A series of experiments have been performed by complete kinematics measurements to study two-proton (2p) correlated emission from the excited states of Ne-17,Ne-18 and S-28,S-29 via the Coulomb excitation by bombarding on Au-197 target. 2p and residua coincident events were picked Out under strict conditions. Visible p-p correlations were observed. It is shown that 2p can be emitted from the high-lying excited states. 2p halo may lead to 2p emission with large spectroscopy factor for the states close to or beyond the threshold.

The foil-excited the spectrum of highly stripped titanium ions between 12-40 nm has been studied. Titanium ions of 80 and 120 MeV were provided by the HI-13 tandem accelerator at the China Institute of Atomic Energy. GIM-957 XUV-VUV monochromator was refocused to get highly-resolved spectra. Our experimental results and the published spectral data of laser-produced plasma show agreement in nearly all cases within +/- 0.03 nm. The spectra contained some weak or strong lines previously unclassified. These spectral lines mainly belong to 2s2p(2) for TiXVIII, 2p(3) for TiXVIII, 2s2p(3) for TiXVII, 2p(6)4p for Ti XII and 2p(6)3d for Ti XII transitions.

Two-electron-one-photon (TEOP) M1 and E2 transition energies, line strengths and transition probabilities between the states of the 2p(3) and 2s(2)2p odd configurations for B-like ions with 18 <= Z <= 92 have been calculated using the GRASP2K package based on the multiconfiguration Dirac-Hartree-Fock (MCDHF) method. Employing active-space techniques to expand the configuration list, we have systematically considered the valence, core-valence and core-core electron correlation effects. Breit interaction and quantum electrodynamical (QED) effects were also included to correct atomic state wavefunctions and the corresponding energies. Influences of electron correlation, Breit interaction and QED effects on transition energies and line strengths of the TEOP M1 and E2 transitions were analysed in detail. The present results were also compared with other theoretical and experimental values.

采用逆运动学方法对放射性核束17F和18Ne与质子进行弹性散射实验 ,得到了实验测量微分截面 .用较准确描述放射性核素性质的CH89参数化的光学势为初始光学势 ,用扭曲波玻恩近似的理论计算程序DWUCK4和自动参数搜索程序ABOD对实验数据进行光学势参数理论拟合 ,得到了与实验数据相符合的光学势参数 .将得到的光学势参数进行分析 ,得到了17F和18Ne实势相互作用均方根半径分别为 3 2 3 9fm和 3 3 1 7fm .

异叶苣苔属(Whytockia)隶属于苦苣苔科(Gesneriaceae)、尖舌苣苔族(Klugieae).70年代以来,该属作为Klugieae中最原始的类群才逐渐被人们所认识,并被认为是解决Klugieae系统发育问题的关键类群,但到目前为止,对该属变异式样及系统发育的认识仍然极其有限.鉴于上述原因,本文结合蜡叶标本和大量的野外观察和采集,从形态学、维管束的结构和分化,形态发生和细胞学等方面,进行异叶苣苔属的系统学和进化研究,并兼论尖舌苣苔族的系统发育. 1.通过对异叶苣苔属大量性状的仔细观察和比较,本文选择了34个性状,并分析了他们的变异式样及其形成原因,为进一步的系统学研究提供依据. 2.地上茎的形态发生研究揭示出异叶苣苔属植物的顶芽完全受到抑制,位于小型叶腋的侧芽萌发,从而替代顶芽生长,地上茎的不分枝是位于大型叶腋的腋芽受到抑制所致,该研究纠正了前人的观察错误. 3.花序及花的形态发生过程表明,异叶苣苔属的花序呈对花从花序轴内侧二列式连续发生,没有任何分枝和附属物.花各部分的发生顺序基本上呈向心式,但在花冠和雄蕊原基之间则为离心式发生.花萼和花冠原基的发生及发育顺序均呈下降式,从而使花萼和花冠在芽中呈下降覆瓦状排列,退化雄蕊是由发育停滞所致.