Nectogaline shrews are a major component of the small mammalian fauna of Europe and Asia, and are notable for their diverse ecology, including utilization of aquatic habitats. So far, molecular phylogenetic analyses including nectogaline species have been unable to infer a well-resolved, well-supported phylogeny, thus limiting the power of comparative evolutionary and ecological analyses of the group. Here, we employ Bayesian phylogenetic analyses of eight mitochondrial and three nuclear genes to infer the phylogenetic relationships of nectogaline shrews. We subsequently use this phylogeny to assess the genetic diversity within the genus Episoriculus, and determine whether adaptation to aquatic habitats evolved independently multiple times. Moreover, we both analyze the fossil record and employ Bayesian relaxed clock divergence dating analyses of DNA to assess the impact of historical global climate change on the biogeography of Nectogalini. We infer strong support for the polyphyly of the genus Episoriculus. We also find strong evidence that the ability to heavily utilize aquatic habitats evolved independently in both Neomys and Chimarrogale + Nectogale lineages. Our Bayesian molecular divergence analysis suggests that the early history of Nectogalini is characterized by a rapid radiation at the Miocene/Pliocene boundary, thus potentially explaining the lack of resolution at the base of the tree. Finally, we find evidence that nectogalines once inhabited northern latitudes, but the global cooling and desiccating events at the Miocene/Pliocene and Pliocene/Pleistocene boundaries and Pleistocene glaciation resulted in the migration of most Nectogalini lineages to their present day southern distribution. Many systematic relationships among Chinese white-toothed shrews of genus Crocidura are presently unresolved. In this paper, a taxonomic revision of Crocidura from Southern China is presented. We studied 338 specimens from Burma, China, Korea, Pakistan, Turkey, Middle and Central Asia, and Russia (Appendix I), 285 of which had complete skulls that were analyzed with principal component and discriminant analyses. Results indicated that 6 species of Crocidura can be recognized in South China. C. fuliginosa occurs in Southwestern and Eastern China, C. attenuata is broadly distributed throughout Southern China, and C. horsfieldii is restricted to the southern part of China. C vorax and C. rapax, usually placed as synonyms of the European C russula, are recognized as 2 valid species whose ranges overlap in Southwestern China. C. shantungensis of Eastern Asia extends to the northern part of Southern China and is distinct from C. suaveolens and C. gmelini of Middle and Central Asia, respectively.

Background: Flying lemurs or Colugos (order Dermoptera) represent an ancient mammalian lineage that contains only two extant species. Although molecular evidence strongly supports that the orders Dermoptera, Scandentia, Lagomorpha, Rodentia and Primates form a superordinal clade called Supraprimates (or Euarchontoglires), the phylogenetic placement of Dermoptera within Supraprimates remains ambiguous. Results: To search for cytogenetic signatures that could help to clarify the evolutionary affinities within this superordinal group, we have established a genome-wide comparative map between human and the Malayan flying lemur (Galeopterus variegatus) by reciprocal chromosome painting using both human and G. variegatus chromosome-specific probes. The 22 human autosomal paints and the X chromosome paint defined 44 homologous segments in the G. variegatus genome. A putative inversion on GVA 11 was revealed by the hybridization patterns of human chromosome probes 16 and 19. Fifteen associations of human chromosome segments (HSA) were detected in the G. variegatus genome: HSA1/3, 1/10, 2/21, 3/ 21, 4/8, 4/18, 7/15, 7/16, 7/19, 10/16, 12/22 (twice), 14/15, 16/19 (twice). Reverse painting of G. variegatus chromosome-specific paints onto human chromosomes confirmed the above results, and defined the origin of the homologous human chromosomal segments in these associations. In total, G. variegatus paints revealed 49 homologous chromosomal segments in the HSA genome. Conclusion: Comparative analysis of our map with published maps from representative species of other placental orders, including Scandentia, Primates, Lagomorpha and Rodentia, suggests a signature rearrangement (HSA2q/21 association) that links Scandentia and Dermoptera to one sister clade. Our results thus provide new evidence for the hypothesis that Scandentia and Dermoptera have a closer phylogenetic relationship to each other than either of them has to Primates.

Cross-species painting (fluorescence in situ hybridization) with 23 human (Homo sapiens (HSA)) chromosome-specific painting probes (HSA 1-22 and the X) was used to delimit regions of homology on the chromosomes of the golden mole (Ghrysochloris asiaticus) and elephant-shrew (Elephantulus rupestris). A cladistic interpretation of our data provides evidence of two unique associations, HSA 1/19p and 5/21/3, that support Afrotheria. The recognition of HSA 5/3/21 expands on the 3/21 synteny originally designated as an ancestral state for all eutherians. We have identified one adjacent segment combination (HSA2/8p/4) that is supportive of Afroinsectiphillia (aardvark, golden mole, elephant-shrew). Two segmental combinations (HSA 10q/17 and HSA 3/20) unite the aardvark and elephant-shrews as sister taxa. The finding that segmental syntenies in evolutionarily distant taxa can improve phylogenetic resolution suggests that they may be useful for testing sequence-based phylogenies of the early eutherian mammals. They may even suggest clades that sequence trees are not recovering with any consistency and thus encourage the search for additional rare genomic changes among afrotheres.

在实验室笼养条件下将28只健康成年树鼬按1:2或1:3的雄雌比例配对饲养,并每隔3周进行一次检查,通过一年的试验饲养,研究结果表明,这批实验树鼬共受孕45次,分娩37窝,流产2窝,死产3窝,总产仔147只,成活139只,另有3只母树鼦怀孕待产.每只母树鼩产仔2~3窝,每窝2~5仔,幼仔初生体重为(10.4±1.2)g,40日龄断奶体重为(66.2±15.7)g.该方法树鼩繁殖率和成活率极高,成本相对较低且操作简单,是一种值得参考和借鉴的新方法.

