The effect of laminarin and Vibrio anguillarum on neutral red retention (NRR) time of lysosome and lysozyme activity in blood cells was investigated in Chinese shrimp, Fenneropenaeus chinensis. In addition the variation, of total haemocyte counts (THC) and differential haemocyte counts (DHC) were dtermined simultaneously The. results showed that the lysosome membrane stability was significantly elongated with the longest NRR time for 180 min at 3 h after lamianrin injection, (p<0.05). The lysozyme activity of haemocytes showed a moderate increase simultaneously. THC also increased and with a highest percentage of semi-granular cell counts at 3 h after laminarin injection. However, the NRR time sharply decreased with the shortest NRR time for 13 min at 1.5 h after V. anguillarum injection. Compared to the control group, the lysozyme activity obviously increased after injection, which was demonstrated at 1.5 h though the THC decreased. The percentage of hyaline cells increased obviously after V. anguillarum injection (p<0.05). The results suggested that the shrimp immune status could be evaluated by using lysosome membrane stability and lysozyme activity. (C) 2008 Elsevier B.V. All rights reserved.

Serine proteinase homologues (SPHs), as one of prophenoloxiase-activating factors (PPAFs), play critical roles in innate immunity of crabs. Based on an EST from the eyestalk full length cDNA library, the complete cDNA (designated as PtSPH) and genomic DNA of SPH from the swimming crab Portunus trituberculatus were cloned in this study. The estimated molecular weight of mature PtSPH (354 amino acids) was 38.7 kDa and its isoelectric point was 5.08. Multiple sequence alignment revealed that PtSPH lacked a catalytic residue with a substitution of Ser in the active site triad to Gly. Phylogenetic analysis indicated PtSPH together with PPAFs of Callinectes sapidus (AAS60227), Eriocheir sinensis (ACU65942), Penaeus monodon (ABE03741, ACP19563) and Pacifastacus leniusculus (ACB41380), formed a distinct cluster which only included clip-SPHs. As the first analyzed genomic structure of PPAFs in crustaceans, two introns were found in the open reading frame region of this gene. The mRNA transcripts of PtSPH could be detected in all the examined tissues, and were higher expressed in the eyestalk than that in gill, hepatopancreas, haemocytes and muscle. Accompanied with the change in phenoloxidase (PO) activity and total haemocyte counts, the temporal expression of PtSPH gene in haemocytes after Vibrio alginolyticus challenge demonstrated a clear time-dependent expression pattern with two peaks within the experimental period of 32 h. These findings suggest that PtSPH is involved in the antibacterial defense mechanism of Portunus tritubercualtus crab. (C) 2010 Elsevier Ltd. All rights reserved.

To obtain these data, 100 blood samples from Chinese rhesus macaques were collected. The normal range of major leukocyte subpopulations, such as T lymphocytes, B lymphocytes, monocytes, myeloid dendritic cells (mDCs) and plasmacytoid dendritic cells (pDCs), were quantitatively analyzed by flow cytometry through BD trucount tubes. The influence of age and sex on the cell counts of leukocyte subpopulations was analyzed. The counts of CD3+ T cells, CD3+CD4+ T cells, CD3+CD8+ T cells and B cells decreased with age, but those of monocytes, mDCs and pDCs had no significant correlation with age. Significant differences existed in the cell counts of most leukocyte subpopulations between the male and female groups except pDCs. Furthermore the values of the females were higher than those of the males. The study provided basic information about the leukocyte subpopulations of Chinese rhesus macaques, and it may be valuable for immunobiological study of Chinese rhesus macaques.