雌蕊的两个心皮在形态发生过程中由离生到合生,该属具中轴胎座的二室子房起源于两个离生心皮的近轴面内卷式愈合,柱头从离生到合生决定于个体发育中愈合部向上推进的程度. 4.为了澄清性状演化极性这一关键问题、本文作了花部器官解剖以及心皮维管束结构及分化过程的研究,结果表明,该属花梗和花萼中均具有分泌道和雄蕊束从中柱独立分出,同独叶苣苔属(Monophyllaea)十分相似:花盘低且薄并且没有维管束,以及子房壁维管束具有不同来源而区别于尖舌苣苔族(Klugieae)中的其它类群.异叶苣苔属的心皮结合程度和胎座形态可分为三种类型,即A型:心皮结合部的中轴区膨大,但四个瓣片的上部分离、B型:除中轴区膨大外,四个瓣片也成对愈合,胎座呈锚形、C型:中轴区不膨大,四个瓣片完全分离,子房上部一室.前者保留了较多的叶性,可能是该属乃至尖舌苣苔族中最原始心皮及胎座形态.后二者代表着两个相反的演化趋势. 5.本文首次报道了异叶苣苔属的染色体数目和核型,染色体数目均为2n=18,x=9.核型分别是2A或3A.核型的演化趋势,即从对称到不对称和随体的丢失同外部形态的变异密切相关。 6.深入地比较和分析了异叶苣苔属和其他类群的关系以及性状的演化趋势,并运用分支分析方法,重建属的系统发育. 7.生态、地理分布和演化的研究表明:异叶苣苔属的多样化中心分别是云南东南部和滇、贵、川交界区域,起源于第三纪或更早期的古热带湿润性森林植物区系,起源地可能是两个现代多样化中心所连接的整个区域,即:滇东南至滇贵川交界区域. 8.本文对异叶苣苔属进行全面的分类学修订,包括7个种,其中一个新种,新组合1个种和一个变种、根据该属的系统发育式样,在属下新设两个组.系统排列如下: 异叶苣苔属Whytockia W. W. Smith 组1:异叶苣苔组Sect. Whytockia Y. Z. Wang sect. nov. 1.河口异叶苣苔W. hekouensis Y. Z. Wang 2.毕节异叶苣苔W. bijieensis Y. Z. Wang, sp. nov. 3.异叶苣苔W. chiritiflora (Oliv.) W.W. Smith 4.峨眉异叶苣苔W. wilsonii (A. Weber) Y. Z. Wang, stat. nov. 5.紫红异叶苣苔W. purpurascens Y. Z. Wang 组2:台湾异叶苣苔组Sect. Synstigma Y. Z. Wang, sect. nov. 6.白花异叶苣苔W. tsiangiana (Hand.-Mazz) A. Weber 7.台湾异叶苣苔W. sasakii (Hayata)B.L.Burtt. 9.在异叶苣苔属系统学和进化研究的基础上,结合前人工作,本文进一步探讨尖舌苣苔族的系统发育.大量的证据充分显示出异叶苣苔属和圆果苣苔属分别是Klugieae最原始的类群,该族中的尖舌苣苔属(Rhynchoglossum),盾座苣苔属(Epithema)和独叶苣苔属(Mono phyllaea),可能是从Whytockia植物的早期类群直接演化而来,十字苣苔属(Stauranthera)和Loxonia也可能直接或间接来自Gyrogyne植物的早期类群.尖舌苣苔族植物可能起源于亚洲大陆或更准确地说,中国西南部地区,在随后的演化过程中,经过分化、迁移,逐渐形成广布于南亚、东南亚及太平洋岛屿至西非、中美洲的现代地理分布式样。

Cubic GaN/GaAs(0 0 1) epilayers and hexagonal inclusions are characterized by X-ray diffraction (XRD), Photoluminescence (PL), Raman spectroscopy, and transmission electron microscopy (TEM). The X-ray {0 0 0 2} and (1 0 (1) over bar 0) pole figures show that the orientation relationships between cubic GaN and hexagonal inclusions are (1 1 1)//(0 0 0 1), <1 1 2 >//<1 0 (1) over bar 0 >. The distribution of hexagonal inclusions mainly results from the interfacial bonding disorder in the grain boundaries parallel to hexagonal <0 0 0 1 > directions and the lattice mismatch in <0 0 0 1 > directions on {1 0 (1) over bar 0} planes. In order to reduce the energy increase in cubic epilayers, hexagonal lamellas with smaller sizes in <0 0 0 1 > directions often nucleate inside the buffer layer or near the interface between the buffer layer and the epitaxial layer, and penetrate through the whole epitaxial layer with this orientation relationship. 本论文对四川蜡瓣花 (Corylopsis willmottiae Rehd. et Wils.)、密花樫木[Dysoxylum densiflorum (Blume) Miq.]、四川溲疏 (Deutzia setchuenensis Franch)及云南豆腐柴 (Premna yunnanensis W. W. Smith)的化学成分进行了研究。通过色谱分离得到44个化合物。主要基于波谱数据鉴定了它们的结构,其中1个为新化合物。 1.从四川蜡瓣花全株的95%乙醇提取物中共分离鉴定了13个化合物,它们是:1-O-(3-O-甲基没食子酸)-岩白菜素(1)、11-O-没食子酰基岩白菜素(2)、 11-O-紫丁香基岩白菜素(3) 、岩白菜素(4)、4-O-没食子酰基岩白菜素(5) 、4,11-O-二没食子酰基岩白菜素 (6)[14]、β-谷甾醇 (7)、acetyl aleuritolic acid (8)、(-)-表没食子儿茶素没食子酸酯(9)、对羟基苯甲酮 (10)、 11-香豆酸酰岩白菜素 (11)[19]、丁香酸 (12)和没食子酸 (13)。