Microcoleus vaginatus Gom., the dominant species in biological soil crusts (BSCs) in desert regions, plays a significant role in maintaining the BSC structure and function. The BSC quality is commonly assessed by the chlorophyll a content, thickness, and compressive strength. Here, we have studied the effect of different proportions of M. vaginatus, collected from the Gurbantunggut Desert in northwestern China, on the BSC structure and function under laboratory conditions. We found that when M. vaginatus was absent in the BSC, the BSC coverage, quantified by the percentage of BSC area to total land surface area, was low with a chlorophyll a content of 4.77 x 10(-2) mg g(-1) dry soil, a thickness of 0.86 mm, and a compressive strength of 12.21 Pa. By increasing the percentage of M. vaginatus in the BSC, the BSC coverage, chlorophyll a content, crust thickness, and compressive strength all significantly increased (P < 0.01). The maximum chlorophyll a content (13.12 mg g(-1)dry soil), the highest crust thickness, and the compressive strength (1.48 mm and 36.60 Pa, respectively) occurred when the percentage of inoculated M. vaginatus reached 80% with a complex network of filaments under scanning electron microscope. The BSC quality indicated by the above variables, however, declined when the BSC was composed of pure M. vaginatus (monoculture). In addition, we found that secretion of filaments and polymer, which stick sands together in the BSC, increased remarkably with the increase of the dominant species until the percentage of M. vaginatus reached 80%. Our results suggest that not only the dominant species but also the accompanying taxa are critical for maintaining the structure and functions of the BSC and thus the stability of the BSC ecosystems.

Background: Serine/threonine kinases (STKs) have been found in an increasing number of prokaryotes, showing important roles in signal transduction that supplement the well known role of two-component system. Cyanobacteria are photoautotrophic prokaryotes able to grow in a wide range of ecological environments, and their signal transduction systems are important in adaptation to the environment. Sequence information from several cyanobacterial genomes offers a unique opportunity to conduct a comprehensive comparative analysis of this kinase family. In this study, we extracted information regarding Ser/Thr kinases from 21 species of sequenced cyanobacteria and investigated their diversity, conservation, domain structure, and evolution. Results: 286 putative STK homologues were identified. STKs are absent in four Prochlorococcus strains and one marine Synechococcus strain and abundant in filamentous nitrogen-fixing cyanobacteria. Motifs and invariant amino acids typical in eukaryotic STKs were conserved well in these proteins, and six more cyanobacteria- or bacteria-specific conserved residues were found. These STK proteins were classified into three major families according to their domain structures. Fourteen types and a total of 131 additional domains were identified, some of which are reported to participate in the recognition of signals or substrates. Cyanobacterial STKs show rather complicated phylogenetic relationships that correspond poorly with phylogenies based on 16S rRNA and those based on additional domains. Conclusion: The number of STK genes in different cyanobacteria is the result of the genome size, ecophysiology, and physiological properties of the organism. Similar conserved motifs and amino acids indicate that cyanobacterial STKs make use of a similar catalytic mechanism as eukaryotic STKs. Gene gain-and-loss is significant during STK evolution, along with domain shuffling and insertion. This study has established an overall framework of sequence-structure-function interactions for the STK gene family, which may facilitate further studies of the role of STKs in various organisms.

Cyanobacteria are an ancient group of gram-negative bacteria with strong genome size variation ranging from 1.6 to 9.1 Mb. Here, we first retrieved all the putative restriction-modification (RM) genes in the draft genome of Spirulina and then performed a range of comparative and bioinformatic analyses on RM genes from unicellular and filamentous cyanobacterial genomes. We have identified 6 gene clusters containing putative Type I RMs and 11 putative Type II RMs or the solitary methyltransferases (MTases). RT-PCR analysis reveals that 6 of 18 MTases are not expressed in Spirulina, whereas one hsdM gene, with a mutated cognate hsdS, was detected to be expressed. Our results indicate that the number of RM genes in filamentous cyanobacteria is significantly higher than in unicellular species, and this expansion of RM systems in filamentous cyanobacteria may be related to their wide range of ecological tolerance. Furthermore, a coevolutionary pattern is found between hsdM and hsdR, with a large number of site pairs positively or negatively correlated, indicating the functional importance of these pairing interactions between their tertiary structures. No evidence for positive selection is found for the majority of RMs, e. g., hsdM, hsdS, hsdR, and Type II restriction endonuclease gene families, while a group of MTases exhibit a remarkable signature of adaptive evolution. Sites and genes identified here to have been under positive selection would provide targets for further research on their structural and functional evaluations.

