To collect information about the genetic diversity of the plankton community and to study how plankton respond to environmental conditions, plankton samples were collected from five stations representing different trophic levels in a shallow, eutrophic lake (Lake Donghu), and investigated by PCR-DGGE fingerprinting. A total of 100 bands (61 of 16S rDNA bands and 39 of 18S rDNA bands) were detected. The DGGE bands unique to any single station accounted for 38% of the total bands, whereas common bands detected at all five stations accounted for only 11%. Using UPGMA clustering and MDS ordination of DGGE fingerprints, stations I and II were found to initially group together into one cluster, which was later joined by station V. Stations III and IV were isolated into two separate groups of one station each. Some differences in grouping relationships were found when analysis was completed on the basis of chemical characteristics and morphological composition, with zooplankton composition showing the greatest variability. However, the most similar stations (I and II) were always initially grouped into one cluster. Moreover, stations that exhibited the same or similar trophic level (stations III and IV), but different concentrations of heavy metals, were further differentiated by the DGGE method. Results of the present study indicated that PCR-DGGE fingerprinting was more sensitive than the traditional methods, as other studies suggested. Additionally, PCR-DGGE appears to be more appropriate for diversity characterization of the plankton community, as it is more canonical, systematic, and effective. Most importantly, fingerprinting results are more convenient for the comparative analyses between different studies. Therefore, the use of the described fingerprinting analysis may provide an operable and sensitive biomonitoring approach to identify critical, and potentially negative, stress within an aquatic ecosystem.

Tank-reared Japanese flounder larvae, Paralichthys olivaceus, had a major feeding peak in the morning and a secondary peak in the afternoon throughout the larval development, with light being the primary factor regulating their feeding activity. The larvae consumed rotifers in preference to Artemia for up to 10 days, after which the food preference shifted to Artemia. Feeding rates of the larvae prior to 10 days post-batch depended on prey density, but in the old larvae, feeding rates were independent of prey density. Maximum feeding rate occurred at 19 degrees C. The occurrence of the attack posture, after its onset at first feeding (2 days post-hatch), increased up to 25 days, began to decrease when the larvae prepared to settle down, then disappeared after settlement. The occurrence frequency of the attack posture was positively related to fish density, but inversely related to starvation duration, and occurred most frequently at 19 degrees C. This posture depended on prey density in larvae prior to 10 days post-hatch, but became independent of prey density as the larvae developed. It was obvious that, for flounder larvae, attack posture was a behavioural character closely related to feeding and subject to larval development and environmental factors. In this paper, a method is developed for determining the effective stiffness of the cracked component. The stiffness matrix of the cracked component is integrated into the global stiffness matrix of the finite element model of the global platform for the FE calculation of the structure in any environmental conditions. The stiffness matrix equation of the cracked component is derived by use of the finite variation principle and fracture mechanics. The equivalent parameters defining the element that simulates the cracked component are mathematically presented, and can be easily used for the FE calculation of large scale cracked structures together with any finite element program. The theories developed are validated by both lab tests and numerical calculations, and applied to the evaluation of crack effect on the strength of a fixed platform and a self-elevating drilling rig.

Stiction in microelectromechanical systems (MEMS) has been a major failure mode ever since the advent of surface micromachining in the 80s of the last century due to large surface-area-to-volume ratio. Even now when solutions to this problem are emerging, such as self-assembled monolayer (SAM) and other measures, stiction remains one of the most catastrophic failure modes in MEMS. A review is presented in this paper on stiction and anti-stiction in MEMS and nanoelectromechanical systems (NEMS). First, some new experimental observations of stiction in radio frequency (RF) MEMS switch and micromachined accelerometers are presented. Second, some criteria for stiction of microstructures in MEMS and NEMS due to surface forces (such as capillary, electrostatic, van der Waals, Casimir forces, etc.) are reviewed. The influence of surface roughness and environmental conditions (relative humidity and temperature) on stiction are also discussed. As hydrophobic films, the self-assembled monolayers (SAMs) turn out able to prevent release-related stiction effectively. The anti-stiction of SAMs in MEMS is reviewed in the last part.

