In this paper, we apply an analytical model [V.V. Kulagin et al., Phys. Plasmas 14, 113101 (2007)] to describe the acceleration of an ultra-thin electron layer by a schematic single-cycle laser pulse and compare with one-dimensional particle-in-cell (1D-PIC) simulations. This is in the context of creating a relativistic mirror for coherent backscattering and supplements two related papers in this EPJD volume. The model is shown to reproduce the 1D-PIC results almost quantitatively for the short time of a few laser periods sufficient for the backscattering of ultra-short probe pulses.

金属硫蛋白(metallothionein,MT)和植物络合素(phytochelatin,PC)是植物中能够与金属离子结合的两大类多肽。二者均富含Cys,但前者是mRNA的编码产物,后者是酶促反应的产物,植物络合素合酶(PCS)则是合成PC的关键酶之一。目前已发现许多植物同时存在金属硫蛋白基因和植物络合素合酶基因。研究重金属胁迫下这两类基因的表达对了解植物的重金属抗性的分子机制具有重要意义,同时还可以为培育能用于植物修复的品种提供新思路。 本论文从大蒜中克隆了一个type 2 MT基因,命名为AsMT2a,将其在大蒜中的表达模式与大蒜的植物络合素合酶基因(AsPCS1)进行了比较,并对二者的过表达转基因拟南芥的重金属抗性进行研究。其主要结果如下: 1. AsMT2a基因全长525 bp,编码79个氨基酸,其中有14个Cys 残基。 推测的氨基酸序列分析表明其Cys的位置和数目与来自其它植物的type 2 MT蛋白的完全一致。 2. RT-PCR的结果显示,大蒜根部AsPCS1的表达在Cd处理的短期(1 hr)内迅速增强,同时PCs含量也大幅度增加。但AsMT2a的表达在Cd处理10 hr后才有明显的增加。说明AsPCS1可能在植物对重金属的急性解毒方面起主要作用,而AsMT2a则在植物对重金属长久耐性中的离子平衡方面起更大作用。暗示在大蒜暴露于Cd胁迫的不同时期AsPCS1和AsMT2a基因可以互相协调而对重金属胁迫作出反应。此外,在不同胁迫条件下,AsPCS1和AsMT2a的表达模式不同,其中Cd、As和热激可以促进根中AsPCS1的表达和PCs的积累。 3.将AsPCS1和AsMT2a转入对砷和镉敏感的酵母菌株 FD236-6A中,RT-PCR的结果显示这两个基因均可在酵母中稳定表达,对转化子的重金属抗性实验表明这两个基因均可提高转化子对砷和镉的抗性。 4.将AsPCS1和AsMT2a 置于 CaMV 35S启动子下转入拟南芥中,RT-PCR结果表明,这两个基因均可在拟南芥中表达。有趣的是,AsPCS1在拟南芥中存在两个转录本,且二者均具有完整的ORF,其推测的氨基酸序列相差38个氨基酸。说明部分AsPCS1在拟南芥中经过了精确的剪切和拼接过程,但其机制尚不清楚。 5.在Cd 胁迫下,AsPCS1的超表达拟南芥的生长好于野生型植株,主要表现在转基因拟南芥的根较长,根数目较多;但在As胁迫下AsPCS1转基因植株与野生型植株没有明显的差别。与此不同的是将AsMT2a转入拟南芥后,转基因植株的As抗性明显增强,同样表现在根长度和根数目上。进一步将AsPCS1和AsMT2a同时转入拟南芥进行超表达,在Cd胁迫下,转基因植株的生长好于野生型植株,且种子萌发率也较高。 6.Cd和As胁迫下,AsPCS1过表达植株的PCs含量增加,同时Cd和As的积累量也明显增加,其中Cd胁迫下Cd含量增加最多,平均比野生型对照增加4倍;而As胁迫下As含量比野生型对照增加1.2倍。在Cd和As胁迫下,AsMT2a过表达植株的Cd和As积累量与野生型相比分别增加1.4倍和0.8 倍。双价基因AsMT2a +AsPCS1过表达植株的 Cd 积累量是野生型的5.8倍,是AsMT2a过表达植株的2.4 倍,是AsPCS1过表达植株的1.2倍。 在克隆AsMT2a的同时,我们还从大蒜中克隆到了一个金属硫蛋白基因家族的新成员,命名为AsMT2b,并对其功能进行了初步探讨。主要结果如下: 1.AsMT2b 全长520 bp,其开读框架为243 bp,编码80个氨基酸,其中含有15个Cys 残基。对推测的氨基酸序列分析表明AsMT2b的N端和C端domain内,Cys的数目和排列方式与其它type 2 MT蛋白明显不同。 其N 端domain内的结构为CXXC——CXC——CXC——CXCC,C端domain 内的结构为CXXC——CXC——CXC。暗示AsMT2b 可能具有与其它MT不同的生物学功能。 2.在较低浓度Cd(200 µM)胁迫下,AsMT2b的表达量随着处理时间(24 hr内)的延长而降低,但随着处理浓度的升高(500 µM)和处理时间延长(48 hr),其表达量又逐步增强,说明AsMT2b可能在胁迫强度增大到一定值时方起作用。 3. 将AsMT2b转入对Cd和As敏感的酵母菌株FD236-6A中,发现AsMT2b对酵母As抗性的提高贡献不大,但可明显提高酵母对Cd的抗性。 4.对AsMT2b的超表达拟南芥的重金属抗性分析表明,与野生型植株比较,转基因植株具有较强的Cd抗性,表现在Cd胁迫下,种子的萌发率较高,根较长,侧根数较多。但在As胁迫下,转基因植株的生长和野生型没有明显差异。可以看出,转AsMT2b的拟南芥对重金属的抗性不同于转AsMT2a的植株,前者的抗Cd性较强,而后者的抗As性较强。 5. Cd胁迫下,AsMT2b过表达拟南芥的Cd含量明显增加,平均比野生型对照植株增加70%,但各个株系的增加幅度不一致。 另外,我们还对CdCl2胁迫下,大蒜幼苗中镉的积累及氧化胁迫和抗氧化能力的变化进行了研究。结果表明在CdCl2 胁迫下,大多数Cd在根部积累,而只有少量的Cd积累于叶片中。5 mM 和10 mM CdCl2 抑制SOD和CAT的活性,但随着处理时间的延长,二者的活性回复到对照水平或高于对照。在CdCl2胁迫下,POD的活性明显增强,同时脂质过氧化产物积累。这些结果说明镉胁迫下,植物细胞中氧化胁迫加剧,而抗氧化酶活性的增强是植物对次生氧化胁迫的一种适应策略。 综上所述,在重金属胁迫下, AsPCS1和AsMT2a之间及AsMT2a和AsMT2b之间均表现出明显的协调反应。这种协调反应可能是植物维持细胞内离子稳态的机制之一。而重金属胁迫下,过表达AsPCS1,AsMT2a或AsPCS1+AsMT2a的拟南芥体内的重金属含量明显增加,表明这些基因可望用于重金属污染土壤的植物修复中。