Eutrophication is becoming a serious problem in coastal waters in many parts of the world. It induces the phytoplankton blooms including 'Red Tides', followed by heavy economic losses to extensive aquaculture area. Some cultivated seaweeds have very high productivity and could absorb large quantities of N, P, CO2, produce large amount of O-2 and have excellent effect on decreasing eutrophication. The author believes that seaweed cultivation in large scale should be a good solution to the eutrophication problem in coastal waters. To put this idea into practice, four conditions should be fulfilled: (a) Large-scale cultivation could be conducted within the region experiencing eutrophication. (b) Fundamental scientific and technological problems for cultivation should have been solved. (c) Cultivation should not impose any harmful ecological effects. (d) Cultivation must be economically feasible and profitable. In northern China, large-scale cultivation of Laminaria japonica Aresch. has been encouraged for years to balance the negative effects from scallop cultivation. Preliminary research in recent years has shown that Gracilaria lemaneiformis (Bory) Daws. and Porphyra haitanensis Chang et Zheng are the two best candidates for this purpose along the Chinese southeast to southern coast from Fujian to Guangdong, Guangxi and Hong Kong. Gracilaria tenuistipitata var. liui Chang et Xia is promising for use in pond culture condition with shrimps and fish.

The construction of the shuttle, expression vector of human tumor necrosis factor alpha (hTNF-alpha) gene and its expression in a cyanobacterium Anabaena sp. PCC 7120 was reported. The 700-bp hTNF cDNA fragments have been recovered from plasmid pRL-rhTNF, then inserted downstream of the promoter PpsbA in the plasmid pRL439. The resultant intermediary plasmid pRL-TC has further been combined with the shuttle vector pDC-8 to get the shuttle, expression vector pDC-TNF. The expression of the rhTNF gene in Escherichia coil has been analyzed by SDS-PAGE and thin-layer scanning, and the results show that the expressed TNF protein with these two vectors is 16.9 percent (pRL-TC) and 15.0 percent (pDC-TNF) of the total proteins in the cells, respectively, while the expression level of TNF gene in plasmid pRL-rhTNF is only 11.8 percent. Combined with the participation of the conjugal and helper plasmids, pDC-TNF has been introduced into Anabaena sg PCC 7120 by triparental conjugative transfer, and the stable transgenic strains have been obtained. The existence of the introduced plasmid pDC-TNF in recombinant cyanobacterial cells has been demonstrated by the results of the agarose electrophoresis with the extracted plasmid samples and Southern blotting with alpha-(32)p labeled hTNF cDNA probes, while the expression of the hTNF gene in Anabaena sp. PCC 7120 has been confirmed by the results of Western blotting with extracted protein samples and human TNF-alpha monoclonal antibodies. The cytotoxicity assays using the mouse cancer cell line L929 proved the cytotoxicity of the TNF in the crude extracts from the transgenic cyanobacterium Anabaena sp. PCC 7120.