其中1为新化合物。 2.从密花樫木根的95%乙醇提取物中共分离纯化了13个化合物,它们是:β-白檀酮(14)、richenone (15)、β-谷甾醇 (7)、cabraleadiol (16)、β-香树脂醇 (17)、龙脑香醇酮 (18)、cabraleadiol monoacetate (19)、cabraleone (20)、3β-hydroxy-5 -pregnen-20-one (21)、3β-hydroxy-5α-pregnan-20-one (22)、cabraleahydroxylactone (23)、川楝子甾醇B (24)、表儿茶素 (25)。 3.从四川溲疏全株95%乙醇提取物中共分离11个化合物,鉴定了其中的9个化合物。它们是:β-谷甾醇 (7)、白桦酯醇(26)、齐墩果酸(27)、hydrangetin (28)、肉桂酸 (29),齐墩果酸-3-O-β-D-吡喃葡萄糖醛酸苷(30)、β-胡萝卜苷 (31)、齐墩果酸-3-O-(β-D-吡喃葡萄糖醛酸-6-正丁酯)(32)、齐墩果酸-3-O-β-D-吡喃葡萄糖醛酸-28-O-β-D-吡喃葡萄糖苷 (33)。 4.从云南豆腐柴95%乙醇提取物中分离得到12个化合物,分别为白桦脂醇 (25)、7-羟基黄烷酮 (34)、松属素 (35)、2’,4’-羟基查儿酮 (36)、高良姜素-3-甲醚 (37) 、高良姜素-3,7-二甲醚 (38)、异甘草素-4-甲醚 (39)、豆蔻明 (40)、乔松酮 (41)、异甘草素 (42)、arjunolic acid (43)、槲皮素3-O-β-D-木糖苷(44)。 5.综述了1976年以来樫木属植物化学成分和活性研究的概况。 Phytochemical investigation on Corylopsis willmottiae, Dysoxylum densiflorum, Deutzia setchuenensis, and Premna yunnanensis, led to the isolation of 44 compounds, 1 of which was new one. 1. One new compound was isolated from 95% ehanolic extrat of the whole plants of C. willmottiae, identified as 11-O-(3-O-methylgalloyl)-bergenin (1). The twelve known compounds isolated were 11-O-galloylbergenin (2), 11-O-syringylbergenin (3), bergenin (4), 4-O-galloylbergenin (5), 4,11-di-O-galloylbergenin (6), β-sitosterol (7), acetyl aleuritolic acid (8), (-)-epigallocatechin 3-O-gallate (9), 1-(4-hydroxyphenyl) ethanone (10), 11-O-coumaroylbergenin (11), syringic acid (12), gallic acid (13). 2. Thirteen compounds were isolated from 95% ethanol extract from the roots of D. densiflorum and identified as β-amyrenone (14), richenone (15), β-sitosterol (7), cabraleadiol (16), β-amyrin (17), hydroxydammarenone-Ⅱ (18), cabraleadiol monoacetate (19), cabraleone (20), 3β-hydroxy-5-pregnen-20-one (21), 3β-hydroxy-5α-pregnan-20-one (22), cabraleahydroxylactone (23), toosendansterol B (24) and (-)-epicatechin (25). 3. Eleven compounds were isolated from ethanol extract of D. Setchuenensis. Nine were identified as β-sitosterol (7), betulin (26), oleanolic acid (27), hydrangetin (28), cinnamic acid (29), oleanolic acid 3-O-β-D-glucuronopyranoside (30), β-daucosterol (31), oleanolic acid 3-O-β-D-glucuronopyranoside-6-O-butyl ester)(32), oleanolic acid 3-O-β-D-glucuronopyranosyl-28-3-O-β-D-glucopyranoside (33). 4. Twelve compounds were isolated from ethanol extract of P. yunnanensis and identified as betulin (26), 7-hydroxyflavanone (34), pinocembrin (35), 2’,4’-dihydroxychalcone (36), galangin 3-methyl ether (37), galangin 3,7-dimethyl ether (38), isoliquiritigenin 4-methyl ether (39), cardamonin (40), pinostrobin (41), isoliquiritigenin (42), arjunolic acid (43), quercetin 3-O-β-D-lyxosopyranoside (44). 5. Chemical constituents and biological activities of the genus Dysoxylum (Meliaceae) were reviewed during 1976-2009.