以探讨添加脂肪酸钙对放牧肉牛(婆罗门和BMY)超数排卵效果及血中雌二醇(E2)和孕酮(P4)水平的影响为目的.将48头供体分为Ⅰ(放牧或对照组)、Ⅱ(放牧+浓缩饲料500 g/d)和Ⅲ(放牧+浓缩饲料500 g/d+脂肪酸钙100 g/d)3组,从同期发情处理开始到冲胚结束试验期32 d,并对供体牛在8个处理时段进行采血,用放射免疫分析法(RIA)分析E2和P4水平.试验结果表明:Ⅰ,Ⅱ,Ⅲ组供体牛超数排卵的排卵率分别为75.4%,74.4%和81.7%(P>0.05),受精率分别为81.4%,55.6%和93.2%(P>0.05);获胚(卵)数分别为7.0枚,6.3枚和7.3枚(P>0.05),可用胚数分别为3.9枚,2.6枚和4.7枚,Ⅲ组显著高于Ⅱ组(P<0.05);E2平均水平分别为0.57 pg/mL,0.47 pg/mL和0.63 pg/mL(P>0.05),P4平均水平分别为3.53 ng/mL,5.03 ng/mL和3.74 ng/mL,Ⅱ组高于Ⅰ,Ⅲ组(P<0.01).表明适量添加脂肪酸钙能改善全放牧肉牛的营养、增强体质,促进排卵与受精,增加超数排卵的可用胚数;能使供体牛的E2和P4水平比对照组有所提高;添加效果冬、春季优于夏、秋季.

