光系统II(PSII)是存在于类囊体膜中的多亚基色素蛋白复合物,是吸收光能、催化光诱导水裂解释放氧气、质子和电子的重要机构。它在体内的基本单位是由外周天线蛋白(LHCII)与PSII核心复合物结合形成的PSII-LHCII超分子复合物,这一结构保证了LHCII吸收的能量能够快速有效的传递到PSII反应中心(RC),进行原初光化学反应。 本论文分为两部分:1、利用捕光色素蛋白复合物(LHCII)与PSII核心复合物在以DGDG、PG、SQDG三种类囊体膜脂形成的脂质体中重组的方法,研究了LHCII与PSII在脂膜上结构与功能的相互作用;2、通过研究光破坏和色素置换对PSII RC的影响,探讨了RC中不同色素的功能。主要结果如下: 1、LHCII与PSII核心复合物的蛋白脂质体研究: 将OECC(粗提核心复合物)、pdOE(纯化核心复合物)、LHCII(大量天线)制剂分别与脂质体重组并研究了其光谱性质。LHCII在与脂质体重组前表现出典型的聚集态光谱特征,重组后吸收和荧光发射峰发生明显蓝移;LHCII、OECC和pdOE三种蛋白脂质体与重组前的样品相比荧光发射强度增加;表明脂环境影响了色素蛋白复合物的聚集状态以及色素和蛋白之间的相互作用。 OECC和pdOE分别与LHCII在脂质体中重组,得到两种重组蛋白(LHCII-OECC和LHCII-pdOE)脂质体,用冰冻蚀刻电镜技术和低温荧光光谱的方法研究其结构和功能特征。LHCII和核心复合物(OECC或pdOE)结合形成PSII-LHCII重组颗粒,并在脂质体中均匀排布,阻止了LHCII晶格状结构的形成。重组蛋白脂质体的吸收光谱既有LHCII的吸收特征,又有核心复合物的特征吸收峰,但低温荧光光谱的主要发射峰是核心复合物的特征峰(684 nm-685 nm),而不是LHCII的特征峰(680 nm);而且激发不同色素得到的荧光发射光谱基本一致,这些结果证明LHCII吸收的能量传递到了核心复合物中,在重组蛋白脂质体中不同色素蛋白复合物在结构和功能上都实现了相互偶联。 通过对OECC或pdOE与LHCII重组形成的蛋白脂质体放氧或DCPIP光还原活性的检测研究了PSII光化学活性特征。LHCII和核心复合物(OECC或pdOE)的重组蛋白脂质体与单独核心脂质体相比,在强光和弱光下光化学活性都明显提高。这从另一个角度证明了核心复合物与LHCII的功能偶联,LHCII的结合使捕光截面积增大,从而使PSII光化学活性增加。 用77K飞秒时间分辨荧光光谱分析了几种蛋白脂质体的能量传递和捕获情况。LHCII、OECC和pdOE三种蛋白脂质体的主要荧光衰减组分分别是670 ps(发射峰在680 nm)、650 ps(发射峰在690 nm)和570 ps(发射峰在685 nm)。LHCII-OECC和LHCII-pdOE脂质体的主要衰减组分分别是940 ps(发射峰在690 nm)和840 ps(发射峰在685 nm),并且出现了一个在核心复合物脂质体和LHCII脂质体中没有的40 ps组分,可以推测,这是LHCII和核心复合物之间达到平衡的时间组分,比激发态衰减的平均寿命要快得多,因此支持了PSII的trap-limited激发能衰减动力学模型。此外,可以看到天线的增大使Chl a荧光衰减的寿命延长,这一特性可能与PSII的光保护机制有关。 LHCII和OECC、LHCII和pdOE在脂质体中都成功的实现了重组,而且在结构和功能上没有明显差异;表明小天线以及23 kDa、17 kDa蛋白可能不是LHCII和核心复合物结合及能量传递所必需的。 2、受体侧光破坏和色素置换对PSII RC的影响: 在800 μmol.m-2 .s-1光照和无外加电子受体、供体的情况下,研究了PSII RC色素的受体侧光破坏情况。Chl a、Pheo和β-Car的光漂白几乎同时发生,其中在680 nm吸收的色素破坏最为显著,670 nm吸收的外周Chl比其他色素更加稳定。荧光发射强度呈先升高后降低的趋势,最大发射峰位逐渐蓝移,表明色素之间的能量传递受到破坏。用β-Car的主要吸收波长488 nm和514.5 nm激发得到两组谱带峰位和强度不同的拉曼光谱,表明在PSII RC中存在两个光谱性质不同的β-Car。光破坏过程中两组谱带的位置和带宽都没有明显变化,表明β-Car的光保护机制不涉及自身构象的变化。 将PSII RC与Cu-Chl a进行色素置换,得到了与Cu-Chl重组的RC(Cu-Chl-RC),含有0.5 Cu-Chl/2Pheo。与对照RC(按同样方式与Chl a置换的RC)和天然RC相比,Cu-Chl含量增加而Chl含量减少,660 nm的吸收增加而670 nm吸收降低,因此推测是外周Chl被替换。色素置换过程对RC的多肽组分及大部分的P680活性没有影响,CD光谱的变化也很小,表明产生CD信号的色素和蛋白环境也没有受到明显影响。但是Cu-Chl-RC的荧光发射强度明显降低,最大发射峰蓝移且峰形发生变化,Cu-Chl可能在重组RC中作为激发态的淬灭剂,阻碍了色素之间的能量传递。

