在患暴发流行性发病鳜鱼的脾、肾、肝、鳃组织细胞的超薄切片中观察到直径为 12 0 150nm的二十面体病毒粒子 ,其截面呈六角形 ,有囊膜。将病鱼内脏与 10倍体积的PBS缓冲液制成组织匀浆液 ,过滤除菌后感染对数生长期的草鱼肾细胞。结果表明 ,该感染液在 2 8℃条件下 ,培养 36 4 8h ,生长旺盛的草鱼肾细胞开始出现CPE ,5 7d感染的单层细胞逐渐脱落死亡。该细胞病毒悬液具有感染性 ,可持续稳定地进行传代培养。感染细胞的病毒培养液 ,经差异离心纯化 ,负染制片 ,电镜下观察到大量的病毒粒子

自1984 年第一例转基因鱼在我国诞生以来,转基因鱼研究取得长足进步。研究表明外源基因可以整合、表达,并能遗传给后代,然而外源基因整合位点难以控制,遗传不稳定等问题仍未解决。所幸我国鱼类核移植技术较为成熟,结合细胞基因转移、基因打靶和核移植技术,有望在不久将来获得稳定整合的纯化转基因鱼。

记述了南鳅属鱼类一新种———异颌南鳅Schisturaheterognathossp.nov.。用作新种描述的模式标本共7尾,其中正模标本1尾,体长800mm,编号8810240;副模标本6尾,体长950~1050mm,编号8810241、8810242、8810150~8810153。1988年采自云南省勐腊县的那着和曼庄,属澜沧江下游支流南腊河。所有模式标本均保存于中国科学院水生生物研究所鱼类标本馆。该新种上下颌厚而强壮,宽阔,无角质;上颌无齿状突起,下颌中部有一细小缺刻或无缺刻;口横列,口宽约等于吻

在优化RAPD检测条件的基础上,采用40对随机引物,比较分析了银鲫复合种、异育银鲫和兴国红鲤相互间扩增DNA片段的异同性。总的来说,银鲫复合种和异育银鲫具有基本一致的扩增产物,而与兴国红鲤的扩增产物多数不同。相似率分析表明,银鲫复合种与兴国红鲤之间的相似率为31.6%,异育银鲫与兴国红鲤之间的相似率为28.6%。在分析中,除发现银鲫复合种、异育银鲫与兴国红鲤间共有的扩增片段外,还发现了银鲫复合种与兴国红鲤间共有的扩增片段以及银鲫复合种所特有的DNA扩增带。本研究不但为银鲫复合种异源遗传成分的整人提供了新的

本文报道了鳜鱼[Sinipercachuatsi(Basilewsky)]鳃上寄生的河鲈锚首吸虫[Ancyrocephalusmogurndae(Gussev)]的食物组成及其能量储存的形式。联苯胺—过氧化物酶染色显示河鲈锚首吸虫摄食宿主的血液,过碘酸Schiff氏反应显示其也摄食宿主的粘液。尽管在河鲈锚首吸虫的肠腔发现残余的细胞,但这些细胞是否为宿主的鳃组织细胞尚待进一步的研究证实。河鲈锚首吸虫体内储存有大量的脂肪,表明有氧代谢的存在并可能是能量供给的主要形式,尽管在虫体的实质中也有糖原的存在。

<正> 福建省气候温暖、潮湿,境内多山,溪河纵横,沟渠塘堰密布,动植物种类繁多,双星藻科植物种类也很丰富。1964—1976年,福建师范大学生物系对福建省双星藻科植物进行了广泛的调查。1977年我们对该科藻类进行了分类研究,共鉴定约90种,其中发现一些新的种类。本文报导了11新种,1新变种,分属于:双星藻属(Zygnema)3种;拟双星藻属(Zygnemopsis)1种,转板藻属(Mougeotia)1种,膝接藻属(Zygogonium)1种及1变

