The effect of metal (Li+, Na+, K+, Ag+) cationization on collision-induced dissociation of ginsenosides was investigated by electrospray ionization mass spectrometry combined with multi-stage mass spectrometry (ESI-MSn). The fragments of sodiated and lithiated molecules give valuable structural information regarding the nature of the aglycone and the sequence and linkage information of sugar moieties. However, the number and relative abundances of fragment ions from lithiated ginsenosides are significantly greater than for the sodiated species, The K+ adducts undergo glycosidic cleavages and very limited cross-ring reactions. The silver ion adducts fragment mainly through glycosidic cleavages. Copyright (C) 2001 John Wiley & Sons, Ltd.

Electrospray ionization (ESI) combined with multiple-stage tandem mass spectrometry (MSn) was used to directly analyze the glycolipid mixture from bacteria Bacillus pumilus without preliminary separation. Full scan ESI-MS revealed the composition of picomole quantities of glycerolglycolipid species containing C-14-C-19 fatty acids, some of which were monounsaturated, Two main components were identified from their molecular masses and fragmentation pathways. The fragmentation pathway of the known compound compared with the investigated compound verified the proposed structure as 1(3)-acyl-2-pentadecanoyl-3(1)-O-[beta-D-glucopyranosyl-(1-->6)-O-beta-D-glucopyranosyl]-sn-glycerols. A comparison of the multiple tandem mass spectra of the different alkali-metal cation adducts indicates that the intensity of fragments and the dissociation pathways are dependent on the alkali-metal type, The basic structures of glycerolglycolipids were reflected clearly from the fragmentation patterns of the sodium cations, The intense fragments of the sugar residue from the precursor ions were obtained from the lithiated adduct ions. ESI-MSn spectra of [M + K](+) ions did not provide as much fragmentation as [M + Na](+) and [M + Li](+) adducts, but their spectra allow the position of glycerol acylation to be determined. On the basis of MS2 spectra of[M + K](+) ions, it was established that all components have a C-15:0 fatty acid at the sn-2 position of the glycerol backbone and C-14-C-19 acids at the sn-1 position of the glycerol backbone. We have developed a two-stage growth one-step process for cultivation of Haematococcus using a self-designed system that mimics an open pond in the natural environment. The characteristics of this process are green vegetative cell growth and cysts transformation and pigment accumulation that proceed spontaneously and successively in one open photobioreactor. Four strains of Haematococcus (H. pluvialis 26; H. pluvialis 30; H. pluvialis 34; H. pluvialis WZ) were cultured in this imitation system for a duration of 12 days. The changes in cell density and medium pH were closely monitored, and the astaxanthin content and yield of the four Haematococcus strains were measured at the end of 12 days of cultivation. Two of the strains, H. pluvialis 26 and H. pluvialis WZ, were selected as strains suitable for mass culture, resulting in the astaxanthin yield of 51.06 and 40.25 mg L-1 which are equivalent to 2.79 and 2.50% of their dry biomass respectively. Based on the laboratory work, 6 batch cultures of H. pluvialis WZ were conducted successfully to produce astaxanthin in two 100 m(2) open race-way pond by two-stage growth one-step process. The astaxanthin content ranged from 1.61 to 2.48 g 100 g(-1) dry wt., with average astaxanthin content of 2.10 g 100 g(-1) dry wt. Compared with the one-stage production of astaxanthin based on continuous culture, the superiority of our process is that it can accumulate much more astaxanthin in red cysts. Compared with two-stage production of astaxanthin, the advantage of our process is that it does not need to divide the production process into two parts using two bioreactors. The presented work demonstrates the feasibility for producing astaxanthin from Haematococcus using a two-stage growth one-step process in open pond, culture systems that have been successfully used for Spirulina and Chlorella mass culture. The future of Haematococcus astaxanthin production has been also discussed. (C) 2009 Elsevier B.V. All rights reserved.