光系统I(photosystem I,PSI)是光合膜上参与光合作用原初反应过程的主要膜蛋白超分子复合体之一。高等植物的PSI是由核心复合体(14个亚基)和捕光色素蛋白复合体I(light-harvesting complex I, LHCI,含4个Lhca蛋白)组成的超分子复合体,它的主要功能是调节光诱导的从囊腔侧的质体兰素(plastocyanin,PC)向基质侧的铁氧还蛋白(ferredoxin,Fd)的电子传递。研究PSI的结构与功能对于揭示植物光合作用高效吸能、传能的分子机理具有重要意义。在本文中,我们首先建立了分离制备PSI及其亚组分的方法(Qin et al., 2007),并在此基础上对PSI在强光破坏的过程中结构与功能的变化进行了比较深入的研究。本论文的主要研究结果如下: 1.快速、高效分离纯化PSI及其亚组分方法的建立。 国际上传统的PSI分离方法(Bassi and Simpson, 1987; Croce et al., 1998; Påsllon et al.1995; Schmid et al. 2002),耗时长较长(分离PSI颗粒一般需要多于20h的蔗糖超速离心过程,而分离PSI的亚组分则需要25-60h的蔗糖超速离心过程)、得率较低,这不便于PSI方面的研究,为此我们首先改进了传统的分离纯化方法。新方法以高等植物菠菜叶片作为原材料,使用Triton X-100作为增溶剂,通过差速离心技术获得的粗制品,然后使用十二烷基麦芽糖苷(n-Dodecyl β-D-maltoside, DDM)增溶PSI粗制品,之后采用100,000×g,垂直转头(Beckman VTi 50)0.1-1 mol/L蔗糖梯度离心3h获得纯度较高的PSI颗粒。然后使用DDM和3-(N, N-Dimethylpalmitylammonio) propanesulfonate (zw 3-16)两种增溶剂处理PSI,后经100,000×g,垂直转头(Beckman VTi 50)蔗糖梯度离心4h获得纯度较高的PSI core、LHCI-680、LHCI-730复合体。采用吸收光谱、荧光光谱技术研究了各样品的基本光谱学特性,采用HPLC分析了各样品的色素组成,结果显示平均每个Lhca蛋白结合1.5-1.6黄体素,1.0紫黄质, 0.8-1.1 β-胡萝卜素,该方法制备的LHCI比传统方法制备的LHCI减少了类胡萝卜素的丢失。这一工作为以后结构与功能的研究工作奠定了良好的基础。 2.PSI复合体及其亚组分的特性研究。 PSI颗粒具有一定的适应环境酸碱变化的能力,在我们的试验条件下PSI颗粒在pH 5-10相对稳定。PSI、LHCI很难通过加入Mg2+、Ca2+、Na+阳离子聚集沉淀。