By sample specificity it is meant that specimens with the same nominal material parameters and tested under the same environmental conditions may exhibit different behavior with diversified strength. Such an effect has been widely observed in the testing of material failure and is usually attributed to the heterogeneity of material at the mesoscopic level. The degree with which mesoscopic heterogeneity affects macroscopic failure is still not clear. Recently, the problem has been examined by making use of statistical ensemble evolution of dynamical system and the mesoscopic stress re-distribution model (SRD). Sample specificity was observed for non-global mean stress field models, such as the duster mean field model, stress concentration at tip of microdamage, etc. Certain heterogeneity of microdamage could be sensitive to particular SRD leading to domino type of coalescence. Such an effect could start from the microdamage heterogeneity and then be magnified to other scale levels. This trans-scale sensitivity is the origin of sample specificity. This paper presents the Hill instability analysis of Tension Leg Platform (TLP) tether it, deep sea. The 2-D nonlinear beam model which is Undergoing Coupled axial and transverse vibrations, is applied. The governing equations are reduced to nonlinear Hill equation by use of the Galerkin's method and the modes superposition principle. The Hill instability charted Lip to large parameters is obtained. An important parameter M is defined and can he expressed as the functions of tether length, the platform surge and heave motion amplitudes. Some example studies are performed for various environmental conditions. The results demonstrate that the nonlinear coupling between the axial and transverse vibrations has a significant effect on the response of structure.. It needs to be considered for the accurate dynamic analysis of long TLP tether subjected to the combined platform surge and heave motions.

凋落物的分解对生物地球化学循环起着重要的作用。本研究以位于中国北方农牧交错区(内蒙古多伦县)的半干旱克氏针茅(Stipa krylovii Roshev.)草原生态系统为背景,针对凋落物分解研究中的几个关键过程和问题进行了探讨。研究内容包括该生态系统中几种常见植物凋落物的分解速率,在分解过程中的养分动态,凋落物的混合效应(即非加性效应)及其机制,全球变化引起的土壤有效N、P和降水增加对凋落物分解的影响及其对草原生态系统碳储量的影响等。 通过对克氏针茅、糙隐子草(Cleistogenes squarrosa Trin.)、双齿葱(Allium bidentatum Fisch.)、野韭(Allium ramosum L. Sp. Pl.)和冰草(Agropyron cristatum Gaertn.)的叶、茎、根凋落物的研究发现,叶和茎的分解速率与凋落物初始N和P含量呈显著正相关,根系的分解速率与初始C/N比呈显著负相关。根系凋落物一般比叶和茎凋落物分解速率快,而且具有更快的养分周转速率。有些凋落物混合在一起分解后对分解速率产生了混合效应(混合凋落物的实际分解速率偏离于基于组分凋落物计算的预期分解速率),同时它们对养分(主要是N、P、Ca、Mg)的固定或释放被延后。混合效应发生与否以及混合效应的方向主要取决于组分凋落物的特点,而与凋落物多样性的高低没有明显的关系。混合凋落物对养分动态的负作用,能降低退化生态系统遭到干扰后养分的损失,而且养分释放时间的延后有助于某些植物更好地生长,有利于退化草原生态系统恢复过程中植被结构的改善。 由于凋落物的质量对分解速率的影响重大,凋落物的化学组成很可能对混合凋落物分解过程中的非加性效应起重要作用,然而迄今为止没有明确的结论。本文研究发现,具有不同初始N、P含量的凋落物(同一物种)混合在一起分解后产生了正的非加性效应,非加性效应的强弱与组分凋落物初始N、P含量的差异大小有关。而且,非加性效应与P含量差异大小的相关性比与N的强,这可能与研究地点土壤中P的含量相对更为缺乏有关。这一研究结果表明,凋落物的化学组成在有些情况下确实与非加性效应有关。 通过施肥和浇水试验,研究了全球变化引起的土壤有效N、P和水分的增加对具有不同生活型和化学组成的两种优势草原植物,即双齿葱和克氏针茅凋落物分解的直接影响。结果发现,向土壤中添加N肥或N、P复合肥,在短期内(100天)加速了这两种植物的分解速率。高质量凋落物(双齿葱)的分解更容易受到土壤水分条件的限制,而低质量凋落物(克氏针茅)的分解对土壤养分有效性更敏感。土壤中有效养分的增加会提高分解凋落物对养分的固定,有利于退化生态系统中养分的保持。双齿葱对内蒙古半干旱典型草原的碳循环和养分动态的贡献,比克氏针茅要更大。研究结果提示人们,对草原生态系统的碳循环和养分动态进行模拟时,需要对不同化学组成的凋落物分别加以考虑。 本文还探讨了土壤有效N长期增加后对两种优势草原植物(克氏针茅和冷蒿(Artemisia frigida Willd))凋落物分解的综合影响,并进一步区分了由土壤有效N增加引起的凋落物质量改变的间接作用和土壤状况改变的直接作用。结果发现,土壤有效N长期增加后对凋落物的分解速率没有明显影响,但是显著加速了分解凋落物对养分的固定,这可能是由于凋落物质量对分解的促进作用和土壤状况对分解的抑制作用相互抵消的原因所造成的。向土壤中添加N素后,地上部植被生产力不可避免的会提高,尽管短期内会加速凋落物的分解速率,但从长远来看,对凋落物的分解速率不会产生影响,所以估计土壤有效N增加后最终会提高草原生态系统的净碳储量。 通过本研究,对中国北方农牧交错区(内蒙古多伦县)半干旱克氏针茅草原生态系统的养分循环过程有了初步的了解,验证了混合凋落物分解过程中非加性效应发生的可能性,并且证明组分凋落物的初始化学组成对这种非加性效应确实起着重要作用。本研究说明全球变化对不同植物种凋落物分解的影响是不同的,而且因研究时间长短的不同对凋落物分解的影响会发生变化。研究结果为草原生态系统碳和养分循环过程的模拟提供了必要的基础资料,为混合凋落物分解过程中非加性效应发生的可能机制提供了强有力的证据,对在全球变化背景下人们对草原生态系统碳和养分循环过程的认识和模拟有着重要的参考价值。