Heterosigma akashiwo (Hada) is a fragile, fish-killing alga. Efforts to understand and prevent blooms due to this harmful species to mitigate the impact on aquaculture require the development of methods for rapid and precise identification and quantification, so that adequate warning of a harmful algal bloom may be given. Here, we report the development and application of rRNA and rDNA-targeted oligonucleotide probes for fluorescence in situ hybridization (FISH) to aid in the detection and enumeration of H. akashiwo. The designed probes were species specific, showing no cross-reactivity with four common HAB causative species: Prorocentrum micans Ehrenberg, P. minimum (Pavillard) Schiller, Alexandrium tarmarense (Lebour) Balech, and Skeletonema costatum (Greville) Cleve, or with four other microalgae, including Gymnodinium sp. Stein, Platy-monas cordiformis (Karter) Korsch, Skeletonema sp.1 Greville and Skeletonema sp.2. The rRNA-targeted probe hybridized to cytoplasmic rRNA, showing strong green fluorescence throughout the whole cell, while cells labeled by rDNA-targeted probe exhibited exclusively fluorescent nucleus. The detection protocols were optimized and could be completed within an hour. For rRNA and rDNA probes, about a corresponding 80% and 70% of targeted cells could be identified and quantified during the whole growth circle, despite the inapparent variability in the average probe reactivity. Cyclic nucleotides (both cAMP and cGMP) play extremely important roles in cyanobacteria, such as regulating heterocyst formation, respiration, or gliding. Catalyzing the formation of cAMP and cGMP from ATP and GTP is a group of functionally important enzymes named adenylate cyclases and guanylate cyclases, respectively. To understand their evolutionary patterns, in this study, we presented a systematic analysis of all the cyclases in cyanobacterial genomes. We found that different cyanobacteria had various numbers of cyclases in view of their remarkable diversities in genome size and physiology. Most of these cyclases exhibited distinct domain architectures, which implies the versatile functions of cyanobacterial cyclases. Mapping the whole set of cyclase domain architectures from diverse prokaryotic organisms to their phylogenetic tree and detailed phylogenetic analysis of cyclase catalytic domains revealed that lineage-specific domain recruitment appeared to be the most prevailing pattern contributing to the great variability of cyanobacterial cyclase domain architectures. However, other scenarios, such as gene duplication, also occurred during the evolution of cyanobacterial cyclases. Sequence divergence seemed to contribute to the origin of putative guanylate cyclases which were found only in cyanobacteria. In conclusion, the comprehensive survey of cyclases in cyanobacteria provides novel insight into their potential evolutionary mechanisms and further functional implications.

蓝藻经过漫长的进化,在地球上具有极其广范的分布,而且对环境胁迫具有极强的耐受性,这其中信号转导系统起了至关重要的作用。蓝藻信号转导系统主要包括二元信号转导系统和丝氨酸/苏氨酸激酶。蓝藻全基因组测序工作的完成为蓝藻信号转导系统的研究奠定了基础。目前国外对蓝藻信号转导系统的研究主要集中在对单个蓝藻的单个信号系统的功能研究。本论文首先采用比较基因组学的方法对蓝藻中的两大信号转导系统从分布、结构及进化等角度进行了系统的分析,进而对其中重要信号转导基因的功能进行了验证。 通过比较基因组学分析发现蓝藻中的信号转导系统的分布、结构特征与物种的生理、生态特征之间关系密切;蓝藻中丝氨酸/苏氨酸激酶具有与真核型激酶类似的催化方式;在信号转导系统的进化过程中存在着基因复制、缺失、附属功能域的获得及随机重排等复杂的进化现象;且二元信号转导系统中相互作用的组氨酸激酶与反应调控蛋白之间并不是完全同步的进化关系,可能有着不同的进化历程。该研究建立了蓝藻信号转导系统中的基因-结构-功能的框架结构,为其功能的研究奠定了基础。 聚球藻PCC7942和集胞藻PCC6803是单细胞的淡水蓝藻,具有天然的外源DNA转化系统,是蓝藻分子遗传学研究的模式生物。通过基因突变的方法对这两株蓝藻中二元信号转导系统的部分重要反应调控蛋白同源基因rre28和syn7942_0095的功能进行了验证,发现高度保守的同源序列在这两株蓝藻中起着不同的作用,且一个组氨酸激酶可能与不同的反应调控蛋白相互作用,说明同源序列的功能在进化的过程中已经发生了分化。 目前对蓝藻信号转导系统中丝氨酸/苏氨酸激酶的功能研究较少。通过基因突变及表达差异分析发现集胞藻PCC6803中的丝氨酸/苏氨酸激酶SpkG参与高盐胁迫的信号传递。通过高盐胁迫条件下SpkG对整个转录图谱影响的研究,发现了60个差异表达基因,涉及转运、能量传递、蛋白加工修饰和信号转导等多个生理过程。该研究首次通过实验验证了蓝藻中的丝氨酸/苏氨酸激酶对环境胁迫的响应,并首次发现由二元信号转导系统和丝氨酸/苏氨酸激酶共同参与的对逆境胁迫的调控网络。 蓝藻兼具细菌和植物的特点,蓝藻成熟的转化体系也为真核生物基因功能的研究提供了新的模式宿主。蓝藻信号转导系统中激酶功能的研究为我们进一步研究真核生物激酶的功能提供借鉴,且对蓝藻信号转导的研究将对植物的抗逆胁迫研究提供重要的理论依据。