经绿胶鉴定我们制备的LHCI-680、LHCI-730是二聚体形式;而把PSI绿胶后再进行第二向十二烷基硫酸钠-聚丙烯酰氨凝胶电泳(SDS-PAGE)电泳,结果发现在稍强烈的绿胶增溶条件下,LHCI-730是以二聚体的形式存在,但是LHCI-680却是以单体的形式出现。这说明LHCI形成的二聚体,尤其是LHCI-680,较容易受到增溶处理而分离成单体形式,解释了以生化分离手段得到的LHCI-680的聚集形式是单体还是二聚体这个目前国际上还有有争议的问题。 3.PSI、LHCI光破坏的基本特点。 采用白光(2500 μmol•m-2•s-1)照射PSI颗粒,通过SDS-PAGE及室温吸收光谱检测光照过程中PSI复合体的变化,结果表明:去氧处理能够大大延缓PSI的光破坏,而PSI脱辅基蛋白不会发生光破坏,这说明PSI发生的光破坏可能与Chl与O2的相互作用有关。采用白光(1000 μmol•m-2•s-1、300 μmol•m-2•s-1)处理LHCI-680、LHCI-730,发现LHCI-680被破坏的速度明显快于LHCI-730被破坏的速度,这是首次在体外分离的水平上揭示了不同LHCI光破坏方面的差异。LHCI-680与LHCI-730在光破坏方面的差异可能与两种天线蛋白结合的类胡萝卜素的种类和数量不同有关,还可能与二者结合的长波长Chl的情况有关,但是具体的原因还有待于进一步的研究。 4.结合不同的捕光色素蛋白复合体(light-harvesting complex,LHC)对PSI光破坏的影响。 为了研究结合不同的捕光天线对PSI光破坏的影响,我们制备了PSI-LHCII、PSI、PSI core三种复合体。使用白光(2500 μmol•m-2•s-1)照射这三种复合体,并通过测定各复合体在光破坏过程中蛋白亚基、吸收光谱、PSI活性及P700含量的变化,对比三者光破坏的速度,结果发现PSI-LHCII在这三种复合体中光破坏速度最快,而PSI和PSI core两种复合体光破坏速度基本一致。我们推测在光照过程中部分光系统II捕光Chl a/b蛋白复合体II(light-harvesting complex II,LHCII)能够向PSI core传递能量,另外PSI-LHCII绿胶分析的结果表明发生了LHCII三聚体向单体的转变,这种强光下发生的LHCII聚合形式的转化可能是高光强下调节光能捕获的一种机制,由于植物体内具有较完整的保护系统,体内PSI-LHCII的光破坏可能与体外情况不同;另外LHCI与PSI core的解离可能发生在强光照射的早期,具有保护PSI core减少光破坏的积极作用。该部分的研究首次观察了结合不同的捕光天线对PSI光破坏的影响。

