染色体黏着是有丝分裂和减数分裂的关键事件,是保证姊妹(或同源)染色体正确分离并分配到子细胞中的关键调控环节之一,它建立于细胞分裂前的S期将新复制的姊妹染色体紧密联系在一起。来自酵母的研究结果已经证明姊妹染色体之间的黏着是由多亚基的蛋白质复合体-黏着素所介导的。在芽殖酵母有丝分裂中,黏着素由Scc1,Scc3,Smc1和Smc3四个亚基组成。减数分裂黏着素的组成与有丝分裂中的相似,只是Scc1被其减数分裂特异的Rec8变体所替换。目前,已经从高等真核生物线虫,果蝇,人,鼠以及拟南芥中分离到了黏着素相关的基因,但是对于这些基因在高等真核生物特别是植物细胞分裂中的功能还知之甚少。即使在酵母中人们对于减数分裂和有丝分裂过程中有关染色体黏着与分离的许多基本问题仍然不清楚,而且许多现象表明减数分裂的详细机制在各种生物中存在重大差异。 我们通过同源克隆的方法证明水稻(和拟南芥)基因组编码4个RAD21/REC8-like基因。这4个基因均以单拷贝存在,在核苷酸水平上没有相似性。它们所编码的蛋白质的相似性主要局限于其N-末端结构域和C-末端结构域。这4个蛋白质的中间区域没有(或者仅有极低的)相似性,但是中间区域都含有潜在的核定位信号,PEST序列,分离酶的识别序列以及多个磷酸化位点。 半定量RT-PCR,原位杂交以及Western杂交结果显示这4个基因都在生殖器官中优势表达,但是它们在花发育过程中的表达动态是不同的。OsRAD21-1和OsRAD21-3都在减数分裂时期的颖花中表达量最高,但是OsRAD21-3还在成熟花粉中高表达;OsRAD21-4在减数分裂前的颖花中表达量最高;OsRAD21-2则在雌雄蕊形成时期表达最强,之后逐渐降低。这些结果暗示这4个基因的功能可能是不同的。 免疫荧光定位分析表明,OsRad21-1和OsRad21-3 特异地定位于有丝分裂的染色体上,其分布动态表明这两个蛋白可能都参与了有丝分裂姊妹染色体之间的黏着。由于水稻四个RAD21/REC8类基因中,只有OsRAD21-3在花粉发育过程中表达,同时水稻花粉的发育成熟要经过两次有丝分裂,推测OsRad21-3蛋白可能参与这两次有丝分裂过程姊妹染色体之间的黏着。OsRad21-4则特异地定位于减数分裂前间期到中期Ⅰ的染色体上,说明它可能特异地介导减数分裂过程姊妹染色体之间的黏着。与其它已知的Rad21/Rec8-like蛋白不同,不论在有丝分裂还是在减数分裂过程中,OsRad21-2蛋白都不定位于染色体上而是特异地定位在核仁中,并且它的动态变化与核仁重建和解体的动态规律在时间上也是相一致的,这说明OsRad21-2是一种新的核仁蛋白质而与染色体的黏着无关。 OsRAD21-4 RNAi转基因水稻植株的花粉活性受到严重影响,种子结实率降低。雄性减数分裂过程中染色体出现多种异常行为:前期Ⅰ染色体异常凝集;同源染色体提早分离;染色体出现片断化。进一步的FISH实验结果证明RNAi株系中同源染色体配对和姊妹染色体臂的黏着均发生异常。因此,OsRad21-4是酵母Rec8的同源蛋白,是正确的减数分裂所必需的。 与表达分析和功能分析所得的结果相一致,进化树分析可以将Rad21/Rec8-like蛋白质分为三个亚家族:(1)Rad21亚家族,参与有丝分裂姊妹染色体黏着;(2)Rec8亚家族,参与减数分裂染色体黏着;(3)Rno亚家族,目前仅发现于高等植物中,是一种核仁蛋白质而与其它的Rad21/Rec8-like蛋白的功能不同,可能不参与染色体之间的黏着。

The genetic diversity and phylogeny of 26 isolates of Bursaphelenchus xlophilus from China, Japan, Portugal and North America were investigated based on the D2/3 domain of 28S rDNA, nuclear ribosomal Internal Transcribed Spacer (ITS) sequences, and random amplified polymorphic DNA (RAPD) analysis. The genetic diversity analysis showed that the D2/3 domain of 28S rDNA of isolates of B. xlophilus from China, Portugal, Japan and the US were identical and differed at one to three nucleotides compared to those from Canada. ITS sequences of isolates from China and Portugal were the same; they differed at one or two nucleotides compared to those of Japanese isolates and at four and 23 nucleotides compared to those front the US and Canada, respectively. The phylogenetic analysis indicated that Chinese isolates share a common ancestor with one of the two Japanese clades and that the Canadian isolates form a sister group of the clade comprised of isolates from China, Portugal,Japan, and the US. The relationship between Japanese isolates and those from China was closer than with the American isolates. The Canadian isolates were the basal group of B. xylophilus. This suggests that B. xlophilus originated in North America and that the B. xylphilus that occurs in China could have been first introduced from Japan. Further analysis based on RAPD analysis revealed that the relationship among isolates from Guangdong, Zhejiang, Shandong, Anhui provinces and Nanjing was the closest, which suggests that pine wilt disease in these Chinese locales was probably dispersed from Nanjing, where this disease first occurred in China.

