In this study, we investigated the effects of animal-plant protein ratio in extruded and expanded diets on nutrient digestibility, nitrogen and energy budgets of juvenile soft-shelled turtle (Pelodiscus sinensis). Four extruded and expanded feeds (diets 1-4) were formulated with different animal-plant protein ratios (diet 1, 1.50:1; diet 2, 2.95:1; diet 3, 4.92:1; diet 4, 7.29:1). The apparent digestibility coefficients (ADCs) of dry matter and crude lipid for diet 1 were significantly lower than those for diets 2-4. There was no significant difference in crude protein digestibility among diets 1-4. The ADC of carbohydrate was significantly increased with the increase in animal-plant protein. Although nitrogen intake rate, faecal nitrogen loss rate and excretory nitrogen loss rate of turtles fed diet 1 were significantly higher than those fed diets 2-4, nitrogen retention rate, net protein utilization and biological value of protein in these turtles were significantly lower than those fed diets 2-4. In addition, energy intake rate, excretory energy loss rate and heat production rate of turtles fed diet 1 were also significantly higher than those fed diets 2-4. Faecal energy loss was significantly reduced with the increase in the animal-plant protein ratio. The ADC of energy and assimilation efficiency of energy significantly increased with a higher animal-plant protein ratio. The growth efficiency of energy in the group fed diet 1 was significantly lower than those in the groups fed diets 2-4. Together, our results suggest that the optimum animal-plant protein ratio in extruded and expanded diets is around 3:1.

The Mg-8Gd-2Y-1Nd-0.3Zn-0.6Zr (wt.%) alloy sheet was prepared by hot extrusion technique, and the structure and mechanical properties of the extruded alloy were investigated. The results show that the alloy in different states is mainly composed of alpha-Mg solid solution and secondary phases of Mg5RE and Mg24RE5 (RE = Gd, Y and Nd). At aging temperatures from 200 degrees C to 300 degrees C the alloy exhibits obvious age-hardening response. Great improvement of mechanical properties is observed in the peak-aged state alloy (aged at 200 degrees C for 60 h), the ultimate tensile strength (sigma(b)), tensile yield strength (sigma(0.2)) and elongation (epsilon) are 376 MPa, 270 MPa and 14.2% at room temperature (RT), and 206 MPa. 153 MPa and 25.4% at 300 degrees C, respectively, the alloy exhibits high thermal stability.

Microstructures and mechanical properties of the Mg-8Gd-xZn-0.4Zr (x = 0, 1 and 3 wt.%) alloys in the as-cast, as-extruded and extruded-T5 conditions, have been investigated. The peak-aged Mg-8Gd-1Zn-0.4Zr alloy during isothermal ageing at 423 K acquires highest mechanical properties, with the highest ultimate tensile strength and yield tensile strength of 314 and 217 MPa, respectively. Addition of Zn has obvious effect on age hardening responses, especially for 1 wt.% Zn addition. It is due to a uniform distribution of beta' phase which can impede the movement of dislocations. However, addition of 3 wt.% Zn to the Mg-8Gd-0.4Zr alloy leads to a precipitation of Mg3Zn3Gd2 phase (W-phase). This phase is incoherent with interface of the matrix and becomes cores of the fracture in tensile test at room or elevated temperature.

Microstructures and mechanical properties of the Mg-5Y-4Gd-xZn-0.4Zr alloys have been investigated. These results show that the Mg-5Y-4Gd-0.5Zn-0.4Zr alloy in the peak-aged condition exhibits the highest tensile strength, and the values of the ultimate tensile strength and yield tensile strength are 370 and 300 MPa, respectively. It is suggested that addition of 0.5% Zn has a great effect on age hardening response. The long periodic stacking structure has been found in these Zn-containing alloys, and the volume fraction of this phase increases with increasing Zn addition. This phase plays an important role in improvement of the mechanical properties, especially for the elongations. The beta' phase precipitates during the ageing process are responsible for the improvement of the mechanical properties of the alloys in the peak-aged condition.

