The process of deoxyribonucleio acid (DNA) sample preparation in scanning tunneling microscope (STM) and atomic force microscope (AFM) is reviewed. The main discussions are devoted to the methods, advantages or drawbacks and improvement of the DNA sample's immobilization and spreading.

Kinetics and its regulation by extrinsic physical factors govern selectin-ligand interactions that mediate tethering and rolling of circulating cells on the vessel wall under hemodynamic forces. While the force regulation of off-rate for dissociation of selectin-ligand bonds has been extensively studied, much less is known about how transport impacts the on-rate for association of these bonds and their stability. We used atomic force microscopy (AFM) to quantify how the contact duration, loading rate, and approach velocity affected kinetic rates and strength of bonds of P-selectin interacting with P-selectin glycoprotein ligand I (PSGL-1). We found a saturable relationship between the contact time and the rupture force, a biphasic relationship between the adhesion probability and the retraction velocity, a piece-wise linear relationship between the rupture force and the logarithm of the loading rate, and a threshold relationship between the approach velocity and the rupture force. These results provide new insights into how physical factors regulate receptor-ligand interactions.
Competitive adsorption between bovine serum albumin and collagen observed by atomic force microscope

Atomic force microscopy (AFM) was used to study the competitive adsorption between bovine serum albumin (BSA) and type I collagen on hydrophilic and hydrophobic silicon wafers. BSA showed a grain shape and the type I collagen displayed fibril-like molecules with relatively homogeneous height and width, characterized with clear twisting (helical formation). These AFM images illustrated that quite a lot of type I collagen appeared in the adsorption layer on hydrophilic surface in a competitive adsorption state, but the adsorption of BSA was more preponderant than that of type I collagen on hydrophobic silicon wafer surface. The experiments showed that the influence of BSA on type I collagen adsorption on hydrophilic surface was less than that on hydrophobic surface.

Thoroughly understanding AFM tip-surface interactions is crucial for many experimental studies and applications. It is important to realize that despite its simple appearance, the system of tip and sample surface involves multiscale interactions. In fact, the system is governed by a combination of molecular force (like the van der Waals force), its macroscopic representations (such as surface force) and gravitational force (a macroscopic force). Hence, in the system, various length scales are operative, from sub-nanoscale (at the molecular level) to the macroscopic scale. By integrating molecular forces into continuum equations, we performed a multiscale analysis and revealed the nonlocality effect between a tip and a rough solid surface and the mechanism governing liquid surface deformation and jumping. The results have several significant implications for practical applications. For instance, nonlocality may affect the measurement accuracy of surface morphology. At the critical state of liquid surface jump, the ratio of the gap between a tip and a liquid dome (delta) over the dome height (y(o)) is approximately (n-4) (for a large tip), which depends on the power law exponent n of the molecular interaction energy. These findings demonstrate that the multiscale analysis is not only useful but also necessary in the understanding of practical phenomena involving molecular forces. (c) 2007 Elsevier Ltd. Phage M13Ko7 Detection With Biosensor Based On Imaging Ellipsometry And Afm Microscopic Confirmation

A rapid detection and identification of pathogens is important for minimizing transfer and spread of disease. A label-free and multiplex biosensor based on imaging ellipsometry (BIE) had been developed for the detection of phage M13KO7. The surface of silicon wafer is modified with aldehyde, and proteins can be patterned homogeneously and simultaneously on the surface of silicon wafer in an array format by a microfluidic system. Avidin is immobilized on the surface for biotin-anti-M13 immobilization by means of interaction between avidin and biotin, which will serve as ligand against phage M13KO7. Phages M13KO7 are specifically captured by the ligand when phage M13KO7 solution passes over the surface, resulting in a significant increase of mass surface concentration of the anti-M13 binding phage M13KO7 layer, which could be detected by imaging ellipsometry with a sensitivity of 10(9) pfu/ml. Moreover, atomic force microscopy is also used to confirm the fact that phage M13KO7 has been directly captured by ligands on the surface. It indicates that BIE is competent for direct detection of phage M13KO7 and has potential in the field of virus detection. (C) 2008 Elsevier B.V. All rights reserved.

