Three genes encoding for fungal cell wall degrading enzymes (CWDE), ech42, nag7O and gluc78 from the biocontrol fungus Trichoderma atroviride were transformed into rice mediated by Agrobacterium tumefaciens singly and in all possible combinations. A total of more than 1800 independently regenerated plantlets in seven different populations (for each of the three genes and each of the four gene combinations) were obtained. Our data indicated that gluc78 gene had negative effects on transformation frequency and plant growth. Some regenerated plants with gluc78 gene were stunted; spontaneously produced brown specks; could not tassel. The combination with either one of the two other genes (ech42, nag70) present in the same T-DNA region reduced the negative effect of gluc78 on plant growth. These results indicated that expression of several genes in one T-DNA region interfered with each other and expression of exogenous gene in recipient plant was a complex behavior. Mitochondrial DNA control region segment I sequences and melanocortin 1 receptor (MC1R) gene polymorphism were examined in ethnic populations in the silk road region of China. Both the frequencies of the MC1R variants and the results of mtDNA data in this region presented intermediate values between those of Europe and East and Southeast Asia, which suggested extensive gene admixture in this area and was in general agreement with previous studies. Phylogenetic analysis of the ethnic populations in the Silk Road region that based on mtDNA data didn't show expected cluster pattern according to their ethnogenesis. We suspect that a high migration rate in female among these closely related populations and other three demographic events might account for it.

The family Cyprinidae is widely distributed in East Asia, and has the important phylogenetic significance in the fish evolution. In this study, the 5' end partial sequences (containing exon 1, exon 2 and indel 1) of S6K1 gene were obtained from 30 representative species in Cyprinidae and outgroup using PCR amplification and sequencing. The phylogenetic relationships of Cyprinidae were reconstructed with neighbor joining (NJ), maximum parsimony (MP), maximum likelihood (ML), and Bayesian methods. Myxocyprinus asiaticus (Catostomidae) was assigned to the outgroup taxon. Similar phylogenetic relationships within the family Cyprinidae were achieved with the four analyses. Leuciscini and Barbini were monophyletic lineages respectively with the high nodal supports. Leuciscini comprises Hypophthalmichthyinae, Xenocyprinae, Cultrinae, Gobioninae, Acheilognathinae and East Asian species of Leuciscinae and Danioninae. Monophyly of East Asian clade was supported with high nodal support. Barbini comprises Schizothoracinae, Barbinae, Cyprininae and Labeoninae. The monophyletic lineage consisting of Danio rerio, D. myersi, and Rasbora trilineata was basal in the tree. In addition, the large fragment indels in intron 1 were analyzed to improve the understanding of Cyprinidae relationships. The results showed that the large fragment indels were correlated with the relations among species. Some conserved regions in intron 1 were thought to be involved in the functional regulation. However, no correlation was found between sequence variations and species characteristic size.

The genes of IRF-1 and IRF-7 have been cloned from the mandarin fish (Siniperca chuatsi). The IRF-1 gene has 4919 nucleotides (nt) and contains 10exons and 9introns, with an open reading frame (ORF) of 903 ntencoding301 aa. The IRF-7 gene has 6057 nt and also contains 10exons and 9introns, with an ORF of 1308 nt encoding 436 aa. The IRF-1 and IRF-7 genes have only one copy each in the genome. The transcription of IRF-1 and IRF-7 in different organs was analyzed by real-time PCR, and both molecules were constitutively expressed. The IRF-I and IRF-7 mRNAs were abundant in gill, spleen, kidney and pronephros. The temporal transcriptional changes for IRF-1, IRF-7 and Mx were investigated within 48 h after poly I: C stimulation in liver, gill, spleen and pronephros. An increased transcription was detected for IRF-1 and IRF-7 12 h post-stimulation, being earlier than the transcription of Mx protein; however, IRF-1 and IRF-7 transcription decreased while the Mx protein was stable at 48 h post-stimulation. The population genetic structure of fish parasitic nematode, Camallanus cotti, collected from the Yangtze River, Pearl River and Minjiang River in China was investigated. From these parasites, the similar to 730 bp of the first internal transcribed spacer of ribosomal DNA (ITS1 rDNA) and the 428 bp of mitochondrial cytochrome c oxidase subunit I (COI) gene were sequenced. For the ITS1 rDNA data set, highly significant Fst values and low rates of migration were detected between the Pearl River group and both the Yangtze River (Fst = 0.70, P < 0.00001; Nm = 0.21) and Minjiang River (Fst = 0.73, P < 0.00001; Nm = 0.18) groups, while low Fst value (Fst = 0.018, P > 0.05) and high rate of migration (Nm = 28.42) were found between the Minjiang and the Yangtze rivers. When different host/locality populations (subpopulations) within each river were considered, subpopulations between the Yangtze River and Minjiang River had low Fst values (<= 0.12) and high Nm values (>3.72), while Pearl River subpopulations were significantly different from the Yangtze River and Minjiang River subpopulations (Fst >= 0.59; Nm < 1). The COI gene data set revealed a similar genetic structure. Both phylogenetic analyses and a statistical parsimony network grouped the Pearl River haplotypes into one phylogroup, while the Yangtze River and Minjiang River haplotypes formed a second group. These results suggested that the Yangtze River and Minjiang River subpopulations constituted a single reproductive pool that was distinct from the Pearl River subpopulations. In addition, the present study did not find host-related genetic differentiation occurring in the same drainage. (C) 2009 Published by Elsevier B.V.