Microstructures and mechanical properties of the Mg-7Y-4Gd-xZn-0.4Zr (x = 0.5, 1.5, 3, and 5 wt.%) alloys in the as-cast, as-extruded, and peak-aged conditions have been investigated by using optical microscopy, scanning electron microscope, X-ray diffraction, and transmission electron microscopy. It is found that the peak-aged Mg-7Y-4Gd-1.5Zn-0.4Zr alloys have the highest strength after aging at 220 A degrees C. The highest ultimate tensile strength and yield tensile strength are 418 and 320 MPa, respectively. The addition of 1.5 wt.% Zn to the based alloys results in a greater aging effect and better mechanical properties at both room and elevated temperatures. The improved mechanical properties are mainly ascribed to both a fine beta' phase and a long periodic stacking-ordered structure, which coexist together in the peak-aged alloys.

Microcoleus vaginatus isolated from a desert algal crust of Shapotou was cultured in BG-11 medium containing 0.2mol l(-1) NaCl or 0.2mol l(-1) NaCl plus 100mg l(-1) sucrose, extracellular polymeric substances (EPS) or hot water-soluble polysaccharides (HWP), respectively. Photosynthetic oxygen evolution rates, photosystem 11 activity (Fv/Fm) and dark respiration of NaCl-stressed cells were enhanced significantly by the added sucrose or EPS under salt stress conditions (0.2mol l(-1) NaCl). Compared with cells treated with salt alone, sodium contents in cells reduced significantly; the content of cellular total carbohydrate did not change, and intracellular sucrose, water-soluble sugar increased significantly following the addition of exogenous carbohydrates. Sucrose synthase (SS) activity of NaCl-stressed cells increased following the addition of sucrose, and sucrose phosphate synthase (SPS) activity of NaCl-stressed cells increased following the addition of exogenous sucrose, EPS or HWP compared with cells stressed with NaCl only. The results suggested that the extruded EPS might be re-absorbed by cells of M. vaginatus as carbon source, they could increase salt tolerance of M. vaginatus through the changes of carbohydrate metabolism and the selective uptake of sodium ions. The ageing behavior of an extruded Mg-7Y-4Gd-0.5Zn-0.4Zr alloy during ageing at 250 degrees C has been investigated. Two types of phases have been observed during the ageing process. One is a lamellar phase with a 14H long periodic stacking structure, the other is the beta' phase with an ellipsoidal morphology. The increased mechanical properties of the peak-aged alloy are mainly ascribed to the presence of both of these phases at peak hardness.

Blends of a liquid crystalline thermotropic copolyester (LCP70) and an amorphous phenolphthalein based poly(ether-ketone)(PEK-C) with two viscosities were prepared by melt blending. The blends' morphology, rheological and mechanical properties were investigated by DSC, SEM, mechanical and rheological tests. It was observed that the optimum composition of the PEK-C/LCP70 blend was 10 wt% LCP for both mechanical and rheological properties. When the LCP content was less than 10%, the LCP phase existed as finely dispersed fibrous domains with a diameter of about 1 mu m in the matrix, and both tensile and flexural properties were improved. In contrast, when the LCP content reached 20% or more, the LCP domains coalesced to ellipsoidal particles with a diameter of about 5 mu m, and the mechanical properties decreased as a result. It is demonstrated that pure PEK-C with a high viscosity which was difficult to process by melt extrusion, could be extruded conveniently when 10% LCP70 was incorporated. It is emphasized that LCP not only can be used as a reinforcing phase but also an effective processing agent for engineering thermoplastics, especially for those with high viscosity and narrow processing window. A new class of liquid crystalline poly(ester-imide)s was synthesized by melt polycondensation. The basic physical properties of the resulting polymers were investigated by differential scanning calorimetry (d.s.c.), wide-angle X-ray diffraction (WAXD), polarized light microscopy, scanning electron microscopy (SEM), thermogravimetric analysis (t.g.a.), and rheological and mechanical testing. All of these poly(ester-imide)s were amorphous, as reflected by the results obtained from the WAXD and d.s.c. studies. Characterization and comparison of these poly(ester-imide)s with the corresponding polyesters suggested that the introduction of imide groups into the polyester chain is favourable for the formation of liquid crystalline phases. These results, together with the rheological studies, suggested that there existed a form of strong inter- or intramolecular electron donor-acceptor interaction which played a significant role in the liquid crystalline properties of the poly(ester-imide)s. The polymer products thus obtained exhibited good mechanical properties, with flexural strengths and moduli as high as 174 MPa and 6.9 GPa, respectively. The morphology of the fracture surfaces of extruded rod samples showed a sheet-like structure which consisted of ribbons and fibres oriented along the flow direction. The glass transition temperatures and thermal stabilities of the polymers were improved by the incorporation of imide groups. Copyright (C) 1996 Elsevier Science Ltd.