The chlorophyll fluorescence kinetics of marine red alga Grateloupia turutunt Yamada, green alga Ulva pertusa Kjellm and brown alga Laminaria japonica Aresch during natural sustained dehydration were monitored and investigated. The pulse amplified modulation (PAM) system was used to analyze the distinct fluorescence parameters during thallus dehydration. Results proved that the fluorescence kinetics of different seaweed all showed three patterns of transformation with sustained water loss. These were: 1) peak kinetic pattern (at the early stage of dehydration fluorescence enhanced and quenched subsequently, representing a normal physiological state). 2) plateau kinetic pattern (with sustained water loss fluorescence enhanced continuously but quenching became slower, finally reaching its maximum). 3) Platform kinetic pattern (fluorescence fell and the shape of kinetic curve was similar to plateau kinetic pattern). A critical water content (CWC) could be found and defined as the percentage of water content just prior to the fluorescence drop and to be a significant physiological index for evaluation of plant drought tolerance. Once thallus water content became lower than this value the normal peak pattern can not be recovered even through rehydration, indicating an irreversible damage to the thylakoid membrane. The CWC value corresponding to different marine species were varied and negatively correlated with their desiccation tolerance, for example. Laminaria japonica had the highest CWC value (around 90%) and the lowest dehydration tolerance of the three. In addition, a fluorescence "burst" was found only in red algae during rehydration. The different fluorescence parameters F-o, F-v and F-v, F-m were measured and compared during water loss. Both F-o and F-v increased in the first stage of dehydration but F-v/F-m. kept almost constant. So the immediate response of in vivo chlorophyll fluorescence to dehydration was an enhancement. Later with sustained dehydration F-o increased continuously while F-v decreased and tended to become smaller and smaller. The major changes in fluorescence (including fluorescence drop during dehydration and the burst during rehydration) were all attributed to the change in F-o instead of F-v This significance of F-o indicates that it is necessary to do more research on F-o as well as on its relationship with the state of thylakoid membrane.

Porphyra yezoensis Ueda (Rhodophyta) is a seaweed of economic importance with a typical dimorphic life cycle consisting of a leafy gametophyte and a filamentous sporophyte. Recently, it has been recognized as a model system for fundamental and applied studies in marine biological sciences. Conchospore, a major spore linking the two distinct multicellular phases in the life cycle, is most widely used in the breeding of P. yezoensis. In this paper, the early-stage development of conchospore, including the attachment and the cell wall formation, was studied with fluorescent reagents staining and Scanning Electron Microscopy detection. Results displayed: (I) the cell wall began to be generated after culturing for 4 h in the attached conchospores; (2) the initially released conchospores were plastids with some filmy, amorphous substance on the surface, and they attached to the fibers firmly via the actively secreted mucilaginous substances after their touch to the fibers; (3) cellulase and pectolase prohibited the attachment of conchospores in the different ways; and (4) only attached conchospores generated cell walls and developed normally, while the suspending ones could not. It indicated that the cellulose played crucial roles in the permanent attachment as the pectin did in the initial attachment. The conchospore attachment seemed to trigger the cell wall formation and the further development. Affects of light on the development of conchospores were also discussed. The results showed that high intensity (200 mu mol.m(-2).s(-1)) and long-wave (>= 580 nm) light facilitated the division rate of conchospores. (C) 2008 Elsevier B.V. All rights reserved.

Based on the study of palaeo-environmental evolution in the shelves of the Eastern China Seas, the concept of ''shelf desertization'' in the late stage of Upper Pleistocene is defined; the environmental background and evolutionary process of shelf desertization are analysed. Study on the records of subbottom profiling and the data of core samples from shelf areas revealed that during low sea-level stages, the sedimentary environment in the exposed shelf plains was dominated by aeolian depositional process under cold and dry climatic conditions, i.e. under the action of strong winter-monsoon winds. Parts of the exposed marine strata were disintegrated, and aeolian sand dunes were formed on the disintegrated marine deposits, from which the finer sediment grains were blown away by wind and deposited in the downwind areas to form the derivative loess deposits. Thus a desertization environmental system was formed in the exposed shelf plains of the Eastern China Seas.