八月瓜属植物五枫藤(Holboellia latifolia Wall.)和驳骨草属植物小驳骨(Gendarussa vulgaris Nees)均为药用植物, 前者化学成分研究不深入, 后者的化学成分未见报道。川西茶藨(Ribes takare D. Don)为茶藨子属植物, 没有化学成分的报道。本论文对三个植物的化学成分和活性成分进行了研究, 主要通过色谱方法分离得到了48 个化合物, 采用波谱分析或与已知标准品对照等手段鉴定了它们的结构, 其中有1 个新的原小檗碱类化合物和3 个新的联苯类化合物,发现了具有细胞毒活性和α-葡萄糖苷酶抑制活性的化合物。1、从五枫藤地上部分的95%乙醇提取物中分离得到了12 个化合物: 五加苷K (1)、hederagenin 3-O- α-L-rhamnopyranosyl-(1→2)- α-L-arabinopyranoside (2)、β-萘乙酸(3) 、3-O-α-L-rhamnopyranosyl-(1→2)-[β-D-glucopyranosyl-(1→3)]-α-L-arabinopyranosyl oleanolic acid 28-O-α-L-rhamnopyranosyl-(1→4)-β-D-glucopyranosyl-(1→6)-β-D-glucopyranosyl ester (4) 、3-O- α-L-rhamnopyranosyl-(1→2)-O- β- D-glucopyranosyl-(1→2)- α-L-arabinopyranosyl oleanolic acid (5) 、3-O-( β-D-glucopyranosiduronic acid)-oleanolic acid 28-O- β-D-glucopyranoside (6)、lup-20(29)-en-3-one (7)、lupeol (8)、β-谷甾醇(9)、齐墩果酸(10)、乌苏酸(11)、β-胡萝卜苷(12)。化合物1 对Lu-06、N-04 和Bre-04 癌细胞株的GI50 分别是0.77µg/mL、1.26 µg/mL 和1.55 µg/mL, 化合物2 对N-04 癌细胞株的GI50 为2.44 µg/mL。2、从小驳骨地上部分的95%乙醇提取物中分离得到了1 个原小檗碱类新化合物13-hydroxyl gusanlung A (25), β-谷甾醇(9)、齐敦果酸(10)、β-胡萝卜苷(12)、棕榈酸(1-)甘油酯(13)、棕榈酸(14)、阿苯哒唑(15)、阿苯哒唑砜(16)、阿苯哒唑亚砜(17)、aurantiamide acetate (18)、华良姜素(19)、芫花素(20)、(-)-丁香树酯醇(21)、gusanlung B (22) 、eupteleasaponinsⅤ acetate (23)、gusanlungA (24)、刺五加苷E (26)、岩白菜素(27)、咖啡酸(28)。化合物25 对肝癌细胞株(HepG2) 的GI50 为2.08 µg/mL。3、从川西茶藨地上部分的95%乙醇提取物中分离鉴定了22 个化合物: β-谷甾醇(9) 、β- 胡萝卜苷(12) 、O-acetyloleanolic aldehyde (29),4,7,8-trimethoxy-2,3-methylenedioxydibenzofuran (30) 、3', 5-dimethoxy-3, 4-methylenedioxybiphenyl (31) 、桦木醇(32) 、6,7-dimethoxy-1-methyl-3,4-dihydroquinolin-2-one (33)、3'-hydroxy-5-methoxy-3,4-methylenedioxybiphenyl (34) 、7-hydroxy-4,8-dimethoxy-2,3-methylenedioxydibenzofuran (35)、桦木醛(36)、没食子酸(37) 、6β- 羟基-4- 烯-3- 酮- 豆甾醇(38) 、5α, 8α-epidioxy-(22E,24R)-ergosta-6, 22-dien-3β-ol (39)、verrucofortine (40)、6-methoxycalpogoniumisoflavone A (41)、2-羟基二苯甲酮(42)、桦木酸(43), 3, 5-二甲氧基苯甲酸-4-O-β-D-吡喃葡萄糖苷(44)、洋芹素(45)、刺槐素(46)、水杨酸(47)、洋芹素-5-O- β-D-葡萄糖苷(48), 化合物30、31 和35 为新的联苯化合物。化合物30的α-葡萄糖苷酶抑制率为10.2% (1.00 mg/mL); 化合物35 的抑制率为17.2% (1.00mg/mL)。4、综述了1960 年以来原小檗碱类化合物药理活性研究进展。 Plants Holboellia latifolia Wall and Gendarussa vulgaris Nees, are used as folkmedicine. Ribes takare D. Don belongs to the genus Ribes. The three plants have notbeen chemically studied in detail. Chemical and bioactive study of three plants led tothe isolation of 48 compounds by chromatography. Their structures were elucidatedon the basis of spectroscopic evidence or comparison with authentic samples. Amongthe 48 componds isolated one protoberberine alkaloid and three biphenyls are newones. Cytotoxic and α-glucosidase inhibitory compounds had been found.1. Twelve compounds were isolated from the 95% ethanol extract of the aerial partof H. latifolia Wall. They were characterized as fellow: eleutheroside K (1),hederagenin-3-O- α-L-rhamnopyranosyl-(1→2)- α-L-arabinopyranoside (2),2-naphthyl acetic acid (3),3-O-α-L-rhamnopyranosyl-(1→2)-[β-D-glucopyranosyl-(1→3)]-α-L-arabinopyranosyl oleanolic acid 28-O-α-L-rhamnopyranosyl-(1→4)-β-D-glucopyranosyl-(1→6)-β-D-glucopyranosyl ester (4), 3-O- α-L-rhamnopyranosyl-(1→2)-O- β- D-glucopyranosyl-(1→2)- α-L-arabinopyranosyl oleanolic acid (5),3-O-( β-D-glucopyranosiduronic acid)-oleanolic acid-28-O- β-D-glucopyranoside (6),lup-20(29)-en-3-one (7), lupeol (8), β-sitosterol (9), oleanolic acid (10), ursolicacid (11), and β-daucosterol (12). Compound 1 showed moderate