Sheet resistance of laser-irradiated Ge2Sb2Te5 thin films prepared by magnetron sputtering was measured by the four-point probe method. With increasing laser power the sheet resistance undergoes an abrupt drop from 10(7) to 10(3) Omega/square at about 580 mW. The abrupt drop in resistance is due to the structural change from amorphous to crystalline state as revealed by X-ray diffraction (XRD) study of the samples around the abrupt change point. Crystallized dots were also formed in the amorphous Ge2Sb2Te5 films by focused short pulse laser-irradiated, the resistivities at the crystallized dots and the non-crystallized area are 3.375 x 10(-3) and 2.725 Omega m, sheet resistance is 3.37 x 10(4) and 2.725 x 10(7) Omega/square respectively, deduced from the I-V Curves that is obtained by conductive atomic force microscope (C-AFM). Self-ordered porous alumina films on a semi-insulated GaAs substrate were prepared in oxalic acid aqueous solutions by three-step anodization. The I-t curve of anodization process was recorded to observe time effects of anodization. Atomic force microscopy was used to investigate structure and morphology of alumina films. It was revealed that the case of oxalic acid resulted in a self-ordered porous structure, with the pore diameters of 60-70 nm, the pore density of the order of about 10(10) pore cm(-2), and interpore distances of 95-100nm. At the same time the pore size and shape change with the pore widening time. Field-enhanced dissolution model and theory of deformation relaxation combined were brought forward to be the cause of self-ordered pore structure according to I-t curve of anodization and structure characteristics of porous alumina films. 本论文主要借助原子力显微镜(AFM)、X-射线光电子能谱(XPS)等实验手段,纳米尺度上在线原位地研究了高温临界组成PMMMA/SAN(50/50,w/w)共混薄膜体系,分析并探讨了此共混薄膜表面相分离及其超薄膜体系的润湿/去润湿和相分离行为。首先,从敲击式AFM中相位图的成像原理出发,建立了一个高温下用原位AFM定性鉴别不同聚合物的纳米尺寸微区的方法,即:先在基底(硅)上铺展一层非常平坦的单一组分(SAN)薄膜,再在此千膜上,通过旋涂由选择性溶剂(冰醋酸)配制的另一组分(PMMA)的极稀溶液,使之不能形成一层连续的完整膜,最后在高温(175℃)下用敲击模式的原子力显微镜(TM-AFM)检测相位图随退火时间的变化。得到在175℃的相位图中,PMMA富相比SAN的富相显得更暗,这为后续工作提供了定性鉴别相区的方法。其次,高温下用原位AFM研究了PMMA/SAN薄膜表面相分离过程,在线观察了相分离的归并过程,定量地得到了临界相分离温度,并给出了特征波矢对时间依赖关系的标度指数,划分了相分离动力学演变的不同阶段。对于膜厚约为130nm的体系,表面相分离的临界温度大约为165℃,其表面相分离的特征波矢与时间的标度关系q+(t)-t-n,在整个实验时间内,随时间的演变过程中,显示了两个不同的指数变化区,即前期很慢的n=0.13和后期的n=1/3。0.13的指数关系可能是由于表面聚合物链的几何受限及表面富集相把部分新生成的另一相覆盖所致,1/3的指数关系可以认为是由普遍的Brownian扩散所致。而对于膜厚为50曲的此组成共混薄膜,得到的标度关系与C汕n线性理论吻合得非常好的Spinodal Deposition(SD)表面相分离的初期,即n=0,这是由于降低膜厚 增加了共混物的相容性,提高了临界温度,从而减缓了相分离过程,使得在我 们观测的时间范围内更易观察到表面相分离的初期。再次,用阶梯式降温的好M和XPS,发现了PMMA/SAN体系原位和离位实验结果存在巨大差别的主要原因之一—润湿温度的存在。对PMMA/SAN(50/50,w/w,-130nm)共混薄膜体系,原位AFM的退火和准淬火实验表明, 虽然在高温和室温、真空和常态、原位和离位,相分离的聚合物共混薄膜表面 形貌变化不大,但是表面物理性质却有很大的区别。原位XPS实验表明,未经 任何处理的样品在185℃退火时,很快在离表面很薄的下面形成一层PMMA含 量远低于本体值的薄层,之后最表面的PMMA也往本体迁移,直至表面SAN 的含量远高于它的本体值。对已在175℃退火20hr的此样品,逐渐降温退火过 程的原位XPS表明,当退火温度降至145oC时,样品表面几乎完全被PMMA覆盖,很好地验证了Conlposto等提出的润湿温度的假设,即对此体系,润湿温 度是原位和离位存在巨大的差异的主要因素之一,这与原位AFM的实验结果也,存在很好的一致性。最后,高温下原位观察了硅基底上PMMA/SAN超薄膜(膜厚-Rg)的去润湿、相分离过程和PMMA在基底上的润湿过程,以及云母基底上的该体系高温下的稳定润湿行为。以硅为基底的PMM刀sAN共混超薄膜在155”c下发生了类 似sPinodal dewetting的去润湿,其原因可能是在垂直于基底的浓度梯度引起的表面组成的涨落,对这种去润湿的动力学还进行了探讨。以硅为基底的PMMA/SAN共混超薄膜在175℃下不仅发生了类似spinodal dewetting的去润 湿,而且还发生了去润湿液滴内部的相分离以及相分离析出的PMMA润湿硅基底的过程,提出了一个简单的模型描述了这种复杂的过程。此外,还研究了不同基底对此共混超薄膜体系的润湿行为进行了研究,以云母为基底的此共混体系在175℃下长时间内是稳定的,其原因可能是云母与PMMA和SAN均有很强的作用力,足于补偿聚合物链在构象嫡上的损失。

