By using 19 Y chromosome biallelic markers and 3 Y chromosome microsatellite markers, we analyzed the genetic structure of 31 indigenous Sino-Tibetan speaking populations (607 individuals) currently residing in East, Southeast, and South Asia. Our results showed that a T to C mutation at locus M122 is highly prevalent in almost all of the Sino-Tibetan populations, implying a strong genetic affinity among populations in the same language family. Furthermore, the extremely high frequency of H8, a haplotype derived from M122C, in the Sino-Tibetan speaking populations in the Himalayas including Tibet and northeast India indicated a strong bottleneck effect that occurred during a westward and then southward migration of the founding population of Tibeto-Burmans. We, therefore, postulate that the ancient people, who lived in the upper-middle Yellow River basin about 10,000 years ago and developed one of the earliest Neolithic cultures in East Asia, were the ancestors of modem Sino-Tibetan populations.

南北纬四十度之间的山区在冰期时为很多植物提供了避难所,因而受到生物地理学研究者的广泛关注。地中海地区和青藏高原(东南部)均是全球生物多样性热点地区,很多生物类群间断分布于这两个地区,研究这种间断分布格局的形成过程具有重要理论意义。雪松属(Cedrus)含有四个关系密切的种,间断分布于地中海地区和喜马拉雅西部。在本研究中,我们从父性遗传的叶绿体和母性遗传的线粒体基因组中选取了八个DNA片段,通过序列分析重建雪松属所有物种的系统发育关系,并探讨该属生物地理格局的形成过程。主要研究结果如下: 所有分析的DNA片段中均未发现种内变异。5个叶绿体片段的合并分析、3个线粒体片段的合并分析以及所有8个细胞质DNA片段的联合分析均只得到一棵最简约树,而且不同分析得到的树图结构高度一致。雪松属分为喜马拉雅(C. deodara)和地中海两支;在地中海雪松支系中,北非雪松(C. atlantica)构成一个亚支,地中海东部的黎巴嫩雪松(C. libani)和塞浦路斯雪松(C. brevifolia)形成另一亚支,两个亚支间为姐妹群关系,并已发生较大的遗传分化。该结果与细胞和生化等研究结果一致。 雪松属的可靠化石发现于哈萨克斯坦西部的渐新世地层、欧洲西南部的中新世地层、欧洲东南部的上新世地层以及撒哈拉的早更新世地层,这种化石分布暗示雪松属可能是在欧亚大陆的高纬度地区起源的。自第三纪开始,全球由广泛的高温气候演变为具有大面积陆地冰盖的寒冷气候,这种气候变化促进了生物群的演替和分化。我们对雪松属的系统发育研究表明:该属植物可能在欧亚大陆高纬度地区起源后,因气候变冷而南迁,并通过隔离分化形成环地中海-喜马拉雅间断分布格局。结合青藏高原和地中海地区的气候和地质历史,我们推测雪松属在中新世以后才分布到喜马拉雅地区,并很可能在第三纪末才迁移到北非。

Black-and-white snub-nosed monkeys (Rhinopithecus bieti) are endemic to the Trans-Himalayas in Northwest Yunnan and Southeast Tibet between the upper Yangtze and Mekong Rivers. Based on field surveys and previous reports, we identified the dark-coniferous forest, the mixed coniferous and broadleaf forest, and oak patches as suitable habitats (SH) for the monkeys. Summer grazing lands (SGL), which were made by local people cutting and burning the dark-coniferous forest at the high altitude belt, replaced SH. To have a general view of the status of the SH in Yunnan, we estimated the areas of SH and SGL from satellite images in 1997, and compared with areas estimated from aerial photo-based maps (ca. 1958). The work resulted in: 1) the area of SH was 4,169 km(2) in 1997; 2) SGL was 1,923 km(2); 3) during the past 40 years, the area of SH decreased by 31% (1,887 km(2)), and SGL increased by 204% (1,291 km(2)); and 4) the mean size of forest patches decreased from 15.6 to 5.4 km(2). In addition, the area of SGL is positively correlated to local human population (R-2 greater than or equal to0.53), implying that the reduction and fragmentation of habitat for Rhinopithecus bieti is a result of population growth of humans, who mostly employ traditional modes of production. Only 11 monkey groups remained in the changing habitat. Considering that forests at lower elevation were also encroached upon by farmlands in a similar way, the forest ecosystem is highly threatened. The destruction will continue unless there is a change in the mode of production in the region.