2005年7~8月以贵州省福泉市草坪与农田、灌草丛为2个调查样地,对采集的大量昆虫标本进行检索、分类、统计处理、区系分析.采集到昆虫标本50 320只,分隶于8目、29科、67属、70种,其中,草坪害虫有8目、19科、29属、32种;农田、灌草丛有8目、28科、56属、61种.研究发现,2个样地中害虫群落种类组成和结构基本相似;鳞翅目害虫(除地老虎等个别种外)是地面害虫的优势类群;鞘翅目害虫是地下害虫的优势类群;草坪害虫区系主要由周边农田,园林和其他天然植被中害虫的迁入形成.

We have cloned a mouse homologue (designated Myak) of the yeast protein kinase YAK1. The 1210 aa open reading frame contains a putative protein kinase domain, nuclear localization sequences and PEST sequences. Myak appears to be a member of a growing family of YAK1-related genes that include Drosophila and human Minibrain as well as a recently identified rat gene ANPK that encode a steroid hormone receptor interacting protein. RNA blot analysis revealed that Myak is expressed at low levels ubiquitously but at high levels in reproductive tissues, including testis, epididymis, ovary, uterus, and mammary gland, as well as in brain and kidney. In situ hybridization analysis on selected tissues revealed that Myak is particularly abundant in the hormonally modulated epithelia of the epididymis, mammary gland, and uterus, in round spermatids in the testis, and in the corpora lutea in the ovary, Myak is also highly expressed in the aqueduct of the adult brain and in the brain and spinal cord of day 12.5 embryos, Mol. The green peach aphid, Myzus persicae, is a major pest of tobacco, Nicotiana tabacum, in Yunnan province, China, where its control still depends on the use of insecticides. In recent years, the local government and farmers have sought to improve the biological control of this tobacco pest. In this paper, we present methods for mass rearing Aphidius gifuensis, a dominant endoparasitoid of M. persicae on tobacco plants in this region. The tobacco cultivar K326 (N. tabacum) was used as the host plant and M. persicae as the host insect. In the greenhouse, we collected tobacco seedlings for about 35 days (i.e., until the six-true-leaf stage), transferred them to 7.5-cm diameter pots, and kept these plants in the greenhouse for another 18 days. These pots were then transferred to an insectary-greenhouse, where the tobacco seedlings were inoculated with five to seven wingless adult M. persicae per pot. After 3 days, the infested seedlings were moved to a second greenhouse to allow the aphid population to increase, and after an additional 4 +/- 1 days when 182 +/- 4.25 aphid adults and nymphs were produced per pot, they were inoculated with A. gifuensis. With this rearing system, we were able to produce 256 +/- 8.8 aphid mummies per pot, with an emergence rate of 95.6 +/- 2.45%; 69% were females. The daily cost of parasite production (recurring costs only) was US$ 0.06 per 1000 aphid mummies. With this technique, we released 109 800 parasitoids in 1998, 196 000 in 1999, 780 000 in 2000, and 5 600 000 in 2001 during a 2-month period each year This production method is discussed with respect to countrywide usage in biological control and integrated control of M. persicae.

This study was designed to determine cytotoxic effects of PBDE-47 and HBCDs individually or with a mixture of both compounds exposure to Hep G2 cells. The results showed PBDE-47 and HBCDs induced increase of nitric oxide synthase (NOS) activity, release of NO. dissipation of mitochondria membrane potential and cell apoptosis. Exposure to HBCDs induced ROS formation. Moreover, preincubation with PTIO (NO scavanger) and N-acetylcysteine (ROS scavanger) partially reversed cytotoxic effects of these compounds. The possible mechanism is that PBDE-47 and HBCDs could boost generation of NO and/or ROS, impact mitochondria, and result in start-ups of apoptosis program. Cells exposed to mixture of both compounds and each of them showed non-apoptotic rate significant difference, but the combination of them caused more adverse effects on cells. These results Suggest that PBDE-47 and HBCDs in single and complex exposure have the cytotoxic activity of anti-proliferation and induction of apoptosis in tumor cells in vitro. Data of classification, origin, pathway and environmental impacts of invasive alien micro-organisms, invertebrates, amphibians and reptiles, fish, birds, mammals, weeds, trees, and marine organisms in terrestrial, aquatic and marine ecosystems of China, were analyzed, based on literature retrieval, field survey and consultation. Some 283 invasive alien species were recorded in China, including 19 invasive alien micro-organisms, 18 aquatic plants, 170 terrestrial plants, 25 aquatic invertebrates, 33 terrestrial invertebrates, 3 amphibians and reptiles, 10 fish, and 5 mammals. Of the invasive alien species, 55.1% originated from North and South America, 21.7% from Europe, 9.9% from Asia, 8.1% from Africa and 0.6% from Oceania. Many institutions and individuals in China lack adequate knowledge of ecological and environmental consequences caused by invasive alien species, with some ignorance of the dangerous invasion in the introduction of alien species. For instance, 50.0% of invasive alien plants were intentionally introduced as pasture, feedingstuff, ornamental plants, textile plants, medicinal plants, vegetables, or lawn plants, 25% of alien invasive animals were intentionally introduced for cultivation, ornament, or biological control, In addition, more efforts are being made in the introduction of alien species, and little attention is paid on the management of introduced alien species, which may cause their escape into natural environment and potential threats to the environment. There were also gaps in quarantine system in China. All microorganisms were unintentionally introduced, through timber, seedling, flowerpot, or soil; 76.3% of alien invasive animals invaded through commodity or transportation facility because of the failure of quarantine. Therefore, quarantine measures should be strictly implemented; and meanwhile the intentional introduction of alien species should be strictly managed and a system of risk assessment should be implemented.