乙型肝炎病毒(Hepatitis Virus type B, HBV)感染是一种严重威胁人类健康的世界性问题。由于HBV感染具有宿主和组织特异性,长期以来一直缺乏有效的体外感染模型,严重制约了HBV感染机制和治疗药物的研究。树鼩(Tree Shrews, Tupaia Belangeri)与人和灵长类亲缘关系较为接近,作为小动物模型在乙型肝炎研究中备受关注。但由于各种原因的限制,HBV树鼩感染模型仍处于“国际公认但实际短缺,国内首创但实际空缺”的状况。因此,复建和优化HBV树鼩体内外感染模型成为我们长期研究方向所必备的前提基础和开端。而树鼩原代肝细胞的分离和培养则是建立HBV体外感染模型的关键的第一步。我们通过与机械分离法的直接比较,验证了两步灌注法在树鼩肝细胞分离中的优越性。进而发现,在分离后的体外培养过程中,二甲基亚砜不仅能够促进和维持原代肝细胞的分化,而且能够显著地抑制纤维状细胞群的出现。同时,肝细胞生长因子(HGF)和表皮生长因子(EGF)能够促进肝细胞在体外长期存活。在此优化的条件下,原代培养可持续4-5周,并且较多的细胞聚集形成类似肝窦结构的形态,从而成功地复制和改进了树鼩原代肝细胞培养体系,为建立HBV体外感染模型提供了基础。与此同时,我们采集了2种来源的HBV,即以乙肝病人血清和HepG2.2.15细胞培养上清为来源。我们采用蔗糖浓度梯度离心法以纯化病人血清病毒,能较好地将HBV颗粒和亚病毒颗粒区分开来。在收集HepG2.2.15细胞株来源的病毒时,我们详细研究了HepG2.2.15细胞的生长曲线和病毒产量曲线,找到病毒产量与细胞生长状态之间的联系,确定了最佳收集病毒时间,并通过放大培养条件,大量收集HepG2.2.15细胞的培养上清。我们采用浓缩和未浓缩的HepG2.2.15病毒、纯化的血清病毒等3种方式制备的病毒感染树鼩原代肝细胞,通过不同水平的指标来检测感染结果。首先,细胞内HBVx基因mRNA的检测结果表明,3种来源的病毒都可以感染树鼩原代肝细胞。同时,细胞上清中分泌的病毒颗粒及病毒蛋白的检测结果表明,HepG2.2.15病毒感染树鼩原代肝细胞的效果比血清病毒的感染效果好。免疫荧光的检测结果进一步证实了HepG2.2.15病毒能够感染树鼩原代肝细胞。因此,在我们的实验中,树鼩原代肝细胞对2种来源和不同制备方式的病毒均具有易感性,且以未浓缩的HepG2.2.15上清病毒感染力优于血清病毒。综上所述,我们现有的结果验证了树鼩原代肝细胞对HBV具有易感性。就感染效率而言,尽管感染效率依赖于病毒滴度及其感染力,我们在病毒细胞比例为0.02:1的条件下,仍能确认易感性,并获得了与国际同类研究相当的感染效率。就本研究的应用前景而言,我们所建立的HBV体外感染模型,能够作为实验室的通用平台,用以来研究HBV的感染机制和抗病毒药物的筛选与评价。

目前多数学者认为树鼩(Tree shrews)是灵长目攀居目树鼩科的一种小型哺乳动物。因其具有体型小、易饲养、价格低、新陈代谢和解剖结构与人类非常接近等优点,树鼩已经成为多种人类疾病研究的良好动物模型。树鼩作为乙型肝炎病毒(HBV)感染的动物模型已经被广泛应用。 自然杀伤性T细胞(Natrual killer T cells, NKT cells)可同时表达NK细胞和T细胞的表面标记,是CD1d限制性的特殊的淋巴细胞亚群,在机体的免疫反应中起重要作用。研究表明,在HBV感染及乙型肝炎的发病过程中,NKT细胞既有抑制和清除病毒的作用,也有加重肝炎及相关病症的负作用。因此,对NKT细胞的研究及恰当地调节将有助于理解和解决在研究乙肝防治中遇到的问题,但在树鼩中,至今尚无NKT细胞的相关报道。 目前公认的检测NKT细胞的手段是用加载了α-GalCer的CD1d四聚体检测。但至今尚无树鼩特异的相关抗体,给树鼩NKT细胞的研究造成很大障碍。因此,获得树鼩特异的CD1d四聚体是研究其NKT细胞特征和功能的前提。本论文作为其中一部分,目的是克隆树鼩CD1d编码区全长序列,为今后抗体的制备奠定基础。 本论文中,我们完成了相关工作并得到了以下结果:1、首次克隆得到了树鼩CD1d基因编码区的全长序列(1002bp),并预测了其氨基酸序列;2、通过比对分析得知其与灵长类的CD1d同源性较高,达到80%以上;同时,通过系统进化树分析,得知树鼩与灵长类亲缘关系较其它物种更近;3、首次分析了树鼩的CD1d mRNA在不同组织中的表达情况,发现树鼩的肝,肺中CD1d表达量较高,外周血和脾脏中也有少量表达。 总之,我们首次克隆得到了树鼩CD1d的编码区的全长序列,为今后制备CD1d四聚体进而研究树鼩的NKT细胞奠定了基础;同时首次分析了CD1d mRNA在树鼩的部分组织中的表达情况,结合以往其它物种中CD1d组织分布情况的检测结果,为树鼩中CD1d分子的研究提供了材料和数据基础。