Three different forms of PS I complexes were isolated from a siphonous marine green alga, Codium fragile, by Triton X-100 sucrose gradient centrifugation. Zone III had a Chl a/b>20, and designated as PS I. core complex CC I because it created only CP I band in mild PAGE. Zone IV and V had absorption at 436 and 674 nm, 467 and 650 nm, and 540 nm, suggesting the presence of Chl a, Chl b, siphonaxanthin and siphonein, Chl a/b were 3.23 and 2.4, respectively. Both CP I and CP I a bands were observed when they were subjected to mild PAGE. Therefore, Zone IV and V were different forms of PS I complexes that consisted of CC I and different amount of light-harvesting complex LHC I. Zone III contained only 66 and 56 ku peptides in SDS-PAGE, while Zone IV and V had 4 different LHC I peptides of 25, 26, 26.2 and 27.5 ku in addition to 66, 56 ku peptides. Fluorescence emission spectra showed that efficient energy transfer were kept among pigments in isolated PS I complexes. Excitation energy absorbed by Chl b, siphonaxanthin and siphonein can be transferred to Chl a.

Three cDNA sequences encoding four SNARE (N-ethylmaleimide-sensitive fusion protein attachment protein receptors) motifs were cloned from sea perch, and the deduced peptide sequences were analyzed for structural prediction by using 14 different web servers and softwares. The "ionic layer" structure, the three dimensional extension and conformational characters of the SNARE 7S core complex by using bioinformatics approaches were compared respectively with those from mammalian X-ray crystallographic investigations. The result suggested that the formation and stabilization of fish SNARE core complex might be driven by hydrophobic association, hydrogen bond among R group of core amino acids and electrostatic attraction at molecular level. This revealed that the SNARE proteins interaction of the fish may share the same molecular mechanism with that of mammal, indicating the universality and solidity of SNARE core complex theory. This work is also an attempt to get the protein 3D structural information which appears to be similar to that obtained through X-ray crystallography, only by using computerized approaches. Granulites from Huangtuling in the North Dabie metamorphic core complex in eastern China preserve rare mineralogical and mineral chemical evidence for multistage metamorphism related to Paleoproterozoic metamorphic processes, Triassic continental subduction-collision and Cretaceous collapse of the Dabieshan Orogen. Six stages of metamorphism are established, based on detailed mineralogical and petrological studies: (I) amphibolite facies (6.3–7.0 kbar, 520–550 °C); (II) high-pressure/high-temperature granulite facies (12–15.5 kbar, 920–980 °C); (III) cooling and decompression (4.8–6.0 kbar, 630-700 °C); (IV) medium-pressure granulite facies (7.7–9.0 kbar, 690–790 °C); (V) low-pressure/high-temperature granulite facies (4.0–4.7 kbar, 860–920 °C); (VI) retrograde greenschist facies overprint (1–2 kbar, 340–370 °C). The P–T history derived in this study and existing geochronological data indicate that the Huangtuling granulite records two cycles of orogenic crustal thickening events. The earlier three stages of metamorphism define a clockwise P–T path, implying crustal thickening and thinning events, possibly related to the assembly and breakup of the Columbia Supercontinent ca. 2000 Ma. Stage IV metamorphism indicates another crustal thickening event, which is attributed to the Triassic subduction/collision between the Yangtze and Sino–Korean Cratons. The dry lower crustal granulite persisted metastable during the Triassic subduction/collision due to lack of hydrous fluid and deformation. Stage V metamorphism records the Cretaceous collapse of the Dabieshan Orogen,possibly due to asthenosphere upwelling or removal of the lithospheric mantle resulting in heating of the granulite and partial melting of the North Dabie metamorphic core complex. Comparison of the Huangtuling granulite in North Dabie and the high-pressure (HP)–ultrahigh-pressure (UHP) metamorphic rocks in South Dabie indicates that the subducted upper (South Dabie) and lower (North Dabie) continental crusts underwent contrasting tectonometamorphic evolution during continental subduction–collision and orogenic collapse. High-pressure granulites are generally characterized by the absence of orthopyroxene. However, the Huangtuling felsic granulite rarely preserves the high-pressure granulite facies assemblage of garnet + orthopyroxene + biotite + plagioclase + K-feldspar + quartz. To investigate the effects of bulk rock composition on the stability of orthopyroxene-bearing, high-pressure granulite facies assemblages in the NCKFMASHTO (Na2O–CaO–K2O–FeO–MgO–Al2O3–SiO2–H2O–TiO2–Fe2O3) system, we constructed a series of P–T–X pseudosections based on the melt-reintegrated composition of the Huangtuling felsic high-pressure granulite. Our calculations demonstrate that the orthopyroxene-bearing, high-pressure granulite facies assemblages are restricted to low XAl [Al2O3/(Na2O + CaO + K2O + FeO + MgO + Al2O3) < 0.35, mole proportion] or high XMg [MgO/(MgO + FeO) > 0.85] felsic–metapelitic rock types. Our study also reveals that the XAl values in the residual felsic–metapelitic, high-pressure granulites could be significantly reduced by a high proportion of melt loss. We suggest that orthopyroxene-bearing high-pressure granulites occur in residual overthickened crustal basement under continental subduction–collision zones and arc–continent collision belts.