P>NF-kappa B is a B-cell specific transcription factor that plays crucial roles in inflammation, immunity, apoptosis, development and differentiation. In the present study, a novel NF-kappa B-like transcription factor Relish was cloned from Chinese mitten crab Eriocheir sinensis (designated as EsRelish) by rapid amplification of cDNA ends (RACE) technique based on expressed sequence tag (EST). The full-length cDNA of EsRelish was of 5034 bp, consisting of a 5' untranslated region (UTR) of 57 bp, a 3' UTR of 1335 bp with two mRNA instability motifs (ATTTA), a polyadenylation signal sequence (AATAAA) and a poly (A) tail, and an open reading frame (ORF) of 3645 bp encoding a polypeptide of 1214 amino acids with a calculated molecular mass of 134.8 kDa and a theoretical isoelectric point of 5.26. There were a typical Rel homology domain (RHD), two nuclear localization signal (NLS) sequences (KR), an inhibitor kappa B (I kappa B)-like domain with six ankyrin repeats, a PEST region and a death domain in the deduced amino acid sequence of EsRelish. Conserved domain, higher similarity with other Rel/NF-kappa Bs and phylogenetic analysis suggested that EsRelish was a member of the NF-kappa B family. Quantitative real-time RT-PCR was employed to detect the mRNA transcripts of EsRelish in different tissues and its temporal expression in hemocytes of E. sinensis challenged with Pichia methanolica and Listonella anguillarum. The EsRelish mRNA was found to be constitutively expressed in a wide range of tissues. It could be mainly detected in the hemocytes, gonad and hepatopancreas, and less degree in the gill, muscle and heart. The expression level of EsRelish mRNA in hemocytes was up-regulated from at 3, 6, 9 and 12 h after P. methanolica challenge. In L. anguillarum challenge, it was up-regulated at 9, 12 and 24 h. The results collectively indicated that EsRelish was potentially involved in the immune response against fungus and bacteria.

Insecticidal activity of proteinous venom from tentacle of jellyfish Rhopilema esculentum Kishinouye was determined against three pest species, Stephanitis pyri Fabriciusa, Aphis medicaginis Koch, and Myzus persicae Sulzer. R. esculentum full proteinous venom had different insecticidal activity against S. pyri Fabriciusa, A. niedicaginis Koch, and M. persicae Sulzer. The 48 It LC50 values were 123.1, 581.6, and 716.3 mu g/mL, respectively. Of the three pests, R. esculentuin full proteinous venom had the most potent toxicity against S. pyri Fabriciusa, and the corrected mortality recorded at 48 It was 97.86%. So, S. pyri Fabriciusa could be a potential target pest of R. esculentum full proteinous venom. (c) 2005 Elsevier Ltd. All rights reserved.

We studied the relationship between genetic diversity of the subterranean Gansu zokor Myospalax cansus and habitat variability in the Loess Plateau, Qinghai Province, China. We used a combination of geographic information systems and molecular techniques to assess the impact of habitat composition and human activities on the genetic diversity of zokor populations in this semi-natural landscape. Although they occurred relatively infrequently in the landscape, woodland and high-coverage grassland habitats were the main positive contributors to the genetic diversity of zokor populations. Rural residential land, plain agricultural land and low-coverage grassland had a negative effect on genetic diversity. Hilly agricultural land and middle-coverage grassland had little impact on zokor genetic diversity. There were also interactions between some habitat types, that is, habitat types with relatively better quality together promoted conservation of genetic diversity, while the interaction between (among) bad habitat types made situations worse. Finally, habitat diversity, measured as patch richness and Shannon's diversity index, was positively correlated with the genetic diversity. These results demonstrated that: (1) different habitat types had different effects on the genetic diversity of zokor populations and (2) habitat quality and habitat heterogeneity were important in maintaining genetic diversity. Habitat composition was closely related to land use thus emphasizing the importance of human activities on the genetic diversity of subterranean rodent populations in this semi-natural landscape. Although the Gansu zokor was considered to be a pest species in the Loess Plateau, our study provides insights for the management and conservation of other subterranean rodent species.