本文以耐旱的牛耳草(Boea hygrometirica)为材料,研究了乙烯合成关键酶之一——ACC氧化酶编码基因(BhACO1)在干旱复苏过程中的诱导表达及其编码蛋白的酶活,分析了乙烯在干旱复苏过程中的积累及其对叶片复苏能力的影响和对干旱诱导基因的调控,探讨了一个受乙烯调控的干旱诱导的引导蛋白编码基因(BhDIR1)的表达及其功能。 利用cDNA微阵列技术从牛耳草干旱2h的叶片中得到一个ACC氧化酶基因片段,经5’-RACE得到全长cDNA,命名为BhACO1。BhACO1包含317个氨基酸,与其它植物中的ACC氧化酶具有80%左右的序列相似性。BhACO1基因受乙烯和干旱诱导、但ACC氧化酶抑制剂氯化钴可抑制其干旱诱导表达。BhACO1基因受ABA、2,4-D、SA、H2O2、CaCl2、EGTA及热害和盐害的诱导,但不受冷害诱导。原核表达的GST-BhACO1融合蛋白在体内体外均表现出ACC氧化酶的活性,而过量表达BhACO1的转基因植物的蛋白提取物也表现出较野生型更强的ACC氧化酶活性。 乙烯在牛耳草叶片干旱复水过程中随着时间延长而逐步积累。外源乙烯可诱导叶片黄化,但不影响叶片在复水后的复苏能力;氯化钴处理可部分地抑制乙烯合成而降低牛耳草叶片在干旱过程中乙烯的释放量,同时导致叶片失去复苏能力。与对照相比,氯化钴处理的叶片在干旱时仍可维持较低的离子渗漏水平,但复水后发生大量离子外渗,表明细胞膜完整性也遭到破坏;光系统ІІ活性下降程度在干旱时与对照相似,但复水后完全丧失。 乙烯诱导牛耳草干旱响应基因BhDohb561,BhLEA2和BhDIR1的表达,但不影响牛耳草干旱响应基因BhCML1,BhGRP1,BhSGP和BhLEA1的表达。除BhLEA1外,上述基因在干旱过程中的诱导表达均可被氯化钴预处理所抑制,尤其是BhSGP最明显。 BhDIR1在牛耳草干旱复水过程中mRNA明显地积累,乙烯、ABA、CaCl2、EGTA、H2O2、SA和热害、冷害、盐害都可诱导其表达。BhDIR1编码一个199个氨基酸的小分子量蛋白质,与松柏等植物中发现的可能参与木质素合成的引导蛋白具有约20-30%的序列相似性。与其它引导蛋白相同,BhDIR1在N’端包含一个外泌的信号肽,GFP定位分析表明BhDIR1定位于细胞膜和壁上。 上述结果表明,乙烯在牛耳草叶片耐脱水复苏反应中有不可或缺的作用,而ACC氧化酶所催化的反应是干旱诱导的乙烯合成中的关键步骤。氯化钴预处理通过抑制干旱过程中的乙烯合成,影响一系列基因的干旱诱导表达导致叶片在生理水平和细胞水平上造成了损伤,或是使牛耳草失去了在复水过程中原有的修复能力而无法恢复生命力。BhDIR1作为乙烯调控的下游靶基因之一,可能通过调控木质素的单体间的连接方式而改变木质素的物理性质来影响细胞壁的机械强度和柔韧性,减少干旱对细胞造成的机械伤害。

This study was conducted to investigate time-dependent changes in oxidative enzymes in liver of crucian carp after intraperitoneally injection with extracted microcystins 600 and 150 mu g kg(-1) body weight. The results showed that activities of antioxidant enzymes, including superoxide dismutase, catalase, glutathione peroxidase and glutathione reductase generally exhibited a rapid increase in early phase (1-3 h post injection), but gradually decreased afterwards (12-48 h) compared with the control, with an evident time-dependent effect. These zigzag changes over time contributed a better understanding on oxidative stress caused by microcystins in fish.

According to the specific property of respective phases in copper ore, optimal extracting condition for, tour phases of copper with five infusion was examined. After a series of experiments, for example, different weight Of sample, artificial mixed single phases sample and repeated experiment, the extracting ratio constants foil all phases in different infusions were obtained. The mathematical model which is on the basis of the stable extracting constant is established. Based on the purpose-control genetic algorithms, we only need to determine the total copper in the sample in different infusions under the selected condition, then the value of respective phase could be calculated. The-relative standard deviation are (%) free oxidized copper phase:1.5, conjunction oxidized copper phase: 17.6,, secondary copper sulfide phase: 1.9, primary copper sulfide phase : 2.7,total copper: 0.8.