Grass carp hemorrhagic virus (GCHV)-induced gene 2 (Gig2) is a novel gene previously identified from UV-inactivated GCHV-treated Carassius auratus blastulae embryonic (CAB) cells, suggesting that it should play a pivotal role in the interferon (IFN) antiviral response. In this study, a polyclonal anti-Gig2 antiserum was generated and used to study the inductive expression pattern by Western blot analysis, showing no basal expression in normal CAB cells but a significant up-regulation upon UV-inactivated GCHV, polyinosinic:polycytidylic acid (Poly I:Q and recombinant IFN (rIFN). However, constitutive expression of Gig2 is observed in all tested tissues from grass carp (Ctenopharyngodon idellus), and Poly I:C injection increases the relative amount of Gig2 protein in skin, spleen, trunk kidney, gill, hindgut and thymus. Moreover, the genomic sequence covering the whole Gig2 ORF and the upstream promoter region were amplified by genomic walking. Significantly, the Gig2 promoter contains three IFN-stimulated response elements (ISREs), nine GAAA/TfTC motifs and five gamma-IFN activating sites (GAS), which are the characteristics of genes responsive to both type I IFN and type 11 IFN. Subsequently, the complete Gig2 promoter sequence was cloned into pGL3-Basic vector, and its activity was measured by luciferase assays in the transfected CAB cells. The Gig2 promoter-driven construct is highly induced in CAB cells after treatment with Poly I:C or rIFN, and the functional capability is dependent on IFN regulatory factor 7 (IRF7), because its activity can be stimulated by IRF7. Collectively, the data provide strong evidence that Gig2 is indeed a novel IFN inducible gene and its expression is likely dependent on IRF7 upon Poly I:C or IFN. (C) 2009 Elsevier Ltd. In Drosophila, Toll signaling cascade, which resembles the mammalian Toll-like receptor (TLR)/IL-1R signaling pathways and regulates the expression of anti-microbial peptide genes, mainly relies on peptidoglycan recognition proteins (PGRPs) for the detection of bacterial pathogens. To explore the effect of zebrafish peptidoglycan recognition protein 6 (zfPGRP6) on Toll-like receptor signaling pathway, RNA interference (siRNA) and real time quantitative PCR (RQ-PCR) methods were used to identify differentially expressed genes regulated by zfPGRP6. The target genes included TLR2, TLR3, TLR5, TLR7, TLR8, IL1R, Sterile-alpha and Armadillo motif containing protein (SARM), myeloid differentiation factor 88 (MyD88) and nuclear factor (NF)-kappa B2 (p100/p52). The results of RQ-PCR showed that RNAi-mediated Suppression of zfPGRP6 significantly down-regulated the expression of TLR2, TLR5, IL1R, SARM, MyD88 and p100/p52. The expression of beta-defensin-1 was also down-regulated in those embryos silenced by zfPGRP6. In challenge experiments to determine the anti-bacterial response to Gram-negative bacteria, RNAi knock-down of zfPGRP6 markedly increased susceptibility to Flavobacterium columnare. This paper reports a new method for detection of ROS scavengers including superoxide dismutase, ascorbic acid and glutathione based on a 'probe' of peroxidase-oxidase biochemical oscillator. The oscillation period and amplitude change with different concentrations of scavengers. The linear ranges of superoxide dismutase, ascorbic acid and glutathione are respectively 1.56 x 10(-4)-1.56 x 10(-3) mg mL(-1), 1.75 x 10(-7) -1.75 x 10(-5) mol L-1 and 9.38 x 10(-7) -7.5 x 10(-5) mol L-1. The selectivity, linearity and precision for superoxide dismutase, ascorbic acid, and glutathione are presented and discussed. The results compared well with other standard methods for determination of superoxide dismutase, ascorbic acid and glutathione. Some possible steps in the overall reaction mechanisms are discussed.

