Temporal and spatial variability in the kinetic parameters of uptake of nitrate (NO3-), ammonium (NH4+), urea, and glycine was measured during dinoflagellate blooms in Changjiang River estuary and East China Sea coast, 2005. Karenia mikimotoi was the dominant species in the early stage of the blooms and was succeeded by Prorocentrum donghaiense. The uptake of nitrogen (N) was determined using N-15 tracer techniques. The results of comparison kinetic parameters with ambient nutrients confirmed that different N forms were preferentially taken up during different stages of the bloom. NO3- (V-max 0.044 h(-1); K-s 60.8 mu M-N) was an important N source before it was depleted. NH4+ (V-max 0.049 h(-1); K-s 2.15 mu M-N) was generally the preferred N. Between the 2 organic N sources, urea was more preferred when K. mikimotoi dominated the bloom (V-max 0.020 h(-1); K-s 1.35 mu M-N) and glycine, considered as a dominant amino acid, was more preferred when P. donghaiense dominated the bloom (V-max 0.025 h(-1); K-s 1.76 mu M-N). The change of N uptake preference by the bloom-forming algae was also related to the variation in ambient N concentrations. Published by Elsevier B.V.

During late spring and early summer of 2005, large-scale (> 15 000 km(2)), mixed dinoflagellate blooms developed along the the coast of the East China Sea. Karenia mikimotoi was the dominant harmful algal bloom species in the first stage of the bloom (late May) and was succeeded by Prorocentrum donghaiense approximately 2 wk later. Samples were collected from different stations along both north-south and west-east transects, from the Changjiang River estuary to the south Zhejiang coast, during 3 cruises of the Chinese Ecology and Oceanography of Harmful Algal Blooms Program, before and during the bloom progression. Nitrogen isotope tracer techniques were used to measure rates of NO3-, NH4+, urea, and glycine uptake during the blooms. High inorganic nitrogen (N), but low phosphorus (P) loading from the Changjiang River led to high dissolved inorganic N:dissolved inorganic P ratios in the sampling area and indicate the development of P limitation. The rates of N-15-uptake experiments enriched with PO43- were enhanced compared to unamended samples, suggesting P limitation of the N-uptake rates. The bloom progression was related to the change in availability of both organic and inorganic N and P. Reduced N forms, especially NH4+, were preferentially taken up during the blooms, but different bloom species had different rates of uptake of organic N substrates. K mikimotoi had higher rates of urea uptake, while P. donghaiense had higher rates of glycine uptake. Changes in the availability of reduced N and the ratios of N:P in inorganic and organic forms were suggested to be important in the bloom succession. Nutrient ratios and specific uptake rates of urea were similar when compared to analogous blooms on the West Florida Shelf.

Screening experiments were conducted in order to find promising synthetic surfactants for harmful algal blooms (HABs) mitigation. The chemically synthesized surfactant cocamidopropyl betaine (CAPB) showed characteristics of relatively high inhibition efficiency, high biodegradability and low cost. The motility inhibition ratios of 10 mg/L CAPB on Cochlodinium polykrikoides and Alexandrium tamarense were about 60% after 5 min. The biodegradation test indicated that the half-life of CAPB in seawater was shorter than one day and 90% was biodegraded after five days under the initial concentration of 100 mg/L at 25degreesC. Further cell lysis experiments revealed the selective lysis effect of CAPB on different HAB organisms. More than 90% of C. polykrikoides lysed at the concentration of 10 mg/L CAPB after 24 h and at 15 mg/L CAPB after 4 h, whereas the lysis effect of CAPB on A. tamarense was slight, no more than 10% after 2 h interaction with 50 mg/L CAPB. This research provided preliminary data for CAPB as a candidate in harmful algal blooms mitigation and pointed out unresolved problems for its practical application in the meantime. The inhibition effect of sophorolipid and removal efficiency of loess on Cochlodinium polykrikoides and Alexandrium tamarense was investigated separately in the laboratory. Based on this, the combination of sophorolipid and loess for harmful algal bloom mitigation was proposed. Algal sedimentation tests in the laboratory and in the field revealed that the combination of sophorolipid and loess showed synergistic effects both on the removal efficiencies and on the mitigation cost. The concentration of 1 g/l loess and 5 mg/l sophorolipid was determined as the optimum ratio for C polykrikoides mitigation. In the field test, the effective concentration of loess and sophorolipid in the combination group was reduced to 10% and 25%, respectively, compared to the non-combination group, and the cost decreased more than 60%. The combination of loess and sophorolipid was considered as a promising novel method in harmful algal bloom mitigation. (C) 2003 Elsevier Ltd. All rights reserved.