Anew class of bifunctional architecture combining the useful functions of superparamagnetism and terbium complex luminescence into one material has been prepared via two main steps by a modified Stober method and the layer-by-layer (LbL) assembly technique. The obtained bifunctional nanocomposites exhibit superparamagnetic behavior, high fluorescence intensity, and color purity. The architecture has been characterized by field-emission scanning electron microscopy (FE-SEM), transmission electron microscopy (TEM), UV-vis absorption and emission spectroscopy, X-ray diffraction, and superconducting quantum interference device (SQUID) magnetometry.

We successfully prepared a new kind of thermoresponsive and fluorescent complex of Tb(III) and PNIPAM-g-P(NIPAM-co-St) (PNNS) core-shell nanoparticle. It was found that Tb(III) mainly bonded to 0 of the carbonyl groups of PNNS, forming the novel (PNIPAM-g-P(NIPAM-co-St))-Tb(III) (PNNS-Tb(III)) complex. The maximum emission intensity of the complex at 545 nm is enhanced about 223 times comparing to that of the pure Tb(III). The intramolecular energy transfer efficiency from PNNS to Tb(III) reaches 50%. When the weight ratio of Tb(III) and the PNNS-Tb(III) complex is 1.2 wt.%, the enhancement of the emission fluorescence intensity at 545 nm is highest.

This work represents the nucleotide sequence of the core histone gene cluster from scallop Chlamys farreri. The tandemly repeated unit of 5671 bp containing a copy of the four core histone genes H4, H2B, H2A and H3 was amplified and identified by the techniques of homology cloning and genomic DNA walking. All the histone genes in the cluster had the structures in their 3' flanking region which related to the evolution of histone gene expression patterns throughout the cell cycle, including two different termination signals, the hairpin structure and at least one AATAAA polyadenylation signal. In their 5' region, the transcription initiation sites with a conserved sequence of 5'-PyATTCPu-3' known as the CAP site were present in all genes except to H2B, generally 37-45 bp upstream of the start code. Canonical TATA and CAAT boxes were identified only in certain histone genes. In the case of the promoters of H2B and H2A genes, there was a 5'-GATCC-3' element, which had been found to be essential to start transcription at the appropriate site. After this element, in the promoter of H2B, there was another sequence, 5'-GGATCGAAACGTTC-3', which was similar to the consensus sequence of 5'-GGAATAAACGTATTC-3' corresponding to the H2B-specific promoter element. The presence of enhancer sequences (5'-TGATATATG-3') was identified from the H4 and H3 genes, matching perfectly with the consensus sequence defined for histone genes. There were several slightly more complex repetitive DNA in the intergene regions. The presence of the series of conserved sequences and reiterated sequences was consistent with the view that mollusc histone gene cluster arose by duplicating of an ancestral precursor histone gene, the birth-and-death evolution model with strong purifying selection enabled the histone cluster less variation and more conserved function. Meanwhile, the H2A and the H2B were demonstrated to be potential good marks for phylogenetic analysis. All the results will be contributed to the characterization of repeating histone gene families in molluscs.