A columnal islands system, which was composed of three layers of self-assembled InAs/GaAs quantum dots (QDs), has been fabricated by solid-source molecular beam epitaxy (MBE) through S-K mode on a (100) semi-insulating GaAs substrate. The effects of the thickness of GaAs space layer, the growth interruption time and the amount of InAs deposition on the emission wavelength of columnal islands were presented. The image of atomic force microscopy (AFM) indicated the columnal islands with high uniformity in size and shape. At room temperature, the emission wavelength of columnal islands with different effective heights was achieved 1.32 and 1.4 mum; however, the emission wavelength of single-layer QDs with normal height was just 1. l mum. It provides a useful and intuitive approach to artificially control the emission wavelength of a QD material system.

The SnO2 material prepared by sol-gel method was modified by thiourea solution in different concentrations (0.05, 0.1 and 0.2 mol dm(-3)). Then the structure and the average grain size of the SnO2 material were investigated by X-ray power diffraction. In order to understand the nature of the species on the SnO2 surfaces, the thermal gravimetric and differential thermal analyzer (TG-DTA) and IR spectra of these modified and unmodified sample were taken. The result indicates that the stability of oxygen adsorbed on thiourea-modified surface was improved and the amount of surface hydroxyl groups adsorbed on this grain surface was decreased. The thiourea adsorbed on SnO2 grain surface is translated to SO42- after sintered at 600 degrees C. SO42- species stabilize the resistance of the SnO2 sensor. (c) 2005 Elsevier B.V. A novel butt-joint coupling scheme is proposed to improve the coupling efficiency for the integration of a GalnAsP MQW distributed feedback (DFB) laser with an MQW electro-absorption modulator (EAM). The proposed method gives more than 90% coupling efficiency, being much higher than the 26% coupling efficiency of the common MQW-MQW coupling technique. The differential quantum efficiency of the MQW-bulk-MQW coupled device is also much higher than that of the MQW-MQW device, 0.106 mW/mA versus 0.02 mW/mA. The EAM-DFB devices fabricated by the proposed method exhibit a very high modulation efficiency (12 dB/V) from 0 to I V. By adopting a high-mesa ridge waveguide and buried polyimide, the capacitance of the modulator is reduced to about 0.28 pF. The experimental results demonstrate that the method can replace the conventional MQW-MQW coupling technique to fabricate high-quality integrated photonic devices. (C) 2007 Elsevier B.V. Two-dimensional ZnO nanowall networks were grown on ZnO-coated silicon by thermal evaporation at low temperature without catalysts or additives. All of the results from scanning electronic spectroscope, X-ray diffraction and Raman scattering confirmed that the ZnO nanowalls were vertically aligned and c-axis oriented. The room-temperature photoluminescence spectra showed a dominated UV peak at 378 nm, and a much suppressed orange emission centered at similar to 590 nm. This demonstrates fairly good crystal quality and optical properties of the product. A possible three-step, zinc vapor-controlled process was proposed to explain the growth of well-aligned ZnO nanowall networks. The pre-coated ZnO template layer plays a key role during the synthesis process, which guides the growth direction of the synthesized products. (C) 2007 Elsevier B.V. All rights reserved.

The fluctuations of the strained layer in a superlattice or quantum well can broaden the width of satellite peaks in double crystal X-ray diffraction (DCXRD) pattern. It is found that the width of the 0(th) peak is directly proportional to the fluctuation of the strained layer if the other related facts are ignored. By this method, the Ge-Si atomic interdiffusion in Ge nano-dots and wetting layers has been investigated by DCXRD. It is found that thermal annealing can activate Ge-Si atomic interdiffusion and the interdiffusion in the nano-dots area is much stronger than that in the wetting layer area. Therefore the fluctuation of the Ge layer decreases and the distribution of Ge atoms becomes homogeneous in the horizontal Ge (GeSi actually) layer, which make the width of the 0(th) peak narrow after annealing.