The formation of memory is believed to depend on experience- or activity-dependent synaptic plasticity, which is exquisitely sensitive to psychological stress since inescapable stress impairs long-term potentiation (LTP) but facilitates long-term depression (LTD). Our recent studies demonstrated that 4 days of opioid withdrawal enables maximal extents of both hippocampal LTP and drug-reinforced behavior; while elevated-platform stress enables these phenomena at 18 h of opioid withdrawal. Here, we examined the effects of low dose of morphine (0.5 mg kg(-1), i.p.) or the opioid receptor antagonist naloxone (1 mg kg(-1), i.p.) on synaptic efficacy in the hippocampal CA1 region of anesthetized rats. A form of synaptic depression was induced by low dose of morphine or naloxone in rats after 18 h but not 4 days of opioid withdrawal. This synaptic depression was dependent on both N-methyl-D-aspartate receptor and synaptic activity, similar to the hippocampal long-term depression induced by low frequency stimulation. Elevated-platform stress given 2 h before experiment prevented the synaptic depression at 18 h of opioid withdrawal; in contrast, the glucocorticoid receptor (GR) antagonist RU38486 treatment (20 mg kg(-1), s.c., twice per day for first 3 days of withdrawal), or a high dose of morphine reexposure (15 mg kg(-1), s.c., 12 h before experiment), enabled the synaptic depression on 4 days of opioid withdrawal. This temporal shift of synaptic depression by stress or GR blockade supplements our previous findings of potentially correlated temporal shifts of LTP induction and drug-reinforced behavior during opioid withdrawal. Our results therefore support the idea that stress experience during opioid withdrawal may modify hippocampal synaptic plasticity and play important roles in drug-associated memory. Micronutrients play an important role in biological processes for wastewater treatment. Many industrial wastewaters lack in nutrients required for microbial growth, and this is one of the problems at many activated sludge plants treating them. In this study, the effects of the micronutrient niacin on the COD removal rates of textile wastewater, together with the effect of Mixed Liquor Suspended Solids (MLSS) on niacin, were studied. Certain improvement effects were found on the removal rates of COD, when 0.5 similar to 2.0 mg/L niacin was added to the textile wastewater. The optimal concentration of niacin was 1.0 mg/L, which was continuously added during textile wastewater treatment, and removal rates were 1.31 times compared to those of the control system. The concentration of MLSS was probably one of the factors influencing treatment efficiency, and the biological performance of treatment system could be optimized through micronutrient niacin supplements.

The study of inland free-living nematodes is relatively imperfect in China, only seventeen papers were previously published. Since the early researches in 20-30s, few works have been accomplished until 80s. Altogether 171 taxa were formerly recorded, among which, over eighty species have been re-combined. A checklist of the former records with notes on their distribution is presented in this paper. Recently, the function of free-living nematodes has received much attention from Chinese zoologists. Hence, the present authors carried out their studies with emphasis on taxonomy of inland nematodes. During the survey of freshwater lakes, two species are found to be nem to science. Aphanonchus orientalis sp. nov. is characterized by having sclerotized vagina, the presence of 10-11 tubular supplements and 42-62 alveoli supplements in males, but no alveoli in females. Daptonema limnobia sp, nov. is distinguished from other species of the genus in the presence of larger and more anteriorly located amphids, shorter bifurcated spicules, smaller apophysis of gubernaculum, shorter terminal setae, and postvulval uterine sac in females.