To investigate the genetic diversity between the populations of woolly flying squirrels (Eupetaurus) from the eastern and western extremes of the Himalayas, partial mitochondrial cytochrome b gene sequences (390-810bp) that were determined from the museum specimens were analyzed using maximum parsimony (MP) and maximum likelihood (ML) methods. The molecular data reveal that the two specimens that were collected in northwestern Yunnan (China) are members of the genus Eupetaurus. Reconstructed phylogenetic relationships show that the populations of Eupetaurus in the eastern and western extremes of the Himalayas are two distinct species with significant genetic differences (12%) and diverged about 10.8 million years ago. Eupetaurus is significantly different from Petaurista and Pteromys. The level of estimated pairwise-sequence divergence observed between Eupetaurus and Petaurista or Pteromys is greater than that observed between Eupetaurus and Trogopterus, Belomys, Glaucomys, or Hylopetes. Considering the divergence time of the two Eupetaurus groups, the glaciations and the uplift of the Himalayas and Qinghai-Tibet plateau during the Pliocene-Pleistocene period might be the major factors affecting the present distribution of Eupetaurus along the Himalayas. The family Sisoridae is one of the largest and most diverse Asiatic catfish families, most species occurring in the water systems of the Qinhai-Tibetan Plateau and East Himalayas. To date published morphological and molecular phylogenetics hypotheses of sisorid catfishes are part congruent, and there are some areas of significant disagreement with respect to intergeneric relationships. We used mitochondrial cytochrome b and 16S rRNA gene sequences to clarify existing gaps in phylogenetics and to test conflicting vicariant and dispersal biogeographical hypotheses of Chinese sisorids using dispersal-vicariance analysis and weighted ancestral area analysis in combination with palaeogeographical data as well as molecular clock calibration. Our results suggest that: (1) Chinese sisorid catfishes form a monophyletic group with two distinct clades, one represented by (Gagata (Bagarius, Glyptothorax)) and the other by (glyptosternoids, Pseudecheneis); (2) the glyptosternoid is a monophyletic group and Glyptosternum, Glaridoglanis, and Exostoma are three basal species having a primitive position among it; (3) a hypothesis referring to Pseudecheneis as the sister group of the glyptosternoids, based on morphological evidence, is supported; (4) the genus Pareuchiloglanis, as presently defined, is not monophyletic; (5) congruent with previous hypotheses, the uplift of Qinghai-Tibetan Plateau played a primary role in the speciation and radiation of the Chinese sisorids; and (6) an evolutionary scenario combining aspects of both vicariance and dispersal theory is necessary to explain the distribution pattern of the glyptosternoids. In addition, using a cytochrome b substitution rate of 0.91% per million years and 0.23% for 16S rRNA, we tentatively date that the glyptosternoids most possibly originated in Oligocene-Miocene boundary (19-24Myr), and radiated from Miocene to Pleistocene, along with a center of origin in the Irrawaddy-Tsangpo drainages and several rapid speciation in a relatively short time. Based on the phylogenetic and biogeographical studies of the glyptosternoid fishes in Qinghai-Tibet area, the following hypothesis is proposed: the speciation of this group has a direct relationship with the three uplift intervals of the Qinghai-Tibet Plateau. This process was explained by the theory of vicariance of biogeography. The ancestor of this group was similar to Bagarus and/or Glyptothorax, which still have a wide distribution. At the moment when the Tethys sea closed, the Indian tectonic plate collided with the Eurasian tectonic plate, so the Glyptothorax-like and Bagarus-like ancestors entered Eurasia and gradually became widely distributed. After the Pleistocene, with the enforced colliding, the gradual uplift of the Qinghai-Tibet Plateau brought about the current water environment, and the Glyptosternoids were generated from Glyptothorax-like fish under this environment. The present Glyptosternum, distributed across the Himalayas is the ancestor of Glyptosternoids. In the three uplift intervals of the plateau, the water system of this region was separated gradually and Glyptosternum-like ancestor was isolated in different rivers and evolved into various species. All this resulted in the speciation and formation of the biogeographical pattern of glyptosternoids.