有害赤潮(Harmful algal blooms, HAB)已成为全球性的海洋灾害,对我国近海养殖业危害尤为巨大,研究赤潮发生、发展的主要环境影响因子,对于控制赤潮的发生以及加强赤潮的防治具有十分重要的意义。本文根据国家自然科学基金“九五”重大项目“中国沿海典型增养殖区有害赤潮发生动力学防治对策研究”项目组近年来对胶州湾东北部养殖海域的监测资料,分析了该海区营养状况,结合该区自然环境特点,探讨了发生于1998年7月和1999年6月两次硅藻赤潮的主要影响因子;并应用胶州现场海水进行了室内模拟培养实验,对影响有害赤潮生消过程的关键环境因子进行了分析探讨。对1998和1999年赤潮多发季节胶州湾东北部增养殖水域海水的营养状况进行了分析,并参照潜在性富营养化的概念,应用NQI指数对该海域的富营养化水平进行了评价。结果表明:(1)胶州湾东北部增养殖海域赤潮多发季节TIN、DIP和活性硅酸盐的含量均较高,远高于同期胶州湾大面观测站各营养盐的平均含量;该海域有机污染物含量相对营养盐含量不高。(2)该海域春季氮、磷营养盐含量充足,水域属富营养化平;夏季该海域具有较高的生物具,磷酸盐水平相对较低,海域多数时间属于磷限制潜在性富营养化水平;但这种限制极易为外界补充所改变,如陆源输入、养殖废水的排放等。(3)该海域为典型增养殖区,养殖废水对海域营养盐的补充有重要意义,因此应当加强对该海域养殖业的综合管理。对分别于1998年7月和1999年6月在胶州湾东北部增养殖海域发生的中肋骨条藻(Skeletonema costatum)赤潮和浮动弯角藻(Eucampia zoodiacus)赤潮生消过程中环境因子的变化进行了探讨。发生赤潮生物量的升高常伴随海水pH值、溶解氧(DO)含量的升高以及各营养盐(氮、磷、硅)含量的降低;认为在该海域较高营养盐含量和较高生物量的情况下,赤潮发生前大规模的降雨过程及随后的气候条件为赤潮生物的快速生长繁殖创造了有利条件;无机磷和活性硅酸盐含量相对于无机氮显得较低,对该海域硅藻赤潮的生消过程起着控制作用。采集胶州湾现场海水进行了实验室模拟培养实验,对影响赤潮生消过程的主要环境因素进行了研究和探讨。结果表明,现场海水进行模拟培养时,排除了潮流、风以及水团间交流混合等因素对水体中浮游生物的影响,培养水体中的浮游植物生物量都存在增高的现象;在氮、磷、硅等营养盐含量充足的条件下,浮游植物可快速进入指数增长期,并较快发展形成赤潮,高浓度的营养盐常导致培养水体中浮游植物的较高的生长速率,浮游植物对氮磷营养盐的吸收受到环境中营养盐含量的影响;初始环境下营养盐的限制可抑制浮游植物的进一步增殖,发生赤潮水体中某种营养盐的含量降低最终导致了赤潮的消亡。