The effects of a PSP producing dinoflagellate Alexandrium tamarense on marine bivalves at their several important life,stages: egg, D - shape larva, eyespot larva, juvenile and adult, were studied! The results show that the hitching survival, activity, filtration and! growth were adversely affected by the alga and the impact was significantly increased with the increase of algal density. The inhibitory effect on egg hatching was most significant, which the hatching rate was only 30% of the control when exposed to the alga at 100 cell/cm(3) after 36 h. Further experiments show that the algal culture, re-suspended cells and cell fragments had the inhibitory effect, while no such effect was from the cell-free medium, cell contents and standard STX. The results indicate that the alga could produce unknown toxins, rather than PSP, associated with the cell surface.

in order to investigate a new method of mitigating the deleterious effect of harmful algal blooms (HABs), the inhibition of the glycolipid biosurfactant sophorolipid on three common harmful algae Alexandrium tamarense, Heterosigma akashiwo and Cochlodinium polykrikoides was studied. The optimum preparation condition for sophorolipid, the inhibition capability of sophorolipid and the interaction mechanism of sophorolipid on the three algal species were investigated. Results showed that sophorolipid prepared by extraction with ethyl acetate exhibited the most prominent inhibition effect and that storage time of one year had little influence on the inhibition effect of sophorolipid. The optimum concentration of 10-20 mg/l sophorolipid inhibited the motility of about 90% of the tested harmful algal cells without recovery. Investigation of the algicidal process revealed that sophorolipid induced ecdysis of A. tamarense, quick lysis of H. akashiwo and swelling of C. polykrikoides in a relatively short time. Investigation of the nucleotides showed that more than 15% of the nucleotides were released from the cytoplasm under the effect of 10-20 mg/l sophorolipid, indicating the irreversible damage on the cellular membrane, which resulted in the disintegration of the harmful algal cells. 本论文从生态环境可持续发展观出发,选用了两种天然高分子聚合物壳聚糖与黄原胶作为改性剂,考察了它们在海水体系中的絮凝性能以及改性当地岸沙絮凝除藻的性能,同时探讨了赤泥复合剂絮凝除藻的性能。研究结果表明: 海水体系中的离子强度阻碍了壳聚糖高分子链的舒展,有效除藻时投加量大于50 mg/L;聚合氯化铝的加入,降低了壳聚糖的投加量,并且10 mg/L壳聚糖与10 mg/L聚合氯化铝协同对100 mg/L当地岸沙进行改性,改性后沙子具有较强的除藻效果,3 min后,强壮前沟藻与海洋小球藻的去除率为80%,沉淀4 h后,两种藻的去除率高达92-96%。 黄原胶单独使用时强壮前沟藻的去除率为32%-55%;氢氧化钙的加入,提高了黄原胶的絮凝活性;当黄原胶:氢氧化钙:土壤/沙子=1:5:15时,黄原胶投加量为20 mg/L时,30 min后,强壮前沟藻的去除率为83%-89%。 赤

Both arsenic pollution and eutrophication are prominent environmental issues when considering the problem of global water pollution. It is important to reveal the effects of arsenic species on cyanobacterial growth and toxin yields to assess ecological risk of arsenic pollution or at least understand naturally occurring blooms. The sensitivity of cyanobacteria to arsenate has often been linked to the structural similarities of arsenate and phosphate. Thus, we approached the effect of arsenate with concentrations from 10(-8) to 10(-4) M on Microcystis strain PCC7806 under various phosphate regimes. The present study showed that Microcystis strain PCC7806 was arsenate tolerant up to 10(-4) M. And such tolerance was without reference to both content