cytotoxicity againstLu-06 (GI50, 0.77 µg/mL), N-04 (GI50, 1.26 µg/mL) and Bre0-4 (GI50=1.55 µg/mL)and compound 2 showed moderate cytotoxicity against N-04 (GI50=2.44 µg/mL).2. A new protoberberine alkaloid, 13-hydroxyl gusanlung A (25), was isolated fromthe aerial part of Gendarussa vulgaris Nees, together with β-sitosterol (9), oleanolicacid (10), β-daucosterol (12), glycerol monopalmitate (13), palmific acid (14),albendazole (15), albendazole sulphone (16), albendazole sufloxide (17), aurantiamideacetate (18), kumatakenin (19), genkwanin (20), (-)-syringaresinol (21), gusanlung B(22), eupteleasaponinsⅤ acetate (23), gusanlung A (24), eleutheroside E (26),bergenin (27) and caffeic acid (28). Compound 25 showed cytotoxicity against HepG2 cells (GI50, 2.08 µg/mL).3. Phytochemical study of the Ribes takare D. Don led to the isolation of three newbiphenyls, 4,7,8-trimethoxy-2,3-methylenedioxydibenzofuran (30), 3', 5-dimethoxy-3,4-methylenedioxybiphenyl (31) and 7-hydroxy-4,8-dimethoxy-2,3-methylenedioxydibenzofuran (35), along with nineteenknown compounds, β-sitosterol (9), β-daucosterol (12), O-acetyloleanolic aldehyde(29), betulin (32), 6,7-dimethoxy-1-methyl-3,4-dihydroquinolin-2-one (33),3'-hydroxy-5-methoxy-3, 4-methylenedioxybiphenyl (34), betulinic aldehyde (36),gallic acid (37), stigmast-4-en-6β-ol-3-one (38), 5α, 8α-epidioxy-(22E, 24R)-ergosta-6,22-dien-3β-ol (39), verrucofortine (40), 6-methoxycalpogonium isoflavone A (41),2-hydroxybenzophenone (42), betulinic acid (43), 3,5-dimethoxygallic acid-4-O- β-D-glucopryranoside (44), apigenin (45), acacetin (46), salicylic acid (47) andapigenin-5-O- β-D-glucopryranoside (48). α-Glucosidase inhibitory rates ofcompound 30 and 35 were respectively 10.2% and 17.2% at a concentration of 1.00 mg/mL).4. Pharmacological activities of protoberberines were summarized.

高等植物种子胚乳贮藏蛋白是种子发芽时的主要氮源,也是人类和动物食用植物蛋白的主要来源。大麦种子胚乳贮藏蛋白主要是醇溶蛋白(hordeins),占大麦胚乳总蛋白的50–60%。根据大麦醇溶蛋白的大小和组成特点,大麦醇溶蛋白被划分为三种类型:富硫蛋白亚类(B,γ-hordeins)、贫硫蛋白亚类(C-hordeins)以及高分子量蛋白亚类(D-hordeins)。B组和C组醇溶蛋白是大麦胚乳的两类主要贮藏蛋白,它们分别占大麦总醇溶蛋白成分的70–80%和10–12%。遗传分析表明,大麦B、C、D和γ-组醇溶蛋白分别是由位于大麦第五染色体1H(5)上的Hor2、Hor1、Hor3和Hor5位点编码。Hor2位点编码大量分子量相同但组成不同的B组醇溶蛋白(B-hordein)。B-hordein的种类、数量和分布是影响大麦酿造、食用及饲养品质的重要因素之一。为深入了解B-hordein基因家族的结构和染色体组织,探明Hor2位点基因表达的发育调控机制,最终达到改良禾谷类作物籽粒品质的目的,本研究以青藏高原青稞为材料,采用同源克隆法,分别克隆B-hordein基因和启动子,通过原核生物表达验证B-hordein基因功能,并利用实时定量PCR探索B-hordein基因表达时空关系,取得如下研究结果: 1. 以具有特殊B组醇溶蛋白亚基组成的9份青藏高原青稞为材料,根据GenBank中三个B-hordein基因序列(GenBank No. X03103, X53690和X53691)设计一对引物,通过PCR扩增,获得23个B-hordein基因克隆并对其进行了序列分析。核苷酸序列分析表明,所有克隆均包含完整的开放阅读框。有11个克隆都存在一个框内终止密码子,推测这11个克隆可能是假基因。推测的氨基酸序列分析表明,所有大麦B-hordein具有相似的蛋白质基本结构,均包括一个高度保守的信号肽、中间重复区以及C-端结构域。不同大麦种重复区内重复基元的数目有较大差异。青稞材料Z07–2和Z26的B-hordeins仅具有12个重复基元结构,更接近于野生大麦。这些重复基元数目的差异导致了重复区序列长度和结构的变异。这种现象极可能是由于醇溶谷蛋白基因在进化过程中染色体的不平衡交换或复制滑动所造成的。对所克隆基因和禾本科代表性醇溶谷蛋白基因进行聚类分析,结果表明所有来自栽培大麦的B-hordeins聚类成一个亚家族,来自野生大麦的B-hordeins以及普通小麦的LMW-GS聚类成另外一个亚家族,表明这两个亚家族的成员存在显著差异。此外,我们发现B-hordein基因推测的C-末端序列具有一些有规律的特征:即具有相同C-末端序列的B-hordein基因在系统发生树中聚类为同一个亚组(除BXQ053,BZ09-1,BZ26-5分别单独聚为一类外)。这个特征将有助于我们对所有B组醇溶蛋白基因家族成员进行分类,避免了在SDS-PAGE电泳图谱上仅依靠大小分类的局限性。 2. 根据上述克隆的青稞B-hordein基因的5’端序列设计三条基因特异的反向引物,以青稞Z09和Z26的基因组DNA为模板,采用SON-PCR和TAIL-PCR技术分离克隆出8个B-hordein基因的上游调控序列(命名为Z09P和Z26P)。序列分析表明,推测的TATA box位于–80 bp,CAAT–like box位于–140 bp处。此外,Z09P和Z26P中有六个序列在–300 bp处均存在一个由高度保守的EM基序和类GCN4基序构成的胚乳盒(Endosperm Box,EB),在约–560 bp处存在一个胚乳盒类似结构。