It was a study of the effect of non-cognitive factors on cognitive aging. The purpose of the study was to research the effect of attitude towards aging on memory performance of the elderly, and trying to explore the mechanism of the attitude towards aging on memory performance. The entire study was divided into three phases and included six experiments. The questionnaire of attitude towards aging was compiled in phase 1. Then the validity and reliability of it was tested in the elderly. The self-esteem scale was also tested as above. These prepare the instruments for the study. It was a static study in phase 2. Through comparing the old groups in different environment, analyzing relationship between different non-cognitive factors and memory performance in the old subjects in identical environment, the paths of the effect of the attitude towards aging on memory performance was explored. It was a dynamic study in phase 3. By changing attitude towards aging of the elderly with direct thinking (positive, negative), the effect on memory performance was observed and the mechanism was explored. Main results: 1. The validity and reliability of the questionnaire towards aging were good. Its construct had two factors (positive, negative). Both reliability of retest and split test were over 0.90. 2. Direct thinking task was able to change the attitude towards aging effectively in old or young adults. The scores of the attitudes towards aging between positive and negative thinking subjects were different significantly. 3. The effect of attitude towards aging on memory performance was significant. The results in static or dynamic condition showed that the elderly with higher score of the questionnaire of attitude towards aging (more positive), their memory self-efficacy and memory performance were higher. In the opposite (negative), they were lower. 4. Mechanism of the effect of the attitude towards aging on memory performance: in static condition there were two paths i.e. direct path and through memory self-efficacy; in dynamic condition, besides the two paths, emotion change constituted another path. 5. Different non-cognitive factors have different effect on psychological function. The results of this study found that self-esteem has no significant correlation with memory performance, but correlates with depression; The factor of education has effect on many non-cognitive factors and memory performance.

Lipopolysaccharide and beta-1,3-glucan-binding protein (LGBP) play a crucial role in the innate immune response of invertebrates as a pattern recognition protein (PRP). The scallop LGBP gene was obtained from Chlamys farreri challenged by Vibrio anguillarum by randomly sequencing cDNA clones from a whole body cDNA library, and by fully sequencing a clone with homology to known LGBP genes. The scallop LGBP consisted of 1876 nucleotides with a canonical polyadenylation signal sequence AATAAA and a poly(A) tail, encoding a polypeptide of 440 amino acids with the estimated molecular mass of 47.16 kDa and a predicted isoelectric point of 5.095. The deduced amino acid sequence showed a high similarity to that of invertebrate recognition proteins from blue shrimp, black tiger shrimp, mosquito, freshwater crayfish, earthworms, and sea urchins, with conserved features including a potential polysaccharide-binding motif, a glucanase motif, and N-glycosylation sites. The temporal expression of LGBP genes in healthy and V. anguillarum-challenged C farreri scallop, measured by real-time semiquantitative reverse transcription polymerase chain reaction (PCR), showed that expression was up-regulated initially, followed by recovery as the stimulation cleared. Results indicated that scallop LGBP was a constitutive and inducible acute-phase protein that could play a critical role in scallop-pathogen interaction. (C) 2004 Elsevier B.V. All rights reserved.

Dicer is a member of the RNAase III family which catalyzes the cleavage of double-stranded RNA to small interfering RNAs and micro RNAs, and then directs sequence-specific gene silencing. In this paper, the full-length cDNA of Dicer-1 was cloned from white shrimp Litopenaeus vannamei (designated as LvDcr1). It was of 7636 bp, including a poly A tail, a 5' UTR of 136 bp, a 3' UTR of 78 bp, and an open reading frame (ORF) of 7422 bp encoding a putative protein of 2473 amino acids. The predicted amino acid sequence comprised all recognized functional domains found in other Dicer-1 homologues and showed the highest (97.7%) similarity to the Dicer-1 from tiger shrimp Penaeus mondon. Quantitative real-time PCR was employed to investigate the tissue distribution of LvDcr1 mRNA, and its expression in shrimps under virus challenge and larvae at different developmental stages. The LvDcr1 mRNA could be detected in all examined tissues with the highest expression level in hemocyte, and was up-regulated in hemocytes and gills after virus injection. These results indicated that LvDcr1 was involved in antiviral defense in adult shrimp. During the developmental stages from fertilized egg to postlarva VII, LvDcr1 was constitutively expressed at all examined development stages, but the expression level varied significantly. The highest expression level was observed in fertilized eggs and followed a decrease from fertilized egg to nauplius I stage. Then, the higher levels of expression were detected at nauplius V and postlarva stages. LvDcr1 expression regularly increased at the upper phase of nauplius, zoea and mysis stages than their prophase. The different expression of LvDcr1 in the larval stages could provide clues for understanding the early innate immunity in the process of shrimp larval development. (C) 2010 Elsevier Ltd. The phase diagram of the dodecyl dimethyl ammonium hydroxyl propyl sulfonate(DDAHPS)/1-pentanol(C5H11OH)/water ternary system has been established. It contains two isotropic monophase regions (L-1 and L-2) and a liquid crystalline region (L.C.). The isotropic phase regions have been investigated by means of Raman spectroscopy and conductivity.