Two new species of Actinolaimidae are described from China. Trachactinolaimus brevicaudatus n. sp. is 3.4-4.4 mm long; a = 40-60, b = 3.5-5.2, c = 20-21 in female and 34-39 in male; odontostyle length is 29-33 mu m; spicules are 67-77 mu m long; and the stoma has four onchia with numerous mural denticles. The female has a longitudinal vulva, and the male has 20 to 23 contiguous ventromedian supplements. Egtitus sinensis n. sp. is 1.7-2.2 mm long; a = 24-33, b = 3.1-3.9, c = 16-19 in female and 0.7-0.9 in male; odontostyle length is 25-29 mu m; spicules are 55-56 mu m long; and the prerectum 53-77 pm long. The cardia is short and blunt conoid, 13-19 mu m long. The male has 12 to 13 ventromedian supplements at intervals of 2-3 mu m.

Aristolochic acids (AAs) are the main bioactive ingredients in the most of Aristolochia plants, which are used to make dietary supplements, slimming pills and Traditional Chinese Medicines (TCMs). Excessive ingestion of AAs can lead to serious nephropathy. Therefore, quantitative analysis and quality control for the plants containing AAs is of great importance. In this paper, capillary electrophoresis (CE) with electrochemical detection (ED) at a 33 mu m carbon fiber microdisk electrode (CFE) has been applied to detect AA-I and AA-II in Aristolochia plants. Under the optimum conditions: detection potential at 1.20 V, 2.0 x 10(-2) mol L-1 phosphate buffer solution (PBS) (pH 10.0), injection time 25 s at a height of 17 cm and separation voltage at 12.5 kV, the AA-I and AA-II were baseline separated within 5 min. Low detection limits for AA-I and AA-II were 4.0 x 10(-8) mol L-1 and 1.0 x 10(-7) mol L-1, respectively. Wide linear ranges were from 4.0 x 10(-8) mol L-1 to 1.9 x 10(-5) mol L-1 and 1.0 X 10(-7) mol L-1 to 5.0 x 10(-5) mol L-1 for AA-I and AA-II, respectively. The proposed method has been successfully applied to analyze AAs contents in plant extracts. The results indicated that the contents of AAs in each part of Aristolochia debilis Sieb. Et Zucc.

Lipophilic extracts from 16 species of seaweeds collected along the Qingdao coastline were screened and evaluated for their antioxidant activities (AA) using the beta-carotene-linoleate assay system. The diethyl ether soluble extracts of all selected seaweeds exhibited various degrees of antioxidative efficacy in each screen. The highest antioxidant capacities among the tested samples were observed for Rhodomela confervoides and Symphyocladia latiuscula and were comparable with that of the well-known antioxidant butylated hydroxytoluene and greater than that of propyl gallate. The lipophilic content of all 16 samples and the chemical composition of 4 selected seaweeds, R. confervoides and S. latiuscula, which had higher AA, Laminaria japonica, which had intermediate AA, and Plocamium telfairiae, which had lower AA, were analyzed by gas chromatography and gas chromatography-mass spectrometry, respectively. Fatty acids and alkanes were found. The present data indicated an increase in antioxidative property with increasing content of unsaturated fatty acid. The result of this study suggests that seaweeds can be considered as a potential source for the extraction of lipophilic antioxidants, which might be used as dietary supplements or in production in the food industry. This is the first report on the antioxidant activities of lipophilic extracts from seaweeds.

Currently, antioxidants are added in the human diet to prevent free radical-induced cell damage, and there has been an explosive interest in the use of antioxidant nutritional supplements. The effects of different factors on the antioxidant activity of phycocyanins (PCs) were studied. The results showed that PCs generated hydroxyl radicals in the light, while scavenging them in the dark. When PCs were denatured by sodium dodecyl sulfate, urea and in alkaline condition, their ability to generate hydroxyl radicals disappeared and that of scavenging them greatly increased. This showed that the phycobilin moiety is the main part of PC involved in scavenging hydroxyl radicals. Trypsin hydrolysis of PCs showed that the apoprotein portion of the molecule also made a significant contribution to the antioxidant activity.

