S) from the medium and the medium pH. An optimal anaerobic incubation period of 32 h gave the maximum H2 evolution in the second phase in the absence of sulphur. Evolution of H,) was greatly enhanced by 13 times when S was deprived from the medium. This result suggests that S plays a critical role in the mediation of H-2 evolution from R subcordiformis. A 14-fold increase in H-2 production was obtained when the medium pH increased from 5 to 8; with a sharp decline at pH above eight. H-2 evolution was enhanced by 30-50% when supplementing the optimal concentrations of 25 mM acetate and 37.5 mM glucose. (C) 2003 Elsevier B.V. All rights reserved.

In order to study the earthquake recurrence and the characteristics of earthquake series, rupture tests of rock samples and plexiglass samples were made. On rock samples, a number of acoustic emission (AE) and strain measuring points were deployed; the load was one side direct shear. The variation characteristics of AE and strain at different detecting points around the extra large fracture were observed and studied. On plexiglass samples, a series of inclined cracks were prefabricated by a small-scale compressive testing machine. The samples were then loaded on a shockproof platen, when the samples were loaded, the stress intensity factor (SIF) was determined by the laser interferometric technique and shadow optical method of caustics. The fracture conditions such as material toughness around the extra large fracture were also studied. From those experimental results and the theory of fracture mechanics, the earthquake recurrence period and the trend of post-seismic development were studied.

The effect of alcohol solution on single human red blood Cells (RBCs) was investigated using near-infrared laser tweezers Raman spectroscopy (LTRS). In our system, a low-power diode laser at 785 nm was applied for the trapping of a living cell and the excitation of its Raman spectrum. Such a design could simultaneously reduce the photo-damage to the cell and suppress the interference from the fluorescence on the Raman signal. The denaturation process of single RBCs in 20% alcohol solution was investigated by detecting the time evolution of the Raman spectra at the single-cell level. The vitality of RBCs was characterized by the Raman band at 752 cm(-1), which corresponds to the porphyrin breathing mode. We found that the intensity of this band decreased by 34.1% over a period of 25 min after the administration of alcohol. In a further study of the dependence of denaturation on alcohol concentration, we discovered that the decrease in the intensity of the 752 cm(-1) band became more rapid and more prominent as the alcohol concentration increased. The present LTRS technique may have several potential applications in cell biology and medicine, including probing dynamic cellular processes at the single cell level and diagnosing cell disorders in real time. Temperature and stress tunabilities of long-period Bragg gratings imprinted in Panda fiber are presented in this letter. It is shown that the temperature and strain response of the resonance peaks for fast and slow axes are different not only in their magnitudes but also in the signs of the slope. Furthermore, the characteristics for different order modes are different both in magnitudes and signs. The complicated phenomena are discussed by using a simplified model.

Based on the Mach-Zehnder effect between the core mode and the cladding modes, the interference fringes are formed by a pair of cascaded long-period fiber gratings (CLPFGs). Theoretical analyses show that the spectral spacing and the wavelength of these fringes are functions of the waveguide dispersion factor gamma, which is a characterizing parameter to LPFG and with theoretical and applicational significance. By measuring the characteristics of the transmission spectra of CLPFGs, the absolute value of gamma can be obtained. At the same time, the thermo-optic coefficient of effective refractive index difference between core and cladding modes, p, can also be obtained by measured the temperature sensitivity of these fringes. In the experiments, \gamma\ and mu were measured by this method to be 0.874 and 4.08 x 10(-5) degreesC(-1), respectively, for LPFGs with period of 450 mum and with a HE14 resonant peak at 1554 nm. A novel optical interleaver scheme based on nested optical glass pairs is proposed. The assembly of pairs behaves as a cascaded Mach-Zehnder interferometer. The interleaver, with simple structure, low cost, and compact size, can be easily implemented with inexpensive material and mature preparation technology. Small channel spacing (<= 50 GHz), high isolation (<-30 dB), a wide, flat passband and stop band (> 2/11 period), and center-frequency tunability can be obtained simultaneously. An optimum design of a 50-GHz tunable interleaver based on this structure is given as an example. Its environmental temperature sensitivity and fabrication tolerance are also analyzed. The novel long-period fiber grating (LPFG) film sensor is composed of the long-period grating coated with solgel-derived sensitive films. The characteristics of the transmissivity of the LPFG film sensor are studied. By analyzing the relation among the sensitivity S-n, the thin film optical parameters, and the fiber grating parameters, the optimal design parameters of the LPFG film sensor are obtained. Data simulation shows that the resolution of the refractive index of this LPFG film sensor is predicted to be 10(-8). Experimentally, a LPFG film sensor for detection Of C2H5OH was fabricated, and a preliminary gas-sensing test was performed. (c) 2006 Optical Society of America.

以亲缘关系极近的近缘种类群、中等距离的远缘种类群为对象,分析生物钟基因period基因的Thr-Gly区段的分子进化特征,发现Thr-Gly区段在果蝇和部分双翅目昆虫中未曾经历性选择和其他定向的正选择。Thr-Gly区段在果蝇nasuta亚群中的分子进化速率为10.4*10~-9同义替换/位点/年。

测定了金色果蝇复合种5个姐妹种和D. rufa的period (per)基因的Thr-Gly区段序列。该区段序列分析表明:DNA序列的碱基组成拥有果蝇其他基因的共同特点;颠换数多于转换数,两两物种间的颠换率与转换率的总比值为2-5,密码子第3位的颠换与转换的比值为2.5-5;同义替换/异义替换(Ks/Ka)值远大于10,且有的物种间根本不存在非同义突变,低的Ka值说明该复合种的per基因Thr-Gly区段在进化过程中可能承受着较强的选择压力。运用所得的核苷酸序列构建Drosophila auraria复合种的系统发育树,为澄清该类群的系统演化关系提供了新的线索。

In this study, the region corresponding to the Thr-Gly region of the period (per) gene in the Drosophila nasuta subgroup of species was sequenced. The results showed, that this region was highly conserved in the D. nasuta subgroup. There were only nine variable sites found in this 300-bp-long region, all located in two small regions highly variable among Drosophila species. No length variation was observed either within this subgroup or in the Yunnan (YN) population of D. albomicans. The deduced amino acid sequences were identical for all 14 taxa in the D. nasuta subgroup, and a stretch of alternating Thr-Gly pairs was not observed in this subgroup. A phylogenetic tree was constructed. The clustering of some species was in general agreement with previous works, but it also raised some question on the phylogenetic relationship between the nasuta species. The data did not implicate the Thr-Gly region playing a role in behavioral isolation in this subgroup of Drosophila.

The objective of this study is to conduct a bibliometric analysis of all biological invasions-related publications in the Science Citation Index (SCI) from 1991 to 2007. The indicator citation per publication (CPP) was used to evaluate the impact of articles, journals, and institutions. In the 3323 articles published in 521 journals, 7261 authors from 1905 institutions of 100 countries participated. As the most productive country of biological invasions research, the US will benefit from more collaboration between institutions, countries, and continents. In addition, analysis of keywords was applied to reveal research trends.
