Processing of a recently acquired seismic line in the northeastern South China Sea by Project 973 has been conducted to study the character and the distribution of gas hydrate Bottom-Simulating Reflectors (BSRs) in the Hengchun ridge. Analysis of different-type seismic profiles shows that the distribution of BSRs can be revealed to some extents by single-channel profile in this area, but seismic data processing plays an important role to resolve the full distribution of BSRs in this area. BSR' s in the northeastern South China Sea have the typical characteristics of BSRs on worldwide continental margins: they cross sediment bed reflections, they are generally parallel to the seafloor and the associated reflections have strong amplitude and a negative polarity. The characteristics of BSRs in this area are obvious and the BSRs indicate the occurrence of gas hydrate-bearing sediments in the northeastern South China Sea. The depth of the base of the gas-hydrate stability zone was calculated using the phase stability boundary curve of methane hydrate and gas hydrate with mixture gas composition and compared with the observed BSR depth. If a single gradient geothermal curve is used for the calculation, the base of the stability zone for methane hydrate or gas hydrate with a gas mixture composition does not correspond to the depth of the BSRs observed along the whole seismic profile. The geothermal gradient therefore changes significantly along the profile. The geothermal gradient and heat flow were estimated from the BSR data and the calculations show that the geothermal gradient and heat flow decrease from west to east, with the increase of the distance from the trench and the decrease of the distance to the island arc. The calculated 2 heat flow changes from 28 to 64 mW/m(2), which is basically consistent with the measured heat flow in southwestern offshore Taiwan.

The effective dielectric response of graded spherical composites having general power-law gradient inclusions is investigated under a uniform applied electric field, where the dielectric gradation profile of the spherical inclusions is modeled by the equation epsilon(i) (r) = c(b+r)(k). Analytical solutions of the local electrical potentials are derived in terms of hyper-geometric function and the effective dielectric response of the graded composites is predicted in the dilute limit. From our result, the local potentials of graded spherical composites having both simple power-law dielectric profile epsilon(i)(r) = cr(k) and linear dielectric profile epsilon(i) (r) = c(b+r) are derived exactly by taking the limits b --> 0 and k --> 1, respectively. In the dilute limit, our exact result is used to test the validity of differential effective dipole approximation (DEDA) for estimating the effective response of graded spherical composites, and it is shown that the DEDA is in excellent agreement with exact result. Based on the mass balance equations of solute transfer in the radial chromatographic column, the theoretical expression to describe the column efficiency and shape of elution profile is obtained under linear isotherm case. Moreover, the tendency for the variation of column efficiency and symmetry of peak profile is systematically discussed. The results showed that in radial chromatography the relationship between the column efficiency and volumetric flow rate is similar with that relationship in axial chromatography; relatively high column efficiency still can be obtained under high flow rate in radial chromatography. Accompanying the increase of retention factor of solutes and injection time, the column efficiency decreases monotonously. The effect of column diameter and column length on the column efficiency interfere with each other. It is more advantageous to increase the column efficiency by applying columns with larger column diameter and shorter column length. According to the discussion of the effect of diffusion on the column efficiency, radial chromatography is proved to be suitable for the separation of samples with relatively high diffusion coefficient, which predicts its obvious advantage in the preparative separation of samples such as proteins and DNA.

Mature human interleukin-11 (HuIL-11) is a cytokine consisting of 178 amino acid residues that results from scission of the N-terminal signal peptide, consisting of 21 amino acid residaues, from the corresponding nascent polypeptide. A DNA fragment encoding a truncated HuIL-11 (trHuIL-11), with an additional 5 amino acid residues removed from the N-terminus, was cloned into vector pGEX-2T between the BamHI site and the EcoRI site. Upon transformation with Escherichia coli BL21, the construct over-produced a glutathione S-transferase (GST)-fused protein in a soluble form after IPTG induction. The fusion protein was initially fractionated with butyl-Sepharose 4 fast flow column and by affinity chromatography using a GSH-Sepharose 4B column. On-site enzymatic release with thrombin gave the target protein at 96% purity as judged by SDS-PAGE and HPLC. Expression of the interleukin as a GST-fused protein thus greatly improved downstream processing. Subsequent biological activity assay suggested that trHuIL-11 had similar activity profile to the naturally produced sample and may be a promising candidate for further development as biopharmaceutical.