Polycarbonate (PC) and poly(acrylonitrile-butadiene-styrene) (ABS) was co-extruded at different weight ratios by a single screw extruder. In order to obtain a finer blend, two times extrusion was carried out. In this case, a ''network'' structure with two-continuous phases was observed for the blends with two compositions of PC/ABS, being 80/20 and 70/30. It is found that the blends with these two compositions just have maximum values on the curves of notched Izod impact strength, flexural modulus and flexural strength vs. composition, respectively. This was never observed in previous publications.

A group of coenocytic marine algae differs from higher plants, whose totipotency depends on an intact cell (or protoplast). Instead, this alga is able to aggregate its extruded protoplasm in sea water and generate new mature individuals. It is thought that lectins play a key role in the aggregation process. We purified a lectin associated with the aggregation of cell organelles in Bryopsis hypnoides. The lectin was ca. 27 kDa with a pI between pH 5 and pH 6. The absence of carbohydrate suggested that the lectin was not a glycoprotein. The hemagglutinating activity (HA) of the lectin was not dependent on the. presence of divalent cations and was inhibited by N-Acetylgalactosamine, N-Acetylglucosamine, and the glycoprotein bovine submaxillary mucin. The lectin preferentially agglutinated Gram-negative bacterium. The HA of this lectin was stable between pH 4 to pH 10. Cell organelles outside the cytoplasm were agglutinated by the addition of lectin solution (0.5 mg ml(-1)). Our results suggest that the regeneration of B. hypnoides is mediated by this lectin. We also demonstrated that the formation of cell organelle aggregates was inhibited by nigericin in natural seawater (pH 8.0). Given that nigericin dissipates proton gradients across the membrane, we hypothesize that the aggregation of cell organelles was proton-gradient dependent.

Tissue culture, SDS-polyacrylamide gel electrophoresis (SDS-PAGE) and spectra analysis were used for studying the aggregation mechanism of protoplasts from Bryopsis hypnoides Lamouroux and the discrepancy between the protoplast-regenerated plants and the wild type. The aggregation of protoplasts from B. hypnoides was observed in natural seawater and artificial seawater with different pH values, and the location and mechanism of the materials causing the aggregation were also studied. Results showed that the protoplasts could aggregate into some viable spheres in natural seawater and subsequently grow into mature individuals. Aggregation of the protoplasts depended exclusively upon the pH value (6-11), and the protoplasts aggregated best at pH 8-9. Some of the extruded protoplasts were separated into two parts by centrifugation: the pellet (PO) and the supernatant (PL). The PO could aggregate in artificial seawater (pH 8.3) but not in PL. No aggregation was found in PO cultured in natural seawater containing nigericin, which can dissipate the proton gradients across the membrane. These experiments suggest that the aggregation of protoplasts is proton-gradient dependent and the materials causing the aggregation were not in the vacuolar sap, but located on the surface or inside the organelles. Furthermore, the transfer of the materials across the membrane was similar to Delta pH-based translocation (Delta pH/TAT) pathway that occurs in the chloroplasts of higher plants and bacteria. Obvious discrepancies in both the total soluble proteins and the ratio of chlorophyll a to chlorophyll b between the regenerated B. hypnoides and the wild type were found, which may be related to the exchange of genetic material during aggregation of the organelles. In the process of development, diatom Amphora coffeaeformis Agardh attached to the protoplast aggregations, retarding their further development, and once they were removed, the aggregations immediately germinated, which showed that diatoms can affect the development of other algae.