Based on the latest seismic and geological data, tectonic subsidence of three seismic lines in the deepwater area of Pearl River Mouth Basin (PRMB), the northern South China Sea (SCS), is calculated. The result shows that the rifting process of study area is different from the typical passive continental margin basin. Although the seafloor spreading of SCS initiated at 32 Ma, the tectonic subsidence rate does not decrease but increases instead, and then decreases at about 23 Ma, which indicates that the rifting continued after the onset of seafloor spreading until about 23 Ma. The formation thickness exhibits the same phenomenon, that is the syn-rift stage prolonged and the post-rift thermal subsidence delayed. The formation mechanisms are supposed to be three: (1) the lithospheric rigidity of the northern SCS is weak and its ductility is relatively strong, which delayed the strain relaxation resulting from the seafloor spreading; (2) the differential layered independent extension of the lithosphere may be one reason for the delay of post-rift stage; and (3) the southward transition of SCS spreading ridge during 24 to 21 Ma and the corresponding acceleration of seafloor spreading rate then triggered the initiation of large-scale thermal subsidence in the study area at about 23 Ma.
Triterpane and sterane biomarkers in the YA13-1 condensates from Qiongdongnan Basin, South China Sea
Resumo:
Triterpanes and steranes in condensates from the YA13-1 gas field, Qiongdongnan Basin, were monitored. The YA13-1 condensates have unusual biomarker distributions dominated by terpanes and steranes derived from higher plants. Anomalously abundant 1 got-oleanane and remarkably abundant bicadinanes are present in the YA13-1 condensates, whereas the 17alpha-hopane contents are extremely low. Taraxastane and significantly abundant 17alpha-diahopanes occur in the condensates. In addition, a number of unknown C-29 and C-30 pentacyclic triterpanes including previously unreported compounds were detected in the condensates, some of which are significantly abundant. The unknown compounds may be terrestrial biomarkers. C-29 homologues are relatively predominant among the regular and rearranged steranes. The diasterane concentrations are markedly higher than those of regular steranes. The maturity of the YA13-1 condensates is relatively high, at the peak to late oil generation stage (corresponding to 0.85-1.10% Ro), based on sterane and terpane and including bicadinane maturity parameters (i.e. T/(T-1 + R) and 2T/R bicadinane ratios). The above maturity assessment result is different from that based on diamondoid maturity parameters (%Ro = 1.60-1.70) [Org. Geochem. 25 (1996) 179], which can be explained by a contribution of hydrocarbons from two sources at different depths. The YA13-1 condensates were probably generated from the Yacheng and Lingshui coal-bearing source rocks buried both in the Qiongdongnan Basin (3400-5000 m) and in the Yinggehai Basin (>5000 m). The possible contribution of lower maturity hydrocarbons from the Yacheng and Lingshui Formations (3400-4100 m) in the Qiongdongnan Basin to the YA13-1 gases and condensates should not be neglected. Heritability and genetic and phenotypic correlations were estimated for juvenile growth traits of Pacific abalone Haliotis discus hannai Ino. The estimates were calculated from shell length and shell width measurements on progeny resulting from 12 half-sib families and 36 full-sib families obtained using artificial fertilization of mating three females to each male. The measurements were taken at 10, 20 and 30 d after fertilization. It was found that heritability estimates based on sire component ranged from 0.23 to 0.36 for shell length and 0.21 to 0.32 for shell width. Heritability estimates from dam component were larger than those from sire component at three ages, indicating presence of maternal effects, non-additive genetic effects and common environmental effects. Phenotypic correlations were significant at three ages (P < 0.05), with values of 0.92, 0.93 and 0.92, respectively. Genetic correlations from the paternal half-sib correlation analysis were highly positive at three ages, with values of 0.50, 0.78 and 0.81, respectively. The results suggest that selective breeding is an effective approach to improving growth traits of Pacific abalone stocks.