而Z09P-2和Z26P-3不存在保守的胚乳盒或其类似结构,预示着这两个启动子所调控的基因表达可能受不同类型反式作用因子的调节,推测该启动子对基因的表达调控具有多样性。 3. 将B-hordein基因的开放阅读框定向克隆到表达载体pET-30a中,将其导入大肠杆菌表达菌株BL21中进行外源基因的诱导表达以验证所克隆基因的功能。结果表明仅含重组子pET-BZ07-2和pET-BZ26-5的BL21细菌有目的表达蛋白产生。在诱导3 h时的蛋白表达量最高;3 mM IPTG诱导的蛋白表达量要高于1 mM IPTG诱导的表达量。这为分离纯化B-hordein蛋白以及进一步研究其对大麦籽粒品质的影响奠定基础。 4. 根据从青稞Z09和Z26中分离克隆的B-hordein基因序列设计一对基因特异的引物,同时,选择大麦α-微管蛋白基因(GenBank no. U40042)为看家基因并设计特异引物,利用实时荧光定量PCR检测了青稞籽粒4个胚乳发育时间段的B-hordein基因表达,荧光定量结果显示:两份材料中B-hordein基因的表达量均随发育过程的进行而逐渐升高。Z09中B-hordein基因在开花后7天开始转录,而Z26开花4天后就有低水平B-hordein的表达,这表明Z26中B-hordein基因可能比Z09表达的较早或者Z09中B-hordein基因表达水平较低以致于不能被检测到。此外,在4个不同的胚乳发育时期中,Z26中B-hordein基因的表达量均高于Z09材料。在开花12天到18天的过程中,Z09和Z26中B-hordein基因的表达水平有一个急剧性的升高。这说明在不同胚乳发育时期,Hor2位点的B-hordein等位基因变异体存在mRNA的差异表达。 Seed endosperm storage proteins in higher plants are the main resources of nitrogen for germinating and plant proteins for human and animals. Barley prolamins (also called hordeins) are the major storage proteins in the endosperm and account for 50–60% of total proteins. Hordeins are classically divided into three groups: sulphur-rich (B, γ-hordeins), sulphur-poor (C-hordeins) and high molecular weight (HMW, D-hordeins) hordeins based on the size and composition. B-hordeins and C-hordeins are two major groups and each respectively account for about 70-80% and 10-12% of the total hordein fraction in barley endosperm. Genetic analysis showed that B-, C-, C-, γ-hordeins are encoded by Hor2, Hor1, Hor3 and Hor5 locus on the chromosome 1H (5). Hor2 locus is rich in alleles that encode numerous heterogeneous B-hordein polypeptides. It is reported that B-hordein species, quantity and distribution are significant factors affecting malting, food and feed quality of barley. To understand comprehensively the structure and organization of B-hordein gene family in hull-less barley and explore the developmental control mechanisms of Hor2 locus gene expression and eventually to better exploitation in crop grain quality improvement, we isolated and cloned B-hordein genes and promotors of hull-less barley from Qinghai-Tibet Plateau by PCR, and testified their expression founction in bacteria expression system and explore their spatial and temporal expression pattern by quantitative real time PCR. Our results are as followed, 1. Twenty-three copies of B-hordein gene were cloned from nine hull-less barley cultivars of Qinghai-Tibet Plateau with special B-hordein subunits and molecularly characterized by PCR, based on three B-hordein genes published previously (GenBank No. X03103, X53690 and X53691). DNA sequences analyses confirmed that the six clones all contained a full-length coding region of the barley B-hordein genes. Eleven clones all contain an in-frame stop codon and they are probably pseudogenes. The analysis of deduced amino acid sequences of the genes shows that they have similar structures including signal peptide domain, central repetitive domain, and C-terminal domain. The number of the repeats was largerly variable and resulted in polypeptides in different sizes or structures among the genes. Twelve such repeated motifs were found in Z07–2 and Z26, and they are close to those of the wild barleys, and it is most probably caused by unequal crossing-over and/or slippage during replication as suggested for the evolution of other prolamins. The relatedness of prolamin genes of barley and