Enzymatic activities and fatty acid methyl esters (FAMEs) in the sediments of two eutrophic lakes in Wuhan city were investigated. The results showed phosphatase and dehydrogenase activities in the lotus zone and plant floating bed zone were significantly lower than those in other sites, and urease activity was the highest where microorganism agents were put in. Fatty acid group compositions indicated the predominance of aerobic bacteria in the surface sediments in shallow lakes. The ratios of FAMEs specific for bacteria and Gram-positive bacteria exibited significant differences between the two lakes. The results of trans to cis indicated that the microorganisms in Lake Yuehu could adapt themselves to environmental stress better. The enzymatic activities and FAMEs showed differences in different sites, indicating that ecological restoration measures and environmental conditions could affect lake sediment to some extent. But the monitoring, work would be done in series to exactly evaluate the effect of the remediation measures.

P>A sampling system for capturing sturgeon eggs using a D-shaped bottom anchored drift net was used to capture early life stages (ELS) of Chinese sturgeon, Acipenser sinensis, and monitor annual spawning success at Yichang on the Yangtze River, 1996-2004, before and just after the Three Gorges Dam began operation. Captured were 96 875 ELS (early life stages: eggs, yolk-sac larvae = eleuthero embryos, and larvae); most were eggs and only 2477 were yolk-sac larvae. Most ELS were captured in the main river channel and inside the bend at the Yichang spawning reach. Yolk-sac larvae were captured for a maximum of 3 days after hatching began, indicating quick dispersal downstream. The back-calculated day of egg fertilization over the eight years indicated a maximum spawning window of 23 days (20 October-10 November). Spawning in all years was restricted temporally, occurred mostly at night and during one or two spawning periods, each lasting several days. The brief temporal spawning window may reduce egg predation by opportunistic predators by flooding the river bottom with millions of eggs. During 1996-2002, the percentage of fertilized eggs in an annual 20-egg sample was between 63.5 to 94.1%; however, in 2003 the percentage fertilized was only 23.8%. This sudden decline may be related to the altered environmental conditions at Yichang caused by operation of the Three Gorges Dam. Further studies are needed to monitor spawning and changes in egg fertilization in this threatened population.

Many unicellular green algae can become yellow or red in various natural habitats due to mass accumulation of a secondary carotenoid, such as lutein, or astaxanthin. The accumulation of secondary carotenoids is generally thought to be a survival strategy of the algae under photo-oxidative stress or other adverse environmental conditions. The physiological role of the carotenoids in stress response is less well understood at the subcellular or molecular level. In this study, a stable astaxanthin overproduction mutant (MT 2877) was isolated by chemical mutagenesis of a wild type (WT) of the green microalga Haematococcus pluvialis Flotow NIES-144. MT 2877 was identical to the WT with respect to morphology, pigment composition, and growth kinetics during the early vegetative stage of the life cycle. However, it had the ability to synthesize and accumulate about twice the astaxanthin content of the WT under high light, or under high light in the presence of excess amounts of ferrous sulphate and sodium acetate. Under stress, the mutant exhibited higher photosynthetic activities than the WT, based on considerably higher chlorophyll fluorescence induction, chlorophyll autofluorescence intensities, and oxygen evolution rates. Cell mortality caused by stress was reduced by half in the mutant culture compared with the WT. Enhanced protection of the mutant against stress is attributed to its accelerated carotenogenesis and accumulation of astaxanthin. Our results suggest that MT 2877, or other astaxanthin overproduction Haematococcus mutants, may offer dual benefits, as compared with the wild type, by increasing cellular astaxanthin content while reducing cell mortality during stress-induced carotenogenesis.