of intra- and extra-cellular phosphate. It seems that arsenate involved the regulation of microcystin synthesis and cellular polyphosphate contributed to microcystin production of Microcystis responding to arsenate, since there was a positive linear correlation of the cellular microcystin quota with the exposure concentration of arsenate when the cells were not preconditioned to phosphate starvation. It is presumed that arsenate could help to actively export microcystins from living Microcystis cells when preconditioned to phosphate starvation and incubated with the medium containing 1 mu M phosphate. This study firstly provided evidence that microcystin content and/or release of Microcystis might be impacted by arsenate if it exists in harmful algal blooms. (C) 2008 Wiley Periodicals, Inc. 有害赤潮(Harmful algal blooms, HAB)已成为全球性的海洋灾害,对我国近海养殖业危害尤为巨大,研究赤潮发生、发展的主要环境影响因子,对于控制赤潮的发生以及加强赤潮的防治具有十分重要的意义。本文根据国家自然科学基金“九五”重大项目“中国沿海典型增养殖区有害赤潮发生动力学防治对策研究”项目组近年来对胶州湾东北部养殖海域的监测资料,分析了该海区营养状况,结合该区自然环境特点,探讨了发生于1998年7月和1999年6月两次硅藻赤潮的主要影响因子;并应用胶州现场海水进行了室内模拟培养实验,对影响有害赤潮生消过程的关键环境因子进行了分析探讨。对1998和1999年赤潮多发季节胶州湾东北部增养殖水域海水的营养状况进行了分析,并参照潜在性富营养化的概念,应用NQI指数对该海域的富营养化水平进行了评价。结果表明:(1)胶州湾东北部增养殖海域赤潮多发季节TIN、DIP和活性硅酸盐的含量均较高,远高于同期胶州湾大面观测站各营养盐的平均含量;该海域有机污染物含量相对营养盐含量不高。(2)该海域春季氮、磷营养盐含量充足,水域属富营养化平;夏季该海域具有较高的生物具,磷酸盐水平相对较低,海域多数时间属于磷限制潜在性富营养化水平;但这种限制极易为外界补充所改变,如陆源输入、养殖废水的排放等。(3)该海域为典型增养殖区,养殖废水对海域营养盐的补充有重要意义,因此应当加强对该海域养殖业的综合管理。对分别于1998年7月和1999年6月在胶州湾东北部增养殖海域发生的中肋骨条藻(Skeletonema costatum)赤潮和浮动弯角藻(Eucampia zoodiacus)赤潮生消过程中环境因子的变化进行了探讨。发生赤潮生物量的升高常伴随海水pH值、溶解氧(DO)含量的升高以及各营养盐(氮、磷、硅)含量的降低;认为在该海域较高营养盐含量和较高生物量的情况下,赤潮发生前大规模的降雨过程及随后的气候条件为赤潮生物的快速生长繁殖创造了有利条件;无机磷和活性硅酸盐含量相对于无机氮显得较低,对该海域硅藻赤潮的生消过程起着控制作用。采集胶州湾现场海水进行了实验室模拟培养实验,对影响赤潮生消过程的主要环境因素进行了研究和探讨。结果表明,现场海水进行模拟培养时,排除了潮流、风以及水团间交流混合等因素对水体中浮游生物的影响,培养水体中的浮游植物生物量都存在增高的现象;在氮、磷、硅等营养盐含量充足的条件下,浮游植物可快速进入指数增长期,并较快发展形成赤潮,高浓度的营养盐常导致培养水体中浮游植物的较高的生长速率,浮游植物对氮磷营养盐的吸收受到环境中营养盐含量的影响;初始环境下营养盐的限制可抑制浮游植物的进一步增殖,发生赤潮水体中某种营养盐的含量降低最终导致了赤潮的消亡。

Nutrient input from the Changjiang River (Yangtze River) has been increasing dramatically since the 1960s. At the mouth of the Changjiang River, the nitrate concentration has increased about three-fold in 40 years, from 20.5 mu mol/L in the 1960s to 59.1 mu mol/L in the 1980s and to 80.6 mu mol/L in 1990-2004. Phosphate concentration increased by a factor of 30%, from 0.59 mu mol/L in the 1980s to 0.77 mu mol/L in 1990-2004. The increasing nitrate input has arisen mostly from the mid and lower reaches of the Changjiang River, where the river meets one of the most strongly developed agriculture areas in China. Responses of the coastal phytoplankton community to the increasing nutrient inputs are also seen in the available monitoring data. First, a trend of increasing phytoplankton standing stock from 1984 to 2002 appeared in the Changjiang River estuary and adjacent coastal waters, especially in late spring. Secondly, the proportion of diatoms in the whole phytoplankton community showed a decreasing trend from about 85% in 1984 to about 60% in 2000. Finally, red tides/harmful algal blooms increased dramatically in this area in terms of both number and scale. About 30-80 red tide events were recorded each year from 2000 to 2005 in the East China Sea. The scale of some blooms has been in excess of 10,000 km(2). (c) 2008 Elsevier Ltd. All rights reserved.