Variations of cellular total lipid, total carbohydrate and total protein content of two dominant bloom-forming species (Skeletonema costatum and Prorocentrum donghaiense) isolated from the Yangtze River Estuary were examined under six different nutrient conditions in batch cultures. Daily samples were collected to estimate the cell growth, nutrient concentration and three biochemical compositions content during 7 days for S. costatum and the same sampling procedure was done every other day during 10 days for P. donghaiense. Results showed that for S. costatum, cellular total lipid content increased under phosphorus (P) limitation, but not for nitrogen (N) limitation; cellular carbohydrate were accumulated under both N and P limitation: cellular total protein content of low nutrient concentration treatments were significantly lower than that of high nutrient concentration treatments. For P. donghaiense, both cellular total lipid content and total carbohydrate content were greatly elevated as a result of N and P exhaustion, but cellular total protein content had no significant changes under nutrient limitation. In addition, the capability of accumulation of three biochemical constituents of P. donghaiense was much stronger than that of S. costatum. Pearson correlation showed that for both species, the biochemical composition of three constituents (lipid, carbohydrate and protein) had no significant relationship with extracellular N concentration, but had positive correlation with extracellular and intracellular P concentration. The capability of two species to accumulate cellular total lipid and carbohydrate under nutrient limitation may help them accommodate the fluctuating nutrient condition of the Yangtze River Estuary. The different responses of two species of cellular biochemical compositions content under different nutrient conditions may provide some evidence to explain the temporal characteristic of blooms Caused by two species in the Yangtze River Estuary. The development of the phytoplankton community was studied in the Jiaozhou Bay during the spring to neap tide in August 2001, through three cruises and a 15 d continuous observation. This investigation indicates that diatom cell abundance increased sharply following the end of a spring tide, from 9 cells/cm(3) to a peak of 94 cells/cm(3). The dominant species composition and abundance show a quick species sequence from spring to neap tide, and the dominant species at the start phase is Skeletomena costatum, then changes to Chaetoceros curvisetus, finally it changes to Eucampia zodiacus. Silicate concentration increases during spring tide, as a result of nutrient replenishment from the water-sediment interface, its initial average concentration in neap tide is 1.39 mumol/dm(3) and reached the peak average concentration of 8.40 mumol/dm(3) in spring tide. But the nitrogen concentration dropped due to dilution by the low nitrogen seawater from the Huanghai Sea, its initial average concentration in neap tide is 67 mumol/dm(3) and decreased to the average concentration of 54 mumol/dm(3) in spring tide. The degree of silicon limitation was decreased and phytoplankton, especially diatoms, responds immediately after nutrient replenishment in the water column. Skeletonmea costatum, as one of the dominant species in the Jiaozhou Bay, shows a quicker response to nutrient availability than Eucampia zodiacus and Chaetoceros curvisetus. It is proposed that dominant species composition and water column stability synchronously determine the development of phytoplankton summer blooms in the Raozhou bay.

in order to investigate a new method of mitigating the deleterious effect of harmful algal blooms (HABs), the inhibition of the glycolipid biosurfactant sophorolipid on three common harmful algae Alexandrium tamarense, Heterosigma akashiwo and Cochlodinium polykrikoides was studied. The optimum preparation condition for sophorolipid, the inhibition capability of sophorolipid and the interaction mechanism of sophorolipid on the three algal species were investigated. Results showed that sophorolipid prepared by extraction with ethyl acetate exhibited the most prominent inhibition effect and that storage time of one year had little influence on the inhibition effect of sophorolipid. The optimum concentration of 10-20 mg/l sophorolipid inhibited the motility of about 90% of the tested harmful algal cells without recovery. Investigation of the algicidal process revealed that sophorolipid induced ecdysis of A. tamarense, quick lysis of H. akashiwo and swelling of C. polykrikoides in a relatively short time. Investigation of the nucleotides showed that more than 15% of the nucleotides were released from the cytoplasm under the effect of 10-20 mg/l sophorolipid, indicating the irreversible damage on the cellular membrane, which resulted in the disintegration of the harmful algal cells. The recovery and fate of three species of dinoflagellates, Alexandrium tamarense, Cochlodinium polykrikoides and Scrippsiella trochoidea, after having been sedimented by yellow clay, were investigated in the laboratory. The effect of burying period in yellow clay pellet and mixing on the recovery of settled algal cells were studied. The morphological changes of algal cells in yellow clay pellet were also tracked. Results showed that there was almost no recovery for A. tamarense and C. polykrikoides, and the cells decomposed after 2-3 days after visible changes in morphology and chloroplasts. There was some recovery for S. trochoidea. Moreover, S. trochoidea cysts were formed in clay pellet during the period of about 14 days, with the highest abundance of 87 000 cysts g(-1) clay and the incidence of cyst formation of 6.5%, which was considered as a potential threat for the further occurrence of algal blooms. S. trochoidea cysts were isolated from yellow clay and incubated to test their viability, and a germination ratio of more than 30% was obtained after incubation for 1 month. These results showed the species specificity of the mitigation effect of yellow clay. It is suggested that cautions be taken for some harmful species and thorough risk assessments be conducted before using this mitigation strategy in the field.