The genus Sarcocheilichthys is a group of small cyprinid fishes comprising 10 species/sub-species widely distributed in East Asia, which represents a valuable model for understanding the speciation of freshwater fishes in East Asia. In the present study, the molecular phylogenetic relationship of the genus Sarcocheilichthys was investigated using a 1140 bp section of the mitochondrial cytochrome b gene. Two different tree-building methods, maximum parsimony (MP) and Bayesian methods, yielded trees with almost the same topology, yielding high bootstrap values or posterior probabilities. The results showed that the genus Sarcocheilichthys consists of two large clades, clades I and II. Clade I contains Sarcocheilichthys lacustris, Sarcocheilichthys sinensis and Sarcocheilichthys parvus, with S. parvus at a basal position. In clade II, Sarcocheilichthys variegatus microoculus is at a basal position; samples of the widespread species, Sarcocheilichthys nigripinnis, form a large subclade containing another valid species Sarcocheilichthys czerskii. Sarcocheilichthys kiangsiensis is retained at an intermediate position. Since S. czerskii is a valid species in the S. nigripinnis clade, remaining samples of S. nigripinnis form a paraphyly. This speciation process is attributed to geographical isolation and special environmental conditions experienced by S. czerskii and stable environments experienced by the other S. nigripinnis populations. This type of speciation process was suggested to be very common. Samples of Sarcocheilichthys sinensis sinensis and Sarcocheilichthys sinensis fukiensis that did not form their own monophyletic groups suggest an early stage of speciation and support their sub-species status. Molecular clock analysis indicates that the two major lineages of the genus Sarcocheilichthys, clades I and II diverged c. 8.89 million years ago (mya). Sarcocheilichthys v. microoculus from Japan probably diverged 4.78 mya from the Chinese group. The northern-southern clades of S. nigripinnis began to diverge c. 2.12 mya, while one lineage of S. nigripinnis evolved into a new species, S. czerski, c. 0.34 mya. The 16S and 18S rRNA genes of planktonic organisms derived from five stations with nutrient gradients in Lake Donghu, China, were studied by PCR-denaturing gradient gel electrophoresis (DGGE) fingerprinting, and the relationships between the genetic diversity of the plankton community and biotic/abiotic factors are discussed. The concentrations of total nitrogen (TN), total phosphorus (TP), NH4-N and As were found to be significantly related (P < 0.05) to morphological composition of the plankton community. Both chemical and morphological analyses suggested that temporal heterogeneity was comparatively higher than spatial heterogeneity in Lake Donghu. Although the morphological composition was not identical to the DGGE fingerprints in characterizing habitat similarity, the two strongest eutrophic stations (I and II) were always initially grouped into one cluster. Canonical correspondence analysis suggested that the factors strongly correlated with the first two ordination axes were seasonally different. The concentrations of TN and TP and the densities of rotifers and crustaceans were generally the main factors related to the DGGE patterns of the plankton communities. The study suggested that genetic diversity as depicted by metagenomic techniques (such as PCR-DGGE fingerprinting) is a promising tool for ecological study of plankton communities and that such techniques are likely to play an increasingly important role in assessing the environmental conditions of aquatic habitats.

The aim of the present study was to purify the common native carp growth hormone (ncGH), produce monoclonal antibodies (mAbs) to common native carp growth hormone (ncGH), and further enhance the sensitivity of enzyme-linked immunosorbent assays (ELISA) for ncGH. Additionally, we investigated changes in serum ncGH levels in carps raised in different environmental conditions. The recombinant grass carp (Ctenopharyngodon idella) growth hormone was purified and used as antigen to immunize the rabbit. The natural ncGH was isolated from the pituitaries of common carp. SDS-PAGE and Western blot utilizing the polyclonal anti-rgcGH antibody confirmed the purification of ncGH from pituitaries. Purified ncGH was then used as an immunogen in the B lymphocyte hybridoma technique. A total of 14 hybridoma cell lines (FMU-cGH 1-14) were established that were able to stably secrete mAbs against ncGH. Among them, eight clones (FMU-cGH1-6, 12 and 13) were successfully used for Western blot while nine clones (FMU-cGH 1-7, 9 and 10) were used in fluorescent staining and immunohistochemistry. Epitope mapping by competitive ELISA demonstrated that these mAbs recognized five different epitopes. A sensitive sandwich ELISA for detection of ncGH was developed using FMU-cGH12 as the coating mAb and FMU-cGH6 as the enzyme labeled mAb. This detection system was found to be highly stable and sensitive, with detection levels of 70 pg/mL. Additionally, we found that serum ncGH levels in restricted food group and in the net cage group increased 6.9-and 5.8-fold, respectively, when compared to controls, demonstrating differences in the GH stress response in common carp under different living conditions.