wheat was assessed in the phylogenetic tree based on their polypeptides comparison. Our phylogenetic analysis suggested that the predicted B-hordeins of cultivated barley formed a subfamily, while the B-hordeins of wild barleys and the two most similar sequences of LMW-GS of T. aestivum formed another subfamily. This result indicated that the members of the two subfamilys have a distinctive difference. In addition, we found the B-hordeins with identical C-terminal end sequences were clustered into a same subgroup (except BXQ053,BZ09-1 and BZ26-5 as a sole group, respectively), so we believe that B-hordein gene subfamilies possibly can be classified on the basis of the conserved C-terminal end sequences of predicted polypeptide and without the limit of SDS-PAGE protein banding patterns. 2. The specific primers were designed according to the published sequences of barley B-hordein genes from Z09 and Z26. Using total DNA isolated from them as the templates, eight clones (designated Z09Pand Z26P) of upstream sequences of the known B-hordein genes was obtained by TAIL-PCR and SON-PCR. Sequences analysis shows that the putative TATA box was present at position –80 bp and CAAT-like box at position –140 bp. Besides, a putative Endosperm Box including an Endosperm Motif (EM) and a GCN4-Like Motif was found at position –300 bp in six clones, and another Endosperm-like box was found at positon –560 bp. While the Endosperm Box or Endosperm-like box was not found in Z09P-2 and Z26P-3. This may indicate that gene expression drived by the two promtors was probably controlled by different trans-acting factors and the genetic control mechanism of corresponding gene expression may be diverse. 3. The B-hordein genic region coding for the mature peptide was cloned into expression vector pET-30a and transformed into bacterial strain BL21 for identifying gene expression fountion. Protein SDS–PAGE analysis showed that only the transformed lysate with the pET-BZ07-2 and pET-BZ26-5 constructs produced proteins related to B-group hordeins of barley, and the mounts of proteins induced by 3 mM IPTG and 3 h were higher than other conditions. This established a base for isolating and putifying B-hordein and further exploring their effects on barley grain quality. 4. The gene-specific primers of B-hordein genes from Z09 and Z26 were used for the quantification of B-hordein gene expression. The α-tubulin gene from Hordeum vulgare subsp. vulgare (GenBank accession number U40042) was used as a control gene. The result shows the transcription of the B-hordein genes in Z09 was found 7 days after flowering, while the transcription of the B-hordein genes in Z26 was found 4 days after flowering, but at a very low level, and it suggested that the B-hordein genes in Z26 probably expressed earlier than those in Z09, or the B-hordein genes in Z09 expressed at so a lower level than Z26 that it can not detected. In addition, B-hordein genes in Z26 accession showed higher expression levels than those in Z09 in four developing stages. Furthermore, a progressive increase in the expression levels of the B-hordein genes between 12 and 18 days after anthesis was observed in both Z09 and Z26. It implies that the B-hordein allelic variants encoded by Hor2 locus exist the differential expression in mRNA levels of during barley endosperm development.