The Taklamakan Desert, lying in the center Tarim Basin of sourthern Xinjiang, is the largest sand sea in China and well known in the world as its inclemency. For understanding the formation and evolution of the Taklamakan Desert, it is very important to identify the provenance of aeolian sediments in the extensive dune fields, but the opinions from earlier studies are quite different. In this study, we examined the major- and trace-element compositions, mineral compositions and grain-size distributions of some Quaternary aeolian and nonaeolian sediments collected from the Taklamakan Desert, together with the variation of chemical and mineralogical compositions of different grain-size fraction. At the same time, we also studied the chemistries of some natural water samples (river water and groundwater) with the items of TDS, pH, Alkalinity, conductivity and major cation and anion compositions. Our results of analysis show some significant opinions as follow: Most of the frequency-distribution curves of grain size of dune sand samples are simgle peak, but that of the river and lacustrine sediment are most double peak or multi-peak. The grain-size distribution of dunefield sand changed gradually from north to south with the major wind direction in large scale, but there are many differences in regional scale. The major, REE, trace element compositions and mineral compositions are very different among the coarse, fine fraction and bulk samples due to the influence of grain-size. Most of the fine fractions are geochemically homogenous, but the coarse fractions and bulk samples are heterogenous. All the surface and ground waters are limnetic or sub-salty, their chemical compositions are mainly controlled by rock-weathering and crystallization- evaporation processes, and mainly come from the evaporate, while the contributions of the carbonate and silicate are little, excluding the influence on oasis water by carbonate. The mineral compositions of selected samples are stable, mainly composed of the strongly resistant mineral types. The mineral maturity of them are more immature at whole compared with other sandy sediments in the world, and they have experienced less degree of chemical weathering and recycling, being lying in the early stage of continental weathering. Among these sediments, the river sediments are relatively primitive. The sources of these sediments are maybe mainly terrigenous, silicic and subaluminous/ metalunious rocks, such as the granodiorite and its metamorphic rock. The geochemical compositions of dunefield sand are similar with those of the river sediments and dune sands near the river way; There are not only the resemblances but also the differences on geochemistry and granularity between north and south dunesands; The sediments from same section have different age but same trace-element compositions; The sediments from the south edge of Tarim Basin are all somewhat geochemically similar with the palaeo-river-sediment on the south edge of studying area. The REE data support the idea that the south dunesands are a little older than the north dunesands, and the tectonic settings of source area are mainly active continental margin based on the major-element compositions, so they indicate that the sediment of Taklamakan Desert maybe come mainly from the rock-weathering production of north part of the Kunlun Mountains. Compared with the sands of other dune field in north of China, the sands of the Taklamakan Desert are distinct by REE composition, but similar with the Luochuan loess, center China, and the two sandy dusts of Beijing, eastern China.

The present maturity of Cambrian and Ordovician source rocks in Tazhong area, Tarim basin, is studied using several organic petrology methods and conodont CAI method. The highest palaeotemperature that the Cambrian-Ordovician undergone is revealed by Laser Roman Microprobe (LRM) analysis and by simulating experiment of the kerogen chemical kinetics. In according to all above study, the thermal history of Cambrian and Ordovician is reconstructed based on numerical simulating approaches. The characteristics of secondary hydrocarbon generating are studied by inclusions analysis. The reflectances of the samples in the drills located in Tazhong area show that the maturities of Cambrian source rocks are in the stages of condense oil-dry gas, and that of Ordovician source rocks range from peak of oil generating to wet gas stage. The palaeotemperature data of Cambrian-Orovician source rocks from well Tacan 1, based on LRM analysis, are in coincidence with that from other methods. Also are the palaeotemperature data of Cambrian-Orovician source rocks in well Tacan 1 based on the simulating experiments of kerogen pyrolysis, similar to the homogenization temperatures of inclusions in the source rocks. Aaccording to the vitrinite inflectance data of the TZ12 well and Tacan 1 well, the paleotemperature gradients are analysized and reconstructed. These data show that the paleotemperature gradient in Tazhong area was the highest during Cambrian-Ordovician period, it was up to 3.5°C/100m. Following, the temperature gradient descended gradually and it reached to the lowest at present (2.2°C/100m). The histories of maturation and hydrocarbon generation of Cambrian and Ordovician source rocks in Tazhong area are researched systematically and quantitatively, the results show that periods of oil generation from Cambrian and Ordovician source rocks lasted for a long time from Ordovician to Carbonferious periods because the central Cambrian stratum in the north slope of Tazhong area is buried differently in depth. The top of the Cambrian entered into the peak of oil generation in middle-late Ordovician, and most area of the north slope of Tazhong area entered into the peak of oil generation in Carbonferious period, and on the uplift belt some of source rocks entered into the peak of oil generation in Permian period. In early Devonian, the central of the Lower Ordovician source rocks near the Manjiaer depression reached the peak of oil generation and near the top of the Tazhong uplift did not reached the peak of oil generation until early Cretaceous. The middle-upper Ordovician entered into the peak of oil generation in early-middle Jurassic. The time of the middle-upper Ordovician in the top of the uplift belt entering into the peak of oil generation was delayed, because the source rock was buried shallowly, and it did not reached the peak of oil generation until middle Cretaceous. Middle-upper Ordovician in the top of the north slope has been in the peak of oil generation now, it is consistent with the maturity (1.0-1.2%Ro) of the source rocks. The characteristics of the inclusions formed by kerogens are different from that by crystal-enclosed organic matters(OM) during secondary hydrocarbon generation of Cambrian and Ordovician source rocks. The secondary hydrocarbon generation mainly occurred in Mesozoic-Cenozoic period, in an area of about 9000km2 in the north slope. The intensity of the secondary hydrocarbon generation of Cambrian and Ordovician is up to 21kg/torg and 36kg/torg) respectively. Using the staged gas chromatography, the high-over maturated carbonate source rocks are analysized to release the adsorbed OM, inclusions OM and crystal-enclosed OM, respectively, and to evaluate their relative contributions to secondary hydrocarbon generation. The three periods of oil and gas migration and petroleum pools formation in Tazhong area are determined according to organic inclusions and solid bitumen.