A near-infrared luminescent macroporous material (PL-Macromaterial) and a near-infrared luminescent/magnetic bifunctional macroporous material (MML-Macromaterial) were synthesized by using polystyrene microspheres (PS) and Fe3O4 @polystyrene core-shell nanoparticles (Fe3O4@PS), respectively, as templates. Both the PL-Macromaterial and the M/PL-Macromaterial show the characteristic emission of the Er 3, ion. Moreover, the M/PL-Macromaterial possesses superparamagnetic properties at room temperature.

A multinary molecular nanocluster, in which a T3 supertetrahedral [Sn4Ga4Zn2Se20](8-) core was neutralized and covalently terminated by four [(TEPA)Mn](2+) (TEPA = tetraethylenepentamine) metal complexes, was synthesized and characterized. The cluster is assembled into, through hydrogen bonding and van de Waals forces, a superlattice that is chemically stable and free of strong covalent coupling. The four different cations were distributed within the cluster in such a manner that both the local charge balance and global charge compensation by the metal complex could be satisfied.

By fusing an electron-deficient ring system with the phenyl ring of a 2-phenylpyridine (ppy)-type ligand, a new and synthetically versatile strategy for the phosphorescence color tuning of cyclometalated iridium(III) and platinum(II) metallophosphors has been established. Two robust red electrophosphors with enhanced electron-injection/electron-transporting features were prepared by using an electron-trapping fluoren-9-one chromophore in the ligand design. The thermal, photophysical, redox and electrophosphorescent properties of these complexes are reported. These exciting results can be attributed to a switch of the metal-to-ligand charge-transfer (MLCT) character of the transition from the pyridyl groups in the traditional Ir-III or Pt-II ppy-type complexes to the electron-deficient ring core, and the spectral assignments corroborate well with the electrochemical data as well as the timedependent density functional theory (TD-DFT) calculations. The electron-withdrawing character of the fused ring results in much more stable MLCT states, inducing a substantial red-shift of the triplet emission energy from yellow to red for the Ir-III complex and even green to red for the PtII counterpart.

Substantial progress has been made recently in extending the supramolecular assembly of biomimetic structures to vesicle-based sophisticated nanocomposites and mesostructures. We report herein the successful preparation of unilamellar surfactant vesicles coated with a monolayer of ring-shaped {Mo-154} polyoxometalate (POM) nanoclusters, (NH4)(28)[Mo-154 (NO)(14)O(448)Hi(4)(H2O)(70)].approximate to 350H(2)O, by coulomb attractions using preformed didodecyldimethylammonium bromide (DDAB) surfactant vesicles as templates. The resultant vesicle-templated supramolecular assemblies are robust (they do not disintegrate upon dehydration) both at room-temperature ambient and vacuum conditions, as characterized by conventional transmission electron microscopy (TEM) and atomic force microscopy (AFM). The flexibility of the complex soft assemblies was also revealed by AFM measurements. The effect of POM-vesicle coulomb attractions on the dimensions of the templating vesicles was also investigated by using dynamic light scattering (DLS).Although origins of the structure stability of the as-prepared supramolecular assemblies are not clear yet, the nanometer scale cavities and the related properties of macroions of the POM clusters may play an important role in it.

Silver nanoparticles were synthesized by the use of a two-armed polymer with a crown ether core [poly(styrene)]-dibenzo-18-crown-6-[poly(styrene)] based on the flexibility of the polymer chains and the complex effect of crown ether with Ag+ and Ag. The size of silver nanoparticles could be tailored by controlling the initial concentrations of the polymer and Ag+, and the molecular weight of the polymer. The emission of silver nanoparticles was blue-shifted, and the intensity of the photoluminescence of silver nanoparticles stabilized by the polymer was significantly increased due to the complex effect between the crown ether embedded in the polymer and the silver nanoparticles.