Background and Aims It is an enduring question as to the mechanisms leading to the high diversity and the processes producing endemics with unusual morphologies in the Himalayan alpine region. In the present study, the phylogenetic relationships and origins of three such endemic genera were analysed, Dolomiaea, Diplazoptilon and Xanthopappus, all in the tribe Cardueae of Asteraceae.Methods The nuclear rDNA internal transcribed spacer (ITS) and plastid trnL-F and psbA-trnH regions of these three genera were sequenced. The same regions for other related genera in Cardueae were also sequenced or downloaded from GenBank. Phylogenetic trees were constructed from individual and combined data sets of the three types of sequences using maximum parsimony, maximum likelihood and Bayesian analyses.Key Results The phylogenetic tree obtained allowed earlier hypotheses concerning the relationships of these three endemic genera based on gross morphology to be rejected. Frolovia and Saussurea costus were deeply nested within Dolomiaea, and the strong statistical support for the Dolomiaea-Frolovia clade suggested that circumscription of Dolomiaea should be more broadly redefined. Diplazoptilon was resolved as sister to Himalaiella, and these two together are sister to Lipschitziella. The clade comprising these three genera is sister to Jurinea, and together these four genera are sister to the Dolomiaea-Frolovia clade. Xanthopappus, previously hypothesized to be closely related to Carduus, was found to be nested within a well-supported but not fully resolved Onopordum group with Alfredia, Ancathia, Lamyropappus, Olgaea, Synurus and Syreitschikovia, rather than the Cardinis group. The crude dating based on ITS sequence divergence revealed that the divergence time of Dolomiaea-Frolovia from its sister group probably occurred 13.6-12.2 million years ago (Ma), and the divergence times of the other two genera, Xanthopappus and Diplazoptilon, from their close relatives around 5.7-4.7 Ma and 2.0-1.6 Ma, respectively.Conclusions The findings provide an improved understanding of the intergeneric relationships in Cardueae. The crude calibration of lineages indicates that the uplifts of the Qiinghai -Tibetan Plateau since the Miocene might have served as a continuous stimulus for the production of these morphologically aberrant endemic elements of the Himalayan flora.

Background and Aims The systematic position of the genus Metagentiana and its phylogenetic relationships with Crawfurdia, Gentiana and Tripterospermum have not been explicitly addressed. These four genera belong to one of two subtribes (Gentianinae) of Gentianeae. The aim of this paper is to examine the systematic position of Crawfurdia, Metagentiana and Tripterospermum and to clarify their phylogenetic affinities more clearly using ITS and trnL intron sequences.Methods Nucleotide sequences from the internal transcribed spacers (ITS) of nuclear ribosomal DNA and the plastid DNA trnL (UAA) intron were analysed phylogenetically. Ten of fourteen Metagentiana species were sampled, together with 40 species of other genera in the subtribe Gentianinae.Key Results The data support several previously published conclusions relating to the separation of Metagentiana from Gentiana and its closer relationships to Crawfurdia and Tripterospermum based on studies of gross morphology, floral anatomy, chromosomes, palynology, embryology and previous molecular data. The molecular clock hypothesis for the tested sequences in subtribe Gentianinae was not supported by the data (P < 0.05), so the clock-independent non-parametric rate smoothing method was used to estimate divergence time. This indicates that the separation of Crawfurdia, Metagentiana and Tripterospermum from Gentiana occurred about 11.4-21.4 Mya (million years ago), and the current species of these three genera diverged at times ranging from 0.4 to 6.2 Mya.Conclusions The molecular analyses revealed that Crawfurdia, Metagentiana and Tripterospermum do not merit status as three separate genera, because sampled species of Crawfurdia and Tripterospermum are embedded within Metagentiana. The speciation and rapid radiation of these three genera is likely to have occurred in western China as a result of upthrust of the Himalayas during the late Miocene and the Pleistocene.

The presumed pair relationships of intercontinental vicariad species in the Podophyllum group (Sinopodophyllum hexandrum vs. Podophyllum pelatum and Diphylleia grayi vs. D. cymosa) were recently, considered to be paraphyletic. In the present paper, the trnL-F and ITS gene sequences of the representatives were used to examine the sister relationships of these two vicariad species. A heuristic parsimony analysis based on the trnLF data identified Diphylleia as the basal clade of the other three genera, but provided poor resolution of their inter-relationships. High sequence divergence was found in the ITS data. ITS1 region, more variable but parsimonyuninformative. has no phylogenetic value, Sequence divergence of the ITS2 region provided abundant, phylogenetically informative variable characters. Analysis of ITS2 sequences confirmeda sister relationship between the presumable vicariad species, in spite of a low bootstrap support for Sinopodophyllum hexandrum vs. Podophyllum pelatum. The combined ITS2 and trnL-F data enforced a sister relationship between Sinopodophyllum hexandrum and Podophyllum pelatum with an elevated bootstrap support of 100%. Based on molecular phylogeny, the morphological evolution of this group was discussed. The self-pollination might have evolved from cross-fertilization two times in this group. The different pollination and seed dispersal systems of Sinopodophyllum hexandrum and Podophlyllum pelatum resulted from their adaptations to different ecological habitats. The divergence time of Sinopodophyllum hexandrum-Podophyllum pelatum is estimated to be 6.52+/-1.89 myr based on the ITS divergence. The divergence of this species pair predated or co-occurred with the recent uplift of the Himalayas 4-3 myr during the late Miocene and the formation of the alpine habitats. Sinopodophyllum hexandrum developed a host of specialized characters in its subsequent adaptation to the arid alpine surroundings. The present study confirmed the different patterns of species relationship between Asian-North American disjuncts. The isolation of plant elements between North America and eastern Asia must have been a gradual process, resulting in the different phylogenetic patterns and divergence times of the disjuncts.

