The electrochemistry of disulfide in cytochrome c on gold electrodes was reported. The observed electrochemical response was used to explain why the electrochemical reaction of cytochrome c is irreversible at gold electrodes. Disulfide bonds in cytochrome c were strongly adsorbed onto the surface of gold electrodes and caused slow rate of electron transfer of the heme group. It was found that the presence of disulfides in cytochrome c was responsible for the lack of electrochemical response of the heme group on a gold electrode. The mechanisms for this effect were studied using electrochemistry and photoelectron spectroscopy. (C) 1999 Elsevier Science B.V. All rights reserved.

A high pressure and high temperature method was used to efficiently extract on a large scale metallofullerenes M@C-2n (M=La,Ce) in a closed vessel under argon gas protection. With pyridine as the HPHT solvent, about 60-80% M@C-2n and 30-55% M@C-82 can be enriched, M@C-82 is dissolved selectively; With toluene as the HPHT solvent, M@C-2n can also be efficiently extracted, especially M@C-74, which is a new member of M@C-2n soluble species. Gas-phase ion-molecule reactions of buckminsterfullerene (C-60) with the ion systems generated from the self-chemical ionization of alkyl methyl ethers (CH3OR, R = n-C2H5, n-C3H7, n-C4H9) were studied in the ion source of a mass spectrometer. The adduct cation [C60C2H5O](+) and protonated molecule [C60H](+) were observed as the major products. The former adduct ion was produced by the reactions of C-60 with the methoxymethyl ion [CH3OCH2](+), and the latter resulted from the proton transfer reactions from protonated alkyl methyl ethers to C-60 It is suggested that the [3+2] cycloadduct to a 6-6 bond of C-60 (a C-C bond common to two annulated six-membered rings) is the most favorable structure among the probable isomers of [C60C2H5O](+). At the self-assembled monolayer (SAM) of a thiol-functionalized viologen modified gold electrode, cytochrome c (cyt c) exhibits a quasi-reversible electrochemical reaction. The heterogeneous electron transfer rate constant of cyt c in 0.1 mol/L phosphate buffer solution(pH 6.96) is 0.164 cm.s(-1) at 500 mV/s. The adsorbed cyt c on the viologen SAM forms a closely packed monolayer, whose average electron transfer rate is 4.85 s(-1) in the scan range of 50 to 500 mV/s. These results suggest that the SAM of viologen-thiol is a relatively stable, ordered and well-behaved monolayer from an electrochemical standpoint and it promotes the electron transfer process of biomolecules on electrode surface well.

The denaturation of cytochrome-e (cyt-c) induced by bromopyrogal red (BPR) was studied by scanning tunnelling microscopy (STM) on the electrochemically pretreated highly oriented pyrolytic graphite (HOPG) surface. STM images reveal that denatured cyt-c molecules exist in variable states including aggregates, globular compact, partially unfolded and combined with BPR molecule. The apparently low image contrast of denatured cyt-c observed in this experiment comparing to that of native cyt-c molecules, and the relative low image contrast of the unfolded part comparing with the compact globular part, are ascribed to the unfavourable tunnelling paths for the conformational variations of denatured cyt-c molecules. Ion/molecule reactions of C-60 with vinyl acetate under chemical ionization conditions have been studied here. Compared with C2H3O+ from acetone, C2H3O+ from vinyl acetate undergoes the reactions more easily, a new heterocycle between C-60 and the studied ion is formed The generation of two sigma-bonds and little angle tensile force of pentatomic ring make it more stable.

The dynamic states of cytochrome c multilayers on electrochemically pretreated highly oriented pyrolytic graphite (HOPG) have been studied by in-situ scanning tunnelling microscopy (STM) under potential control of both the tip and the substrate in cytochrome c and phosphate buffer solution. The dynamic characterization of cytochrome c multilayers and relatively stable adsorbed single cytochrome c molecules scattered on HOPG imply that physically adsorbed multilayers were more easily influenced by the STM tip than those of chemically adsorbed single molecules. In-situ STM images of chemically adsorbed cytochrome c molecules with discernible internal structures on HOPG revealed that morphologies of cytochrome c molecules also suffered tip influence; possible tip-sample-substrate interactions have been discussed.