The impacts of Prorocentrum donghaiense Lu and Alexandrium catenella Balech, causative species of the large-scale HAB in the East China Sea, were studied under laboratory conditions. According to bloom densities, the effects of monoculture and mixture of the two species were examined on the egg-hatching success of Argopecten irradians Lamarck, and the population growth of Brachionus plicatilis Muller and Moina mongolica Daday. The results showed that monoculture of A. catenella had a significant inhibition on the egg hatching success of A. irradians, and the population growth of B. plicatilis and M. mongolica. The median effective densities ( EDSo) inhibiting the egg hatching success of A. irradians for 24 h and the population growth of B. plicatilis and M. mongolica for 96 h were 800, 630, and 2 400 cells/cm(3), respectively. Monoculture of P. donghaiense has no such inhibitory effect on the egg hatching success of A. irradians; P. donghaiense at lower suitable densities could sustain the population growth of B. plicatilis (1 x 10(4) similar to 3 x 10(4)cells/cm(3)) and M. mongolica (2 x 10(4) similar to 5 x 10(4) cells/cm(3)); P. doaghaiense at higher densities had significantly adverse effect on the population growth of B. plicatilis (4 x 10(4) similar to 10 x 10(4) cells/cm(3)) and M. mongolica (10 x 10(4) cells/cm(3)). When the two algae were mixed according to bloom densities, P. donghaiense at suitable densities to some extent could decrease the toxicity of A. catenella to B. plicatilis and M. mongolica. The results indicated that the large-scale HAB in the East China Sea could have adverse effect on zooplankton, and might further influence the marine ecosystem, especially when there was also Alexandrium bloom.

Red tides (high biomass phytoplankton blooms) have frequently occurred in Hong Kong waters, but most red tides occurred in waters which are not very eutrophic. For example, Port Shelter, a semi-enclosed bay in the northeast of Hong Kong, is one of hot spots for red tides. Concentrations of ambient inorganic nutrients (e.g. N, P), are not high enough to form the high biomass of chlorophyll a (chl a) in a red tide when chl a is converted to its particulate organic nutrient (N) (which should equal the inorganic nutrient, N). When a red tide of the dinoflagellate Scrippsiella trochoidea occurred in the bay, we found that the red tide patch along the shore had a high cell density of 15,000 cells ml(-1), and high chl a (56 mu g l(-1)), and pH reached 8.6 at the surface (8.2 at the bottom), indicating active photosynthesis in situ. Ambient inorganic nutrients (NO3, PO4, SiO4, and NH4) were all low in the waters and deep waters surrounding the red tide patch, suggesting that the nutrients were not high enough to support the high chl a >50 mu g l(-1) in the red tide. Nutrient addition experiments showed that the addition of all of the inorganic nutrients to a non-red-tide water sample containing low concentrations of Scrippsiella trochoidea did not produce cell density of Scrippsiella trochoidea as high as in the red tide patch, suggesting that nutrients were not an initializing factor for this red tide. During the incubation of the red tide water sample without any nutrient addition, the phytoplankton biomass decreased gradually over 9 days. However, with a N addition, the phytoplankton biomass increased steadily until day 7, which suggested that nitrogen addition was able to sustain the high biomass of the red tide for a week with and without nutrients. In contrast, the red tide in the bay disappeared on the sampling day when the wind direction changed. These results indicated that initiation, maintenance and disappearance of the dinoflagellate Scrippsiella trochoidea red tide in the bay were not directly driven by changes in nutrients. Therefore, how nutrients are linked to the formation of red tides in coastal waters need to be further examined, particularly in relation to dissolved organic nutrients. (C) 2008 Elsevier B.V. To investigate harmful effects of the dinoflagellate Alexandrium species on microzooplankton, the rotifer Brachionus plicatilis was chosen as an assay species, and tested with 10 strains of Alexandrium including one known non-PSP-producer (Alexandrium tamarense, AT-6). HPLC analysis confirmed the PSP-content of the various strains: Alexandrium lusitanicum, Alexandrium minutum and Alexandrium tamarense (ATHK, AT5-1, AT5-3, ATC102, ATC103) used in the experiment were PSP-producers. No PSP toxins were detected in the strains Alexandrium sp1, Alexandrium sp2. Exposing rotifer populations to the densities of 2000 cells ml(-1) of each of these 10 Alexandrium strains revealed that the (non-PSP) A. tarnarense (AT-6) and two other PSP-producing algae: A. lusitanicum, A. minutum, did not appear to adversely impact rotifer populations. Rotifers exposed to these three strains were able to maintain their population numbers, and in some cases, increase them. Although some increases in rotifer population growth following exposures to these three algal species were noted, the rate was less than for the non-exposed control rotifer groups. In contrast, the remaining seven algal strains (A. tamarense