Glaciers in west China are the sources of the major great rivers in Asia, and the solid water resources are crucial to China and South Asia. Black carbon (BC) results in very complex climate effects not only in the atmosphere, but accelerates the melting after its deposit on the surface of snow/ice. As the main distributed area of glaciers in China, the Tibetan Plateau (TP) and Xinjiang region are abutted by South Asia, Central Asia, and Russia, and east China, and the atmospheric environment would be influenced by the BC emitted from these regions. Whereas, the BC’s temporal and spatial distributions for concentration in the mid and top troposphere in west China, its transport, and its radiative forcing after deposited on the snow/ice surface are not well understood at the present. In the field, we collected samples from surface snow, snow pits, ice core, and aerosol in the glaciers, analyzed BC content mainly by the thermo-oxidized method in the laboratory, and discussed temporal and spatial distributions for BC concentrations in glaciers, the transport, and its impacts on the environment. Several conclusions were derived as follows: 1_Spatial distribution and the impact on albedos for BC concentrations in snow/ice: the BC concentrations in the surface snow for the investigated glaciers could be placed in areas, the Tianshan Mountains > the central TP > the Pamirs > the Qilian Mountians > the Himalayas. This distribution could be attributed to the elevation of the glaciers, the topography of the TP, and more regional emissions. Probably significant impacts on the albedos of the glacier surface could be caused by BC deposits, and the estimated reduced albedos on the glaciers are 9.8% (the Zhadang glacier), 8.7% (the Miao’ergou Riverhead No.3 glacier), and 6.8% (the Kuitun River Haxilegen No.48 glacier), and 6.2% (the Dongkemadi glacier), and 5.3% (the La’nong glacier), and 4.2% (the Muztagata glacier), etc. 2_The temporal variance of BC concentrations in ice of the East Rongbuk Glacier (ERG) and its climatic implications: major cations and anions (e.g., SO42- and Ca2+) concentrations in aerosols during summer monsoon seasons showed their close relationships with the sources of air masses, in which the variance of SO42- concentrations suggested the atmospheric environment over the ERG was significantly influenced by the aerosols from South Asia. BC record based on an ice core suggested its deposit was dominantly transported by monsoons in summers and by westerlies in other seasons, and the BC from South Asia in summers dominated the varying trend of its concentrations in the ice core and caused higher concentrations in summers than those in other seasons. In the past 50 yrs, BC concentrations showed fluctuations, whereas showed an increasing tread in the most recent decade, and exceeded 50 μg kg-1 in the summer of 2001; correspondingly, the radiative forcing caused by BC showed an increasing trend since 1990s, and exceeded 4.5 W m-2 in the summer of 2001. 3_Cabonaceous aerosols in the Nam Co region: organic carbon (OC) concentration accounted for ~95% and BC for ~5% in the total carbonaceous aerosol concentration, which was significantly influenced by summer precipitations. OC was dominantly derived from fossil fuel burning and BC from both fossil fuel and biomass burning. Trajectory analysis and aerosol optical depth suggested the atmospheric environment in the Nam Co region was most probably influenced by the emissions from South Asia. The potential source regions of air pollutants in the Nam Co regions in summers might be Bangladesh and east India, and in winters might be the Indo-gangetic basin. The scavenging ratio of atmospheric BC by rainfalls was less than those at other sites. West China is a less-developed region for industry, where BC concentrations in the atmosphere and snow/ice could be significantly influenced by the emissions from the abutted regions with rising industries (South Asia, Central Asia, and Russia). For example, snow/ice BC concentrations in the glaciers of the Parmirs, the Tianshan Mountains, and the Qilian Mountains in the northeast margin of the TP might be more influenced by the emissions from Centrial Asia (transported by the westerlies), those in the glaciers of the Himalayas might be more influenced by the emissions from South Asia (transported by the monsoons and the westerlies), and atmospheric carbonaceous aerosols might also be more influenced by the emissions from South Asia (transported by the monsoons and the westerlies). The BC concentrations in some glaciers might cause significant impacts on the albedos for the glaciers, and therefore enhanced the radiative forcings, for example, the ERG. The research on the relationships among atmospheric and snow/ice BC and its radiative forcing, variance of snow cover, mass balance of glaciers, and climate forcing would be needed in future.