The changes of the synchronous fluorescence spectra and the electrochemical behaviour of cytochrome c with the urea concentration are studied. It has been found that with the increase of urea concentration, there occur sequentially the deaggregation of cytochrome c molecules, the increase of exposure extent of the heme group to the solvent, the disruption of Fe-S bond of the heme group and the change in the electrochemical behaviour of cytochrome c. It is suggested that the reason why the electrochemical reaction of cytochrome c is irreversible is that cytochrome c molecules exist in the concentrated solution as oligomers which are electrochemically inactive.

Amorphous samples of polyether ketone with cardo(PEK-C) have been studied in the solution state by C-13, H-1 high-resolution NMR, The H-1 and C-13 1D NMR spectra were assigned using two dimensional chemical shift correlated spectroscopy, 2D homonuclear correlated(COSY) and heteronuclear correlated (HETCOR) spectroscopy present important information. In this work, the structural units of PEK-C was determined by NMR. For some peaks, these assignments are confirmed by two dimensional long-range heteronuclear correlation experiments, A little modification is made on the original C-13 peak assignments for the main chain, The symmetry and the isotacticity of the chain structure for PEK-C are obvious on NMR data.

The direct electrochemistry of cytochrome c was studied at nanometer-sized rare earth element dioxide particle-modified gold electrodes. It was demonstrated that rare earth element oxides can accelerate the electrochemical reaction of cytochrome c and the reversibility of the electrochemical reaction of cytochrome c was related to the size of rare earth element oxide particles.

The conformational transition of horse heart cytochrome c induced by bromopyrogal red (BPR) in very low concentration has been firstly investigated by dynamic spectroelectrochemical technique, both at the BPR adsorbed platinum gauze electrode and at a bare platinum gauze electrode in a solution containing BPR. The effect of BPR on the structure of cytochrome c was studied by UV-visible and Fourier transform IR spectroscopy. The unfolded cytochrome c behaves simply as an electron transfer protein with a formal potential of -142 mV vs. a normal hydrogen electrode. The difference between the formal potentials of the native and unfolded cytochrome c is coupled to a difference in conformational energy of the two states of about 40 kJ mol(-1), which agrees well with the result reported. The stability and slow refolding of the unfolded cytochrome c are discussed.

Synchronous fluorescence spectra of cytochrome c solutions were studied. It was found that synchronous fluorescence spectra of tyrosine and tryptophan residues in cytochrome c molecules can be separated using different wavelength intervals. The changes in synchronous fluorescence spectra of cytochrome c solutions with the solution pH are different from that of free tyrosine and tryptophan and reflect the pH-induced conformational transitions of cytochrome c molecules. The states of cytochrome C molecules in aquous solution were studied with synchronous fluorescence spectroscopy, It was found that the synchronous fluorescent spectra of cytochrome C were contributed by tyrosine and tryptophan residues separately at Delta lambda = 20 nm and Delta lambda = 80 nm, The peak position in synchronous fluorescent spectra of tyrosine residues in cytochrome C molecule does not change with its concentration, but that of tryptophan residue changes with its concentration, Only one peak at 340.0 nm was observed in the dilute solution of cytochrome C, With increasing the concentration of cytochrome C, a new peak at 304. 0 nm appeared. The peak at 340.0 nm disappeared and only one peak at 304.0 nm was observed at a higher concentration of cytochrome C, It may originate from the change of aggregation states of cytochrome C molecules and it was considered that the peak at 340.0 nm was attributed to the monomer and peak at 304.0 nm was due to the dimmer or oligomers. When urea was added into cytochrome C solution in which both monomer and dimmer or oligomers exist, cytochrome C molecules do not denature in the range of the specific concentrations of urea. The concentration of monomer of cytochrome C molecules increased and that of aggregation slates decreased by adding urea, Therefore, the synchronous fluorescence spectroscopy can be used to identify monomer and aggregation states of cytochrome C molecules.

In chemical ionization mass spectrometry (CIMS) gas phase C-60(+) or C-60 can react with fragment ions from three chloromethane and four multichloroethane molecular ions via ion-molecule reactions. A dozen of gas-phase adduct ions of C-60 are observed, and most of them contain chlorine atoms. The results of the comparison and analysis show that the relative intensities of adductions are not directly proportional to the corresponding fragment ions in the MS of reagents,which implies that some fragment ions containing radicals are more reactive with C-60(+) or C-60. This indicates that the alkene-like C-60(+) or C-60 can act as a radical sponge in addition reactions.