Slide-debris flow is debris flow which is transformed from landslide consecutively in a short time, it comprises of two phases: First, Landslide starts to slide; Second, landslide changes to debris flow. Slide-debris flow which brings great property and life loss happens frequently at home and abroad. In order to forecast the happening possibility and scope of slide-debris flow, transfromation mechanism of Slide-debris flow must be studied. Research on transformation mechanism of slide-debris flow is intersectant science of landslide kinetics and debris flow starting theory, It is a fringe problem as well as front problem of geological hazard. This paper takes Qingning slide-debris flow in Da County, Sichuan Province for example and has studied the mechanism of its instability and transfromation into debris flow through indoor test (including usual soil test and ring shear test) and digital modeling method.The research gets the following conclusions. Qingning Landslide took place mainly because of confined water head arising from rainfall infiltration. Before Landslide occurring, it rained continuously for 22 days, accumulated precipitation arrived at 521.6mm.Investigation shows that strata of Qingning Landslide contains quaternary loose accumulation, slip soil and highly weathered bedrock, which is a good condition for formation of confined water in the slope. Further more, groundwater seepage in the slope body and corresponding slope safety factor before landslide occurring have been computed through finite element method. The result shows that because of infiltration of rainfall, confined water head in the slope arose sharply, accordingly, the safety factor of the slope declined quickly. The result also shows that force put on the slide body by the rock mass detached from Dazhaiyan mountain was the direct factor for landslide occurring. Qingning slide-debris transformation mode has been summarized, the process the landslide changed into debris flow is divided into three phases in the prospective of macroscopic geological condition: landslide occurring, transformation and debris flow. Landslide occurring phase is from slope’ local creeping slide to Landslide occurring; transformation phase contains slide body sliding on the slide bed after slide occurring and sliding on the slope after shearing opening; debris flow phase is that slide body breaks up completely and flows downward into the ditches. The transformation mechanism of Qingning slide-debris flow has been studied through indoor ring shear test of slip soil. The result shows that transformation mechanism contains two points: first, during slide body sliding on the slide bed and slope after shearing opening, shearing shrinkage, grain crushing and grain layering brought about declining of its volume and produced excess pore water pressure, and because producing velocity of excess pore water pressure is much greater than its dissipating velocity, shear strength of slide body decreased sharply because of accumulated pore water pressure. Second, grains crushing and grains layering during slide body sliding brought about thick liquefied layer at the bottom of the slidebody, liquefied layer contained high water content and its shear strength was very low, its thickness increased as the sliding displacement increasing. Liquefied layer makes slide body sliding fast and easily break down to debris flow. Excess pore water pressure and liquefied layer made shear strength of slidebody became very low, furthermore, water in the pit of slope joining in the slidebody was also a facter that made slidebody accelerate the transformation. Influence of slide body thickness and fine grains content to transformation of slide-debris flow has been studied through ring shear test. The result reaches two conclusions. First, thickness of slide body affects transformation of slide-debris flow by two ways, porewater pressure and effect of “soft base” increases as thickness of slide body increasing.so the thicker slide body is ,the easier transformation is. Second, actual dissipating velocity of porewater pressure should be considered when studying the influence of fine grains content to tranformation of slide-debris flow. There should be a critical content of fine grains which makes the difference of producing and dissipating velocity of water pore pressre greatest, this value is the best for slide-debris transformation. The whole process of slide-debris flow transformation is reproduced through discrete element method. Transformation mechanism of slide-debris flow is studied through monitoring various parameters including pore water pressure, grain crushing and grain layering in the slide body during the transformation. The result confirms and supplements the transformation mechanism of slide-debris flow got from ring shear test well.