Natural gas pays more important role in the society as clean fuel. Natural gas exploration has been enhanced in recent years in many countries. It also has prospective future in our country through "85" and "95" national research. Many big size gas fields have been discovered in different formations in different basins such as lower and upper Paleozoic in Erdos basin, Tertiary system in Kuche depression in Tarim basin, Triassic system in east of Sichuan basin. Because gas bearing basins had been experienced multiple tectogenesis. The characteristics of natural gases usually in one gas field are that they have multiple source rocks and are multiple maturities and formed in different ages. There has most difficult to research on the gas-rock correlation and mechanism of gas formation. Develop advanced techniques and methods and apply them to solve above problems is necessary. The research is focused on the critical techniques of geochemistry and physical simulation of gas-rock correlation and gas formation. The lists in the following are conclusions through research and lots of experiments. I 8 advanced techniques have been developed or improved about gas-rock correlation and gas migration, accumulation and formation. A series of geochemistry techniques has been developed about analyzing inclusion enclave. They are analyzing gas and liquid composition and biomarker and on-line individual carbon isotope composition in inclusion enclave. These techniques combing the inclusion homogeneous temperature can be applied to study on gas-rock correlation directly and gas migration, filling and formation ages. Technique of on-line determination individual gas carbon isotope composition in kerogen and bitumen thermal pyrolysis is developed. It is applied to determine the source of natural is kerogen thermal degradation or oil pyrolysis. Method of on-line determination individual gas carbon isotope composition in rock thermal simulation has being improved. Based on the "95"former research, on-line determination individual gas carbon isotope composition in different type of maceral and rocks thermal pyrolys is has been determined. The conclusion is that carbon isotope composition of benzene and toluene in homogenous texture kerogen thermal degradation is almost same at different maturity. By comparison, that in mixture type kerogen thermal pyrolysis jumps from step to step with the changes of maturity. This conclusion is a good proof of gas-rock dynamic correlation. 3. Biomarker of rock can be determined directly through research. It solves the problems such as long period preparing sample, light composition losing and sample contamination etc. It can be applied to research the character of source rock and mechanism of source rock expulsion and the path of hydrocarbon migration etc. 4. The process of hydrocarbon dynamic generation in source rock can be seen at every stage applying locating observation and thermal simulation of ESEM. The mechanism of hydrocarbon generation and expulsion in source rock is discussed according to the experiments. This technique is advanced in the world. 5. A sample injection system whose character is higher vacuum, lower leaks and lower blank has been built up to analyze inert gas. He,Ar,Kr and Xe can be determined continuously on one instrument and one injection. This is advanced in domestic. 7. Quality and quantity analysis of benzene ring compounds and phenolic compounds and determination of organic acid and aqueous gas analysis are applied to research the relationship between compounds in formation water and gas formation. This is another new idea to study the gas-rock correlation and gas formation. 8. Inclusion analysis data can be used to calculate the Paleo-fluid density, Paleo-geothermal gradient and Paleo-geopressure gradient and then to calculate the Paleo-fluid potential. It's also a new method to research the direction of hydrocarbon migration and accumulation. 9. Equipment of natural gas formation simulation is produced during the research to probe how the physical properties of rock affect the gas migration and accumulation and what efficiency of gas migrate and factors of gas formation and the models of different type of migration are. II study is focused on that if the source rocks of lower Paleozoic generated hydrocarbon and what the source rocks of weathered formation gas pool and the mechanism of gas formation are though many advanced techniques application. There are four conclusions. 1.The maturity of Majiagou formation source rocks is higher in south than that in north. There also have parts of the higher maturity in middle and east. Anomalous thermal pays important role in big size field formation in middle of basin. 