The aim of the present study was to purify the common native carp growth hormone (ncGH), produce monoclonal antibodies (mAbs) to common native carp growth hormone (ncGH), and further enhance the sensitivity of enzyme-linked immunosorbent assays (ELISA) for ncGH. Additionally, we investigated changes in serum ncGH levels in carps raised in different environmental conditions. The recombinant grass carp (Ctenopharyngodon idella) growth hormone was purified and used as antigen to immunize the rabbit. The natural ncGH was isolated from the pituitaries of common carp. SDS-PAGE and Western blot utilizing the polyclonal anti-rgcGH antibody confirmed the purification of ncGH from pituitaries. Purified ncGH was then used as an immunogen in the B lymphocyte hybridoma technique. A total of 14 hybridoma cell lines (FMU-cGH 1-14) were established that were able to stably secrete mAbs against ncGH. Among them, eight clones (FMU-cGH1-6, 12 and 13) were successfully used for Western blot while nine clones (FMU-cGH 1-7, 9 and 10) were used in fluorescent staining and immunohistochemistry. Epitope mapping by competitive ELISA demonstrated that these mAbs recognized five different epitopes. A sensitive sandwich ELISA for detection of ncGH was developed using FMU-cGH12 as the coating mAb and FMU-cGH6 as the enzyme labeled mAb. This detection system was found to be highly stable and sensitive, with detection levels of 70 pg/mL. Additionally, we found that serum ncGH levels in restricted food group and in the net cage group increased 6.9-and 5.8-fold, respectively, when compared to controls, demonstrating differences in the GH stress response in common carp under different living conditions.

We present an analysis of extensive nutrient data sets from two river-dominated coastal ecosystems, the northern Adriatic Sea and the northern Gulf of Mexico, demonstrating significant changes in surface nutrient ratios over a period of 30 years. The silicon:nitrogen ratios have decreased, indicating increased potential for silicon limitation. The nitrogen:phosphorus and the silicon:phosphorus ratios have also changed substantially, and the coastal nutrient structures have become more balanced and potentially less limiting for phytoplankton growth. It is likely that net phytoplankton productivity increased under these conditions and was accompanied by increasing bottom water hypoxia and major changes in community species composition. These findings support the hypothesis that increasing coastal eutrophication to date may be associated with stoichiometric nutrient balance, due to increasing potential for silicon limitation and decreasing potential for nitrogen and phosphorus limitation. On a worldwide basis, coastal ecosystems adjacent to rivers influenced by anthropogenic nutrient loads may experience similar alterations.

Concentrations and ratios of nutrients in Jiaozhou Bay, China, have changed much in the past decades, with trends indicating an increase in nitrogen and a decrease in silicate. Statistical analysis has shown that the long-term variations of nutrients are associated with agricultural activities, precipitation, and anthropogenic factors. Stoichiometric calculations indicate that the nutrient structure has become more and more unbalanced. There has been almost no possibility for nitrogen limitation since the 1980s, the probability of P limitation has increased, and the probability of Si limitation has also increased markedly from the 1980s to the 1990s. As a consequence of changes in nutrient structure, a decrease in the abundance of net phytoplankton was evident, whereas total chlorophyll a levels have remained roughly unchanged at around 3.55 mu g/L. Thus, it is likely that smaller species have taken the niche vacated by the larger species. Changes in phytoplankton size and species composition may ultimately lead to various functional and structural changes at the system level.

Three years of eddy covariance measurements were used to characterize the seasonal and interannual variability of the CO2 fluxes above an alpine meadow (3250 m a.s.l.) on the Qinghai-Tibetan Plateau, China. This alpine meadow was a weak sink for atmospheric CO2, with a net ecosystem production (NEP) of 78.5, 91.7, and 192.5 g C m(-2) yr(-1) in 2002, 2003, and 2004, respectively. The prominent, high NEP in 2004 resulted from the combination of high gross primary production (GPP) and low ecosystem respiration (R-e) during the growing season. The period of net absorption of CO2 in 2004, 179 days, was 10 days longer than that in 2002 and 5 days longer than that in 2003. Moreover, the date on which the mean air temperature first exceeded 5.0 degrees C was 10 days earlier in 2004 (DOY110) than in 2002 or 2003. This date agrees well with that on which the green aboveground biomass (Green AGB) started to increase. The relationship between light-use efficiency and Green AGB was similar among the three years. In 2002, however, earlier senescence possibly caused low autumn GPP, and thus the annual NEP, to be lower. The low summertime R-e in 2004 was apparently caused by lower soil temperatures and the relatively lower temperature dependence of R-e in comparison with the other years. These results suggest that (1) the Qinghai-Tibetan Plateau plays a potentially significant role in global carbon sequestration, because alpine meadow covers about one-third of this vast plateau, and (2) the annual NEP in the alpine meadow was comprehensively controlled by the temperature environment, including its effect on biomass growth.