The extruded protoplasm from the coenocytic green alga, Bryopsis hypnoides Lamouroux, was able to reform a cell wall and develop further into a mature alga in seawater. In this paper, the influence of albumen on the ability of aggregation and on the photosynthesis of protoplasm was examined. Results show that the protoplasm of B. hypnoides could aggregate in either albumen or chicken egg, which is similar to that in seawater. However unlike in seawater, the aggregation from B. hypnoides in albumen and chicken egg failed to develop into a mature individual. Interestingly, the protoplasm of B. hypnoides could maintain its photosynthetic O-2 evolution in albumen and chicken egg, while the time in chicken egg was longer than that in albumen.

The cell organelles of the coenocytic alga Codium fragile (Sur.) Hariot aggregated rapidly and protoplasts were formed when its protoplasm was extruded out in seawater. Continuous observation showed that there were long and gelatinous threads connecting the cell organelles. The threads contracted, and thus the cell organelles aggregated into protoplasmic masses. The enzyme digestion experiments and Coomassie Brilliant Blue and Anthrone stainings showed that the long and gelatinous threads involved in the formation of the protoplasts might include protein and saccharides as structure components. Nile Red staining indicated that the protoplast primary envelope was non-lipid at first, and then lipid materials integrated into its surface gradually. The fluorescent brightener staining indicated that the cell wall did not regenerate in the newly formed protoplasts and they all disintegrated within 72 h after formation. Transmission electron microscopy of the cell wall of wild C. fragile showed electron-dense material embedded in the whole cell wall at regular intervals. The experiments indicated that C. fragile would be a suitable model alga for studying the formation of protoplasts.

In recent years, bivalve feces and powdered algae have been used as the food sources of holothurians in China. In this study, growth and energy budget for sea cucumber Apostichopus japonicus (Selenka) with initial wet body,veights of 32.5 1.0 g (mean +/- SE, n=45) when fed with five different granule diets containing dried bivalve feces and/or powdered algae in water temperature 13.2-19.8 degrees C and salinity 30-32ppt were quantified in order to investigate how diets influence growth and energy distribution and to find out the proper diet for land-based intensive culture of this species. Results showed that diets affected the food ingestion, feces production, food conversion efficiency and apparent digestive ratios, hence the growth and energy budget. Sea cucumbers fed with dried feces of bivalve showed poorer energy absorption, assimilation and growth than individuals fed with other four diets; this could be because feces-drying process removed much of the benefits. Dried bivalve feces alone, therefore, were not a suitable diet for sea cucumbers in intensive cultivation. The mixed diets of feces and powered algae showed promising results for cultivation of sub-adult Apostichopus japonicus, while animals fed with powdered algae alone, could not obtain the best growth. According to SGR of tested animals, a formula of 75% feces and 25% powdered algae is the best diet for culture of this species. Extruded diets were used in the present experiment to overcome shortcomings of the traditional powdered feeds, however, it seems a conflict exists between drying bivalve feces to form extruded diets and feeding sea cucumbers with fresh feces which contain beneficial bacteria. Compared with other echinoderms, in holothurians the energy deposited in growth is lower and the energy loss in feces accounts for the majority of the ingested energy.