In this paper, we report for the first time on the synthesis of ZnO nanocrystallites in conjugated polymer (PPV) nanofibers by the coupling of the in situ/blend methods and electrospinning. These composite nanofibers were characterized by fluorescence microscopy, atomic force microscope (AFM), field emission scanning electron microscopy (FE-SEM), transmission electron microscopy (TEM), photoluminescence (PL) spectra, Fourier transform infrared (FT-IR) spectroscopy, and X-ray powder diffraction (XRD).

The reversible fabrication of positive and negative nanopatterns on 1-hexadecanethiol (HDT) self-assembled monolayers (SAMs) on Au(111) was realized by bias-assisted atomic force microscopy (AFM) nanolithography using an ethanol-ink tip. The formation of positive and negative nanopatterns via the bias-assisted nanolithography depends solely on the polarity of the applied bias, and their writing speeds can reach 800,um/s and go beyond 1000 mu m/s, respectively. The composition of the positive nanopatterns is gold oxide and the nanometer-scale gold oxide can be reduced by ethanol to gold, as proved by X-ray photoelectron spectroscopy (XPS) analysis, forming the negative nanopatterns which can be refilled with HDT to recover the SAMs.

The assembly and disassembly of RecA-DNA nucleoprotein filaments on double-stranded DNA (dsDNA) or single-stranded DNA (ssDNA) are important steps for homologous recombination and DNA repair. The assembly and disassembly of the nucleoprotein filaments are sensitive to the reaction conditions. In this work, we investigated different morphologies of the formed nucleoprotein filaments at low temperature under different solution conditions by atomic force microscopy (AFM). We found that low temperature and long keeping time could induce the incomplete disassembly of the formed nucleoprotein filaments.

NaYF4:Yb3+, Er3+ nanoparticles were successfully prepared by a polyol process using diethyleneglycol (DEG) as solvent. After being functionalized with SiO2-NH2 layer, these NaYF4:Yb3+, Er3+ nanoparticles can conjugate with activated avidin molecules (activated by the oxidation of the oligosaccharide chain). The as-formed NaYF4:Yb3+, Er3+ nanoparticles, NaYF4:Yb3+, Er3+ nanoparticles functionalized with amino groups, avidin conjugated amino-functionalized NaYF4:Yb3+, Er3+ nanoparticles were characterized by X-ray diffraction (XRD), transmission electron microscopy (TEM), atomic force microscopy (AFM), Fourier transform infrared (FT-IR), UV/Vis absorption spectra, and up-conversion luminescence spectra, respectively.

An electrochemiluminescence (ECL) sensor based on Ru(bpy)(3)(2+)-graphene-Nafion composite film was developed. The graphene sheet was produced by chemical conversion of graphite, and was characterized by atomic force microscopy (AFM), scanning electron microscopy (SEM), and Raman spectroscopy. The introduction of conductive graphene into Nafion not only greatly facilitates the electron transfer of Ru(bpy)(3)(2+), but also dramatically improves the long-term stability of the sensor by inhibiting the migration of Ru(bpy)(3)(2+) into the electrochemically inactive hydrophobic region of Nafion. The ECL sensor gives a good linear range over 1 x 10(-7) to 1 x 10(-4) M with a detection limit of 50 nM towards the determination of tripropylamine (TPA), comparable to that obtained by Nafion-CNT.

The structural evolution of a single-layer latex film during annealing was studied via grazing incidence ultrasmall-angle X-ray scattering (GIUSAXS) and atomic force microscopy (AFM). The latex particles were composed of a low-T-g (-54 degrees C) core (n-butylacrylate, 30 wt %) and a high-T-g (41 degrees C) shell (t-butylacrylate, 70 wt %) and had an overall diameter of about 500 nm. GIUSAXS data indicate that the q(y) scan at q(z) = 0.27 nm(-1) (out-of-plane scan) contains information about both the structure factor and the form factor. The GIUSAXS data on latex films annealed at various temperatures ranging from room temperature to 140 degrees C indicate that the structure of the latex thin film beneath the surface changed significantly. The evolution of the out-of-plane scan plot reveals the surface reconstruction of the film. Furthermore, we also followed the time-dependent behavior of structural evolution when the latex film was annealed at a relatively low temperature (60 degrees C) where restructuring within the film can be followed that cannot be detected by AFM, which detects only surface morphology.