2. The amount of gas generation in high-over maturity source rocks in lower Paleozoic is lager than that of most absorption of source rocks. Lower Paleozoic source rocks are effective source rocks. Universal bitumen exists in Ordovician source rocks to prove that Ordovician source rocks had generated hydrocarbon. Bitumen has some attribution to the middle gas pool formation. 3. Comprehensive gas-rock correlation says that natural gases of north, west, south of middle gas field of basin mainly come from lower Paleozoic source rocks. The attribution ratio of lower Paleozoic source rocks is 60%-70%. Natural gases of other areas mainly come from upper Paleozoic. The attribution ratio of upper Paleozoic source rocks is 70%. 4. Paleozoic gases migration phase of Erdos basin are also interesting. The relative abundance of gasoline aromatic is quite low especially toluene that of which is divided by that of methyl-cyclohexane is less than 0.2 in upper Paleozoic gas pool. The migration phase of upper Paleozoic gas may be aqueous phase. By comparison, the relative abundance of gasoline aromatic is higher in lower Paleozoic gas. The distribution character of gasoline gas is similar with that in source rock thermal simulation. The migration phase of it may be free phase. IH Comprehensive gas-rock correlation is also processed in Kuche depression Tarim basin. The mechanism of gas formation is probed and the gas formation model has been built up. Four conclusions list below. 1. Gases in Kuche depression come from Triassic-Jurassic coal-measure source rocks. They are high-over maturity. Comparatively, the highest maturity area is Kelasu, next is Dabei area, Yinan area. 2. Kerogen thermal degradation is main reason of the dry gas in Kuche depression. Small part of dry gas comes from oil pyrolysis. VI 3.The K12 natural gas lays out some of hydro-gas character. Oil dissolved in the gas. Hydro-gas is also a factor making the gas drier and carbon isotope composition heavier. 4. The mechanism and genesis of KL2 gas pool list as below. Overpressure has being existed in Triassic-Jurassic source rocks since Keche period. Natural gases were expulsed by episode style from overpressure source rocks. Hetero-face was main migration style of gas, oil and water at that time. The fluids transferred the pressure of source rocks when they migrated and then separated when they got in reservoir. After that, natural gas migrated up and accumulated and formed with the techno-genesis. Tectonic extrusion made the natural gas overpressure continuously. When the pressure was up to the critical pressure, the C6-C7 composition in natural gas changed. The results were that relative abundance of alkane and aromatic decreased while cycloalkane and isoparaffin increased. There was lots of natural gas filling during every tectonic. The main factors of overpressure of natural gas were tectonic extrusion and fluid transferring pressure of source rocks. Well preservation was also important in the KL2 gas pool formation. The reserves of gas can satisfy the need of pipeline where is from west to east. IV A good idea of natural gas migration and accumulation modeling whose apparent character is real core and formation condition is suggested to model the physical process of gas formation. Following is the modeling results. 1. Modeling results prove that the gas accumulation rule under cap layer and gas fraction on migration path. 2. Natural gas migration as free phase is difficult in dense rock. 3. Natural gases accumulated easily in good physical properties reservoirs where are under the plugging layer. Under the condition of that permeability of rock is more than 1 * 10~(-3)μm~(-1), the more better the physical properties and the more bigger pore of rock, the more easier the gas accumulation in there. On the contrary, natural gas canonly migrate further to accumulate in good physical properties of rock. 4. Natural gas migrate up is different from that down. Under the same situation, the amount of gas migration up is lager than that of gas migration down and the distance of migration up is 3 times as that of migration down. 5. After gas leaks from dense confining layer, the ability of its dynamic plug-back decreased apparently. Gas lost from these arils easily. These confining layer can confine again only after geology condition changes. 6. Water-wetted and capillary-blocking rocks can't block water but gases generally. The result is that water can migrate continuously through blocking rocks but the gases stay under the blocking rocks then form in there. The experiments have proved the formation model of deep basin gas.