Amorphous SiO2 thin films with about 400-500 nm in thickness were thermally grown on single crystalline silicon. These SiO2/Si samples were firstly implanted at room temperature (RT) with 100 keV carbon ions to 2.0 x 10(17),5.0 X 10(17) or 1.2 x 10(18) ions/cm(2), then irradiated at RT by 853 MeV Pb ions to 5.0 x 10(11), 1.0 X.10(12) 2.0 x 10(12) or 5.0 x 10(12) ions/cm(2), respectively. The variation of photoluminescence (PL) properties of these samples was analyzed at RT using a fluorescent spectroscopy. The obtained results showed that Pb-ion irradiations led to significant changes of the PL properties of the carbon ion implanted SiO2 films. For examples, 5.0 x 10(12) Pb-ions/cm(2) irradiation produced huge blue and green light-emitters in 2.0 x 10(17) C-ions/cm(2) implanted samples, which resulted in the appearance of two intense PL peaks at about 2.64 and 2.19 eV. For 5.0 x 10(17) carbon-ions/cm(2) implanted samples, 2.0 x 10(12) Pb-ions/cm(2) irradiation could induce the formation of a strong and wide violet band at about 2.90 eV, whereas 5.0 x 10(12) Pb-ionS/cm(2) irradiation could,create double peaks of light emissions at about 2.23 and 2.83 eV. There is no observable PL peak in the 1.2 x 10(18) carbon-ions/cm(2) implanted samples whether it was irradiated with Pb ions or not. All these results implied that special light emitters could be achieved by using proper ion implantation and irradiation conditions, and it will be very useful for the synthesis of new type Of SiO2-based light-emission materials.

The reaction of LnCl3.2LiCl with 1 equiv of MeCpNa in THF gives the complexes [(THF)2Li(mu-Cl)2]2[MeCpLn(THF)] (Ln = Nd (1), La (2)) in good yield. These precursors react further with 2 equiv of LiNPh2 to produce the new complexes [Li(DME)3][MeCpLn(NPh2)3] (Ln = La (3), Pr (4), Nd (5)). They have been characterized by elemental analyses and IR and NMR spectra, as well as by structural analyses of 1 and 3. The chloride 1 crystallizes in the monoclinic space group P2(1)/n (No. 14) with a = 12.130 (5) angstrom, b = 17.343 (5) angstrom, c = 17.016 (5) angstrom, beta = 108.54 (3)-degrees, V = 3393.87 angstrom3, Z = 4, and D(c) = 1.45 g/cm3. Least-squares refinement led to a final R value of 0.051 (I greater-than-or-equal-to 3-sigma(I(o))) for 2004 independent reflections. Complex 3 crystallizes in the monoclinic space group P2(1)/c (No. 14) with a = 18.335 (6) angstrom, b = 16.576 (5) angstrom, c = 17.461 (6) angstrom, beta = 96.04 (3)-degrees, V = 5277.17 angstrom3, D(c) = 1.26 g/cm3, Z = 4, and R = 0.057 (I greater-than-or-equal-to 2.5-sigma(I(o))) for 3378 reflections. The structure of 3 consists of discrete ion pairs [Li(DME)3]+ and [MeCpLa(NPh2)3]- with average La-N and La-C(ring) distances of 2.459 (8) and 2.84 (1) angstrom, respectively.

以木榄(Bruguiera gymnorriza)、白骨壤(Avicennia marina)、桐花树(Aegiceras corniculata)、秋茄(Kandelia candel)和海漆(Excoecaria agallocha)为对象,以光合作用对环境因子的响应为主线,建立了从叶片水平到群体冠层水平上的光合产量模型,探讨了从器官、个体到群体的光合产量对环境因子响应的定量关系。 将Farquhar提出的单叶片光合作用生理生化模型与气孔导度B-B模型相结合,建立了光合作用-气孔导度耦合模型。模型模拟结果与实际测量结果具有较好的一致性。在温度为25.0℃,光合有效辐射为1000μmol•m-2s-1 的条件下,当外部CO2浓度倍增到720μmol•mol-1时,白骨壤、木榄、桐花树、秋茄、海漆的光合速率分别提高22.56%,17.13%,18.43%,18.63%和18.41%。在大气CO2浓度和光合有效辐射通量密度不变的条件下,光合作用速率对温度的响应呈单峰型曲线,即有一个最适温度,5种红树植物的最适温度值均为26.5℃左右。大气CO2浓度和温度固定不变(分别为350μmol•mol-1和25.0 ℃)时,光合作用对光合有效辐射的响应符合Michaelis-Menten反应曲线,模型在PAR<1800μmol•m-2s-1时模拟精度较高(P<0.01)。 在典型晴天条件下,5种红树植物的光合速率日变化都出现两个极大值(分别在11时和15时左右),中午前后光合速率较低,模型模拟光合速率日变化与实测数值日变化趋势一致。本模型能较好地模拟5种红树植物光合产量以及对环境因子的响应,模拟预测精度较高(P<0.01)。 以Ross和Nilson叶倾角分布模型为基础,分别建立了直接辐射和散射辐射在冠层内传输的子模型。冠层内的消光系数均有明显的日变化,且上午8时之前和下午16时之后随时间变化较大。在典型晴天条件下,单位土地面积日合成干物质总量(折合为CH2O)白骨壤为15.840g•m-2d-1,对于木榄、桐花树、秋茄、海漆其相应的值分别为 22.254 g•m-2d-1, 23.610 g•m-2d-1,24.525 g•m-2d-1和25.996 g•m-2d-1 。