ATHK, AT5-1, AT5-3, ATC102, ATC103; also Alexandrium sp1 and Alexandrium sp2) all have adverse effects on the rotifers. Dosing rotifers with respective algal cell densities of 2000 cells ml-1 each, for Alexandrium spl, Alexandrium sp2, and A. tamarense strains ATHK and ATC103 showed mean lethal time (LT50) on rotifer populations of 21, 28, 29, and 36h, respectively. The remaining three species (A. tamarense strains AT5-1, AT5-3, ATC102) caused respective mean rotifer LT50S of 56, 56, and 71 h, compared to 160 h for the unexposed "starved control" rotifers. Experiments to determine ingestion rates for the rotifers, based on changes in their Chlorophyll a content, showed that the rotifers could feed on A. lusitanicum, A. minutum and A. tamarense strain AT-6, but could graze to little or no extent upon algal cells of the other seven strains. The effects on rotifers exposed to different cell densities, fractions, and growth phases of A. tamarense algal culture were respectively compared. It was found that only the whole algal cells had lethal effects, with strongest impact being shown by the early exponential growth phase of A. tamarense. The results indicate that some toxic mechanism(s), other than PSP and present in whole algal cells, might be responsible for the adverse effects on the exposed rotifers. (C) 2004 Elsevier B.V. The effects of Alexandrium tamarense (strain ATHK) on early development of the bay scallop Argopecten irradians concentricus were studied under laboratory conditions. The algal culture was verified by HPLC to produce paralytic shellfish poisoning (PSP) at a level of 37.48 fmol/cell. Survival of the scallop larvae was not affected when they were grown with A. tamarense at concentrations of 500-10,000 cells/ml for 48 h. However, the activity of D-shape larvae was inhibited after 48-h exposure to A. tamarense at the algal cell density of 10,000 cells/ml. Scallop growth was inhibited significantly by A. tantarense during a 14-day exposure starting at the eye-spot larval stage. The size of juvenile scallops in the group of 10,000 cells/ml was only about 32% of that of the controls, although no obvious effect of A. tamarense was found on the rate of larval metamorphosis. All juvenile scallops survived in algal concentrations of 600-2400 cells/ml, however, attachment rates were significantly lower than control values after a 5-h exposure to A. tamarense at concentrations >600 cells/ml, while they were not obviously reduced after only 1 h of exposure. At concentrations >600 cells/ml, the climbing ability of juveniles was clearly reduced by exposure to A. tamarense after only 1 h. The climbing rate and height were only 55% and 45%, respectively, of those of the controls, when exposed to A. tantarense at a concentration of 600 cells/ml. The results indicated that A. tamarense blooms may have detrimental impacts on shellfish at early life stages, therefore, special attention should be paid to the toxic algal blooms in shellfish breeding area. Very little is known about how global anthropogenic changes will affect major harmful algal bloom groups. Shifts in the growth and physiology of HAB species like the raphidophyte Heterosigma akashiwo and the dinoflagellate Prorocentrum minimum due to rising CO2 and temperature could alter their relative abundance and environmental impacts in estuaries where both form blooms, such as the Delaware Inland Bays (DIB). We grew semi-continuous cultures of sympatric DIB isolates of these two species under four conditions: (1) 20 degrees C and 375 ppm CO2 (ambient control), (2)20 degrees C and 750 ppm CO2 (high CO2),(3) 24 degrees C and 375 ppm CO2 (high temperature), and (4) 24 degrees C and 750 ppm CO2 (combined). Elevated CO2 alone or in concert with temperature stimulated Heterosigma growth, but had no significant effect on Prorocentrum growth. P-Bmax (the maximum biomass-normalized light-saturated carbon fixation rate) in Heterosigma was increased only by simultaneous CO2 and temperature increases, whereas P-Bmax in Prorocentrum responded significantly to CO2 enrichment, with or without increased temperature. CO2 and temperature affected photosynthetic parameters alpha, Phi(max), E-k, and Delta F/F'(m) in both species. Increased temperature decreased and increased the Chl a content of Heterosigma and M Prorocentrum, respectively. CO2 availability and temperature had pronounced effects on cellular quotas of C and N in Heterosigma, but not in Prorocentrum. Ratios of C:P and N:P increased with elevated carbon dioxide in Heterosigma but not in Prorocentrum. These changes in cellular nutrient quotas and ratios imply that Heterosigma could be more vulnerable to N limitation but less vulnerable to P-limitation than Prorocentrum under future environmental conditions. In general, Heterosigma growth and physiology showed a much greater positive response to elevated CO2 and temperature compared to Prorocentrum, consistent with what is known about their respective carbon acquisition mechanisms. Hence, rising temperature and CO2 either alone or in combination with other limiting factors could significantly alter the relative dominance of these two co-existing HAB species over the next century.
